Contribution of Marine Microbes to Particulate Organic Matter in the Korea Strait

  • To assess the relative contribution of bacterial and phytoplankton biomasses to particulate organic matter (POM) in the water column, microbial abundance and biomass were from two transects in the western channel of the Korea Strait in 1996. Bacterial abundance had a mean value of $5.9{\times}10^5$ cells/ml and chlorophyll-a averaged 0.14 ${\mu}g/l$. Bacterial abundance in the Korea Strait showed a positive relationship with chlorophyll-a concentration, while the distribution of POM did not covary with chlorophyll-a. Particulate organic carbon (POC) and nitrogen (PON) concentrations were greater in August than in October. Bacterial carbon (POC) and nitrogen (PON) concentrations were greater in August than in October. Bacterial carbon and nitrogen biomasses were 7.29 ${\mu}gC/l$ and 1.24 ${\mu}gN/l$, respectively, during the study periods. Bacterial biomass was larger in October than in August due to the autumn phytoplankton bloom. Phytoplankton biomass based on chlorophyll-a was 7.67 ${\mu}gC/l$ for carbon and 1.10${\mu}gN/l$l for nitrogen. The ratio of bacterial carbon (BC) to phytoplankton carbon (Cp) averaged 0.95 in the Korea Strait in 1996. Bacteria may play a more significant role in the dynamics of POM than phytoplankton do in August, with BC/Cp ratio of 1.26. The ratio of BC to Cp increased with a decrease in chlorophyll-a concentration. Averaged over all the samples in both cruises, the contribution of microbial biomass to POC and PON was about 43% and 51%, respectively. Bacterial assemblage constituted a significant fraction of POC (21%) and PON (27%). Phytoplankton accounted for 22% of POC and 24% of PON. Microbial biomass played a more important role in the dynamics of POC and PON in October than in August due to a significant increase in microbial biomass in the southern transect (transect-B) in October by the autumn phytoplankton bloom. This study showed that marine microbes may constitute a significant part in the reservoir of POM in the Korea Strait.

Removal of Organic and Nutrients in Fish Market Wastewater using Sequencing Batch Reactor (SBR) (SBR공정을 이용한 수산물 위판장 폐수에서 유기물 및 질소 제거)

  • This research work aims at treating saline wastewater generated from a fish market using four Sequencing Batch Reactors (SBR) operated under different conditions. The effect of C/N ratio (3, 6) and salt concentration (0.5~2%) on organic and nitrogen removal was studied. The synthetic wastewater prepared with glucose ($C_6H_{12}O_6$) as the primary carbon source along with ammonium chloride ($NH_4Cl$) was used in the three reactors. The fill, anoxic, aeration, settle and draw conditions were 2 hr, 4 hr, 4 hr and 2 hr respectively. The fourth reactor was operated at different conditions to investigate the practical feasibility of SBR application to handle fish market wastewater generated in Ulsan city that had fluctuating loading characteristics. Though the unacclimated sludge was initially affected by the salt concentration, the acclimated sludge removed 95% of the organics irrespective of the NaCl concentration and C/N ratio. However, the removal of nitrogen was affected more by C/N ratio than the salt concentration. While handling fish market wastewater, though the organic and nitrogen loading rate were varying between $0.009{\sim}0.259gCOD_{OH}/gVSS/day$ and 0.005~0.034 gN/gVSS/day, the effluent concentrations were far less than the effluent standard of $120mgCOD_{OH}/L$ and 60 mgN/L respectively, except when loading rates were fluctuating and 4 times higher than the average.

Vegetation Structures and Soil Properties of Calanthe discolor and C. sieboldii Population (새우난초(Calanthe discolor)와 금새우난초(C. sieboldii) 개체군의 식생구조와 토양특성)

  • This study was carried out to analyze the vegetation and soil characteristic, and ordination between Calanthe discolor and C. sieboldii population, Jeju Island. The C. discolor population was classified into Cryptomeria japonica dominant population, Miscanthus sinensis dominant population, and C. discolor typical population. The C. discolor population was located in elevation of 156m to 430m, and C. sieboldii was located in elevation of 424m to 604m in Jeju Island. In the study sites, soil organic matter, nitrogen, exchangeable potassium, exchangeable calcium, exchangeable magnesium, cation exchange capacity, and soil pH were 30.05~53.58%, 0.74~1.64%, 0.80~1.95cmol$^+$/kg, 4.35~22.53cmol$^+$/kg, 4.00~6.63cmol$^+$/kg, 25.60~51.33cmol$^+$/kg, and 4.83~5.70, respectively. C. discolor population was found in the low elevation and steep sloped area that has less organic matter, total nitrogen, and cation exchange capacity than C. sieboldii population. C. discolor typical population was found in the high elevation area that has more organic matter and total nitrogen in the C. discolor. Miscanthus sinensis dominant population was found in the shade of west direction of japanese black pine. C. sieboldii was found in a high elevation and a gentle sloped area that has high percentage organic matter, total nitrogen, and cation exchange capacity.

