Endocrine Disrupting Organotin Compounds are Potent Inducers of Imposex in Gastropods and Adipogenesis in Vertebrates

  • The persistent and ubiquitous environmental contaminant, tributyltin chloride (TBT), induces not only imposex in gastropods but also the differentiation of adipocytes in vitro and increases adipose mass in vivo in vertebrates. TBT is a nanomolar affinity ligand for retinoid X receptor (RXR) in the rock shell(Thais clavigera) and for both the RXR and the peroxisome proliferator activated receptor $\gamma(PPAR\gamma)$ in the amphibian (Xenopus laevis), mouse, and human. The molecular mechanisms underlying induction of imposex by TBT have not been clarified, though several hypotheses are proposed. TBT promotes adipogenesis in the murine 3T3-L1 cell model and perturbs key regulators of adipogenesis and lipogenic pathways in vivo primarily through activation of RXR and $PPAR\gamma$. Moreover, in utero exposure to TBT leads to strikingly elevated lipid accumulation in adipose depots, liver, and testis of neonate mice and results in increased adipose mass in adults. In X. laevis, ectopic adipocytes form in and around gonadal tissues following organotin, RXR or $PPAR\gamma$ ligand exposure. TBT represents the first example of an environmental endocrine disrupter that promotes adverse effects from gastropods to mammals.

Determination of Dibutyltin in Sediments Using Isotope Dilution Liquid Chromatography-Inductively Coupled Plasma Mass Spectrometry

  • A method is described for the determination of dibutyltin (DBT) in sediment by isotope dilution using liquid chromatography inductively-coupled plasma/mass spectrometry (LC-ICP/MS). To achieve the highest accuracy and precision, special attentions are paid in optimization and evaluation of overall processes of the analysis including extraction of analytes, characterization of the standards used for calibration and LC-ICP/MS conditions. An approach for characterization of natural abundance DBT standard has been developed by combining inductively-coupled plasma/optical emission spectrometry (ICP/OES) and LC-ICP/MS for the total Sn assay and the analysis of Sn species present as impurities, respectively. An excellent LC condition for separation of organotin species was found, which is suitable for simultaneous DBT and tributyltin (TBT) analysis as well as impurity analysis of DBT standards. Microwave extraction condition was also optimized for high efficiency while preventing species transformation. The present method determines the amount contents of DBT in sediments with expanded uncertainty of less than 5% and its result shows high degree of equivalence with reference values of an international inter-comparison and a certified reference material (CRM) within stated uncertainties.

기체크로마토그래프/질량분석기에 의한 물시료 중 Organotin의 추출 및 유도체 반응에 관한 연구 (The Extraction and Derivatization of Organotins in Water Sample by Gas Chromatograph/Mass Spectrometer)

  • 본 연구에서는 GC/MS를 이용하여 수질시료로부터 7종의 유기주석화합물을 분석하는 방법을 연구하였다. Hydrogenation과 ethylation의 반응시의 pH, 반응시간, 반응시약의 농도 등을 변화시켜 최적 반응조건을 조사하였다. n-Hexane을 용매로 사용한 액체-액체 추출 후 hydrogenation에 의한 유기주석염화물의 추출 회수율은 61-112% 범위로 나타났다. 물시료에서 ethylation한 후 n-hexane으로 추출한 경우의 추출 회수율이 74-113% 범위로 나타나 hydrogenation의 경우보다 높은 추출율을 보였다. 또한 ethylation후 styrene-divinylbenzene (SDB) 재질의 disk형의 고체상을 사용하고 추출 용매로 methylene chloride를 사용한 경우 61-97% 범위의 회수율을 나타냈다. Hydrogenation과 ethylation에 의한 검출한계는 각각 0.05-0.5 ng/ml와 0.02-0.05 ng/ml로 나타났다.

구룡포항 어패류에 함유된 유기주석화합물 분석 (Analysis of Organotin Compounds in Fish and Shellfish from Guryongpo Harbor)

  • 구룡포항에서 수획한 어패류에 함유된 유기주석화합물인 BT 화합물의 농도를 분석한 결과, 어류에 함유된 TBT, DBT, MBT의 농도는 각각 $15.68{\sim}52.93\;ng/g$(평균 28.65 ng/g), $13.95{\sim}72.16\;ng/g$(평균 26.99 ng/g), $37.73{\sim}133.59\;ng/g$(평균 85.18 ng/g)으로 분석되었다. 또한 어류의 근육보다는 내장에 BT 화합물이 더 많이 축적되어 있는 것으로 나타났다. 패류에서의 TBT, DBT, MBT 농도는 각각 $42.15{\sim}56.13\;ng/g$(평균 50.94 ng/g), $34.16{\sim}52.82\;ng/g$(평균 42.79 ng/g), $41.25{\sim}72.60\;ng/g$(평균 53.71 ng/g)으로 분석되어 패류의 BT 화합물 함유량이 어류의 BT 화합물 함유량보다 다소 높았다. 또한 어류에 함유된 PT 화합물의 농도를 분석한 결과 TPT와 DPT의 농도가 각각 $0.47{\sim}3.90\;ng/g$(평균 2.33 ng/g), $ND{\sim}0.56\;ng/g$(0.15 ng/g)이었으며 MBT는 검출되지 않았다. 패류의 PT 화합물의 농도는 TPT가 $10.24{\sim}21.67\;ng/g$(평균 17.45 ng/g, DPT가 $3.32{\sim}4.78\;ng/g$(평균 3.91 ng/g), MPT가 $0.11{\sim}0.36\;ng/g$(평균 0.25 ng/g)으로 분석되었다. 한편 어류에 함유된 유기주석화합물의 비율을 볼 때, BT 화합물의 경우 MBT 비율이 높게 나타난 반면 PT 화합물에서는 TPT 비율이 높게 나타났다. 전체적으로 볼 때 구룡포항에서 수획한 어패류의 유기주석화합물 농도는 국내 다른 항구의 어패류보다 높지 않은 것으로 나타났다.

