Comparative transcriptome analysis of the protective effects of Korean Red Ginseng against the influence of bisphenol A in the liver and uterus of ovariectomized mice

  • Background: Bisphenol A (BPA), known as an endocrine disruptor, is widely used in the world. BPA is reported to cause inflammation-related diseases. Korean Red Ginseng (KRG) has been used safely in human for a long time for the treatment of diverse diseases. KRG has been reported of its mitigating effect on menopausal symptoms and suppress adipose inflammation. Here, we investigate the protective effect of orally administered KRG on the impacts of BPA in the liver and uterus of menopausal mice model. Methods: The transcriptome analysis for the effects of BPA on mice liver was evaluated by Gene Expression Omnibus (GEO) database-based data (GSE26728). In vivo assay to evaluate the protective effect of KRG on BPA impact in ovariectomized (OVX) mice were designed and analyzed by RNA sequencing. Results: We first demonstrated that BPA induced 12 kinds of gene set in the liver of normal mice. The administration of BPA and KRG did not change body, liver, and uterine weight in OVX mice. KRG downregulated BPA-induced inflammatory response and chemotaxis-related gene expression. Several gene set enrichment analysis (GSEA)-derived inflammatory response genes increased by BPA were inhibited by KRG in OVX mice. Conclusion: Our data suggest that BPA has commonly influenced inflammatory response effects on both normal and OVX mice. KRG protects against BPA impact of inflammatory response and chemotaxis in OVX mouse models. Our comparative analysis will provide new insight into the efficacy of KRG on endocrine disrupting chemicals and OVX mouse.

Selection of the Fittest Anti-osteoporotic Mixed Compositions Consist of Morindae Radix and Cistanchis Herba Aqueous Extracts on Ovariectomized Mice (난소적출 마우스를 이용한 골다공증 개선효과를 나타내는 파극천(巴戟天)과 육종용(肉蓗蓉) 열수(熱水) 추출물(抽出物)의 복합 최적 조성 선택 실험)

  • Objectives: To select the optimal ranges showing obvious synergic anti-osteoporotic potential after adjust mixed formula consisted of Morindae Radix (MR) and Cistanchis Herba (CH) as compared with those of each single formula or risedronate sodium (RES) using bilateral ovariectomized (OVX) female mice. Methods: Fourteen groups, total eight sham or 104 OVX mice were selected based on the body weights at 34 days after OVX surgery. After that, 9 types mixed compositions, single formula of MR and CH, and RES were orally administered for 35 days. And we measured changes in body weight and gain, femur weight, bone mineral density (BMD), bone strength (failure load) and mineral content - calcium (Ca) and inorganic phosphorus (IP), osteocalcin contents and bone specific alkaline phosphatase (bALP) activities of all mice. Results: The OVX-induced estrogen-deficient osteoporotic signs were significantly inhibited by 35 days of continuous oral treatment of all treated mice as compared with OVX control mice. Especially, MR:CH 1:3 and 1:1 mixed formula treated mice showed significantly more favorable inhibitory activities against estrogen-deficient osteoporosis symptoms as compared to those of each single formula of MR and CH. Although RES also ameliorated the decreases of the femur BMD, strength and trabecular bone architectures through the inhibited the increases of bone turnover, but they did not critically influenced on the bone formations. Conclusions: The results suggest that MR:CH 1:3 mixed formula showed somewhat lower anti-resorptive effects as compared to those of RES, but they also showed bone formation effects. therefore, it is expected that MR:CH 1:3 mixture will be promising as a potent protective agents for relieving the osteoporosis in menopausal women.

Effect of Exopolymers of Aureobasidium pullulans on Improving Osteoporosis Induced in Ovariectomized Mice

  • Treatment with exopolymers of Aureobasidium pullulans SM-2001 containing $\beta-1,3/1,6-glucan$ inhibited osteoclastogenesis of bone marrow stem cells in a co-culture system with calvariae osteoblastic cells. In addition, the treatment increased mineral deposition in osteoblastic cells. These two observations prompted us to evaluate whether the exopolymers could be used as an anti-osteoporotic agent, and efficacy of the exopolymers to prevent bone loss was compared with alendronate, a bisphosphonate, in ovariectomized mice prone to osteoporosis. Administration of the exopolymers to the ovariectomized mice resulted in improved effects on femur weight and histomorphometric changes of femur such as trabecular bone volume (TBV), trabecular bone thickness (TBT), and cortical bone thickness (CBT). In conclusion, the exopolymers treatment inhibited bone loss from osteoporosis induced by ovariectomy, and the effect was comparable to alendronate administration.

