Pine Needle Extract Applicable to Topical Treatment for the Prevention of Human Papillomavirus Infection

  • Most cervical cancers are associated with high-risk human papillomavirus (HPV) infection. Currently, cervical cancer treatment entails surgical removal of the lesion, but treatment of infection and preventing tissue damage are issues that still remain to be addressed. Herbal medicine and biological studies have focused on developing antiviral drugs from natural sources. In this study, we analyzed the potential antiviral effects of Pinus densiflora Sieb. et Zucc. leaf extracts against HPV. The pine needle extracts from each organic solvent were analyzed for antiviral activity. The methylene chloride fraction (PN-MC) showed the highest activity against HPV pseudovirus (PV). The PN-MC extract was more effective before, rather than after treatment, and therefore represents a prophylactic intervention. Mice were pre-treated with PN-MC via genital application or oral administration, followed by a genital or subcutaneous challenge with HPV PV, respectively. The HPV challenge results showed that mice treated via genital application exhibited complete protection against HPV. In conclusion, PN-MC represents a potential topical virucide for HPV infection.

Antibacterial and phytochemical properties of Aphanamixis polystachya essential oil

  • Now a day's rise of new antibiotic resistant bacterial strains is a global threat. Ethnic people of India have been employing Aphanamixis polystachya (Wall.) R. Parker wood extract in healing cancerous wounds. The aim of this study was to evaluate the antimicrobial activity and to identify the medicinally potent chemicals in the essential oil extract of A. polystachya. The antibacterial properties of various organic extracts were evaluated against a range of bacteria (gram-positive and gram-negative bacteria) based on the disc diffusion method and GC-MS based analysis for finding active oil extract components. All extracts of A. polystachya leaves showed potential antibacterial activity, notably ethyl acetate, while petroleum ether extracts revealed highly sensitive activity against all tested bacteria (zones of inhibition ranging from 8.83 to 11.23 mm). In addition, the petroleum ether extract had the lowest MIC value (32 to $256{\mu}g/mL$) against E. coli, S. lutea, X. campestris, and B. subtilis bacteria. The major compounds detected in oil [${\beta}$-elemene (16.04 %), ${\beta}$-eudesmol (12.78 %), ${\beta}$-caryophyllene (19.37 %), ${\beta}$-selinene (11.32 %), elemol (5.76 %), and ${\alpha}$-humulene (5.68 %)] are expected to be responsible for the potent antimicrobial activity. The results of this study offer valuable insights into the potent role of A. polystachya essential oil extract in pharmaceutical and antibiotic research.

해수산 클로렐라(Chlorella ellipsoidea) 유기용매 추출물의 항염증 효과 (Anti-Inflammatory Effect of Chlorella ellipsoidea Extracted from Seawater by Organic Solvents)

  • Chlorella has been reported to have certain beneficial physiological effects, including hypocholesterolemic, antihypertensive, antioxidative, and anti-tumor activities in animal and human studies. The aim of this study was to determine the anti-inflammatory activities of an 80% methanol extract(CE-Met), hexane fraction (CE-Hex), and ethyl acetate fraction (CE-EA) of Chlorella ellipsoidea in lipopolysaccharide (LPS)-stimulated RAW 264.7 murine macrophage. Treatment with various concentrations of the C. ellipsoidea extract resulted m a significant, dose-dependent reduction in nitric oxide (NO) production by LPS-induced macrophages. The C. ellipsoidea extract significantly inhibited LPS-induced NO production accompanied by an attenuation of IL-6 and TNF-$\alpha$ formation in macrophages. These results suggest potent inhibit이y effects on the production of inflammatory mediators by a C. ellipsoidea extract. Thus, C. ellipsoidea extract may be a potent anti-inflammatory agent for troubled skin.