유기주석 노출에 의한 Mysid, Neomysis awatschensis의 독성 영향 (The Toxic Effects of Mysid, Neomysis awatschensis Exposed to Organotin)

  • Tests for the toxicity of tributyltin (TBT) were conducted on mysid collected from Dadepo beach, Pusan, Korea. The toxic effects of tributyltin on the survival, growth and oxygen consumption of the mysid, Neomysis awatschensis have been evaluated. Mysids were exposed to several concentrations of TBT (0, 0.56, 1.15, 3.07 and 6.12 $\mu\textrm{g}$/L) for 6 weeks. Survival rate was decreased with increases in concentration and exposure time and the reduction of more than 40% occurred at TBT concentration greater than 1.15 $\mu\textrm{g}$/L after 6 weeks. Growth rate was significantly decreased at concentrations greater than 1.15 $\mu\textrm{g}$/L. Oxygen consumption rate was also decreased in a concentration-dependent way and significantly decreased to 39,47 and 69% of the control at 1.15, 3.07 and 6.12 $\mu\textrm{g}$/L, respectively. These results indicate that the contamination of aquatic environment by TBT has the potential to significantly reduce coastal and estuaries recruitment of mysids.

Synthesis of Radioiodinated Carbocyclic Cytosine Analogues

  • The synthesis of carbocyclic analogues of normal nucleosides has grown exclusively since they have shown potential antiviral and antitumor activities. Radiolabeled cis-1-[4-(hydroxy-methyl)-cyclopent-2-enyl]-5-$[^{124}I]$-iodocytosine (carbocyclic d4IC) and cis-1-[4-(hydroxy-methyl)-cyclopent-2-enyl]-5-(2-$[^{124}I]$iodovinyl)cytosine(carbocyclic d4IVC) were synthesized. The synthetic route employed Pd(0)-catalyzed coupling reaction together with organotin and exchange reaction for radioiodination as key reactions. Carbocyclic $[^{124}I]$d4IC gave more than 75% radiochemical yield with greater than 95% radiochemical purity. Carbocyclic $[^{124}I]$d4IVC gave more than 80% radiochemical yield with greater than 95% radiochemical purity.

  • The free-living nematode, Caenorhabditis elegans (C. elegans) has been adopted as a multicellular biosensor of biological toxicity for alkylphenol, organotin compounds and heavy metals. To adopt as a biosensor, suitability to assess must be fulfilledthrough several criteria; the organism must be sensitive to the testing toxicants, easy to manage in the laboratory and available throughout the year. C. elegans widely used as a simple multicellular organism in developmental biology studies and satisfies all these criteria, and its culture conditions, developmental staging, anatomy and genetic properties are well defined. In addition, researchers can take advantage of the worm's short life cycle, low cost and little individual variation. Moreover, genomic sequencing of C. elegans has recently been completed. With these aspectsof the organism, C. elegans become a more potent model organism for basic and applied bioassays.

Sliane 중합을 통한 면섬유의 개질에 관한 연구 (Modification of Cotton Fibers via In-Situ Polymerization of Silane Monomers)

  • 다양한 유기관능기를 가진 Silane가요제의 기능 양면성과 독특한 화학적 구조는 이들이 면섬유의 물성을 향상시키기 위한 in-situ 중합 및 가교처리의 단량체로 사용될 수 있는 가능성을 시사하므로, 본 연구에서는 이를 위한 기초 실험으로 수용액 상의 silane 단량체의 거동과 in-situ중합의 가능성을 조사하였다. 관능기를 달리하는 methyltriethoxysilane (MES), vinyltriethoxysilane (VES), vinyltriacetoxysilane (VAS)과 epoxy (glycidoxy) propyltrimethoxysilane (EMS) 등이 silane단량체로 선택되었다. Silane수용액의 안정성과 용해도는 단량체의 농도가 증가함에 따라 감소하였으며, pH에 의해서도 크게 영향을 받아 PH 3과 4.5사이에서 가장 안정함을 나타내었다. 10분간의 중기 고착과정에 의해서 충분한 양의 단량체가 면섬유 안으로 확산되었으며, 섬유의 방추성은 반응성이 높은 organotin촉매제를 사용하여 열처리한 후 증가되었다.

  • 몇가지 유기할로겐화물, 즉 브롬화에틸, 염화부틸, 브롬화페닐 및 염화벤질등을 탄화수소중에서 그리냐르법으로 무수사염화주석과 반응시켰다. 이때 소량의 에테르를 그리냐르반응단계에서 계내에 투입하엿을 때 보다 용이하게 반응이 진행되어 좋은 수율로 유기주석화합물들을 얻을 수 있었다.
