This study was conducted to establish the tissue culture system for Korean domestic Miscanthus sinensis, which is used in various purposes such as forage, and bio-energy resources. With the mature seed of Miscanthus, optimum concentrations of plant growth regulators were identified for an efficient callus induction and regeneration. Among the treatments of 1~10 $mg{\cdot}L^{-1}$ 2,4-D, IBA, or NAA, callus induction rate was highest (85.3%) on MS medium containing 5 $mg{\cdot}L^{-1}$ 2,4-D. Under the condition, the callus were efficiently induced and proliferated with comparably lower frequencies of callus browning. In shoot regeneration, the treatment of NAA combined with BAP seemed to contribute more efficient conditions to shoot regeneration than those of NAA with Kinetin or 2-iP. Especially, regeneration efficiency and number of regenerated plants were 83.7% and 5.5 in 3 $mg{\cdot}L^{-1}$ NAA with 5 $mg{\cdot}L^{-1}$ BAP, respectively, which were higher frequencies than those in NAA with Kinetin or 2-iP. In results, 5 $mg{\cdot}L^{-1}$ 2,4-D and 3 $mg{\cdot}L^{-1}$ NAA combined with 5 $mg{\cdot}L^{-1}$ BAP were efficient for embryogenic callus induction and regeneration of Miscanthus. This system would be useful for mass-propagation and developing new cultivars via tissue culture of Miscanthus sinensis.
Experiments were performed for investigate the immune activities on human B and T cell growth and secretion of their cytokines. Also, antibodies in serum were investigated in female ICR mouse by feeding the extracts of Acer mono at dose of 30 and 100 mg/mL of one day orally for 15 days. The cytotoxicity of the samples on normal human cell (HEL299) was below 24.15% in adding the all extracts. Both human immune B and T cells were increased up to about 40% by the high pressure extraction process. The secretion of cytokines (IL-6, TNF-${\alpha}$) on human B and T cells were increased up to 20-50% by adding the high pressure extraction process compare to the hot water extraction process. Also, total serum IgG levels increased by feeding Acer mono extacts. It can be conclude that optimum condition for efficient extraction of Acer mono as functional materials is solvent extraction process using water with high pressure at below $100^{\circ}CA$ than typical process.

'Baekjung' adaptable to high temperature was made by crossing between monokaryon derived from selfing of brown strain and monokaryon derived from Korea white strain. In the condition that temperature is maintained at $10^{\circ}C$ without low temperature of $4^{\circ}C$ suppressing treatment and wrapping during cultivation period, it showed good productivity than Uri1ho(control). The optimum temperature of mycelial growth was $30^{\circ}C$ and that of fruiting body initiation and development were $14^{\circ}C$ and $7^{\circ}C$, respectively. The days for the fruiting and yield were 7 days and $277{\pm}11.2$ g per 1,100 ml bottle, respectively. This variety needed high concentration of carbon dioxide up to 4,000 ppm for the good quality.

This study was conducted to find optimum mixed sowing ratio of green manure crops to reduce the use of chemical fertilizer as well as to increase the crop yield potential which will foster the utilization of green manure crops in the upland field in view of environment friendly agriculture. According to the study, the mixed ratio, 50:50, of hairy vetch and green barley showed highest nitrogen production yield in the soil due to the relatively higher organic nitrogen supply from the hairy vetch plant as well as nitrogen fixation from the air rather than other mixed ratio. In the 50:50 mixed ratio of hairy vetch and green barely total nitrogen amount in the soil showed 17.2kg per 10a, but in the other treatment ratio such as 75:25, 25:75. total nitrogen fixation amount were 16.7, 16.9 respectively. We also conducted the experiment to compare the effect of the mixed sowing treatment of green manure crops on the production of corn cultivated as a succeeding plant of hairy vetch. According to the result, the mixed ratio, 50:50, of hairy vetch and green barley treatment showed highest yield potential of corn as 153kg per 10a in seed weight which is due to the relatively higher organic nitrogen supply from the hairy vetch plant as well as nitrogen fixation from the air rather than other mixed ratio. In the mixed sowing treatment of hairy vetch 100 and barley 0 ratio, the corn production showed 148kg per 10a which is 5kg lower than that of hairy vetch 50 and barley 50 ratio, but showed statistically no difference between those two treatment. Otherwise, different treatments, such as hairy vetch 75 and barley 25, 25 and 75, 0 and 100 showed statistically different each other. Therefore, it was concluded that green manure crops, such as hairy vetch, green barley and rye were very effective crops to increase the soil fertility and gave the positive effect to the crops to give vegetative and propagative growth condition and, in turn, increased the yield potential. We have to make policy to enhance the utility of green manure crops in the upland crops as well as faddy field for the soil fertility and crop yield production which will guarantee prominent quality of environment friendly agriculture products.
