The physical and chemical changes exhibited by high density polyethylene (HDPE) after treatment with hydrogen at a pressure of 90 MPa followed by rapid release of the hydrogen were studied. X-ray diffraction, differential scanning calorimetry, thermo gravimetric analysis, and attenuated total reflectance (ATR)-fourier transform infrared (FTIR) were used for this purpose. As a result, it was found that the degree of crystallinity of HDPE decreased after hydrogen pressure treatment, while the average thickness of lamellae that constitute the crystals and the melting temperature of the crystalline region actually increased. The decomposition temperature also increased by about 3℃. In addition, it was found that the hydrogen bonding network between -OH groups in the HDPE sample was strengthened and partial chain scission occurred. These cut chains were found to be terminated by oxidative degradation such as cross-linking between chains, -C=O, -C-O, and -CHO, or by the formation of -CH3 at the chain ends, as confirmed by ATR-FTIR.
Oxidative stress induces apoptosis in many cellular systems including glioblastoma cells, with caspase-8 activation was regarded as a major contribution to $H_2O_2$-induced cell death. This study focused on the role of the autophagic protein p62 in $H_2O_2$-induced apoptosis in U87MG cells. Oxidative stress was applied with $H_2O_2$, and cell apoptosis and viability were measured with use of caspase inhibitors or autophagic mediators or siRNA p62, GFP-p62 and GFP-p62-UBA (del) transfection. We found that $H_2O_2$-induced U87MG cell death was correlated with caspase-8. To understand the role of p62 in MG132-induced cell death, the levels of p62/SQSTM1 or autophagy in U87MG cells were modulated with biochemical or genetic methods. The results showed that the over-expression of wild type p62/SQSTM1 significantly reduced $H_2O_2$ induced cell death, but knockdown of p62 aggravated the process. In addition, inhibition of autophagy promoted p62 and active caspase-8 increasing $H_2O_2$-induced apoptosis while induction of autophagy manifested the opposite effect. We further demonstrated that the function of p62/SQSTM1 required its C-terminus UBA domain to attenuate $H_2O_2$ cytotoxity by inhibition of caspase-8 activity. Our results indicated that p62/SQSTM1 was a potential contributor to mediate caspase-8 activation by autophagy in oxidative stress process.
Objectives: It was investigated the cytoprotective efficacies of isorhamnetin, a flavonoid originally derived from Hippophae rhamnoides L., against oxidative stress-induced apoptosis in C2C12 myoblasts. Methods: The effects of isorhamnetin on cell growth, apoptosis and reactive oxygen species (ROS) generation were evaluated by trypan blue dye exclusion assay, 4',6-diamidino-2-phenylindole staining and flow cytometry. The levels of apoptosis-regulatory and nuclear factor erythroid 2-related factor 2 (Nrf2) signaling pathway-related proteins, and caspase activities (caspase-3 and -9) were determined by Western blot analysis and colorimetric assay, respectively. Results: Our results revealed that treatment with isorhamnetin prior to hydrogen peroxide ($H_2O_2$) exposure significantly increased the C2C12 cell viability and, indicating that the exposure of C2C12 cells to isorhamnetin conferred a protective effect against oxidative stress. Isorhamnetin also effectively attenuated $H_2O_2$-induced apoptosis and ROS generation, which was associated with the restoration of the upregulation of Bax and downregulation of Bcl-2 induced by $H_2O_2$. In addition, $H_2O_2$ enhanced the activation of caspase-9 and -3, and degradation of poly (ADP-ribose)-polymerase, a typical substrate protein of activated caspase-3; however, these events were almost totally reversed by pretreatment with isorhamnetin. Moreover, isorhamnetin increased the levels of heme oxygenase-1, a potent antioxidant enzyme, associated with the induction of Nrf2. Conclusions: Our data indicated that isorhamnetin may potentially serve as an agent for the treatment and prevention of muscle disorders caused by oxidative stress.
Chios Gum Mastic (CGM) is a natural resin extracted from the leaves of Pistacia lentiscus, a plant endemic to the Greek island of Chios. It has been used by traditional healers, and it has antibacterial, antifungal properties, and therapeutic benefits for the skin. The CGM reduces the formation of dental plaque and bacterial growth in oral saliva, and recent studies have demonstrated the role of antioxidant activity of CGM. Although CGM has been widely investigated, its protective effect against oxidative-damage to keratinocytes, as well as the relationship between CGM and autophagy, has not been investigated. The aim of this study was to assess the protective effect of CGM against $H_2O_2$-induced oxidative stress and to evaluate the autophagic features induced by CGM in human keratinocytes. The pretreatment with CGM significantly reduced apoptosis in $H_2O_2$-exposed HaCaT cells. It promoted the degradation of caspase-3, caspase-8, and caspase-9; and it induced the formation of the processed PARP. The treatment with CGM caused an increase in vesicle formation compared to control group. The level of p62 was reduced and the conversion of LC3-I to LC3-II was increased in CGM treated HaCaT cells. Also, the treatment with CGM increased cleavage of ATG5-ATG12 complex. In summary, CGM helps the cells to survive under stressful conditions by preventing apoptosis and enhancing autophagy. Besides, the present investigation provides evidence to support the antioxidant potential of CGM in vitro and opens up a new horizon for future experiments.