Effect of swimming exercise on anti-angiogenesis of white adipose tissue in high-fat diet-fed female ovariectomized mice (고지방식이를 섭취한 난소절제 암컷 쥐의 수영운동이 백색지방조직의 항혈관신생에 미치는 효과)

  • This study investigated whether swimming exercises improves obesity through regulation of angiogenesis in white adipose tissue. Female mice with high-fat diets were divided into sham-operated group (Sham), ovariectomized group (OVX), and swim-trained ovariectomized group (OVX + Swim). Compared to the Sham, OVX increased body weight, adipose tissue mass and size of adipocyte. However, these factors (: such as body weight, adipose tissue mass and size of adipocyte) of OVX + Swim decreased compared with OVX. Compared with the Sham, OVX increased the mRNA expression of angiogenic activator and MMPs and decreased the mRNA expression of angiogenic inhibitors in white adipose tissue. But OVX + Swim decreased the mRNA expression of angiogenic activator and MMPs and increased the mRNA expression of angiogenic inhibitors in white adipose tissue, compared with the OVX. Theses results suggested that swimming exercises the angiogenesis in white adipose tissue, resulting to improve obesity in high-fat diet-fed female OVX mice.

Effects of Herbal Medicine on Tibia Bone Changes in Ovariectomized Rats and SAM P6 Mice (한약이 난소제거 흰쥐와 SAM P6 생쥐의 골소주에 미치는 영향)

  • The effects of herbal medicine on travecular bone area were studied using ovariectomized rat as an animal model of Type I osteoporosis and SAM P6 as that of Type II. We counted red blood cells(RBC), hemoglobin(Hb), and hematocrit(Hct) using Couter`sR method. Each traditional boiling water extract of Achyrathis Radix, Psoraleae Fructus, Rehmanniae Radix Preparat, and Cornii Fructus and a systemic water extract of Astragali Ractiex was given 5g/kg/day, p.o., for 30 days in a group of 4-5 ovariectormized rats. One ml of blood was taken by tail vein at day 0, 7, 14, 21, and 30 days after administration of the extract. The traditional hot water extract of Cervi parvumn Corni (Cervi) was given the same dose as described above for 14 days in a group of 10 SAM P6 mice and systemic water extract of Astragali Radix was administered as the same dose as above for 30 days in 10 SAM P6 mice. Travecular bone area was measured 5 mcm decalcified and stained thin bone slice by image analysis using a digitalizer. In Type I, ovariectomized rats, administration of Astragali Radix, Rhemanniae Radix Preparat, and Corni Fructus decreased in RBC, Hb and Hct. In Type II, administration of Cervi increased in RBC and Hct and that of Astragali Radix was also elevated RBC. In Type I, any administration of herbal medicine used in this study did not elevate travecular bone area significantly except Corni Fructus showed a trend of increase in travecular bone area. However, Type II, Cervi and Astragali Radix increased in both mean and total travecular bone area. Thus, there are significant difference in response of herbal medicine in different types of osteoporosis.

Synergistic effect of soy isoflavone and swimming exercise on improvement of liver function in ovariectomized mice (대두 이소플라본과 수영운동이 난소절제 쥐의 간 기능 개선에 미치는 시너지 효과)

