Technology of High Purity Powder Sintering by Ti Scrap Recycling (티타늄 스크랩 재활용에 의한 고순도 분말 소결 기술)

  • In this study, Ti powder was fabricated from Ti scrap by the Hydrogenation-Dehydrogenation (HDH) method. Hydrogenation reactions of Ti scrap occurred at near 450 $^{\circ}C$ with a sudden increase in the reaction temperature and the decreasing pressure of hydrogen gas during the hydrogenation process in the furnace. The dehydrogenation process was also carried out at 750 $^{\circ}C$ for 2hrs in a vacuum of $10^{-4}$ torr. After the HDH process, a deoxidation treatment was carried out with the Ca(purity: 99.5) at 700 $^{\circ}C$ for 2hrs in the vacuum system. It was found that the oxidation content of Ti powder that was deoxidized with Ca showed noticeably lower values, compared to the content obtained by HDH process. In order to fabricate Ti compacts, Ti powder was sintered at $1100\sim1400^{\circ}C$ for 2hrs under a vacuum of $10^{-4}$ torr. The relative density of compact was 94.9% at 1300 $^{\circ}C$. After sintering, all of the Ti compacts showed brittle fracture behavior, which occurred in an elastic range with short plastic yielding up to a peak stress.

  • Objective: The objectives of this study were to investigate the direct antioxidative effect of 90 Kda heat shock protein (Hsp90) obtained from duck muscle. Methods: The interaction of Hsp90 with phospholipids and oxidized phospholipids was studied with surface plasmon resonance (SPR), and their further oxidation in the presence of Hsp90 was evaluated with thiobarbituric acid reactive substances (TBARS) assay. The scavenging effect on the 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azinobis (3-ethylbenzthiazoline-6-sulfonic acid (ABTS) was measured, and the electron paramagnetic resonance (EPR) spectroscopy in combination with 5-tert-Butoxycarbonyl-5-methyl-1-pyrroline-N-oxide and 2-phenyl-4,4,5,5,-tetramethylimidazoline-1-oxyl-3-oxide (PTIO) was utilized to determine the abilities of Hsp90 in scavenging hydroxyl and PTIO radicals. Results: SPR showed Hsp90 could bind with both phospholipids and oxidized phospholipids, and prevent their further oxidation by the TBARS assay. The DPPH and ABTS scavenging activity increased with Hsp90 concentration, and could reach 27% and 20% respectively at the protein concentration of 50 μM. The EPR spectra demonstrated Hsp90 could directly scavenge ·OH and PTIO· radicals. Conclusion: This suggests that Hsp90, a natural antioxidant in meat, may play an important role in cellular defense against oxidative stress, and may have potential use in meat products.

Protective Effect of Dietary Buchu (Chinese chives) Against Oxidative Damage from Aging and Ultraviolet Irradiation in ICR Mice Skin

  • Protective effect of skin by antioxidative dietary buchu (Chinese chives, Allium tuberosum Router), was evaluated in ICR mice fed diets containing 2% or 5% buchu for 12 months. Lipid peroxidation and protein oxidation in skin, with or without ultraviolet B (UVB) irradiation, activities of antioxidative enzymes, total glutathione concentrations, and non-soluble collagen contents were measured. Dietary buchu decreased significantly in TBARS and protein carbonyl levels in skin compared to the control group, and were lower in those fed 5% than 2% buchu diet group. ICR mice exhibited an age-dependent decrease in antioxidative enzyme activities and total glutathione concentrations on the control diet, but in the groups fed buchu diet the enzyme activities and glu-tathione concentrations remained at youthful levels for most of the study. SOD, glutathione peroxidase, and catalase activities as well as total glutathione concentrations increased with time in the skins of the mice fed buchu diets. Lipid peroxidation and protein oxidation provoked by UVB irradiation on ICR mice skin homogenates were also significantly inhibited by dietary buchu. The buchu diets also decreased the formation of non-soluble collagen in mice skin, compared to the control group. These results suggest that antioxidative components and sulfur-compounds in buchu may confer protective effect against oxidative stress resulting from aging and exposure to ultraviolet irradiation.