인체 상피세포주와 암세포주에 대한 소목 추출물의 증식억제 효과 (Antiproliferative Effects of Caesalpinia sappan Extract on Human Epithelial Cell Line HaCaT and Cancer Cell Lines)

  • Caesalpinia sappan L. (Leguminosae) is an oriental medicinal herb distributed in China and Taiwan, and its heartwood has been traditionally used as an analgesic, a therapy for thrombosis or tumor. This study was to investigate the proliferation and inhibition effects of Caesalpinia sappan extracts against human epithelial cell and cancer cell lines. The methanol extract of dried C. sappan heartwood was evaporated (KS-6), and then sequentially extracted by hexane (KS-01), chloroform (KS-02), ethyl acetate (KS-03), n-butanol (KS-04), and water (KS-05). After 48 hr of exposure, these fractions at a concentration of $50{\mu}g/ml$ significantly increased, and reduced cell proliferation in both human normal epithelial and cancer cell lines. The ethyl acetate fraction (KS-03) among organic solvent fractions was 120.2% of the most proliferation activity ($50{\mu}g/ml$) against HaCaT human epithelial cell. However, fractions from chloroform, butanolic and methanolic extract had 7.2, 28.7 and 20.8% of antiproliferative effect on HaCaT cell, respectively. In cell proliferation effects of C. sappan extract on HeLa, SiHa and C33A human cervical cancer cells, chloroform fraction (KS-2) was the most antiproliferative activity, its antiproliferative rate (dosage, $50{\mu}g/ml$) relative to control was 25.8, 12.2 and 17.4% for SiHa, HeLa and C33A, respectively. The results indicated that the six extract fractions could induce cell cycle stimulate or arrest in some way. Finally, further investigation is needed to assess the molecular mechanisms mediated anticancer activities of this plant.

Evaluation of Chemical Composition and In vitro Digestibility of Appennine Pasture Plants Using Yak (Bos grunniens) Rumen Fluid or Faecal Extract as Inoculum Source

  • Pastures of the Apennines of Central Italy contribute to feed resources of high altitude (above 1,300 m sea level) grazing systems. The objective of this study was to evaluate the effectiveness of faecal extract from the yak (Bos grunnienes) as an alternative microbial inoculum to rumen fluid for estimation of digestibility of several forage species. Forage samples produced at high altitude were tested in this study: four legumes (Lathyrus sativus L., Lotus corniculatus L., Onobrychis viciaefolia L. and Trifolium pratense L.), three forbs (Achillea millefolium L., Potentilla reptans L. and Teucrium flavum L.) and one grass (Brachipodyum pinnatum L.) were incubated with yak rumen fluid or faecal extract. A large variability in chemical composition was observed among the species collected. Rumen liquor and faecal samples were collected from adult healthy yak. The $Daisy^{II}$ incubator was used to evaluate the nutrient digestibility of forages using rumen liquor as control and faecal extract as alternative microbial inoculum sources. Filter bags containing samples of browse species were added to the four digestion vessels along with their respective inoculum and then incubated for 48 h and dry matter (DM), organic matter (OM), crude protein (CP), neutral and detergent fiber (NDF) digestibility was determined. There was a significant relationship between estimates, indicating that faecal liquor has the potential to be used instead of rumen fluid for estimation of in vitro digestibility of plants. It is concluded that the $Daisy^{II}$ incubator results are appropriate for the determination of in vitro digestibility of nutrients using faecal liquor to define the potential for adaptation of yak to new pastures.

희렴(Siegesbeckia pubescens) 추출물의 어병세균 Streptococcus iniae에 대한 항균활성 (Antibacterial Effect of Siegesbeckia pubescens Extract against Fish Pathogenic Streptococcus iniae)

  • Antibacterial activity of 80% methanol extract from 13 commercial herb medicines was measured against fish pathogens Streptococcus iniae, causing several diseases in various kind of fish. Siegesbeckia pubescens showed the strongest antibacterial activity against S. iniae. Methanol extract of S. pubescens was further extracted using several organic solvents having different polarity. Extract from n-hexane and ethyl acetate fraction showed strong activity. Minimal inhibitory concentration, MIC of S. pubescens extract was measured and resulted showing 8 μg/mL with n-hexane fraction and 32 μg/mL with ethyl acetate fraction against S. iniae. The growth of S. iniae was fully inhibited by adding 50 μg/mL (final concentration) of n-hexane or ethyl acetate fraction in the liquid media. It is needed that, from these results, purification and isolation of responsible active compound(s) of these activities and further study on the synergy effect using combination with commercial antibiotics against fish pathogenic bacteria.

