Aspergillus oryzae 배양물이 육계의 생산성, 분변의 미생물 성상 및 암모니아 가스 발생량에 미치는 영향 (Effect of Dietary Supplementation of Aspergillus oryzae Ferment on Growth Performance of Broiler Chicks and Microbial Population and Fecal Ammonia Production)

  • 본 연구는 질소원 결핍 배지에서 배양되어 in vitro 실험상에서 항균 활력이 있는 것으로 증명된 Aspergillus oryzae 배양물을 질소원 공급 배지와 비교하여 육계의 증체, 사료 섭취량, 영양소 이용율, 유해 가스 생산 및 분내 미생물 균총 변화 등에 미치는 영향을 조사하기 위하여 시행되었다. 총 168수 2일령 육계를 7 처리구(대조구, 질소원 공급 배지에서 자란 Aspergillus oryzae 배양물(NAOF) 0.05, 0.1, 0.5%, 질소원 결핍 배지에서 자란 Aspergillus oryzae 배양물(NNAOF) 0.05, 0.1, 0.5%에 배치하였다. 본 실험 결과 Aspergillus oryzae 첨가로 사료의 기호성이 증가되었으며, Aspergillus oryzae 배양시 질소원 결핍 처리는 일반적인 배양 방법에 비하여 증체와 사료 기호성에 더욱 효과적으로 작용하였다. 영양소 소화율은 질소원 처리 구간의 유의적 차이는 없었지만, 대조구에 비하여 Aspergillus oryzae 배양물을 첨가시에 영양소의 소화율이 전반적으로 향상되었는데, 이러한 결과는 육계의 소장 및 대장의 pH 조절에 관여하고 유익균의 생장을 촉진하며, 유해균의 증식을 억제할 수 있는 효소나 대사산물이 분비되어 긍정적으로 작용하여 사료의 기호성 및 영양소의 소화율을 증진시킨 결과라고 추정할 수 있다. 처리에 따른 분내 총균수 변화는 유의적(p<0.05)인 차이를 보였으며, 균수는 NNAOF>NAOF>대조구순으로 나타났다. E. coli와 살모넬라에서는 Aspergillus oryzae 배양물 처리구가 대조구에 비해 현저하게(p<0.05) 감소되었고, 특히 NNAOP 처리구에서 가장 낮게 나타났다. 분내 암모니아 가스 발생량을 조사한 결과, 실험실적으로 검사할 수 있는 발생량은 대조구>NAOF>NNAOF 순으로 나타났다. 본 연구 결과는 NNAOF을 이용한 육계 생산은 생산성 향상과 더불어 환경 오염을 예방할 수 있는 방법임을 시사하므로 더욱 심도있는 연구가 필요한 것으로 판단된다.

Xanthomonas oryzae pv. oryzae로 부터 aspartate aminotransferase 유전자의 분리 및 생화학 특성 (Cloning and Biochemical Characterization of Aspartate Aminotransferase from Xanthomonas oryzae pv. oryzae)

  • Xoo로 부터 aspartate aminotransferase로 추정되는 유전자를 분리한 다음 발현시켜 생화학 특성을 조사하였다. 분리된 유전자는 His6 pET-21(a) 운반체에 삽입시켰으며 E. coli BL21(DE3)에서 발현시켰다. 재조합된 Asp-AT는 affinity chromatography를 이용하여 분리하였으며 SDS-PAGE분석에서 43kDa의 단일 밴드를 나타내었다. 분리된 효소는 amino donor 로서 L-aspartate에 대하여 효소활성도가 가장 높았고, L-leucine 및 L-cysteine에 대하여서도 상당한 활성도를 나타내었다. 효소의 최적 pH는 약 7.5 근처에서 나타났고 효소의 안정성은 산성조건 보다는 알칼리 조건에서 훨씬 높았다. 최적 온도는 약 $35-40^{\circ}C$로 나타났고 $55^{\circ}C$에서 20분간 열처리한 이후의 잔여 활성도는 약 78%로 나타났다. 여러 중금속 중에서 망간 이온에 의해 효소활성이 촉진되었다.