The preparation of potassium hexatitanate whisker by flux method was investigated. In this study, 8 types synthesis of flux such as $V_2O_5$, $Bi_2O_3$, $B_2O_3$, $Pb_3O_4$, KCl, $K_4P_2O_7$, $K_2WO_4$ and $K_2MoO_4$ were tested to find a suitable flux for the synthesis of potassium hexatitanate whisker. Effects of various reaction variables such as reaction temperature, time, $TiO_2$ mole ratio to $K_2CO_3$, flux mole ratio to the mixture of $K_2CO_3$ and $TiO_2$, and slow-cooling treatment on the crystallization of potassium hexatitanate whisker were investigated. $K_2MoO_4$ and $K_2WO_4$ were better flux than others tested for the synthesis of potassium hexatitanate. In the presence of $K_2MoO_4$ or $K_2WO_4$ flux, the optimum condition for the synthesis of potassium hexatitanate whisker was that reaction temperature of $1000{\sim}1100^{\circ}C$, reaction time of 5 hours, $TiO_2$ mole ratio to $K_2CO_3$ of 6.0, and flux mole ratio to mixture ($K_2O+nTiO_2$) of 4.0. Slow-cooling treatment showed good effect on the growth of long fibrous potassium hexatitanate.
Environmental Specimen Banks (ESBs) are playing pivotal role in monitoring the effect of environmental pollution on the ecosystem based on the retrospective analysis of the representative samples collected regularly and stored in cryogenic condition. In Korea, National Environmental Specimen Bank (NESB) was established in 2009 and the standard operating procedures (SOPs) for sampling, and cryogenic milling and storage had been prepared during 2007-2010. Since then, the tentative SOPs for the seven kinds of specimens (shoots of Red Pine (Pinus densiflora) and Korean Pine (Pinus koraiensis), leaves of Mongolian Oak (Quercus mongolica) and Zelkova Tree (Zelkova serrata), eggs of Feral Pigeon (Columba livia var. domestica), muscles and organs of Common Carp (Cyprinus carpio), and Freshwater Bivalve (Unio (Nodularia) douglasiae)) have been put to test in the field and laboratory as well against the practicality and feasibility. The SOPs were improved by reflecting the findings from the research and the following discussion regarding the selection of specimen (Feral Pigeon suffering from a control management), sample size (a problem of decreasing number of sampling trees related to increasing sampling time) and period (a problem related to a bud growth), and sampling methods etc.. In addition, barcoding system for the management of the specimen information, and monitoring system of the cryogenic storage to regulate the optimum temperature and the liquid nitrogen level were also developed for the efficient and effective control of the samples. Lastly, the safety guide and emergency protocol were augmented to guarantee a safe work environment with the cryogenic facility. These improvements of the SOPs are expected to contribute to more stable operation of the NESB.
The purpose of this experiment was to investigate the development of new spot disease on the leaf and stem of rosemary (Rosmarinus officinalis) in commercial greenhouses at Jeonju and Namwon in Korea. Incidence of target spot on rosemary was higher at the end of the rainy season with high humidity. Those symptoms were black ring spots (3-5 mm in diameter) and withering on green leaves and stems. Conidiophores and conidia were formed on the infected tissue in moist chamber and conidia were shown as the cylindrical and oval types in chain, ranged from 55 to $275{\mu}m$ in length, and 7 to $14{\mu}m$ in width. Conidia with eight to ten pseudosepta were formed on the conidiapore. The optimum growth temperature of isolates was $30^{\circ}C$ on the PDA medium under the dark condition. In the pathogenesis test, the target spot and withering symptoms were appeared on the leaves and stems 3 days after inoculation showing similar symptoms compared to those of in nature. The same fungus was re-isolated from infected lesion, indicating that Corynespora cassiicola caused leaf target spot and twig blight on rosemary. The rDNA ITS nucleotide sequences of the pure cultured isolate from the diseased area on rosemary showed 100% similarity to the sequences of C. cassiicola available in the GenBank database (JQ595296, JQ595297, FJ852715 and AY238606). Therefore, we report that the target spot of leaves and stems in rosemary was caused by C. cassiicola.

