Histological Changes of Pituitary Gland Associated with Ovarian Follicular Cyst in Korean Native Cattle (난포낭종(卵胞囊腫)에 나환(羅患)된 한우(韓牛) 뇌하수체(腦下垂體)의 조직학적변화(組織學的變化))

  • To investigate the changes of pituitary gland associated with ovarian cyst in Korean native cattle, pituitaries and ovaries were collected from 54 Korean native cattle at abbatoir. Pituitaries were stained with HerIant pituitary stain method and all the tissues were examined under light microscope. The results obtained were summarized as follows; 1. The delta cells and beta cells in the pars distalis were dull blue and violet in colors respeetively. Basophil size in the follicular phase and pregnant groups was larger than those of luteal phase and ovarian dysfunction groups. 2. The numbers of delta cells in the pars distalis of follicular cyst group were larger than those of remaining groups (p<0.01). 3. The distribution of delta cells in the acidophil zone was greater than that in the basophil zone of the follicular cyst and follicular phase groups. 4. The granulations of delta cells were more intensive in follicular cyst, follicular phase and pregnancy groups than in luteal phase and ovarian dysfunction groups (p<0.01). 5. The numbers of beta cells in follicular phase and ovarian dysfunction groups were larger than those in luteal phase and follicular cyst groups respectively (p<0.01). 6. In all groups, the distribution of beta cells was greater in basophilic zone than in acidophilic zone (p<0.01).

Ovarian stimulation and liver dysfunction: Is a clinical relationship possible? A case of hepatic failure after repeated cycles of ovarian stimulation

  • Liver damage induced by ovarian stimulation has been demonstrated in some cases reported in the literature. However, there has never been a fruitful debate on this topic. The present manuscript tried to fill this gap. We reported a case of a 35-year-old nulliparous woman admitted to our obstetric emergency room for severe pre-eclampsia. She had been subjected to four cycles of controlled ovarian stimulation for intrauterine insemination. At 32 weeks of gestation, she developed severe pre-eclampsia, which led to HELLP syndrome complicated by fatal liver failure. The etiological link between ovarian stimulation and HELLP syndrome is intriguing. Further investigations are needed to understand whether repeated ovarian stimulation may represent a risk factor in pre-eclamptic patients.

  • In reproduction, mitochondria produce bioenergy, help to synthesize biomolecules, and support the ovaries, oogenesis, and preimplantation embryos, thereby facilitating healthy live births. However, the regulatory mechanism of mitochondria in oocytes and embryos during oogenesis and embryo development has not been clearly elucidated. The functional activity of mitochondria is crucial for determining the quality of oocytes and embryos; therefore, the underlying mechanism must be better understood. In this review, we summarize the specific role of mitochondria in reproduction in oocytes and embryos. We also briefly discuss the recovery of mitochondrial function in gametes and zygotes. First, we introduce the general characteristics of mitochondria in cells, including their roles in adenosine triphosphate and reactive oxygen species production, calcium homeostasis, and programmed cell death. Second, we present the unique characteristics of mitochondria in female reproduction, covering the bottleneck theory, mitochondrial shape, and mitochondrial metabolic pathways during oogenesis and preimplantation embryo development. Mitochondrial dysfunction is associated with ovarian aging, a diminished ovarian reserve, a poor ovarian response, and several reproduction problems in gametes and zygotes, such as aneuploidy and genetic disorders. Finally, we briefly describe which factors are involved in mitochondrial dysfunction and how mitochondrial function can be recovered in reproduction. We hope to provide a new viewpoint regarding factors that can overcome mitochondrial dysfunction in the field of reproductive medicine.

A Retrospective Study on the Influence of Kamisoyo-san (Jiaweixiaoyao-san) on Improving Ovarian Function in Child-bearing Period Infertile Women with Ovarian Dysfunction (가미소요산(加味逍遙散)이 가임기 난소기능저하 불임여성의 난소기능개선에 미치는 영향에 관한 후향적 연구)

  • Objectives: This study was performed to elicit the effectiveness of a herbal formula, Kamisoyo-san (Jiaweixiaoyaosan) on enhancing ovarian function in infertile woman with ovarian dysfunction. Methods: 28 patients who initially visited between November 2006 and February 2009 and were administered Kamisoyo-san until May 2009 were retrospectively evaluated for their ovarian function by means of basal FSH (b-FSH), menstrual cycle, body mass index (BMI), and body fat ratio. To identify the major factor of improving ovarian function, 28 patients were classified into two groups by criteria of patent factors, such as pre-administration b-FSH, patient age, and treatment duration, respectively. Two groups were compared in terms of pregnancy percentage, b-FSH, menstrual cycle, BMI, and body fat ratio. Results: Post-administration b-FSH significantly decreased in comparison with pre-administration (p=0.004). The higher group (b-FSH $\geq$ 25mIU/mL) in pre-administration b-FSH was more effective in decrease of post-administration b-FSH than the lower one (10mIU/mL < b-FSH < 25mIU/mL). Conclusion: Kamisoyo-san may have a therapeutic effect on the infertility of child bearing period woman with ovarian dysfunction.