  • Soy isoflavones are attracting attention from postmenopausal women because of their beneficial effects on menopausal symptoms. This study was investigated whether a combination of soy isoflavone genistein and swimming exercise (Gen+SE) would have a beneficial synergistic effect on obesity and improvement of liver function compared to the genistein only (Gen) and swimming exercise only (SE) in ovariectomized mice. Ovariectomized mice were randomly divided into control group (Con), Gen, SE, and Gen+SE, and were fed a high-fat diet for 8 weeks. As a result of examining the body weight, weight of white adipose tissue, lipid accumulation of liver, and serum ALT and AST levels, both Gen and SE decreased compared to Con, and Gen+SE decreased more than compared to Gen and SE. The expression of inflammatory cytokines MCP-1, IL-6 and TNF-𝛼 genes in liver decreased in both Gen and SE compared to Con, and were further decreased in Gen+SE compared to Gen and SE. But The expression of adiponectin showed opposite results. The expression of fatty acid oxidation related genes in liver increased in both Gen and SE compared to Con, and were more effectively than increased in Gen+SE compared to Gen and SE. Therefore this study suggests that the interaction between soy isoflavone and swimming exercise is very effective controlling obesity and recovering decreased liver function, and this is caused by promoting fatty acid oxidation in the liver in ovariectomized mice.

Anti-Osteoporosis Effects of the Fruit of Sea Buckthorn (Hippophae rhamnoides) through Promotion of Osteogenic Differentiation in Ovariectomized Mice

  • The fruit of Hippophae rhamnoides has been widely used for medicinal purposes because of its anti-inflammatory, antioxidant, antiplatelet, and antimicrobial effects. Since there are no clear reports on the therapeutic efficacy of H. rhamnoides in osteoporosis, this study aimed to confirm the potential use of H. rhamnoides for the treatment of osteoporosis through its osteogenic differentiation-promoting effect in ovariectomized mice. Through an in vitro study, we compared the effects of the EtOH extract of H. rhamnoides fruits (EHRF) on the differentiation of C3H10T1/2, a mouse mesenchymal stem cell line, into osteoblasts based on alkaline phosphatase (ALP) staining and the relative expression of osteogenesis-related mRNAs. The EHRF significantly stimulated the differentiation of mesenchymal stem cells into osteoblasts and showed 7.5 times (* p < 0.05) higher osteogenesis than in the untreated control. A solvent fractionation process of EHRF showed that the hexane-soluble fraction (HRH) showed 10.4 times (** p < 0.01) higher osteogenesis than in the untreated control. Among the subfractions derived from the active HRH by preparative HPLC fractionation, HRHF4 showed 7.5 times (* p < 0.05) higher osteogenesis than in the untreated naïve cells, and HRH and HRHF4 fractions showed 22.6 times (*** p < 0.001) stronger osteogenesis activity than in the negative control. Osteoporosis was induced by excision of both ovaries in 9-week-old female ICR mice for in vivo analysis, and two active fractions, HRH and HRHF4, were administered orally for three months. During the oral administration period, body weight was measured weekly, and bone mineral density (BMD) and body fat density were measured simultaneously using a DEXA machine once a month. In particular, during the in vivo study, the average BMD of the ovariectomized group decreased by 0.0009 g/cm2, whereas the average BMD of the HRH intake group increased by 0.0033 g/cm2 (* p < 0.05) and that of the HRHF4 intake group increased by 0.0059 g/cm2 (** p < 0.01). The HRH and HRHF4 intake groups significantly recovered the mRNA and protein expression of osteogenic genes, including ALP, Osteopontin, Runx2, and Osterix, in the osteoporosis mouse tibia. These findings suggest that the active fractions of H. rhamnoides fruit significantly promoted osteoblast differentiation in mesenchymal stem cells and increased osteogenic gene expression, resulting in an improvement in bone mineral density in the osteoporosis mouse model. Taken together, H. rhamnoides fruits are promising candidates for the prevention and treatment of osteoporosis.

A comparative study on the osteoporotic animal models in ddY mouse: ovariectomized and neurectomized models (ddY 마우스를 이용한 골다공증 모델에 대한 비교연구: 난소적출 및 신경절단 모델)

  • In order to compare the induced time of osteoporosis between ovariectomized and neurectomized models in ddY mice. Experimental groups were divided into Sham, ovariectomized (OVX group) and neurectomized (NX group) group. The changes of body weight, tibia weight and histomorphometry of epiphyseal regions of tibia that were generally used as criteria index in osteoporosis, were evaluated at 2 and 4 weeks after operations with other generally used index-changes of serum osteocalcin. Also, calcium and phosphorus levels in the ash tibia were demonstrated with their ratio (Ca/P ratio). From the result of this study, evidences which reflect osteoporotic states of animals such as decrease of absolute and relative tibia weight, histomorphometrical index of epiphyseal region of tibia including trabecular bone volume %, and calcium and phosphorous contents in tibia, were generally detected from 4 weeks after ovariectomy and 2 weeks after neurectomy with increase of serum osteocalcin levels. In conclusion, it is considered that more rapid and favorable osteoporosis was induced in neurectomized model compared to that of ovariectomized model.

