OPTIMIZATION OF CULTURE CONDITIONS FOR PRODUCTION OF PNEUMOCOCCAL CAPSULAR POLYSACCHARIDE TYPE I

  • Streptococcus pneumoniae (pneumococcus), the most common cause of bacterial pneumonia, has an ample polysaccharide(PS) capsule that is highly antigenic and is the source of PS vaccine. This investigation was undertaken to optimize the culture conditions for the production of capsular PS by type 1 pneumococcus. Among several culture media, brain heart infusion (BHI) and Casitone based media were found to support luxuriant growth of pneumococcus type 1 at the same level. Because BHI medium is rather expensive and more complex than the Casitone based media, the Casitone based media was used to study optimization of the culture condition. The phase of growth which accomodated maximum PS production was logarithmic phase. Concentrations of glucose greater than 0.2% did not enhance growth or PS production. Substitution of nitrogen sources with other resources or supplemention of various concentrations of metal ion (with the exception of calcium ion) had adverse effects on growth and PS production. On the other hand, low level aeration was beneficial for increased PS production. Addition of 3 mg/I concentration of methionine, phenylalanine, and threonine were found to enhance growth and PS production. The synergistic effect of all the favorable conditions observed in pneumococcal growth assays provided a two-fold cumulative increase in capsular PS production.

쌀가루를 첨가한 양배추 크림수프의 제조조건 최적화 (Optimization of Mixing Condition of Cabbage Cream Soup)

  • The principal objective of this study was to determine the optimal mixing condition of two different amounts of cabbage and rice flour for the preparation of a cabbage cream soup. The experimental design was based on the central composite design methodology of response surface, which included 10 experimental points, including two replicates for the cabbage and rice flour. Physiochemical and sensory properties were measured, and these values were applied to the mathematical models. A canonical form and perturbation plot showed the influence of each ingredient on the mixed final product. Water content and pH values increased with increasing quantities of rice flour. Neither cabbage or rice flour affected the L and a values, but the b value increased with greater quantity of both ingredients. Viscosity increased with increasing added cabbage. Sensory evaluation results were significant in the predicted model for flavor (p<0.05), concentration (p<0.01) and overall quality (p<0.01). As a result, the optimum formulations by numerical and graphical methods were calculated as 111.79 g cabbage and 8.99 g rice flour.

  • Effect of light intensity on ALA production was, investigated. The culture condition and medium optimization were also examined for the biosynthesis of ALA using Rhodobacter sphaeroides, non-sulfur bacteria, and investigated for enhancement of the production of ALA. In the dark condition, extracellular ALA formation and cell growth were not observed. Optimum light intensity for cell growth and ALA production were 4 kLux and 5 kLux, respectively.

Cordycepin 생성을 위한 배양조건 및 배지조성의 최적화 (Optimization of Culture Condition and Media Composition on the Production of Cordycepin by Cordyceps militaris.)

  • Cordyceps militaris의 액체 배양시 cordycepin(3'-deoxyadenosine) 생성에 영향을 주는 물리화학적 조건 및 배지 조성의 영향을 조사하였다. 사용된 17종 균주 중에서 C. militaris KCTC 6862, C. militaris KCTC 16932 및 C. militaris DGUM 32003이 우수한 cordycepin생산성을 보여주었다. Cordycepin생산에 적합한 최적 온도와 PH는 각각 $24^{\circ}C$와 pH 6.0-10범위이었다. YM배지에 탄소원으로 glucose를, 질소원으로 tryptone을 각 1% 첨가하였을 매 우수한 cordycepin 생산을 보여주었다. Tryptone을 질소원으로 하는 YMG 액체배지에서 5일간 배양하였을 경우, 39mg/l의 cordycepin의 농도가 측정되었다. 0.1%의 다양한 인산원을 각각 첨가한 결과, cordycepin생산을 저해하였다.

