Overcoming F1 Sterility of Intersectional OA Lily Hybrids by Somatic Chromosome Doubling and Production of Subsequent Progeny (체세포 염색체 배가법을 이용한 백합 OA 종간 잡종의 불임극복과 후대획득)

  • This study was carried out to develop the efficient methods for the subsequent progeny of intersectional hybrid between Oriental and Asiatic lily hybrids. The pollen fertility and germination ability of 3 different allotetraploids (OAOA) after somatic chromosome doubling was ranged from 0 to 80 percent on artificial pollen germination medium. The number of BC$_1$ progeny using allotetraploid of F$_1$ OA-hybrid as male and female parent was different. The efficiency of BC$_1$ progeny production was increased when F$_1$OA-hybrids was used as male rather than as female parent. And in back crosses of F$_1$ OA-hybrids with the Asiatic and Oriental hybrids, Asiatic hybrids showed higher efficiency on BC$_1$ progeny production. The ploidy level between 2x or 4x Asiatic hybrid and allotetraploid F$_1$ OA-hybrid was determined and showed higher progeny production in 2x-4x crosses rather than 4x-4x.

Heterosis and Combining Ability in $F_1$ Hybrids of Korea Local and Oriental Tobacco Varieties (Nicotiana tabacum L.) (재래종과 Orient종 연초 $F_1$의 잡종강세와 조합능력)

  • In order to obtain the basic information for the successful breeding of aromatic tobacco (Nicotiana tabacum L.), the heterosis, combining ability and gene distributions in 28 Fl hybrids by diallel crosses between Korea local and Oriental varieties were investigated. The yield were higher, stalk height, leaf length and leaf width were larger and days to flower was shorter for the Fl hybrids when compared to mid-parent. Estimates of general combining ability were significant for all characters investigated, and specific combining ability were significant for yield, stalk height, leaves Per Plant, leaf width and days to flower. In vr-wr graph, partial dominane was exhibited by all characters investigated.

High Frequency Bulblet Formation in Scale and Stem Thin Cell Layer Explant Cultures of Lilium Oriental Hybrids (오리엔탈 백합의 인편 및 줄기의 박판 세포층 절편으로부터 고빈도 자구형성)

  • An efficient system for in vitro bulblet formation of Lilium oriental hybrids(cvs). Casa Blanca and Siberia is described. Transverse thin cell layer(tTCL)(1mm thick) explants of 'Casa Blanca' formed bulblets at a frequency of 97.7% when cultured on Murashige and Skoog (MS) medium supplemented with 1mg/L 2,4 dichlorophenoxyacetic acid(2,4-D) (On average 15.6 bulblets were formed per explant). The frequency of bulblet formation was drastically reduced when the explant ghickness was thinner than 1 mm. Explants from the outermost layer of bulb scale produced greater frequency of bulblet formation than middle or innermost layer. Among auxins supplemented to culture medium at 1 mg/L, 2,4-D led to greater frequency of bulblet formation on explants than dicamba, picamba, or phenylacetic acid(PAA). tTCL explants from the middle region of the outermost layer bulb scale yielded greater frequency of bulblet formation than the upper or lower region. tTCL stem explants of 'Siberia' formed bulblets at a frequency of 95.3% when cultured on MS medium with 1 mg/L 2,4-D(On average 9.1 bulblets were formed per explant). The system estabilished in this study will be useful for in vitro rapid propagation and genetic transformation of Lilium Oriental hybrids.

Detection and Molecular Characterization of a Stolbur Phytoplasma in Lilium Oriental Hybrids

  • Stolbur Phytoplasma was detected from Lilium Oriental hybrids showing flattened stem and flower clustering. The presence of phytoplasma was demonstrated using polymerase chain reaction(PCR) assays with phyto-plasma-universal(P1/P6)and stolbur phytoplasma-specific 16F1/R1-S primer pairs amplifying phytoplasma 16S rDNA regions. Nucleotide suquences of the phytoplasma 16S rDNA were determined. Nucleic acid extracted from lily amplified 1.5 kb DNA with a phytoplasma universal primer pair. In nested PCR, 1.1 kb PCR product was obtained using specific primer pair, indicating an isolate of stolbur phytoplasma. Nucleotide sequence of phytoplasma 16S rDNA reported in this study showed 99.5% and 99.1% identities with two known stolbur phytoplamas (16Sr XII-A). Also, it exhibited a sequence homology of 98.0% with phormium yellow leaf (16Sr XII-B), and 97.9% with Australian grapevine yellows (16Sr XII-B). Meanwhile, it showed 98.1% identity with strawberry green petal phytoplama, (16Sr1-C), and 94.7 % with American aster yellows (16Sr1-B). Homology percentage of the 16S rDNA nucleotide sequence suggests that this phytoplama could be classified into the stolbur phytoplasma, subgroup A (16Sr XII-A), as a type strain stolbur.

Effects of bulblet size, low temperature treatment and time of incubation on stem emergence during of Lilium oriental hybrids. (오리엔탈 백합의 순화재배시 자구크기, 저온처리, 배양기간이 경출현에 미치는 영향)

  • The experiment were carried out to establish the optimal condition of bulblet size, low temperature treatment and time of incubation in acclimatization on stem emergence of Lilium oriental hybrids. The large size bulblet was better than middle and small size bulblets in percentage of stem emergence and weight of bulblets. The highest percentage of stem emergence was 95% in large size bulblet 'Acapulco' but small size bulblet of 'Casablanca' was no stem emergence. Low temperature treatment(5℃)for breaking dormancy was needed at least more than 9weeks. Weight of bulblets and percentage of stem emergence was not good in 5℃ treatment for 6 weeks. The best stem emergence showed 5 month growing in vitro and the weigh test bulblets during acclimatization of bulblet

Cytogenetic Studies in Hybrids from a Pair of Sibling Drosophila Species

  • The cytogenetic pattern of autosome and sex chromiosome after Giemas staining were examined in the hybrids between two sibling species, Drosophila melanogaster and D. simulans. The analysis of karyotype in the hybrid female between D. melanogaster females and D. simulans males could be easily distinguished the characteriation of eight chromosomes from bothe species, especially with regard to X chromosomes. The lagging duplication of Y chromosome was investigated in the interspecific hybrid males from the cross between female of Drosophila melanogaster(OR) and males of D. simulasn (K18). On the other hand, the X chromatids of D. simulans were loosely separated in the early stage of anaphase.

Effect of Mulching Materials on Growth and Flowering of Oriental Hybrids Lilies in Alpine Area (피복재료가 고랭지 재배 Oriental 백합의 생장과 개화에 미치는 영향)

  • Bulbs of Lilium Oriental hybrids cv. 'Casablanca' and 'Marco Polo' were grown in a plastic house in alpine area of Pyongchang Kangwondo. Saw-dust, black film, reflective film, transparent film, and white/black double film were used as mulching materials to ascertain the growth of foliage, the quality of flower, and the size of bulbs. Foliage weights of both lilies were higher when lilies were grown with mulching materials than control. There were no differences of bulb weight among the control and mulching treatments, except delayed bulb growth in black film and white/black double film. Mulching cultivation with saw-dust and reflective film stimulated foliage growth, root growth and bulb production, but black film delayed the growth and development. Transparent and white/black double films showed no effects on foliage growth and bulb development than the control. As mulching materials tended to delay lily growth, including foliage and bulb, an extension of growing term might be better to produce high quality lilies than common cultivation in alpine area.