Yield and Seed Quality as Affected by Water Deficit at Different Reproductive Growth Stages in Soybean

  • The effect of water deficits on soybean [Glycine max (L.) Merr.] could appear on seed quality through changes of morphological plant characteristics. Two Korean genotypes, Hwangkeum (determinate growth habit) and Muhan (indeterminate growth habit), were used to examine the influences of treatment stage and method of water deficit during reproductive growth period on yield and seed quality of soybean. Water deficit at R5 or R6 stages was as damaging to seed quality as double water-deficit treatments at R2+R5 or R2+R6. However, seed from double water-deficit treatment tended to have lower oxidation-reduction potential compare to the corresponding single water-deficit treatment. In comparison with Muhan, Hwangkeum had significantly greater oxidation-reduction potential value. Seed yield per plant in both genotypes depended greatly on seed yield of branches. However, the proportion of number of branch seed to total seed umber in Hwangkeum was increased as the water deficit was applied during later reproductive stage, whereas, in Muhan the proportion was lower. Water-deficit treatments including the single and double water-deficit treatments and non-stressed treatment were able to be classified into five groups for Hwangkeum and four groups for Muhan based on the influences on yield components, number of pod, number of seed, and single seed weight, using principal component analysis. In both genotypes, R2+R5 water-deficit treatment decreased number of pod and seed, but increased single seed weight. On the contrary, R6 or R2+R6 stress increased the pod and seed number, but decreased single seed weight.

Oxidation-Induced Conformational Change of a Prokaryotic Molecular Chaperone, Hsp33, Monitored by Selective Isotope Labeling

  • Hsp33, a prokaryotic molecular chaperone, exerts holdase activity in response to oxidative stress. In this study, the stepwise conformational change of Hsp33 upon oxidation was monitored by NMR. In order to overcome its high molecular weight (33 kDa as a monomer and 66 kDa as a dimer), spectra were simplified using a selectively [$^{15}N$]His-labeled protein. All of the eight histidines were observed in the TROSY spectrum of the reduced Hsp33. Among them, three peaks showed dramatic resonance shifts dependent on the stepwise oxidation, indicating a remarkable conformational change. The results suggest that unfolding of the linker domain is associated with dimerization, but not entire region of the linker domain is unfolded.

Effect of Anodized Carbon Fiber Surfaces on Mechanical Interfacial Properties of Carbon Fibers-reinforced Composites (탄소섬유의 양극산화가 탄소섬유 강화 복합재료의 기계적 계면 특성에 미치는 영향)

  • In this work, the effect of anodic oxidation on surface characteristics of high strength PAN-based carbon fibers was investigated in mechanical interfacial properties of composites. The surface properties of the carbon fibers were determined by acid-base values, scanning electron microscopy (SEM), X-ray photoelectron spectroscopy (XPS), and contact angles. And their mechanical interfacial properties of the composites were studied in interlaminar shear strength (ILSS) and critical stress intensity factor ($K_{IC}$). As a result, the acidity or the $O_{ls}/C_{ls}$ ratio of carbon fiber surfaces was increased, due to the development of the oxygen functional groups. Consequently, the anodic oxidation led to an increase in surface free energy of the carbon fibers, mainly due to the increase of its specific (or polar) component. The mechanical interfacial properties of the composites, including ILSS and $K_{IC}$, had been improved in the anodic oxidation on fibers. These results were explained that good wetting played an important role in improving the degree of adhesion at interfaces between fibers and epoxy resin matrix.

  • Microstrip line was fabricated on the oxidized porous silicon layer which has nearly electrically and chemically identical properties with thermally oxidized silicon layer. Thick oxidized porous silicon layer of few tenth of micrometers was prepared by thermal oxidation of porous silicon layer on silicon substrate. Multi-step thermal oxidation process was used to obtain high Quality and thick oxidized silicon layer and to release thermal stress. Microstrip line was fabricated on the oxidized porous silicon layer. Its microwave characteristics were measured and the availability for MMIC substrate was investigated.