야관문 에탄올 추출물의 항산화 및 Metalloproteinase 저해 활성 (Antioxidant and Metalloproteinase Inhibitory Activities of Ethanol Extracts from Lespedeza cuneata G. Don)

  • 야관문의 항산화 활성과 항주름 활성을 조사하기 위하여 30% 에탄올 추출물과 각종 용매 분획물을 이용하여 DPPH 및 ABTS radical 소거활성과 matrix metalloproteinase MMP1과 MMP3 생성억제 효과를 조사하였다. 에탄올 추출물과 각종 용매 분획물 중 에틸아세테이트 분획물에서 페놀 및 플라보노이드 함량이 가장 높았다. 야관문 추출물의 항산화 활성과 피부 주름을 유발하는 MMP1, MMP3의 생성을 억제 효과는 페놀 및 플라보노이드 함량과 높은 상관관계를 보였으며, 페놀과 플라보노이드 함량이 가장 높은 에틸아세테이트 분획물의 항산화 활성과 MMPs 생성 억제 효과가 가장 높았다.

Propolis 추출물의 유지 산화 억제 효과 비교 (Antioxidative Effect of Propolis Extract on Palm Oil and Lard)

  • Propolis를 몇 가지 용매로 추출하여 분획물을 얻고 이를 palm oil과 lard에 단독으로 혹은 synergist와 함께 첨가하여 항산화 효과를 Rancimat을 이용하여 측정하였다. 용매별 추출,수율은 68.1%(75% ethanol), 75.5%(99% ethanol), 67.4%(methanol), 86.7%(chloroform), 72.6%(ethyl acetate), 65.6%(butanol)였다. Methanol 추출물이 항산화 효과가 가장 우수 하였고 palm oil 보다는 lard에서 더욱 효과적이었다. Ethanol(75%) 추출물을 chloroform, ethyl acetate, butanol 및 water 순으로 분획 하였을 때 ethyl acetate 분획물은 조추출물보다 항산화 효과가 상승하였다. 또 ethyl acetate fraction 600 ppm에 synergist로 ascorbic acid(200ppm)을 palm oil에 첨가 했을 때와 ${\dalta}-toocopherol(200ppm)$을 lard에 첨가했을 때 높은 항산화 상승 효과를 보였다.

Evaluation of the Oral Acute Toxicity of Black Ginseng in Rats

  • We studied the acute oral toxicity of black ginseng (BG) produced by heat process in rats. Single acute BG extract doses of 0, 5, 10, and 15 g/kg dissolved in saline were administered by oral gavage and the animals were kept under observation for 14 days. The single administration of BG extract up to 15 g/kg did not produce mortality, behavioral change or abnormal clinical signs in the rats. These results indicated that the oral $LD_{50}$ of the BG extract in the rats is higher than 15 g/kg. Compared to the control group, no treatment-related biologically significant effects of BG extract were noted in the measurements of the body weight or food intake. At the end of the period, the biochemical parameters and hematological parameters were analyzed in the plasma and blood. A histopathological examination of the liver and kidney was also conducted. Only the blood nitrogen urea and potassium levels in the biochemical indices showed significant differences at 10 and 15 g/kg doses of BG extract compared to the control group. These changes were not considered to be due to the toxicity. None of the other clinical chemistry parameters were affected. Therefore, these results indicate that the BG by heat processing is virtually nontoxic.

추출방법에 따른 미숙사과 추출물의 유용성분 함량 및 항산화 활성 (Effective Component Contents and Antioxidative Activities of Unripe Apple by Extraction Methods)

  • The purpose of this study was to determine the optimum extraction method suitable for the availability of biological activities in unripe apples known to be rich in functional components. Unripe apples 'Hongro' picked on May 28th, 2019 were extracted by various extraction methods (hot water, ethanol, enzymatic pre-treatment, ultrasonic wave, and subcritical water) and their extracts were investigated yield, effective component contents, and antioxidant activities. Overall, the yields by the extraction solvent were higher in water than in organic solvent(ethanol) because water-soluble compounds were eluted from a polar solvent. Total phenol contents of the ultrasonic wave (ethanol) extracts were significantly higher in 6 times than hot water extract. Contents of flavonoid were highest in the ethanol extract at 29.14 mg QE/g. Contents of tannin and ursolic acid were also significantly higher in the ultrasonic wave (ethanol) extract. The DPPH radical and ABTS radical scavenging activities were the strongest in the ultrasonic wave (ethanol) extract. Correlation between effective components and antioxidant activities was high in the total phenol content with ABTS and the ursolic acid content with DPPH (p<0.01). The above results suggested that ultrasonic wave (ethanol) extract of unripe apples has the potential to act as a functional material.
