Bacillus alkalophilshaggy JY-827가 생산하는 항우식균물질의 생산조건과 분리, 정제 및 특성 (Production Condition, Purification and Properties of Anticaries Microbial Agent by Bacilluse alkalo-philshaggy JY-827)

  • The study was performed to investgate the excellent microbial anticaries substance which is more effective that the chlorhexidine in the dental caries treatment. A typi-cal strain which produced the most excellent antimicrobial subatance was selected. and identified novel alkalophillic Bacillus alkalophilshaggy JY-827. For the maximal production of themicrobial antibiotic against Streptococcus mutans from B. alkalophilshaggy JY-827, the optimal culture condition was in the medium containing glucose 15g/ L, pepton 10g/L and $K_2$$HPO_4$ 2g/L the highest production of antibiotic against S.mutans was obtained at $25^{\circ}C$ and pH 11.0 for 5 days. The antibiotic from B. alkalophilshaggy JY-827 was purified by organic solvent extraction, silica gel and sephadex LH-20 column chromatograpies, and then crystallized with methanol. The crystallin compoma-tion of this antibiotic was as a curcular shape. The melting point and rm[$\alpha$]$D^{20}$ were 152-154$^{\circ}C$ and +55。, respec-tively. Based on Instumental analyses such as FT-IR, $^{1}$H-NMR $^{13}$ C-NMR and GC-mass, the antibiotic was identified as aminoglycoside. It was obtained as amorphous white power, and soluble in water power, and soluble in water, methanol but insoluble in ether, chroloform. This antibiotic inhibited the growth of S.mutans to about 3 day at the concentration of $2.5$\times$10^{-7}$ /M. It was stable at the alkalli condition but unstable within the acid condition. It was also stable up to $70^{\circ}C$.

Isolation and Partial Characterization of Phytotoxic Mycotoxins Produced by Sclerotinia sp., a Potential Bioherbicide for the Control of White Clover(Trifoliorum repens)

  • Sclerotinia sp. (isolate BWC98-105) causes stem blight and root rot in Leghum sp., and is presently being evaluated as a potential mycoherbicide for the control of Trifoliorium repens. Bioassays have shown that Sclerotinia sp. produces phytotoxic substance which is biologically active against T. repens. Two biologically active compounds, designated as compoundsI and II, were produced in vitro from the culture filtrate of BWC98-105 isolate Sclerotium sp. Compounds I and II were purified by means of liquid-liquid extraction and $C_{18}$ open column chromatography (300 ${\times}$ 30 mm, i.d). To determine the purity, the purified compounds were analyzed by RP-HPLC. The analytical RP-HPLC column was a TOSOH ODS-120T (150 ${\times}$ 4.6 mm i.d, Japan), of which the flow rate was set at 0.7 mL/min using the linear gradient solvent system initiated with 15 % methanol to 85 % methanol for 50 min with monitoring at 254 nm. Under these RP-HPLC conditions, compounds I and II eluted at 3.49 and 4.13 min, respectively. Compound II was found to be most potent and host specific. However, compound I had a unique antibiotic activity against phytopathogenic bacteria like bacterial leaf blight (Xanthomonas oryzae) on rice, where it played a less important role in producing toxicity on T. repens. No toxin activity was detected in the water fraction after partitioning with several organic solvents. However, toxin activity was detected in the ethyl acetate and butanol fractions. In the leaf bioassay using compound II, the disease first appeared within 4-5 h as water soaked rot, which subsequently developed into well-defined blight affecting the whole plant.

Propolis 추출물의 유지 산화 억제 효과 비교 (Antioxidative Effect of Propolis Extract on Palm Oil and Lard)

  • Propolis를 몇 가지 용매로 추출하여 분획물을 얻고 이를 palm oil과 lard에 단독으로 혹은 synergist와 함께 첨가하여 항산화 효과를 Rancimat을 이용하여 측정하였다. 용매별 추출,수율은 68.1%(75% ethanol), 75.5%(99% ethanol), 67.4%(methanol), 86.7%(chloroform), 72.6%(ethyl acetate), 65.6%(butanol)였다. Methanol 추출물이 항산화 효과가 가장 우수 하였고 palm oil 보다는 lard에서 더욱 효과적이었다. Ethanol(75%) 추출물을 chloroform, ethyl acetate, butanol 및 water 순으로 분획 하였을 때 ethyl acetate 분획물은 조추출물보다 항산화 효과가 상승하였다. 또 ethyl acetate fraction 600 ppm에 synergist로 ascorbic acid(200ppm)을 palm oil에 첨가 했을 때와 ${\dalta}-toocopherol(200ppm)$을 lard에 첨가했을 때 높은 항산화 상승 효과를 보였다.