Acer tegmentosum Maxim Prevents Bone Loss by Inhibiting Osteoclastogenesis and Promoting Osteoblast Mineralization in Ovariectomized Mice

  • Osteoporosis is a worldwide disease leading to significant economic and societal burdens globally. Osteoporosis is caused by unbalanced bone remodeling between the rate of osteoclast bone resorption and osteoblast bone formation. Acer tegmentosum Maxim (AT) is a traditional herbal medicine containing multiple biological activities such as anti-oxidant and anti-inflammatory purposes. However, its role in osteoporosis has not been fully studied. Therefore, we investigated whether AT has a potent inhibitory effect on osteoporosis and its mechanism through a systemic evaluation in ovariectomized (OVX) mice. OVX mice were orally administrated with the AT at doses of 50, 100, and 200 mg/kg for 10 weeks. Histological images and histomorphometry analyses were performed by H&E and Toluidine blue satin, and the expression levels of receptor activator for nuclear factor-kB ligand (RANKL), nuclear factor of activated T cells cytoplasm 1 (NFATc1), c-Fos, and matrix metalloproteinase 9 (MMP9) related to the osteoclast differentiation were investigated using immunohistochemical analysis. Administration of AT prevented bone loss and the alternations of osteoporotic bone parameters at the distinct regions of the distal femur and spongiosa region in OVX mice. Further, administration of AT increased periosteal bone formation in a dose-dependent manner. Meanwhile, AT inhibited not only the expression of NFATc1 and c-Fos, which are two major regulators of osteoclastogenesis but also reduced bone resorbed encoding expression of MMP9 and RANKL. Our results indicated that administration of AT prevented bone loss and the alternations of osteoporotic bone parameters at the distinct regions of the distal femur and spongiosa region in OVX mice. Also AT has the bone protective effect through the suppression of osteoclast and promotion of osteoblast, suggesting that it could be a preventive and therapeutic candidate for anti-osteoporosis.

Combined effects of soy isoflavone and lecithin on bone loss in ovariectomized mice

  • BACKGROUND/OBJECTIVES: Isoflavones (ISFs) are effective in preventing bone loss, but not effective enough to prevent osteoporosis. Mixtures of soy ISF and lecithin (LCT) were prepared and characterized in an attempt to improve the bone loss. MATERIALS/METHODS: The daidzein (DZ) and genistein (GN) solubility in soy ISF were measured using liquid chromatography-mass spectrometry. The change in the crystalline characteristics of soy ISF in LCT was evaluated using X-ray diffraction analysis. Pharmacokinetic studies were conducted to evaluate and compare ISF bioavailability. Animal studies with ovariectomized (OVX) mice were carried out to estimate the effects on bone loss. The Student's t-test was used to evaluate statistical significance. RESULTS: The solubility of DZ and GN in LCT was 125.6 and 9.7 mg/L, respectively, which were approximately 25 and 7 times higher, respectively, than those in water. The bioavailability determined by the area under the curve of DZ for the oral administration (400 mg/kg) of soy ISF alone and the soy ISF-LCT mixture was 13.19 and 16.09 ㎍·h/mL, respectively. The bone mineral density of OVX mice given soy ISF-LCT mixtures at ISF doses of 60 and 100 mg/kg daily was 0.189 ± 0.020 and 0.194 ± 0.010 g/mm3, respectively, whereas that of mice given 100 mg/kg soy ISF was 0.172 ± 0.028 g/mm3. The number of osteoclasts per bone perimeter was reduced by the simultaneous administration of soy ISF and LCT. CONCLUSIONS: The effect of preventing bone loss and osteoclast formation by ingesting soy ISF and LCT at the same time was superior to soy ISF alone as the bioavailability of ISF may have been improved by the emulsification and solvation of LCT. These results suggest the possibility of using the combination of soy ISF and LCT to prevent osteoporosis.
