Application of Geo-Statistic and Data-Mining for Determining Sampling Number and Interval for Monitoring Microbial Diversity in Tidal Mudflat (갯벌 미생물 다양성 모니터링 시료 채취 개수 및 간격 선정을 위한 지구통계학적 기법과 데이터 마이닝 적용 연구)

  • Tidal mudflat is a reservoir for diverse microbial resources. Microbial diversity in tidal mudflat sediment can be easily influenced by various human activities. It is necessary to take representative samples to monitor microbial diversity in tidal mudflat sediments. In this study, we analyzed the microbial diversity and chemical characteristics of vegetation and non-vegetation tidal mudflat regions in the Kangwha tidal mudflat using geo-statistics and data-mining. According to the geo-statistical analysis, most correlation range values for the vegetation region were smaller than those for the non-vegetation region, which suggested that the shorter number and interval of sampling are required for the vegetation tidal mudflat environment due to its higher degree of chemical and biological complexity and heterogeneity. The data-mining analysis suggested that the organic content and nitrate were the major environmental factors influencing microbial diversity in the vegetation region while pH and sulfate were the major influencing factors in the non-vegetation region. Using the geo-statistical and data-mining integration approach, we proposed a guideline for determining the sampling interval and number to monitor microbial diversity in tidal mudflat.

The Effect of Aquatic Macrophytes on the Biogeochemistry of Wetland Sediments (습지 식물이 퇴적물의 생지화학적 반응에 미치는 영향)

  • This research investigates the influences of the presence of aquatic macrophytes on the changes of biogeochemistry in the sediments through the comparative analysis of porewater and sediments. From the in situ measurements, elevated SO$_4{^{2-}}$ concentrations were observed in the rhizosphere during the growing season, which was resulted from the oxidation of reduced sulfide in the sediments by the oxygen release from the plant roots. There was sufficient AVS in the sediments to induce observed SO$_4{^{2-}}$ concentrations. The amount of oxygen in the oxidation of AVS to produce observed SO$_4{^{2-}}$ concentrations is 0.85 g/m$^2$ day which is relevant to the results of other researches. The AVS concentrations in the vegetated sediments increased with the depth whereas there is higher mass of AVS in the surface of the non-vegetated sediments. This shows that evapotranspiration induces the transportation of SO$_4{^{2-}}$ in the surface water into the anaerobic sediments. In addition, the elevated organic content caused by the presence of plants increased $\beta$-glucosidase activities which play an important role in the carbon cycle of the sediments.

Paleoenvironmental Changes in the Northern East China Sea and the Yellow Sea During the Last 60 ka

  • A borehole core ECSDP-102 (about 68.5 m long) has been investigated to get information on paleoenvironmental changes in response to the sea-level fluctuations during the period of late Quaternary. Several AMS $\^$14/C ages show that the core ECSDP-102 recorded the depositional environments of the northern East China Sea for approximately 60 ka. The Yangtze River discharged huge amounts of sediment into the northern East China Sea during the marine isotope stage (MIS) 3. In particular, $\delta$$\^$13/Corg values reveal that the sedimentary environments of the northern East China Sea, which is similar to the Holocene conditions, have taken place three times during the MIS 3. It is supported by the relatively enriched $\delta$$\^$13/Corg values of -23 to -21$\textperthousand$ during the marine settings of MIS 3 that are characterized by the predominance of marine organic matter akin to the Holocene. Furthermore, we investigated the three Holocene sediment cores, ECSDP-101, ECSDP-101 and YMGR-102, taken from the northern East China Sea off the mouth of the Yangtze River and from the southern Yellow Sea, respectively. Our study was focused primarily on the onset of the post-glacial marine transgression and the reconstructing of paleoenvironmental changes in the East China Sea and the Yellow Sea during the Holocene. AMS $\^$14/C ages indicate that the northern East China Sea and the southern Yellow Sea began to have been flooded at about 13.2 ka BP which is in agreement with the initial marine transgression of the central Yellow Sea (core CC-02). $\delta$$\^$18/O and $\delta$$\^$13/C records of benthic foraminifera Ammonia ketienziensis and $\delta$$\^$13/Corg values provide information on paleoenvironmental changes from brackish (estuarine) to modem marine conditions caused by globally rapid sea-level rise since the last deglaciation. Termination 1 (T1) ended at about 9.0-8.7 ka BP in the southern and central Yellow Sea, whereas T1 lasted until about 6.8 ka BP in the northern East China Sea. This time lag between the two seas indicates that the timing of the post-glacial marine transgression seems to have been primarily influenced by the bathymetry. The present marine regimes in the northern East China Sea and the whole Yellow Sea have been contemporaneously established at about 6.0 ka BP. This is strongly supported by remarkably changes in occurrence of benthic foraminiferal assemblages, $\delta$$\^$18/O and $\delta$$\^$13/C compositions of A. ketienziensis, TOC content and $\delta$$\^$13/Corg values. The $\delta$$\^$18/O values of A. ketienziensis show a distinct shift to heavier values of about 1$\textperthousand$ from the northern East China Sea through the southern to central Yellow Sea. The northward shift of $\^$18/O enrichment may reflect gradually decrease of the bottom water temperature in the northern East China Sea and the Yellow Sea.