Column-switching HPLC를 이용한 성장기용 조제식 중 비타민 $D_3,\;K_1$의 동시분석 (Simultaneous Determination of Vitamin $D_3\;and\;K_1$ in Infant Formula by Column-switching High Performance Liquid Chromatography with UV Detection)

  • 본 연구에서는 성장기용 조제식 중에 영양강화를 위해 첨가하는 비타민 $D_3$$K_1$의 함량측정을 위해 lipase를 이용하여 효소 가수분해 후 유기용매로 비타민을 추출하여 동시에 분석하는 신속분석법을 위와 같이 수행하였다. 추출된 비타민을 전처리 컬럼에서 2번의 switching time을 설정하여 농축컬럼에 홉착시킨 후 gradient mode로 2가지 이동상으로 분석컬럼에서 각각의 성분을 모두 분리하여 동시에 검출하는 방법을 사용하였다. 시료로 사용된 성장기용 조제식은 표기함량보다 다소 높게 분석되었으나, 국제표준인증물질을 시료로 사용하여 본 연구의 실험방법에 의해 측정된 값은 인증된 표준값내의 결과를 보여주었다. 따라서, 비타민 $D_3$ 또는 $K_1$을 강화한 분유, 이유식 등의 분말 유제품 중에서 함량을 측정하고자 할 때 한정된 장비와 인력으로 각각의 2가지 실험방법을 수행하기가 어렵거나 시간단축이 필요한 경우, 본 연구에서 수행한 실험방법과 같이 시료전처리를 간단하고 신속하게 수행할 뿐만 아니라 역상컬럼과 column-switching HPLC에 의해 비타민 $D_3$ 또는 $K_1$을 동시에 분석함으로써 보다 효율적인 분석을 진행 할 수 있을 것으로 사료된다.

아시클로비어와 수용성 고분자를 이용한 고체분산체 제조 및 생체외 방출 (Preparation and In Vitro Test of Sold Dispersion using Acyclovir and Water Soluble Polymer)

  • Acyclovir(ACV) is an important antiviral drug used extensively against infections caused by herpes viruses, especially herpes simplex and varicella zoster. Because of high crystallinity and large particle size, solubility of intact ACV is very low in water(1.3 mg/ml). The goal of this work is to enhance the solubility of ACV. To make solid dispersion, Polyethyleneglycol, Hydroxyprophylmethylcelluose and Polyvinylpyrrolidone were used as polymer carriers in this work. Polymer carriers and drug were dissolved in acetic acid. And then spray drying method and freeze drying method were used as solvent extraction. Morphology, crystallization and functional group were characterized using SEM, XRD and FT-IR. The result of in vitro test showed the sample using PVP as polymer carrier had higher dissolution rate(up to 466%) than intact ACV.

인삼 앱솔루트 정유의 추출, 성분 분석 및 항균 활성 (Extraction, Component Analysis and Antibacterail Activity of Panax ginseng Absolute Essential Oil)

  • 웰빙 시대로 지칭되는 오늘날의 사회 특성으로부터 천연물에 대한 관심은 점점 증대되고 있다. 본 연구에서는 한국을 대표하는 생약재이자 향료식물인 인삼에 포함된 향료를 유기용매로 추출하여 앱솔루트 정유를 제조하고 그 성분을 분석하였다. 헥산과 에탄올 등 유기용제를 이용하여 제조된 인삼 앱솔루트 정유의 수율은 0.146%였으며, 주요성분은 neoclovene, panasinsene 및 calarene 등, sesquiterpene류가 주를 이루었다. 그 외에 110여 종의 인삼 향기 물질을 GC-MS 분석으로 확인하였다. 또한 일반세균 3종(대장균, 화농균, 녹농균), 효모균, 여드름균 및 비듬균에 대한 인삼정유 성분의 항균력을 측정한 결과 여드름균에 대한 강한 항균작용을 확인할 수 있었다.