Progesterone assays as an aid for improving reproductive efficiency in dairy cattle II. Use of plasma or milk progesterone profiles for differential diagnosis of ovarian cysts (Progesterone 농도측정(濃度測定)에 의한 유우(乳牛)의 번식효율증진(繁殖效率增進)에 관한 연구(硏究) II. 혈액(血液) 및 유즙(乳汁)중 progesterone 농도측정(濃度測定)에 의한 난소낭종(卵巢囊腫)의 감별진단(鑑別診斷))

  • In 60 dairy cows with inactive ovaries, follicular cyst, luteal cyst, persistent corpus luteum and silent heat as diagnosed by rectal palpation, and those that had not resumed ovarian cycles until 60 days postpartum, progesterone concentrations for differential diagnosis of reproductive disorders were measured and were compared in matched plasma, skim milk and milk fat samples at 10 days interval. The incidence rate of reproductive disorders were as follows; inactive ovaries 20(33.3%), silent heat 11(18.3%), follicular cyst 7(11.7%), luteal cyst 7(11.7%), persistent corpus luteum 7(11.7%), pyometra 4(6.7%), vaginitis 2(3.3%), cystic corpus luteum 1(1.7%), and endometritis 1(1.7%), respectively. Cows having a progesterone concentration in plasma and skim milk < 1.0 ng/ml, and in milk fat < 80.0 ng/ml were considered to have inactive ovaries or follicular cyst. Those with concentrations in plasma and skim milk ${\geq}1.0ng/ml$, and in milk fat ${\geq}80.0ng/ml$ were regarded as the cases of luteal cyst or persistent corpus luteum. Progesterone concentrations in above cows did not differ significantly between the time of initial determination and the 10 days after initial determination. But progesterone concentrations in cows with silent heat did differ significantly between the time of initial determination and the 10 days after initial determination(P<0.05). The accuracy of rectal palpation for making a differential diagnosis of ovarian dysfunction, as defined on basis of progesterone concentrations, were as follows; follicular cyst 55.6%, luteal cyst 50.0%, inactive ovaries 90.5% and persistent corpus luteum 60.0%, respectively. It may be concluded that progesterone determinations at 10 days interval is practical as an aid to diagnosing ovarian dysfunction, particularly follicular cyst and luteal cyst.

  • Inborn errors of metabolism (IEM) are very rare and genetically transmitted diseases and have man y different symptoms related with multisystemic involvement. More rarely, endocrinopathies can be an early and first symptom of IEM, but presents with signs of later complications in adolescent or adulthood. The mechanisms of endocrine dysfunction in IEM are poorly understood. Hypogonadotropic hypogonadism is common in hemochromatosis, adrenoleukodystrophy, galactosemia, and glycogen storage disease. Many girls with classic galactosemia are at high risk for premature ovarian insufficiency (POI), despite an early diagnosis and good control. Mitochondrial diseases are multisystem disorders and are characterized by hypo- and hypergonadotrophic hypogonadism, thyroid dysfunction and insulin dysregulation. Glycogen storage disorders (GSDs), especially type Ia, Ib, III, V are assocciated with frequent hypoglycemic events. IEM is a growing field and is not yet well recognized despite its consequences for growth, bone metabolism and fertility. For this reason, clinicians should be aware of these diagnoses and potential endocrine dysfunction.

Studies on the improvement of reproductive efficiency in Korean native cows -Plasma progesterone concentrations for monitoring ovarian activity in the postpartum period- (한우(韓牛)의 번식효율(繁殖效率) 증진(增進)에 관한 연구(硏究) -혈중(血中) progesterone농도(濃度) 측정(測定)에 의한 분만(分娩)후 난소기능(卵巢機能) 회복상태(回復狀態)의 검토(檢討)-)

