  • Ke Ho Lee and Kun Hyung Chang (Army Research and Testing Lab.): Studies on koji for optimum condition of growth and identification of Aspergillus spp. Cpart 2). Kor. J. Microbiol. Vol. 2, No. 1, p. 17-18 (1964). The experiments reported in this paper are concerned with the identifications, the growth conditions and lethal temperatures of the three strains of Aspergillus spp. which have been isolated from the soils and Meju (fermented soy bean in Korea). The results obtained in the light of the manual of Raper and Thom for identification of the Genus, Aspergillus, have been shown that the three strains of Aspergillus spp. are pertained to Aspergillus flavusoryzae group. The optimum temperature, pH and the lethal temperatures of the Aspergillus spp. have been measured.

  • This research aims the production of anti-tumor substances through in vitro culture of hairy roots transformed by Agrobacterium rhizogenes in Artemisia sylvatica MAX and the effect of culture conditions on optimum growth of hairy roots. We investigated the optimum medium, pH, carbon source, sucrose, light, Fe and polyamine conditions of various lines of hairy roots (NK3, NK4, YX. NK3-10) induced from Artemisia sylvatica to increase the optimum growth of hairy roots. MS medium was the best for optimum growth of hairy root clone, NK3-S10. The optimum culture period was 4 weeks for NK3-S10. The optimum sucrose concentration was 3.5%. The optimum concentration of FeSO$_4$, spermine and spermidine was 0.1 mM, 10 mM and 100 mM, respectively.

  • This study is on the investigation of hydrogen production and substrate removal by photosynthetic bacteria. After using of Rhodospillum rubrum KS-301 and IFO 3986, which are photosynthetic bacteria as strains, R. rubrum KS-301 was turned out a better strain. And result of experiment in which glucose and sodium lactate, components of wastewater, were used limiting substrates, showed that the productivity of hydrogen was indifferent with the kind of substrates. In batch experiments using free cells and immobilized whole cells, the decrease in hydrogen productivity was observed in the latter case. From the results of these experiments, specific growth rate of cells, specific utilization rate of glucose, and specific production rate of hydrogen were calculated. And each rate was expressed in the form of Monod equation of which parameters were estimated. Also the optimum condition of hydrogen production for free cells was $30^{\circ}C$, pH 7, and 12,000 Lux, and the optimum immobilized condition was as follows: initial immobilized cell concentration 1.0g/L, sodium alginate concentration 2% and light intensity 12,000 Lux.

Isolation of Photosynthetic Bacteria and the Cell Growth on Soybean-Curd Whey Wastes (광합성 세균의 분리 동정 및 두부 유청 폐수에서의 배양조건 검토)

  • For the purpose of fixing solar energy and utilizing water wastes from food industries for SCP(single cell protein), 170 strains of photosynthetic bacteria were isolated from 56 samples. Among 170 strains, B-Ps-106 strain was selected as the most suitable strain and identified as a variant or a relation of Rhodopseudomonas sphaeroides and its growth was better under anaerobic light condition than aerobic condition. The optimum conditions of the cell growth of B-Ps-106 were investigated on soybean-curd whey media. The optimum pH for cell growth was $8.5{\sim}9.0$. The optimum temperature was $30^{\circ}C$ and the optimum light density was above $0.72\;cal/cm^2/min$. The most favorable concentration of $K_2HPO_4\;and\;(NH_4)_2SO_4$ was both 0.9 g/l when added to soybean-curd whey media.

Optimum Salinity and Temperature Condition for Mass Culture of the Brackish Water Flea, Diaphanosoma celebensis (기수산 물벼룩, Diaphanosoma celebensis의 대량배양을 위한 최적 염분 및 수온 조건)

  • This study investigated the optimum salinity and temperature conditions for mass culture of the brackish water flea, Diaphanosoma celebensis. Community and individual cultures of flea were maintained in 1 L beakers and 3 mL vessels (of a 12-well culture plate), respectively, and fed green algae, Tetraselmis suecica. In salinity experiments ranging from 5 to 34 psu, continuous growth of flea populations was found up to 34 psu. However, the specific growth rate and life span of females showed decreasing tendencies with the increase of salinity. The highest maximum density and offspring number were 33.6 individuals (ind.)/mL and 55.3 ind. at 10 psu, respectively. In the temperature experiments ranging from 20 to $40^{\circ}C$, population growth of D. celebensis increased continuously until $35^{\circ}C$ and then decreased over $40^{\circ}C$. The specific growth rate was significantly higher at 25 and $30^{\circ}C$ than at 20 and $40^{\circ}C$. Female life span tended to decrease with temperature increase. The highest maximum density and offspring number were 52.3 ind./mL and 46.0 ind. at $30^{\circ}C$, respectively. These results suggest that the optimum salinity and temperature for mass culture of D. celebensis may be 10 psu and $30^{\circ}C$, respectively.

