Granulosa Cell Tumor of Ovary in a Yorkshire terrier Dog (요크셔테리어에서 발생한 난소의 Granulosa cell tumor)

  • A 12-year-old, intact female Yorkshire terrier was presented with progressive abdominal distention. On radiographic and ultrasonographic evaluation, a large mass was detected in right upper abdomen and it had several discrete anechoic follicles and large cysts. The mass was removed by complete ovariohysterectomy and the resected specimen was histopathologically examined. Based on the gross and histopathologic findings, the mass was definitely diagnosed as malignant granulosa cell tumor (GCT). Until now, the dog has been successfully managed more than a year without complications.

  • In order to define the clinical characteristics of Korean women with polycystic ovary syndrome, clinical symptoms, biochemical features and ultrasonographic findings were determined in ninety PCO patients. Frequency of clinical manifestations were as follows:acne 42%, overweight 31 %, oily skin 14 %, hirsutism 10%. Relatively low frequencies of hirsutism is recognized in patients with Korean PCO syndrome. Mean(${\pm}$S.E.) of each hormone was:testosterone 1.18${\pm}$1.07ng/ml, LH 21.47${\pm}$${\pm}$2.6mIU/ml, FSH 7.26${\pm}$2.67mIU/ml, LH/FSH ratio 2.94${\pm}$1.29, prolactin 25.48${\pm}$46.33ng/ml, DHEA-S 333.78${\pm}$309.60ng/dl, 17-0HP 1.72${\pm}$1.74ng/ml. Mean 17-OHP after ACTH stimulation test was 5.07${\pm}$12.01ng/ml. Ultrasonographically measured mean ovarian volume were $11.02{\pm}5.92cm^3$ in right and $9.23{\pm}5.64cm^3$ in left and small sized multiple subcapsular follicular cysts were noted in 43 patients (47.9%) with PCO syndrome.

Survey on the Incidence of Reproductive Disorders in Dairy Cattle

  • Intensive genetic selection has resulted in modern dairy cow with very high milk yields but reduced fertility, due mainly to an increase in reproductive disorders. The study was conducted to estimate the proportion of reproductive disorders among dairy cattle. The factors analyzed were; milk yield, cow parity, periparturient disorders, and reproductive status. The result of this study showed the incidence of reproductive disorders in high yielding dairy cows was prevalent. Repeat breeding was the major postpartum reproductive problem comprising (42.3%) of the samples. Higher reproductive disorders were also associated with lower parity. These disorders subsequently reduced reproductive performance by prolonged intervals from calving to first artificial insemination and from calving to pregnancy. Abnormalities in the reproductive system such as repeat breeder, silent heat, anestrous and ovarian cysts, as well as lower fertility have been recorded during the summer months (June to August). In conclusion, the study revealed that incidence of repeat breeder is mostly prevalent in dairy cows. Moreover, the incidence of reproductive disorders was higher in summer season and hence appropriate preventive and or therapeutic measures as per the type of abnormalities need to be undertaken.

Effect of the Interval from Calving to First Insemination and Days Open on the Subsequent Reproductive Performance in Dairy Cows (젖소에서 분만 후 첫 수정 간격 및 임신 간격이 이후의 번식능력에 미치는 영향)