  • Plasma progesterone concentrations in 44 suckling Korean native cows were determined to monitor ovarian activity postpartum by radioimmunoassay. Blood samples were collected in 3 day intervals from 15 to 80 days postpartum. The ovaries and uterus were examined in 6 day intervals by rectal palpation. Results are summaried as follows: 1. The cows were qualified into four categories; Type I(normal): cyclic changes in plasma progesterone concentrations appear within 45 days postpartum(35 cows, 79.5%), Type II(cycle delayed): delayed resumption of ovarian activity by 55 days postpartum(5 cows, 11. 4%), Type III(cycle ceased with low progesterone): plasma progesterone concentrations remained low(${\leq}1ng/ml$) until 80 days postpartum(3 cows, 6.8%), Type IV(cycle ceased with high progesterone): plasma progesterone concentrations after 30 days remained high(${\geq}4.0ng/ml$) until 80 days postpartum(1 cow, 2.3%). 2. Out of the cows classified Type II and III, 7 cows had inactive ovaries and a cow had follicular cyst by rectal palpation. The cow of Type IV was diagnosed as bearing persistent corpus luteum by rectal palpation. 3. About 82% of the cows showed significant rises in plasma progesterone concentrations prior to 50 days postpartum and the cows of Type II and III resumed cycles in 31. 6(range 17~55) days after calving. However, 43.2% resumed cycles in 50 days postpartum by estrus signs. These results indicated that plasma progesterone concentrations assessed by radioimmunoassay can be utilized for monitoring postpartum ovarian activity and would be helpful for the early detection of ovarian dysfunction in the Korean native cow.

Neuropeptide Y improves cisplatin-induced bone marrow dysfunction without blocking chemotherapeutic efficacy in a cancer mouse model

  • Cisplatin is the most effective and widely used chemotherapeutic agent for many types of cancer. Unfortunately, its clinical use is limited by its adverse effects, notably bone marrow suppression leading to abnormal hematopoiesis. We previously revealed that neuropeptide Y (NPY) is responsible for the maintenance of hematopoietic stem cell (HSC) function by protecting the sympathetic nervous system (SNS) fibers survival from chemotherapy-induced bone marrow impairment. Here, we show the NPY-mediated protective effect against bone marrow dysfunction due to cisplatin in an ovarian cancer mouse model. During chemotherapy, NPY mitigates reduction in HSC abundance and destruction of SNS fibers in the bone marrow without blocking the anticancer efficacy of cisplatin, and it results in the restoration of blood cells and amelioration of sensory neuropathy. Therefore, these results suggest that NPY can be used as a potentially effective agent to improve bone marrow dysfunction during cisplatin-based cancer therapy.

Effect of Sofosbuvir on rats' ovaries and the possible protective role of vitamin E: biochemical and immunohistochemical study

  • Hepatitis C virus (HCV) infection is a major health problem worldwide and its eradication is mandatory. Direct acting HCV polymerase inhibitors, such as Sofosbuvir (SOF), is an effective regimen. However, it has some side effects like mutagenesis, carcinogenesis, and the impairment of testicular function. It is important to evaluate the safety of SOF on the ovary, as there are no studies yet. Increasing the production of Reactive Oxygen Species (ROS), causes oxidative stress, which affects ovulation process, female reproduction, and fertility. Accumulation of SOF in the cells was demonstrated to promote ROS generation. Vitamin E (Vit E) is an antioxidant agent that has an essential role in the female reproductive system, its deficiency can cause infertility. We explored the effect of SOF treatment alone and co-treated with Vit E on ovarian ROS level and ovarian morphology experimentally using biochemical and immunohistochemical studies. Significant changes in oxidative stress markers; nitric oxide and malondialdehyde lipid peroxidation, antioxidant enzymes; catalase, super oxide dismutase, and reduced glutathione, proliferating markers; proliferation cell nuclear antigen and Ki-67 antigen and caspase 3 apoptotic marker were demonstrated. It was shown that where SOF induced oxidative stress, it also aggravated ovarian dysfunction. The essential role of Vit E as an antioxidant agent in protecting the ovarian tissue from the effect of oxidative stress markers and preserving its function was also displayed. This could be guidance to add Vit E supplements to SOF regimens to limit its injurious effect on ovarian function.

  • Granulosa cells, which surround the oocyte within the ovarian follicle, play an essential role in creating conditions required for the development of oocytes and follicles. The solute carrier family (SLC) is comprised of influx transporters of steroidal hormones, various drugs, and several other substrates. The differential expression of selected DEGs was confirmed using in situ hybridization analysis. SLC23A3 and SLC39A10 were highly expressed in the ovary. The SLC39A10 gene was expressed in the primordial follicle stage, but SLC23A3 was expressed in the growing follicle stage. Contrastingly, the expression of SLC23A3 was increased in granulosa cells at the growing follicle stage. The differential expressions of SLC23A3 and SLC39A10 between the primordial and primary follicles were additionally confirmed by using follicle isolations. The gene expression profile from the present study may provide insight for future studies on the mechanism(s) involved in primordial-primary follicular transition and suggestions to promote follicular development in ovarian dysfunction.