Biodiesel is an environmentally-friendly fuel with low smoke emission because it contains about 10% oxygen. Biodiesel fuel prepared by transesterification of vegetable oil or animal fats is susceptible to auto-oxidation. The rate of auto-oxidation depends on the number of methylene double bonds contained within the fatty acid methyl or ethyl ester groups. Biodiesel may be easily oxidized under several conditions, i.e., upon exposure to sunlight, temperature, oxygen environment. Maintenance of the fuel quality of biodiesel requires the development of technologies to increase the resistance of biodiesel to oxidation. Treatment with antioxidants is a promising approach for extending the shelf-life or storage time of biodiesel. The chemical properties of various amine-based antioxidants were evaluated after synthesis of the antioxidants by condensation of phenylenediamine with alkylamines at room temperature. In general, the oxidative stability can be assessed based on various experimental parameters. Such parameters may include temperature, pressure, and the flow rate of air through the samples. The Rancimat method (EN14112) was selected because it is a rapid technique that requires very little sample and provides good precision for oxidative degradation analysis. Specifically, the EN 14112 technique provides enhanced efficiency for oxidative stability evaluation when a larger ester head group is utilized. Therefore, this technique was employed for evaluation of the oxidation stability of biodiesel by the Rancimat method (EN14112).
In this study, the effects of smoking cessation and relative antioxidant activities on the oxidative stress were determined by using in vitro method. Thirty healthy smokers who were free of any disease and smoked more than 1 pack per day for the past 10 years participated in this study. For smoking cessation, smokers were asked to wear nicotine patch (21mg nicotine/ patch) everyday for 30 days and then to replace at the same time of the day. Smoking cessation program in conjunction with nicotine patch replacement was also conducted every week, one hour/each session, for 4 weeks. Canthaxanthin, $\beta-carotene$, and $\alpha-tocopherol$ were added into red blood cells at pre and post smoking cessation. As indicators of oxidative stress, hemoglobin degradation, lipid peroxidation, and percent hemolysis were determined at both pre and post smoking cessation. After 30 days of smoking cessation, the subjects gained an average of 5 pounds, varying 2 to 8 pounds, by suggesting that behavioral problems rather than nicotine itself are more important for gaining weight in ex-smokers. The total hemoglobin concentrations in blood were similar in pre and post smoking cessation, but smoking cessation resulted in a decrease in the percentage of methemoglobin from 0.96% to 0.85% Smoking cessation also caused to decease malondialdehyde (MDA) values ($26.7{\pm}7.8$ vs. $23.6{\pm}4.5$ (without oxidation), $179.3{\pm}21$ vs. $161.2{\pm}28$ nmol/ml (with oxidation) (p<0.05)), not percent hemolysis. Various antioxidants with smoking cessation significantly decreased MDA values(p<0.05), in contrast to marginal decrease of MDA in smoking cessation only. Three antioxidants used in this stu study were similarly effective in inhibiting MDA production, but relative effectiveness of canthaxanthin or $\alpha-tocopherol$ was greater than that of $\beta-carotene$ (p<0.05), in case of oxidation induced. The percent hemolysis was greatly decreased when antioxidants were added into the blood of ex-smokers (p<0.05) but no statistical significance in relative effectiveness of antioxidants was observed.

Coolant rubber hoses for automobile radiators can be degraded and thus failed due to the influence of contacting stresses of air and coolant liquid under the thermal and mechanical loadings. In this study, test analysis was carried out for evaluating the degradation and failure mechanisms of coolant hose materials. Two kinds of EPDM rubber materials applicable to the hoses were adopted: commonly-used ethylene-propylene diene monomer(EPDM) rubbers and EPDM rubbers with high resistance against electro-chemical degradation (ECD). An increase of surface hardness and a large reduction of failure strain were shown due to the formation of oxidation layer for the specimens which had been kept in a high temperature air chamber. Coolant ageing effects took place only by an amount of pure thermal degradation. The specimens degraded by ECD test showed a swelling behavior and a considerable increase in weight on account of the penetration of coolant liquid into the skin and interior of the rubber specimens. The ECD induced material softening as well as drastic reduction in strength and failure strain. However EPDM rubbers designed for high resistance against ECD revealed a large improvement in reduction of failure strain and weight. This study finally established a procedure for reliability analysis and evaluation of the degradation and failure mechanisms of EPDM rubbers used in coolant hoses for automobile radiators.