  • The yeast strain was isolated from food industry waste water and its identification and biological characteristics were investigated. The optimum condition for cultivations and its activities for the reduction of B.O.D. on the food industry waste water were also confirmed. The results are as follows; 1) The isolated was identified as Candida curvata. 2) Candida curvata grew well in all of the experimented media, so and it can be regarded as a useful strain in the treatment of food industry waste water. 3) There was only a slight difference in the induction period between sterilized cultivation and unsterilized cultivation. But in the ice cream waste water, the period was considerably longer in unsterilized cultivation. 4) Specific rate of growth of Candida curvata in sugar waste water was 0.50/hr, ice cream waste water 0.50/hr, and beer waste water 1.0/hr. 5) Increasing of innoculum reduced the induction period in unsterilized cultivation. 6) The amount of dried yeast from sugar waste water were $175mg/{\ell}$, ice cream waste water $628mg/{\ell}$, and beer waste water $857mg/{\ell}$. Crude protein content in the dried yeast from sugar waste water were 52%, ice cream waste water 54%, and beer waste water 54%. 7) The rate of BOD reduction in sugar waste water were 49%, ice cream waste water 80%, and beer waste water 64%.

  • The purpose of this study is to analyze the behavior of swine slurry wastewater from bogen, in the treatment of Aerobic Liquid-Composting treatment by Aerobic Liquid-Composting using a mixer and Micro Air Diffusion pH level was at the beginning and its rise was seemingly related to VFA. It appears that removal of BOD and COD are more effective by Aerobic than by Anaerobic. In terms of removal efficiency, it shows 70.9% of BOD and 39% of COD in M.A+Mix and 67.8% of BOD and 19% of COD in M.A. $NH_3-N$ decreases in all conditions, which is caused by both the characteristic of nitrogen and the rise of pH. $NO_3-N$ increases in all conditions. It is judged that the accumulation of $NO_3-N$ affects the reduction of the ratio of denitrification. In the result of the analysis of Manure in swine slurry of liquified fertilizer ingredients, content of Manure in swine slurry of liquified fertilizer ingredients in aerobic conditions (M.A+Mix) is higher than anaerobic conditions.

Distribution Characteristics of Polychlorinated Biphenyls in Sediments inside Jeju Harbor (제주항내 퇴적물의 PCBs 분포특성)

  • In this paper, polychlorinated biphenyls (PCBs) were measured in surface sediments collected three times (June, October, December, 2013), inside Jeju Harbor as major harbors of Jeju Island. The concentration of PCBs inside Jeju Harbor was in the range of 1.62~4.45 (mean) ng/g on a dry weight basis and the levels were very low. In the analysis of PCBS homologue patterns, high-chlorinated PCB congeners were dominant in surface sediments inside Jeju Harbor, indicating that their sources were originated from shipping activity. In the relationships between PCBs concentrations and particle size (mud, sand and gravel) in surface sediments, PCBs concentrations were higher in the sediments with higher mud content, indicating that higher PCBs were distributed with increasing sediments of fine gradules. The PCBs concentrations in surface sediments in this study were very low, compared with ER-L (effect range-low) and TEL (threshold effects level) among sediment quality guideleines (SQGs) applied in foreign countries, such as USA, Canada, and Australia, etc), indicating that their biological effects on the bottom organisms in marine environment were expected to be very low.

Improvement of Kimchi Fermentation by Using Acid-Tolerant Mutant of Leuconostoc mesenteroides and Aromatic Yeast Saccharomyces fermentati as Starters