Culture Characteristics and Optimal Conditions for Mycelial Growth of Calocybe indica (Calocybe indica의 배양적 특성과 균사 배양 적합 조건 설정)

  • Calocybe indica is an edible mushroom commercially cultivated in India and other tropical countries. In this study, the culture characteristics and optimal conditions of milky mushroom strains were determined. The growth temperature and pH range of milky mushrooms was extensively investigated between 15-35 ℃ and pH 3-11. For efficient cultivation, 20 types of nutrient sources were selected that consisted of one of 21 types of carbon sources, 6 organic nitrogen sources, 6 inorganic nitrogen sources, 13 amino acids, 6 organic acids and 12 inorganic salts. The impact of each of the selected nutrition sources and their concentration on growth was investigated. The optimum pH and temperature were determined to be pH 6.0 and 15 ℃, respectively. The optimum concentration of medium elements for the mycelial growth of C. indica was determined to be as follows: carbon source, 2% maltose; organic nitrogen source, 1% yeast extract; inorganic nitrogen source, 0.1% NaNO3; amino acid, 0.7% asparagine; organic acid, 0.07% acetic acid; inorganic salt, 0.07 mM MnSO4.

Vegetation Structures of Warm Temperate Evergreen Broad-leaved Forest in Gageodo, Korea (가거도 상록활엽수림의 식생구조)

  • This study used a phytosociolocal approach in classifying the vegetation communities of evergreen broad-leaved forest in Gageodo island of Korea, and the results are as follows: Quercus acuta community was classified into Dendropanax morbifer subcommunity, Daphniphyllum macropodum subcommunity, and Quercus acuta typical subcommunity. In the study sites, soil organic matter, total nitrogen, available phosphorous, exchangeable K, exchangeable Ca, exchangeable Mg, cation exchange capacity, and soil pH were ranged from 15.23~18.89%, 0.73~0.99%, 4.31~15.06 (mg/kg), 1.09~1.13 ($cmol^+$/kg), 21~7.15 ($cmol^+$/kg), 1.33~2.10 ($cmol^+$/kg), 33.04~38.28 ($cmol^+$/kg) and 4.47~4.83 respectively. The Dendropanax morbifer subcommunity were found in sites with high percentage of organic matter, total nitrogen, and low percentage of exchangeable Mg and Ca while the Daphniphyllum macropodum subcommunity were found in sites with low percentage of organic matter, total nitrogen, exchangeable Mg and Ca. The Quercus acuta typical subcommunity were found in sites with high percentage of organic matter, total nitrogen, exchangeable Mg and Ca. It is urgently needed to work for the preparation of an island management plan to the levels of species through habitat conservation.

  • Nitrogen pollution in urban and rural groundwater is a common problem and poses a major threat to drinking water supplies based on groundwater. In this work, the kinetics of nitrification-denitrification coupled reactions are modeled and new reaction modules for the RT3D code describing the fate and transport of nitrogen species, dissolved oxygen, dissolved organic carbon, and biomass are developed and tested. The proposed nitrogen transformations and transport model showed very good match with the results of other public codes.

Effects of Application Amount of Organic Compound Fertilizer on Lettuce Growth and Soil Chemical properties under Plastic film house (시설재배지에서 유기복합비료 시용량에 따른 상추 생육 및 토양화학성에 미치는 영향)

  • The Project supporting organic fertilizer started in 1999 as a national policy. In farmhouse, over application of mixed organic compound fertilizer(OC) caused salt accumulation in plastic film house soil. To replace inorganic fertilizer with OC fertilizer, this study was investigated the effect of OC application on yield and soil chemical properties for lettuce cultivation in plastic film house. The OC fertilizer was applied at 50(OC50+N50), 100(OC100), and 150(OC150) % level of the basal amount of nitrogen fertilizer in soil testing recommendation. And these were compared to NPK(nitrogen, phosphat, and potash fertilizer) and PK treatment. The yield of lettuce in OC100 was similar to that of NPK treatment. In OC 50, 100 and 150 treatments, pH had a tendency to increase than that of NPK treatment. Nitrate nitrogen(NO3-N) and electrical conductivity(EC) were similar to NPK treatment. These showed that nutrients from OC fertilizer were less likely to accumulate in soil than NPK. Also, use efficiency of nitrogen in OC100 treatment were similar to NPK treatment. These results suggest that OC application as the basal dressing at the 100% level could be best to prevent a nutrient accumulation of soil and to increase the yield and commercial quality for lettuce.