Antioxidant and Antimicrobial Activities of Camellia Oleifera Seed Oils

  • The antioxidant and antimicrobial activities of Camellia oleifera seed oil were studied. Four kinds of seed oil samples were prepared, crude oil and refined oil, extracted by cold pressing method (CPC, CPR), and organic solvent extraction (OSC, OSR). Antioxidant activity analysis was measured in 2,2-azinobis (3-ethylbenzothiazoline-6-sulfonic acid)-diammonium salt, ferric reducing Ability of Plasma, and 2,2-diphenyl-1-picrylhydrazyl assays. Besides, the percentage of inhibition of red blood cells hemolysis induced by 2,2'-azobis(2-amidnopropane) dihydrochlorid, the lag time of LDL conjugated dienes formation in vitro, and the inhibitors of loss in tryptophan fluorescence were all used to estimate the antioxidant activity of the samples. The total phenolic contents (TPC) were detemined by Folin-Ciocalteu method. The TPC of the C. oleifera seed oils can be arranged in descending order: CPC ($1.9172{\mu}g/mL$) > OSC ($1.5218{\mu}g/mL$) > CPR ($1.0611{\mu}g/mL$) > OSR ($0.6782{\mu}g/mL$). And the oils were investigated for activity against Escherichia coli, Bacillus subtilis, Saccharomyces cerevisiae and Aspergillus niger. The results showed the antioxidant activity of crude oil by cold pressing method was stronger than others, and all oils did inhibit activity of the top three bacteria expert A. niger. The further significance of the study contributes to measure the antioxidant and antimicrobial activity of the potential health benefits by the different methods of preparation and the oil of C. oleifera seeds acting as free radical scavenger, pharmaceuticals and preservatives may offer some information in medicine and cosmetic not just in food field.

염화비닐의 요중 대사물질인 thiodiglycolic acid의 분석을 위한 전처리 조건 (Pretreatment method of urinary thiodiglycolic acid as metabolite of vinyl chloride)

  • The analysis of thiodiglycolic acid in urine has been used as an index of biological exposure to vinyl chloride. Unfortunately thiodiglycolic acid has a strong hydrophilic character, because it has two carboxylic groups, so that it can only be extracted with organic solvent with a great difficulty. Underivatized thiodiglycolic acid tends to tail because of non-specific interaction with the inert support. Therefore, esterification is the obvious first choice for derivatization of thiodiglycolic acid, particularly for gas chromatography. In this study, the focus of interest is to compare two method of esterifications (methylation and silylation). Methylation is to make the methyl ester of thiodiglycolic acid by reaction with diazomethane. Silylation is to make the trimethylsilyl ester of thiodiglycolic acid by reaction with N-trimethylsily-ldiethylamine. The results and conclusions are as the following: 1. The detection limit (sensitivity) of methylated thiodiglycolic acid was $5.00{\mu}g/m{\ell}$ and silylated thiodiglycolic acid was $3.07{\mu}g/m{\ell}$ by gas chromatography with flame ionization detector. 2. The optimal liquid-liquid extraction of thiodiglycolic acid was as following: To each of the tubes, $15m{\ell}$ of urine, concentrated sulfuric acid (pH 1 - 2) and 5 gsodium sulfate were added. The samples was extracted three times with $5m{\ell}$ ethylacetate each time. 3. The methylated thiodiglycolic acid was more stable than silylated thiodiglycolic acid in extractional solvent which contained humidity. 4. The precision (pooled coefficient of variation for 4 days) of the analysis was 0.07324 in methylated thiodiglycolic acid with external standard calibration, and 0.07033 in methylated thiodiglycolic acid with internal standard calibration. 5. The precision (pooled coefficient of variation for 4 days) of the analysis was 0.10914 in silylated thiodiglycolic acid with external standard calibration, and 0.13602 in silylated thiodiglycolic acid with internal standard calibration. From the above results, the analysis of methylated thiodiglycolic acid was more sensitive (limit of detection) than silylated thiodiglycolic acid by gas chromatography. However, the methylated thiodiglycolic acid was stable in the humidity and was separated sharply on chromatogram. Also, analysis of methylated thiodiglycolic acid was more precise (pooled coefficient of variation for 4 days) than silylated thiodiglycolic acid. In conclusion, it is established that the analysis of methylated thiodiglycolic acid is appropriate for biological monitoring of exposure to vinyl chloride.