  • This retrospective study evaluated the effects of the interval from calving to first insemination (ICFI) and days open (DO) on the reproductive performance in dairy cows. In the first analysis, data from 705 cows were grouped based on the ICFI: short (30-60 days, n=217), medium (61-90 days, n=309), and long (91-150 days, n=179). The occurrence of endometritis was greater in the long group than in short and medium groups (p<0.05). The occurrence of ovarian cysts increased with increasing ICFI (p<0.05), while body condition score (BCS) during the 5 month postpartum period was lower with increasing ICFI (p<0.01). The hazard of pregnancy by 365 days in milk (DIM) was lower (hazard ratio [HR]=0.70, p<0.0001) in the long group, but higher (HR=1.41, p<0.0001) in the short group compared with the medium group. In the second analysis, data from 436 cows were grouped based on the DO: short (30-90 days, n=154), medium (91-180 days, n=183), and long (181-360 days, n=99). The occurrence of a retained placenta was greater in the long group than in the medium group (p < 0.05). Ovarian cysts occurred more frequently in medium and long groups than in the short group (p=0.08). BCS was lower in the short group compared with medium and long groups at month 1 postpartum (p<0.05). Milk yield (kg/day) was greater in the medium group compared with the short group at months 2 to 5 postpartum (p<0.05). The hazard of first insemination by 150 DIM was lower in the long group than in the short group (HR=0.73; p<0.02). The hazard of pregnancy by 365 DIM was lower (HR=0.64, p<0.0001) in the long group than in the medium group, while the hazard did not differ between short and medium groups (p>0.05). Moreover, the culling rate was greater in the long group than in the short group (p<0.05). In conclusion, a longer ICFI and DO resulted in reduced reproductive performance in dairy cows, which was attributable to an increase in postpartum diseases, a lower BCS and a greater milk yield.

Effects of Milk Yield on the Postpartum Health and Reproductive Performance of Dairy Cows (젖소에서 산유량이 분만 후 건강과 번식 능력에 미치는 영향)

  • This study evaluated the effects of milk yield on the postpartum health and reproductive performance of dairy cows. In total, data were collected from 1,060 cows on six dairy farms, including their milk production, body condition score (BCS), postpartum disorders, and reproductive performance. The lactation data were grouped based on the 305-day milk yield into control (< 10,000 kg, n = 445) and high milk yield (${\geq}$ 10,000 kg, n = 615) groups. The milk fat and protein, and BCS were lower during the first 5 months postpartum in the high milk yield group compared with the control group (p < 0.01). Ovarian cysts were more frequent (p < 0.0001) in the high milk yield group (28.6%) than in the control group (15.3%), whereas endometritis tended to be less frequent in the high milk yield group (29.6%) than in the control group (35.1%, p = 0.06). A higher proportion of cows tended to receive reproductive hormones (p = 0.06) in the high milk yield group (62.4%) than in the control group (56.6%). The probability of a pregnancy after first insemination tended to be lower (odds ratio = 0.78, p = 0.07) in the high milk yield group (30.2%) than in the control group (35.2%). Furthermore, the hazard of pregnancy by 210 days in milk was lower in the high milk yield group (hazard ratio = 0.84, p = 0.04) than in the control group, which resulted in a 20-day increase in the median interval to pregnancy. In conclusion, high milk yield was related to lower milk fat and protein, lower BCS, an increased incidence of ovarian cysts, and increased use of reproductive hormones, which resulted in decreased reproductive performance of dairy cows.

Multi-Parameter Approach for Evaluation of Genomic Instability in the Polycystic Ovary Syndrome