Growth and fatty acid composition of three heterotrophic Chlorella species

  • Some Chlorella species grow heterotrophically with organic substrate in dark condition. However, heterotrophic Chlorella species are limited and their optimum culture conditions are not fully known. In this study, three heterotrophic Chlorella species, two strains (C4-3 and C4-4) of C. vulgaris and one Chlorella sp. (C4-8) were examined on optimum culture conditions such as carbon source, temperature, and concentrations of nitrogen and phosphorus in Jaworski's medium (JM). And the growth and fatty acid composition of Chlorella were analyzed. For three heterotrophic Chlorella species, glucose (1-2%) as a carbon source only increased the growth and the range of optimum culture temperature was $26-28^{\circ}C$. Doubled concentrations of the nitrogen or phosphorus in JM medium also improved the growth of Chlorella. Chlorella cultured heterotrophically showed significantly higher growth rate and bigger cell size than those autotrophically did. C. vulgaris (C4-3) cultured heterotrophically showed the highest biomass in dry weight ($0.8g\;L^{-1}$) among three species. With respect to fatty acid composition, the contents of C16:0 and n-3 highly unsaturated fatty acid (HUFA) were significantly higher in autotrophic Chlorella than in heterotrophic one and those of total lipid were not different between different concentrations of nitrogen and phosphorus in JM medium. Among three Chlorella species in this study, C. vulgaris (C4-3) appeared to be the most ideal heterotrophic Chlorella species for industrial application since it had a high biomass and lipid content.

  • Effect of light intensity on ALA production was, investigated. The culture condition and medium optimization were also examined for the biosynthesis of ALA using Rhodobacter sphaeroides, non-sulfur bacteria, and investigated for enhancement of the production of ALA. In the dark condition, extracellular ALA formation and cell growth were not observed. Optimum light intensity for cell growth and ALA production were 4 kLux and 5 kLux, respectively.

Growth of Spirulina platensis in Effluents from Wastewater Treatment Plant of Pig Farm

  • Spirulina platensis NIES 39 was grown in effluents from a wastewater treatment plant utilizing activated sludge process at a pig farm to reduce pollutants and to produce feed resources. The addition of 2 g/l NaCl was required for the growth of S. platensis in the effluents with about 100 mg/l ammonia nitrogen concentration. The growth was stimulated by the addition of 2 g/l sodium bicarbonate and 4 mg/l ferrous sulfate. The microorganism grew well at pH 8~11, and the optimum pH for the growth was 10. The algal concentration of 1.1 g/l was attained after 8 days of growth at the optimum condition with the removal of 95% ammonia nitrogen and 34% phosphorus.

  • Effect of temperature, pH and soil depth on germination of Dianthus chinensis, Dianthus barbatus, and Perennial pennant were investigated in growth chamber and soil condition at the ratio of 7:3 (natural soil and organic soil) in laboratory condition. the optimum temperature for seed germination was recorded for $20^{\circ}C-\;25^{\circ}C$. Maximum germination was observed for Dianthus barbatus (76%) where as in soil condition Perennial pennant (51%) showed maximum germination at 1 cm soil depth. Similarly, optimum pH for seed germination was at pH 6 in all the species. So in lower pH (at pH4) seed germination was inhibited. Germination of these selected species at different environmental condition help to construct the ecological sound proof wall to mitigate the noise especially in urban areas.

Control of Organic Acid Inhibition in Escherichia coli Culture with Eledtroudialysis (유기산의 저해를 조절하기 위한 Escherichia coli의 전기투석배양)

  • E. coli growth is inhibited by organic acids produced in the broth. In order to reduce the inhibition, an electrodialysis unit was used. Model solutions (acetic acid plus distilled water or M-9 medium) were tested in the unit for investigating the optimum condition of current and voltage. Electrodialysis cultures were performed with the optimum condition where the highest current efficiency could be attained. The distilled water plus acetic acid gave us a higher current efficiency than the M-9 plus acetic acid. Electrodialysis efficiently removed acetic acid and so enhanced the specific growth rate of E. coli compared with the control experiment without clectrodialysis.