Effects of Dietary Supplementation of a Meju, Fermented Soybean Meal, and Aspergillus oryzae for Juvenile Parrot Fish (Oplegnathus fasciatus)

  • In this study, dietary supplementations of Korean Meju, fermented soybean meal (F-SBM) by Aspergillus oryzae, and A. oryzae itself were evaluated on growth performance, feed utilization, immune responses and phosphorus availability in juvenile parrot fish, a marine aquaculture fish species. Four isonitrogenous and isocaloric diets were formulated to contain 8% soybean meal (control diet), 4% Meju (50% soybean meal was replaced by Meju), 4% F-SBM (50% soybean meal was replaced by F-SBM), or 0.08% A. oryzae itself. One of the four experimental diets was fed to triplicate groups of fish for 8 weeks. At the end of the feeding trial, no significant differences were found in growth performances and feed utilization. Red blood cell counts in the fish fed the A. oryzae diet were significantly higher than that of fish fed the control diet. The antioxidant activity in Meju diet was significantly higher than that of the control and A. oryzae diets. Fish fed Meju and F-SBM diets showed numerically higher antioxidant activity of serum compared to that of fish fed the control diet, even though it was not significant. Liver superoxide dismutase activity of fish fed the test diets was significantly higher than that of fish fed the control diet. The apparent digestibility coefficients of protein of fish fed all the diets were not significantly different. Phosphorus absorption was numerically increased in fish fed F-SBM and A. oryzae diets compared to that of fish fed the control diet. This study indicates that the fermentation process of soybean meal does not impair growth performance and feed utilization in parrot fish. The fermentation process could enhance the availability of phosphorus in soybean meal and non-specific immune responses of parrot fish.

Effect of co-inoculation of Brevibacterium iodinum RS16 and Methylobacterium oryzae CBMB20 on the early growth of crop plants in Saemangeum reclaimed soil

  • The objective of this study was to determine the effect of single and co-inoculation of plant growth promoting bacteria (PGPB) on early plant growth in Saemangeum reclaimed soil. Plant growth promoting Brevibacterium iodinum RS16 and Methylobacterium oryzae CBMB20 were inoculated on maize (Zea mays L.) and sorghum-sudangrass hybrid (Sorghum bicolor L.) grown in Saemangeum reclaimed soil. Single and co-inoculation of B. iodinum RS16 and M. oryzae CBMB20 increased plant height, dry biomass accumulation and macro-nutrient accumulation of maize and sorghum-sudangrass hybrid. M. oryzae CBMB20 treatment increased plant height in maize by 41.2% at 30 days after sowing (DAS), shoot dry weight and total dry weight compared to non-inoculated treatment. Macro-nutrient accumulation (N and P) in maize roots was significantly increased with co-inoculation treatment, K and Ca content was significantly increased at B. iodinum RS16 treatment compared to non-inoculated treatment. Macro-nutrient accumulation (P, K, Ca and Mg) in shoot was higher with M. oryzae CBMB20 treatment compared to non-inoculated treatment. In case of sorghum-sudangrass hybrid, co-inoculation treatment showed 33.7% increase in plant height compared to non-inoculated treatment at 30 DAS. M. oryzae CBMB20 treatment increased root dry weight and total dry weight, macro-nutrient accumulation in roots and N, Ca and Mg accumulation in shoot compared to non-inoculated treatment. P and K accumulation in shoot was significantly increased at co-inoculation treatment compared to non-inoculated treatment. This pot culture experiment demonstrated that single and co-inoculation of B. iodinum RS16 and M. oryzae CBMB20 increased the early growth and nutrient accumulation of maize and sorghum-sudangrass hybrid.

재조합 Alkaline Protease를 대량 생산하는 Aspergillus oryzae 균주개발 (Breeding of Aspergillus oryzae for the Alkaline Pretense Overproducing Strain.)