환삼덩굴의 용매분획별 항균성 및 항산화성 (Antimicrobial and Antioxidative Activities of Solvent Fraction from Humulus japonicus)

  • 환삼덩굴의 생리활성물질을 검색하기 위하여 물과 메탄올로 추출하여 항균성과 trypsin 저해활성 및 항산화 효과를 조사하였으며, 메탄올 추출물은 다시 계통 분획하여 분획별 활성을 조사하였다. 세균 6종과 효모 2종을 대상으로 항균성을 조사한 결과, 물 추출물 보다는 메탄올 추출물의 항균성이 우수하였으며, Escherichia coli를 제외한 모든 시험균주에 항균효과를 나타내었다. 분획별 항균효과를 비교한 결과, 부탄올 분획이 모든 시험균주에 항균효과를 나타내었으며, 균종별로는 Staphylococcus aureus와 Saccharomyces cerevisiae에 대한 항균효과가 우수하였다. 부탄올 분획의 미생물에 대한 최소저해농도(MIC)는 $0.1{\sim}0.4%$ 범위였다. 환삼덩굴 추출물의 trypsin 저해활성을 조사한 결과, 물 추출물은 저해활성을 나타내지 않았으나, 메탄올 추출물은 저해활성을 나타내었다. 분획별 저해활성은 클로로포름 분획에서 가장 높았으며 클로로포름 분획의 50% 저해농도($IC_{50}$)는 1mg/ml였으며, EI(enzyme-inhibitor) 복합체 형성시간은 20분에 90% 이상이었다. 환삼덩굴의 메탄올 추출물은 돈지와 대두유를 기질로한 항산화성 시험에서 과산화물의 생성을 억제하였다. 메탄올 추출물의 분획별 활성은 클로로포름 분획에서 가장 높게 나타났으며, 클로로포름 분획의 농도를 0.2% 첨가하였을 때, 유지의 유도기간은 돈지는 15일, 대두유는 9일로서 산화안정성이 연장되었다.

리기다소나무재(材)의 유기용매(有機溶媒) 추출물(抽出物)과 그 tall oil 간의 조성(組成)의 차이(差異) (Difference of the Chemical Compositions Between Petroleum Ether Extract and its Tall Oil in Pitch Pine, Pinus Rigida Mill)

  • 본(本) 실험(實驗)은 리기다소나무(Pinus rigida Mill)를 petroleum ether로 추출(抽出)한 추출액(抽出被)과 흑액(黑液)으로부터 얻은 tall oil과의 조성(組成)의 변화(變化)를 조사(調査)하기 위하여 실시(實施)하였다. 생재(生材)의 유기용매(有機溶媒) 추출액(抽出液)과 tall oil 을 DEAE-Sephadex 칼럼과 aluminum oxide 칼럼을 사용하여 수지산(樹脂酸)과 지방산(脂肪酸)으로 분리(分離)한 후 이를 개스크로마토 그래프로 분석(分析)하였다. 본(本) 실험(實驗)에서 얻는 결과(結果)는 다음과 같다. 생재(生材)의 유기용매(有機溶媒) 추출액(抽出液힘)과 tail oil은 주로 수지산(樹脂酸)으로 구성(構成)되어 있고 특히 abietic-type acide의 함량(含量)이 매우 높게 나타났으며, tall oil의 조성(組成)은 생재(生材)의 유기용매(有機溶媒) 추출물(抽出物)에 비(比)해 지방산(脂肪酸)의 함량(含量)이 증가(增加)한 반면 수지산(樹脂酸)은 감소(減少)하였고, 증해시(蒸解時) 고온(高溫)과 증해약품(蒸解藥品)에 의하여 불포화(不飽和) 지방산(脂肪酸)의 이중결합(二重結合) 위치(位置)의 변화(變化) 및 이성질화(異性質化)에 의하여 미확인(未確認) 물질(物質)의 양(量)이 증가(增加)하였다.