The effect of water activity on degradation of pigments in dried lavers, Porphyra tenera Kjellm. was examined when stored at room temperature for fifty days. Chlorophyll pigment was extracted with methanol-petroleum ether mixture solvent(2:1 v/v), partitioned in ether, and analysed spectrophotometrically at 662 nm as chlorophyll a. The degradation products of chlorophyll were isolated on sugar-starch column(85:15 w/w) with n-propanol-petroleum ether solution(1:200 v/v) as a developing solvent. The isolated green colored zones were analysed individually at the wavelengths of 650, 662, and 667 nm as allomerized product, chlorophyll a retained, and pheophytin formed respectively. Carotenoida were also extracted with the methanol mixture solvent, partitioned in ether, and finally redissolved in acetone after the evaporation of ether in a rotary vacuum evaporator. The total carotenoid content was measured as lutein at 450 nm. From the results, it is noted that the rate of chlorophyll degradation reached a minimum at 0.11 to 0.33 water activity while progressively increased at higher moisture levels resulting in rapid conversion of chlorophyll to pheophytin. At lower activity, autocatalysed oxidizing reaction like allomerization seemed prevailing the acid catalysed conversion reaction. The loss of carotenoid pigment was also greatly reduced at the range of 0.22 to 0.34 water activity with much faster oxidative degradation at both higher and extremely lower moisture levels. These two moisture levels indicated above at which the both pigments exhibited maximum stability are considerably higher than the BET monolayer moisture which appeared 7.91 percent on dry basis at Aw=0.10 calculated from the adsorption isothermal data of the sample at $20^{\circ}C$. The rate of pigment loss in heat treated samples at 60 and $100^{\circ}C$ for 2 hours prior to storage somewhat decreased, particularly at higher moisture levels although the final pigment retention was not much stabilized.
Gasoline generates organic acid and polymer (gum) by hydrocarbon oxidation depending on the storage environment such as temperature and exposure to sunlight, which can cause metal corrosion, rubber and resin degradation and vehicle malfunction caused by accumulation in fuel supply system. The gasoline which has not been used for a long time in bi-fuel (LPG-Gasoline) vehicle causes problems, and low octane number gasoline have evaporated into the field, but the exact cause has not been studied yet. In this study, we suggest a plan of quality management by investigating the gasoline oxidation behavior. In order to investigate the oxidation behavior of gasoline, changes of gasoline properties were analyzed at various storage conditions such as storage time, storage vessel type (vehicle fuel tank, PE vessel and Fe vessel) and storage circumstances (sunlight exposure and open system, etc.). Currently distributing gasoline and bioethanol blended fuel (blended 10%) were stored for 18 weeks in summer season. The sample stored in PE vessel was out of quality standard (octane number, vapor pressure, etc.) due to the evaporation of the high octane number and low boiling point components through the vessel cap and surface. Especially, the sunlight exposure sample stored in PE vessel showed rapid decrease of vapor pressure and increase of gum. Bioethanol blended fuel showed similar results as gasoline.
Objectives : Oxidative stress due to excessive accumulation of reactive oxygen species (ROS) is one of the risk factors for the development of several chronic diseases, including neurodegenerative diseases. Methods : In the present study, we investigated the protective effects of cheongnoemyeongsin-hwan (CNMSH) against oxidative stress‑induced cellular damage and elucidated the underlying mechanisms in neuronal-derived SH-SY5Y cells. Results : Our results revealed that treatment with CNMSH prior to hydrogen peroxide (H2O2) exposure significantly increased the SH-SY5Y cell viability, indicating that the exposure of the SH-SY5Y cells to CNMSH conferred a protective effect against oxidative stress. CNMSH also effectively attenuated H2O2‑induced comet tail formation, and decreased the phosphorylation levels of the histone ${\gamma}H2AX$, as well as the number of apoptotic bodies and Annexin V‑positive cells. In addition, CNMSH exhibited scavenging activity against intracellular ROS generation and restored the mitochondria membrane potential (MMP) loss that were induced by H2O2, suggesting that CNMSH prevents H2O2‑induced DNA damage and cell apoptosis. Moreover, H2O2 enhanced the cleavage of caspase-3 and degradation of poly (ADP-ribose)-polymerase, a typical substrate protein of activated caspase-3, as well as DNA fragmentation; however, these events were almost totally reversed by pretreatment with CNMSH. Furthermore, CNMSH increased the levels of heme oxygenase-1 (HO-1), which is a potent antioxidant enzyme, associated with the induction of nuclear factor-erythroid 2-related factor 2 (Nrf2). According to our data, CNMSH is able to protect SH-SY5Y cells from H2O2-induced apoptosis throughout blocking cellular damage related to oxidative stress through a mechanism that would affect ROS elimination and activating Nrf2/HO-1 signaling pathway. Conclusions : Therefore, we believed that CNMSH may potentially serve as an agent for the treatment and prevention of neurodegenerative diseases caused by oxidative stress.