  • Background: The polycystic ovary syndrome (PCOS), characterized by hyperandrogenism and chronic anovulation, is a common endocrine disorder in women. PCOS, which is associated with polycystic ovaries, hirsutism, obesity and insulin resistance, is a leading cause of female infertility. In this condition there is an imbalance in female sex hormones. All the sequelae symptoms of PCOS gradually lead to cancer in the course of time. It is heterogeneous disorder of unknown etiology so it is essential to find the exact cause. Materials and Methods: In this study both invasive and non-invasive techniques were employed to establish the etiology. Diagnosis was based on Rotterdam criteria (hyperandrogenism, ovulatory dysfunction, PCOM) and multiparameters using buccal samples and dermatoglypic analysis and cytogenetic study for 10 cases and four age and sex matched controls. Results: In clinical analysis we have observed the mean value of total testosterone level was 23.6nmol/L, total hirsutism score was from 12-24, facial acne was found in in 70% patients with 7-12 subcapsular follicular cysts, each measuring 2-8 mm in diameter. In dermatoglypic analysis we observed increases in mean value ($45.9^{\circ}$) of ATD angle when compared with control group and also found increased frequency (38%) of Ulnar loops on both fingers (UU), (18%) whorls on the right finger and Ulnar loop on left finger (WU) and (16%) arches on right and left fingers (AA) were observed in PCOS patients when compared with control subjects. Features which could be applied as markers for PCOS patients are the presence of Ulnar loops in middle and little fingers of right and left hand. The buccal micronucleus cytome assay in exfoliated buccal cells, we found decrease in frequency of micronuclei and significant increases in frequency of karyolysed nuclei in polycystic ovarian syndrome patients. Chromosome aberration analysis revealed a significant increase in frequency of chromosome aberrations (CAs) in PCOS patients when compared with controls. Conclusions: From this present work it can be concluded that non-invasive technique like dermatoglypics analysis and buccal micronucleus cytome assays with exfoliated buccal cell can also be effective biomarkers for PCOS, along with increased CAs in lymphocytes as a sign of genetic instability. There is a hypothesis that micronuclei and chromosomal aberrations could have a predictive value for cancer. From this present work it can be concluded to some extent that non-invasive technique like dermatoglypics and buccal cell analysis can also be effective for diagnosis.

Treatment of Reproductive Dysfunctions and Reproductive Monitoring Using Ultrasonography in Dairy Cow (초음파 검사에 의한 젖소 번식 검진과 번식 장애 치료)

  • This study was carried out principally to obtain the basic data for the improvement of the reproductive performance and production using plasma progesterone assay and ultrasonography in dairy cow. The results obtained from this studies were as follows. The results of reproductive examination in 85,983 cows were ovarian diseases 40,399 (47.0%), uterine diseases 11,912 (13.9%), pregnancy or pregnant failures 26,587 (30.9%), adhesion of reproductive tracts 172 (0.2%), freemartin 8 (0.01%), and others 6,905 (8.3%), respectively. The treatment status of reproductive dysfunction in 30,241 cows were silent heat or error of estrus detection 14,909 (49.3%), follicular cysts 3,750 (12.4%), luteal cysts 907 (3.0%), inactive ovaries 665 (2.2%), granulosa cell tumor of ovary 3 (0.01%) and endometritis 6,986 (23.1%), respectively. The indices of reproductive efficiency after the periodical examination of reproductive status were as follows; the mean intercalving inteual was reduced from 475 days at the first examination to 381 days at the last examination of reproductive status, the mean interval calving to conception was reduced from 186 to 98 days, the mean interval calving to first service was reduced from 106 to 66 days, the cows showing heat by 60 days postpartum were increased from 32 to 90%, the mean conception rate to first service was increased from 42 to 64%, and the mean service per conception was reduced from 2.6 to 1.8 times, respectively.

  • Sex differentiation of the brown croaker Miichthys miiuy (Basilewsky) is described from hatching to the 120th day post-hatching (dph) (water temperature $24^{\circ}C$). Primordial germ cells (PGCs) were observed on the 20th dph (10.4 mm total length (TL), 0.14 g body weight (BW), and began to protrude into the peritoneal cavity from the 40th dph (19.4 mm TL, 0.39 g BW). On the 65th dph (31.3 mm TL, 0.93 g BW, $1,560D^{\circ}$ (degree-days)), initial ovarian differentiation was identified by the PGCs with condensed chromatin, and their transformation into meiotic oocytes. By the 120th dph (4.60 mm TL, 1.38 g BW, $2,880D^{\circ}$), the oocytes were in the perinucleolus stage and had increased from 20 to $40{\mu}m$ in diameter. While ovaries gradually grew after sex was differentiated, testes continued to multiply from the 65th dph. On the 80th dph (37.9 mm TL, 1.39 g BW, $1,920D^{\circ}$), the beginning of testis lobule formation was indicated by the occurrence of spermatogonial cysts enveloped by somatic cells in some of the testes. On the 120th dph, the testis lobules of some of the fish contained all germ cell stages through to the spermatocytes. Therefore, the sex differentiation type of the brown croaker is identified as gonochoristic.