The Government is trying to increase total food grain yield to meet national self sufficiency by means of increasing unit yield as well as extending crop land, and this year he set the target of 321,000 hectare of forest to clear for cropping. This study was carried to investigate the most efficient method of clearing hillock by bulldozer, and successful method to develope yielding potential of newly cleared land in short term. Since the conventional land clearing method is just earth leveling and root removing neglecting top soil treatment, the growth of crop was poor and farmer tends not to care the land. The top-soil-furrowing method is applied through out this study, that is advantageous especially for the land having shallow top soil and low fertility like Korean forest. In this study, various operating method were tried to find out most efficient method separately in connection with the land slope less than 25 percent and over, and several fertilizing methods to develop yielding potential. The results are as follows; 1) For the natural land slope utilization method, applicable to the land having less than 25 percent slope, reverse operating was more efficient than using forward gear of bulldozer. The operating time was 3 hour 32 minutes and 36 seconds using forward gear was 2 hour 32 minutes and 30 seconds for reverse gear operation per 1,000 square meter. 2) Bulldozer having angle blade adjustment needed 7hr 15min. for constructing of terrace per 10a compaire with the one having angle & tilt adjustment needed 6hr 4min for same operations. Specially there is significant difference for operation time of first period (earth cutting) such as bulldozer having angle blade adjustment needed 3hr 56min compaired with the one having angle & tilt adjustment 3hr 59min. In construction of terrace, the bull-dozer having tilting and angle blade adjustment was most suitable and performed efficiently. 3) For the fertilizer application treatment, the grass (Ladino clover) yield in first year was almost same as ordinary field's in the plot applied(N.P.K+lime+manure) while none fertilizer plot showed one tenth of it, and (N.P.K.+lime) applied plot yielded on third. 4) The effect of different land clearing method to yield showed significant difference between each treatment especially in the first year, and the conventional method was the lowest. In the second year, still conventional terracing plot yielded only half of ordinary field while the other plots showed as same as ordinary field's. 5) The downward top soil treatment plot showed most rapid improvement in soil structure during one year physio chemically, it showed increase in pH rate and organic composition, and the soil changed gradually from loam to sand-loam and the moisture content increased against the pF rate, and it gives good condition to grow hay due to the increase of field water capacity with higher available water content. 6) Since the soil of tested area was granite, the rate of soil errosion was increased about 2 to 5 percent influencing in soil structure more sand reducing clay content, and an optimum contour farming method should be prepared as a counter measure of errosion.
Effects of light intensity and temperature on photosynthesis and respiration of autogenous wild SUMBODY plants, Dystaenia takesimana, known as a prospective source of forage in Ulleung island, Korea, were investigated. Results were as follows : 1. Light saturation point at $20^{\circ}C$ was 34 to 38 klux and light compensation point was 4 to 6 klux. Apparent photosynthetic rate at light saturation was 9 to 12 mg $CO_2/dm^2/hr$. 2. Optimum temperature for photosynthesis was $20^{\circ}C$ and $Q_{10}$ values of two temperature ranges, 20 to $30^{\circ}C$ and 30 to $40^{\circ}C$, were 0.8 and 0.9 for photosynthesis, while $Q_{10}$ for respiration were 1.6 and 1.7. 3. Native plants sampled in area of higher altitude had a higher apparent photosynthtic rate at lower temperature and the plants sampled in area of mixed with bushes had lower a light compensation point. These rusults suggest that the Inland weather condition of Korea during spring and fall seasons might be. suitable for the SUMBODY growth but inadequate during summer.
Korean ginseng(Panax gmseng C.A. Meyer) is very difficult to obtain stable production of qualified ginseng roots because of variable stresses in soil environments. In environment stresses, soil condition is the most important factor, among which nutrients, especially inorganic materials such as N, P, K, Ca, Mg, Fe, etc., influence greatly on the ginseng growth. However, present ginseng field soils in Korea contain so much amount of such inorganic materials that a variety of remarkable disorders were noted in many ginseng plantations, resulting in decrease of qualitative ginseng production. Therefore, it is required to search for genetic resources and genes tolerant to salt stress for the development of ginseng cultivars. Selection of stress-tolerant ginseng lines in fields is very difficult because it is almost impossible to control properly the environmental conditions of soil. On the contrary, it can be studied with ease to search for stress-tolerant ginseng lines through in vitro culture because of easy manipulation of stress conditions. Murashige & Skoog(MS) media with 2.5 folds of $KNO_3,\;NH_4NO_3,\;MgSO_4\;7H_2O,\;KH_2PO_4,\;and\;CaCl_2\;2H_2O$ was established for the selection of ginseng lines tolerant to salt stress under the embryo culture.