Removal of Dissolved Organic Matters in Drinking Water by GAC adsorption using RSSCT (RSSCT를 이용한 GAC의 상수원수 내 용존유기물질 제거)

  • Granular activated carbon (GAC) has been identified as a best available technology (BAT) by the United States Environmental Protection Agency (USEPA) for removal disinfection by-product (DBP) precursors, such as dissolved organic carbon (DOC) and dissolved organic nitrogen (DON). Rapid small-scale column test (RSSCT) were used to investigate four types of carbon (F400, Norit1240, Norit40S, and Aquasorb1500) for their affinity to absorb natural organic matter (NOM). DOC, $UV_{254}$, and Total dissolved nitrogen (TON) concentrations were measured in the column effluent to track GAC breakthrough. DOC and $UV_{254}$ breakthrough occurred at around 3500 bed volumes (BVs) of operation for all GACs investigated. The $UV_{254}$ breakthrough curves showed 33% to 48% at 8000 BVs, when the DOC was 48% to 65%. All GACs showed greater removal in DOC than $UV_{254}$. The NORIT1240 GAC was determined to have the highest adsorption capacity for DOC and $UV_{254}$. The removal of nitrate (NOTN) had not broken through over BVs. The initial TON breakthrough curves were started around 50%, when the DOC breakthrough was only 10 % at 500 BVs. The curves were gradually increased after 3500 BVs and approximately 69% through 81% of TON breakthrough occurred at 8000 BVs. All of the GACs were able to remove TON, in the case of this investigation the majority of the TON was present as DON. Because nitrate nitrogen was seldom removed and ammonium nitrogen ($NH_3-N$) was not detected in the effluent from RSSCTs even though raw water. The carbon usage rate of DOC was from 2 to 6 times less than that of TON. The NORIT1240 GAC demonstrated the best performance in terms of DOC removal, while the F400 GAC was best in terms of TON removal. Excitation emission matrix(EEM) analysis was used to show that GAC adsorption successfully removed most of Humic-like DOC and Fulvic-like DOCs. However, soluble microbial product(SMP)-like DOC in the absence of raw water were detected in the NORIT40S and Aquasorb1500 GAC. The authors assumed that this results is due probably to the part of GAC in the RSSCT which was converted into biological activated carbon(BAC). To compare with organics removal by GAC according to preloading, the virgin GACs had readily accessible sites that were adsorbed DOC more rapidly than preloaded GACs, but the TDN removal had not showed differences between those GACs.

Effects of Green Manures and Complemental Fertilization on Growth and Nitrogen Use Efficiency of Chinese Cabbages Cultivated in Organic Systems (녹비작물과 추비방법이 유기재배 배추의 생육과 질소 이용효율에 미치는 영향)

  • This study was conducted to evaluate the effects of green manures and complemental fertilization with oil cake or liquid fertilizer on growth and nitrogen use efficiency of Chinese cabbage cultivated in organi systems. Field experiments were carried out at the National Institute of Agricultural Science in Suwon, South Korea from 2012 to 2014. Two green manure crops, Crotalaria and Hairy vetch, was cultivated in summer and in winter, respectively. The application methods of the green manure consisted of three tillage systems (overall tillage, partial tillage and no tillage). Oil cake and liquid fertilizer were used as complemental fertilizer. The results showed that when used as covering material in the upland soil without tillage, green manure fertilization was more effective in increasing growth and yield of Chinese cabbage than when incorporated into soil. It was possible to grow and harvest Chinese cabbage in the spring season by the application of hairy vetch as winter green manure. The higher yield of Chinese cabbage with green manure application was caused by the lower incidence rate of soft rot and tip-burn. The yield of the Chines cabbage that received green manure applications over two consecutive seasons followed by the supplemental fertilization with oil cake was similar to that of the conventional chemical fertilization. Following a single season green manure application in summer, however, the yield of cabbage was only about 70% of the conventional treatment. Green manure cultivation with additional liquid fertilization produced a yield similar to the conventional fertilization treatment, soil inorganic nitrogen concentration remained stable and the nitrogen use efficiency increased in the green manure applied soil. In conclusion, the organic cultivation of Chinese cabbage in the autumn season could be outperformed in the upland soil receiving two seasons (winter and summer) of green manure fertilization followed by the supplemental fertilization with liquid fertilizer.

Impact of Herbicide Oxadiazon on Microbial Activity and Nitrogen Dynamics in Soil Environment

  • Influence of herbicide oxadiazon on soil microbial activity and nitrogen dynamics was evaluated. Soil samples were treated with oxadiazon at field and tenfold field rates and incubated. Organic amendment was added as an additional substrate for soil microorganisms. Tenfold field rate oxadiazon stimulated substrate-induced respiration (SIR) and dehydrogenase activity (DHA) in amended soil as compared to unamended soil and control treatment. Soil urease activity was not affected by oxadiazon treatment. In both amended and unamended soils, treatment of the herbicide at higher rate had not significant influence on $NH_4$-N and $NO_3$-N concentrations. Higher dose of oxadiazon was degraded in both soils, but dissipation rate in amended soil was higher than unamended soil, with half-lives ($t_{1/2}$) of 23.1 and 138.6 days, respectively. Recommended field rate did not affect microbial activity and nitrogen dynamics in soil ecosystem. Results showed influence of oxadiazon on cycling processes of nitrogen in soil was not significant however its effect on microbial activity was a tendency depending on addition of organic amendment to soil.