Expression of NGF in Estradiol Valerate-Induced Polycystic Ovary and CHO Cells (Estradiol Valerate에 의해 유도된 다낭성난소와 CHO세포에서 NGF발현)

  • Polycystic ovary syndrome (PCOS) is hormonal imbalance condition as the endocrine and metabolic disorder that induces the infertility and various complications in reproductive age women. Estradiol valerate (EV) is used hormone replacement therapy in menopausal women and is reported that excessive administration of EV induces the PCOS. Nerve growth factor (NGF) is the factor to regulate the survival and maturation of developing neuronal cell and is also synthesized in ovary. And NGF is overexpressed in EV-induced polycystic ovary (PCO) as previously reported. Therefore, this study examined the possibility of NGF as can be used the biological marker in diagnosis of PCOS, the hormonal imbalance condition, using PCO and CHO (chinese hamster ovarian) cell lines. The concentration of EV treatment is optimized a 1 mg as not influence on the proliferation of CHO cell but 2 mg and 3 mg of EV treatment have the inhibition effect at initial stage. The morphological change was not observed in CHO cell after dose dependent manner treatment of EV. Expression of NGF mRNA and protein is significantly increased at 30 min after EV treatment in CHO cells compared to that of control. And NGF protein expression is strongly increased in PCO tissue, which observed many follicular cysts compared to normal ovary tissue. Taken together, overexpression of NGF may be act as a molecule to induce an abnormal development of follicle, suggesting that NGF can be used as a biological marker in diagnosis of PCOS.

Reproductive Cycle of a Rockfish, Sebastes schlegeli (조피볼락의 생식주기)

  • To clarify the annual reproductive cycle in a rockfish, Sebastes schlegeli, monthly changes in gonadosomatic index (GSI), hepatosomatic index (HSI) and histological feature of gonads and plasma levels of sex steroid hormones ($estradiol-l7{\beta},\;17{\alpha},\;20{\beta}-dihydroxy-4-pregnen-3-one,\;testosterone\;and\;11-ketotestosterone$) were investigated. The annual reproductive cycle in females could be divided into 5 periods as follows: 1) recovery period (June to September): serum level of $estradiol-l7{\beta}$ increased gradually; 2) vitellogenesis period (Septemer to february) : vitellogenic oocytes were obsewed, GSI sustained high value, and serum level of $estradiol-l7{\beta}$ increased; 3) gestation period (February-April): developing larva showed in the ovary, and serum levels of $17{\alpha},\;20{\beta}-dihydroxy-4-pregnen-3-one$ and testosterone increased; 4) partrition period (April to May) : larva were delivered, and value of GSI and serum levels of hormones decreased rapidly; 5) resting period (May to June) : value of GSI and serum levels of $estradiol-l7{\beta}$ and testosterone remained low. The annual reproductive cycle in males could be divided into 6 periods; 1) early maturation period (April to June): value of GSI and serum levels of hormones incresed gradually, cyst of spermatogonia incresed in number, and a small number of cyst of spermatocyte was observed; 2) mid-maturation perid (June to September); value of GSI and serum levels of hormones increased, and germ cells in many cysts were undergoing active sperrnatogenesis; 3) late maturation period (September to November) : value of GSI and serum levels of hormones remained high and spermatozoa were released into the lumina of the seminal lobules; 3) spermatozoa dischaging period (Nobember to December) : the lumina of the seminal lobules were enlarged and filled with mature spermatozoa; 4) degeneration period (December to Februauy)i value of GSI decresed and cyst of spermatocyte were decresed in number; 5) resting period (December to April) : no histological changes of testes were observed, and value of GSI and serum levels of hormones remained low. In November, the lumina of the seminal lobules were filled with mature spermatozoa and sperm masses were present in the ovarian cavity. Thus, copulation in this species occurred in November and December.