  • As the operating conditions in a supercritical oxidation reactor are set in high temperature with high pressure causing a reactor suffering from the harsh circumstances. It means the reactor adopts itself with Fe-Cr alloy in acidic atmosphere with low pH value and Ni alloy in basic atmosphere with high pH value due to its superior corrosion resistance. The study, whose target waster water is pertinent to the latter part, has selected Ni alloy such as ostenite type stainless steel 304 and 316, superstainless steel AL6XN, Inconel 625, MAT 21, and titanium Gr. 5 in order to measure corrosion resistance against those samples under the same conditions of temperature and pressure applied for a supercritical oxidation reactor. The result shows the identifiable difference in corrosion resistance by observing the surface states through a scanning probe microscope as well as measuring the weight loss through making the samples above deposited in wastewater for two-week and four-week stay. The purpose of this corrosion experiment is to identify the most corrosion-resistant material among sample species pre-selected according to pH concentration of wastewater in pursue of applying for a reactor exposed to the extreme corrosion environment. It is because such a reactor made of a verified material enables to safeguard a stable operation under the supercritical wastewater processing facility.

Role of tetrahydrobiopterin in dopaminergic cell death: Relevance to Parkinson's disease

  • Parkinson's disease (PD) is a progressive neurodegenerative disorder affecting $1\%$ of the population above the age of 65 and is characterized by a selective loss of dopaminergic neurons in the substantia nigra pars compacta. Although the underlying cause of dopaminergic cell death or the mechanism by which these cells degenerate is still not clearly understood, oxidative stress, mitochondrial dysfunction, and protein misfolding are thought to play important roles in the dopaminergic degeneration in PD. Tetrahydrobiopterin (BH4) is synthesized exclusively in the monoaminergic, including dopaminergic, cells and serves as an endogenous and obligatory cofactor for syntheses of the potential oxidative stressors dopamine and nitric oxide. In addition to its contribution toward the syntheses of these two potentially toxic molecules, BH4 itself can directly generate oxidative stress. BH4 undergoes oxidation during the hydroxylation reaction as well as nonenzymatic autooxidation to produce hydrogen peroxide and superoxide radical. We have previously suggested BH4 as an endogenous molecule responsible for the dopaminergic neurodegeneration. BH4 exerts selective toxicity to dopamine-producing cells via generation of oxidative stress, mitochondrial dysfunction, and apoptosis. BH4 also induces morphological, biochemical, and behavioral characteristics associated with PD in vivo. BH4 as well as enzyme activity and gene expression of GTP cyclohydrolase I, the rate-limiting enzyme in BH4 synthesis pathway, are readily upregulated by cellular changes such as calcium influx and by various stimuli including stress situations. This points to the possibility that cellular availability of BH4 might be increased in aberrant conditions, leading to increased extracellular BH4 subsequent degeneration. The fact that BH4 is specifically and endogenously synthesized in dopaminergic cells, Is readily upregulated, and generates oxidative stress-related cell death provides physical relevance of this molecule as an attractive candidate with which to explain the mechanism of pathogenesis of PD.

SIRT1 Suppresses Activating Transcription Factor 4 (ATF4) Expression in Response to Proteasome Inhibition

  • The synthetic machinery of ATF4 (activating transcription factor 4) is activated in response to various stress conditions involved in nutrient restriction, endoplasmic reticulum homeostasis, and oxidation. Stress-induced inhibition of proteasome activity triggers the unfolded protein response and endoplasmic reticulum stress, where ATF4 is crucial for consequent biological events. In the current study, we showed that the $NAD^+$-dependent deacetylase, SIRT1, suppresses ATF4 synthesis during proteasome inhibition. SIRT1 depletion via transfection of specific siRNA into HeLa cells resulted in a significant increase in ATF4 protein, which was observed specifically in the presence of the proteasome inhibitor MG132. Consistent with SIRT1 depletion data, transient transfection of cells with SIRT1-overexpressing plasmid induced a decrease in the ATF4 protein level in the presence of MG132. Interestingly, however, ATF4 mRNA was not affected by SIRT1, even in the presence of MG132, indicating that SIRT1-induced suppression of ATF4 synthesis occurs under post-transcriptional control. Accordingly, we propose that SIRT1 serves as a negative regulator of ATF4 protein synthesis at the post-transcriptional level, which is observed during stress conditions, such as proteasome inhibition.