  • Alkaline protease를 대량생산하는 Aspergillus oryzae를 만들기 위하여 A. oryzae의 alkaline pretense 유전자 alpA를 고발현시키는 plasmid pTAalp를 제조하고 이 plasmid로 A. oryzae M-2-3 균주를 형질전환시켰다. 16개의 형질전환체를 얻어 이들의 protease생산성을 skim milk 분해에 의한 halo 형성능에 의하여 확인하였다. 또한 protease 생산성이 증가한 형질전환체는 pTAalp가 multi-copy로 염색체 안에 integration되어 있음을 Southern blot에 의하여 확인하였고, 이들의 배양액을 polyacrylamide gel전기영동에 의하여 분석한 결과, 형질전환체 No. 14에서는 전체 분비단백질의 80-90%가 alkaline protease 임을 알 수 있었다. 간장원료 분해실험의 결과 No. 14에 의한 원료분해액은 간장 양조용 대조균에 의한 분해액보다 TN이 증가하였으며 원료분해율도 1.4-1.5배로 증가되었다.

Foliar Colonization and Growth Promotion of Red Pepper (Capsicum annuum L.) by Methylobacterium oryzae CBMB20

  • In order to exploit Methylobacterium oryzae CBMB20 as of plant growth promoting agent, different inoculation methods have been evaluated. The present study aimed to evaluate soil, foliar, and soil+foliar inoculations of M. oryzae CBMB20 to improve the growth, fruit yield, and nutrient uptake of red pepper (Capsicum annuum L.) under greenhouse conditions. The population range of green fluorescent protein (gfp)-tagged M. oryzae CBMB20 using the three inoculation methods was 2.5-2.9 ${\log}_{10}$ cfu/g in the rhizosphere and 4.5-6.0 ${\log}_{10}$ cfu/g in the phyllosphere of red pepper plants. Confocal laser scanning microscopy results confirmed the colonization of M. oryzae CBMB20 endophytically on leaf surface. Plant height, fruit dry weight, and total biomass were significantly higher ($p{\leq}0.05$) in all M. oryzae CBMB20 inoculation methods as compared to non-inoculated control. Furthermore, uptake of mineral nutrients such as N, P, K, Ca, and Mg in red pepper plants in all M. oryzae CBMB20 inoculation methods was higher than in non-inoculated control. Comparative results of inoculation methods clearly demonstrated that soil+foliar inoculation of M. oryzae CBMB20 lead to the highest biomass accumulation and nutrient uptake which may be due to its efficient colonization in the red pepper rhizosphere and phyllosphere.

  • Interspecific hybrids between Aspergillus oryzae var oryzae and A. nidulans 514 were obtained by nuclear transfer technique. Several autotrophic mutants isolated from conidiospores of the two strains were mutagenized with ultraviolet and N-methyl-N-nitrosoguanidine. Optimal conditions for formation of intergeneric hybrids were investigated. Frequencies of hybrid formation by nuclear transfer were $3{\times}10^{-5}{\sim}1{\times}10^{-5}$. From observation of genetic stability, conidial size, DNA content, and nuclear stain, it was suggested that their karyptypes are aneuploid. The hybrids showed 1.1~1.4 fold higher xylanase activities than parental strains did. The xylanase of Aspergiilus sp. TAVD514-3 was purified and some of it's enzymatc characteristics were investigated. The enzyme was purified about 85 fold with an overall yield of 17% from the culture medium by ammonium sulfate fractionation, Sephadex G-75 gel permeation chromatography, and CM-sephadex A-50 ion exchange chromatography. The purified enzyme functions optimally at pH 9.0 and 80$^{\circ}C$. The enzymatic activity was increased by the presence of $Mg^{2+}$ and $Mn^2$ ions.