  • Saccharomyces fermentati and Leuconostoc mesenteroides were isolated from a traditional kimchi, and then the Leu. mesenteroides was mutated to the acid-tolerant mutant Leu. mesenteroides M-l00. In the result of growth properties in MRS broth with various pHs adjusted with HCl and acid solution (latic acid:acetic acid=1:2), an acid-tolerant mutant Leu. mesenteroides M-100 showed more increased ability for growth than its wild strain at various temperatures. The strains were used as starters for the fermentation of kimchi. The experiments were performed with classified experimental groups (Group I, control kimchi; Group II, addition of YK-19 only; Group III, addition of M-100 only; Group IV, addition of mixture of M-100 and YK-19), and their pH, total acidity, reducing sugars content, organic acid productivity, organoleptic tests, and microfloral changes were compared. As a result, in pH and acidity, the optimal ripening period of Group IV was about 13.5 days (i.e. from the 8.5 to 22 days of fermentation). This result indicates that the optimal ripening period of Group IV was about 1.5 times longer than that of Group I. Furthermore, Group IV was edible to 28 days of fermentation. In addition, high contents of succinc acid was observed in Group IV. Group IV was also highly ranked on the organoleptic test. During the fermentation of kimchi, the number of Leuconostoc sp. in group I reduced after 7 days; however, the number of Leuconostoc sp. in Group II, III, and IV decresed after 14 days of fermentation. An especially high number of Leu. sp. was observed in Group IV, and this gave better flavor of kimchi than any other during the whole fermentation period. Citric acid, tartaric acid, succinic acid, fumaric acid, and lactic acid were detected in the kimchi, and a significant increase in the concentration of lactic acid during fermentation was observed in the all experimental groups.

  • This study was carried out to offer the raw data of floricultural resource by scientific analyzing the growth and soil environment in Viola orientalis habitat, and establish the ecological characteristics in developing the relative model. In the results of soil factors analysis, there showed that acidity was pH 5.1, organic matter content of 9.1%, available $P_2O_5$ of 40.6mg/kg, exchangeable $K^+$ of $0.2cmol^+/kg$, exchangeable $Ca^{2+}$ of $3.5cmol^+/kg$, exchangeable $Mg^{2+}$ of $0.8cmol^+/kg$, cation exchange capacity(C.E.C) of $13.7cmol^+/kg$ and electrical conductivity(EC) of 0.4dS/m. The growth characteristics were surveyed that height was 12.4cm, leaf width of 2.5cm, leaf length of 3.0cm, flower width of 2.5cm, peduncle of 2.3cm and chlorophyll of $38.5{\mu}g{\cdot}mg^{-1}$.

Effects of Irrigation Interval, Medium Composition and Drainage on the Growth Response of Korea Native Fern Rumohra aristata at Indoor (실내에서 관수주기, 상토종류, 배수층의 유$\cdot$무에 따른 자생 가는쇠고사리의 생육반응)

  • It was aimed to promote as a material for interior landscape by validating Rumohra aristata, in an indoor environment, especially irrigation interval, medium composition and drainage at indoor. 1. The result of physico-chemical analysis of medium composition showed that porosity, pH and Ex-Ca, Ex-Mg and Ex-K were high with peatmoss: vermiculite: perlite(1:1:1) and water contents, organic matter content, total nitrogen and cation exchange capacity were high with sand: leaf mold(1:1). 2. Growth and indoor adaptability of Rumohra aristata were better with irrigation at 2 interval per week than irrigation at 7 interval per week regardless of drainage. 3. In the case of medium composition, of growth and indoor adaptability were higher with sud: leaf mold (1:1) than peatmoss: vermiculite: perlite(1:1:1). 4. Fronds fresh weight and dry weight decreased when irrigation interval increased and were higher with sand: leaf mold(1:1) than peatmoss: vermiculite: perlite(1:1:1) treatment.

Physicochemical Adsorption Characteristics of MTBE and Cadmium on Clay Minerals (점토광물에 대한 MTBE와 카드뮴의 물리화학적 흡착 특성)

  • This study was performed to investigate adsorption characteristics of MTBE and Cd depending upon types of clay minerals md their physicochemical properties. The adsorption characteristics were examined by batch adsorption test on various experimental parameters such as adsorption time, ratio of solution to soil, concentration of contaminants, content of organic matter, pH, and zeta potential. The adsorption efficiency of MTBE or Cd for three types of clays decreased in response to the increase of the ratio of solution to soil whereas their adsorbed amounts increased. MTBE was greatly adsorbed in the decreasing order of vermiculite, bentonite, and CTAB-bentonite while Cd was adsorbed in the decreasing order of bentonite, vermiculite, and CTA-bentonite. An equilibrium isotherm for MTBE was well fitted to Freundlich plotting whereas that for Cd was closely corresponded to Langmuir isotherm. The adsorbed amount of MTBE on bentonite and vermiculite showed the maximum at 1% and 5% of humic acid, thereafter diminished while the adsorbed amount of MTBE on CTAB-bentonite increased in proportion to humic acid. Conversely, the adsorbed amount of Cd on the addition of humic acid continued to increase regardless of types of adsorbents. For all types of adsorbents, adsorbed quantity and adsorption efficiency of Cd have been coincidently increased at pH 8 and they were further enhanced at pH 10 showing 90% adsorption efficiency. Upon pH rose, the zeta potential on each adsorbent began to decrease, while increasing Cd concentration led to decline of zeta potential, which in turn ascribed to lowering dispersion stability that could consequently enhance adsorption capability.