Rhizopus oryzae CCS01로 제조된 쌀누룩을 이용한 곡류 막걸리의 품질 특성 (Quality Characteristics of Cereal Makgeolli Rice Nuruk Prepared Rhizopus oryzae CCS01)

  • 본 연구에서는 시판누룩에서 분리한 Rhizopus oryzae CCS01로 쌀누룩을 제조하고 이를 이용하여 곡류 막걸리 제조하였다. R. oryzae CCS01로 제조한 쌀누룩은 시판누룩보다 탄수화물 함량은 높았으나, 수분 및 회분, 조단백질, 조지방 함량은 낮았고 당화력은 약 2.2배 정도 높게 나타났다. R. oryzae CCS01로 제조된 쌀누룩으로 제조한 막걸리는 pH와 알코올 함량은 시판누룩보다 높게 나타났으나, 당도 및 산도, 젖산균수, 효모균수, 갈변도는 낮게 나타났다. 특히 쌀누룩으로 제조한 찹쌀(GUR) 막걸리의 경우 다른 막걸리들 보다 알코올 함량이 가장 높았다. 이 결과로 R. oryzae CCS01로 제조된 쌀누룩을 이용하여 새로운 곡류 막걸리 제조가 가능할 수 있을 것으로 판단되었다.

Expression of hpa1 Gene Encoding a Bacterial Harpin Protein in Xanthomonas oryzae pv. oryzae Enhances Disease Resistance to Both Fungal and Bacterial Pathogens in Rice and Arabidopsis

  • Xanthomonas oryzae pv. oryzae causing bacterial leaf blight disease in rice produces and secretes Hpa1 protein that belongs to harpin protein family. Previously it was reported that Hpa1 induced defense responses when it was produced in tobacco. In this study, we expressed hpa1 gene in rice and Arabidopsis to examine the effects of Hpa1 expression on disease resistance to both fungal and bacterial pathogens. Expression of hpa1 gene in rice enhanced disease resistance to both X. oryzae pv. oryzae and Magnaporthe grisea. Interestingly, individual transgenic rice plants could be divided into four groups, depending on responses to both pathogens. hpa1 expression in Arabidopsis also enhanced disease resistance to both Botrytis cineria and Xanthomonas campestris pv. campestris. To examine genes that are up-regulated in the transgenic rice plants after inoculation with X. oryzae pv. oryzae, known defense-related genes were assessed, and also microarray analysis with the Rice 5 K DNA chip was performed. Interestingly, expression of OsACS1 gene, which was found as the gene that showed the highest induction, was induced earlier and stronger than that in the wild type plant. These results indicate that hpa1 expression in the diverse plant species, including monocot and dicot, can enhance disease resistance to both fungal and bacterial plant pathogens.

Aspergillus oryzae로 제조한 Soybean koji 식이가 40-60대 중년여성의 손톱 개선에 미치는 영향 (Effects of Soybean koji Prepared with Aspergillus oryzae on the Improvement of the Nails of in their 40s to 60s Women)

  • 본 연구에서는 노화시작으로 손톱 상태가 나빠지는 중년 여성이 Aspergillus oryzae로 배양한 콩 koji를 섭취하므로 손톱의 건강상태가 얼마나 개선되는지를 알아보았다. 8주 동안 40~60대 여성을 대상으로 대조군은 삶은 비 발효 콩을, 시험군은 Aspergillus oryzae로 발효한 콩 koji를 섭취하도록 하였다. 연구결과 시험군의 손톱표면의 들뜸이나 갈라짐 상태가 개선되고, 수분량은 대조군에 비해 5.1배(p<.01), 수분 손실량은 대조군 감소량보다 1.4(p<.05)배, 손톱두께는 대조군에 비해 1.9(p<.05)배, 표면의 주름상태는 대조군에 비해 약 2.4(p<.01)배 정도 각각 개선됨을 보였다. 따라서 콩 koji 섭취가 손톱건강상태 개선에 영향을 미치는 것으로 나타났으며. 연구를 통하여 콩 koji가 이너뷰티 소재로서 활용이 가능할 것으로 기대한다.