Optimization of cell growth and TAPS production by Pichia ciferrii mutant in batch culture

  • Batch culture of mutant derived from Pichia ciferrii ATCC 14091 was investigated for producing the intracellular tetraacetylphytosphingosine (TAPS). Composite experimental design was used to optimize the composition of the culture medium for maximizing the productivity of TAPS. In this experiment, various culture parameters were investigated that were the effects of temperature, the initial culture pH, the carbon-to-nitrogen ratio, the concentration of trace elements, and the concentration of cofactors. The optimal temperature for cell growth and TAPS synthesis appeared to be %25^{\circ}C$. An initial pH value of 7.5 gave the best results. Under the best condition, the maximum TAPS concentration indicated 7.2 g/L and its productivity was 0.06 g/L-hr in a 2.5 L jar

Optimization of Mulberry Jelly Making by Response Surface Methodology

  • Recently, though mulberry's superiority as a functional food has been proven, its use as a food material is limited. Therefore, in this study, to develop jelly using mulberry that is compatible with Korean tastes as health functional food by grafting the method to manufacture jelly consumed as a dessert or a snack in the west, according to the central composite design, mulberry jelly was produced by varying the content of citric acid ($X_1$), sucrose ($X_2$), and gelatin ($X_3$) at 5 levels. And by applying the response surface methodology, rheology and sensory preference experiment results were analyzed, the optimization of mulberry jelly manufacturing condition was carried out, and studies on the analysis of composition were performed. As the sensory preference of mulberry jelly, except the flavor, the remaining hardness, elasticity, sweetness, color, and the overall quality were found to be significant. And similarly, it was found to be influenced greatly by gelatin content generally. Based on the overlapped part of categories, in the range of factors that satisfy all the sensory categories, the value located in the middle was calculated, the optimization point was obtained, and it was found to be 6.2 g citric acid, 141.0 g sugar, and 13.5 g gelatin.

Hericium erinaceum 액체배양의 최적화 (Optimization of Submerged Cultivation of Hericium erinaceum)

  • H. erinaceum은 식용은 물론 각종 약리효과를 나타내어 현재 산업적으로 매우 주목되고 있는 기능성 소재들 중의 하나이다. 본 연구에서는 H. erinaceum의 기능성 소재화 연구의 일환으로, 지금까지 체계적으로 연구된 바 없는 H. erinaceum의 액체배양을 시도하고. 이 버섯 액체배양의 최적화를 수행하였다. 16종 버섯 배지의 플라스크 배양을 통해 우수 기본 배지를 선발하였고, 이의 반응표면분석에 의해 배지 및 배양조건의 최적화를 수행한 결과, 최적 배지 조성은 glucose 3%, yeast extract/peptone(1:1) 0.2%, $KH_2PO_4/MgSO_4$(1:1) 0.2%이었으며, 최적 배양조건은 $22.28^{\circ}C$, pH 5.67, 접종량 13.42%에서 얻어졌다. 이러한 최적 배지 및 배양조건하에서 얻어진 균사체량은 약 9 g/L 이었으며, 이는 기본배지에서 배양 8일 후 얻어진 최대값인 5.40 g/L에 비하면 약 1.7배 향상된 결과이었다. 하지만 H. erinaceum의 액체배양시 낮은 균체량과 비교적 느린 생육이 관찰되어 보다 높은 생산성의 향상을 위해서는 이에 적합한 새로운 형태의 fermenter의 선정 및 새로운 배양 system의 적용이 필요한 것으로 나타났다.

Monascus purpureus로부터 다량의 색소생성 변이주의 분리 및 색소생성의 최적조건 (Isolation of Pigment Overproducing Mutant from Monascus purpureus and Optimization of Pigment Production)

  • Monascus purpureus KCCM 60016을 이용하여 다량의 색소생산 변이주를 개발하고 생산조건에 대하여 조사하였다. 자외선 조사로 M. purpureus KCCM 60016으로부터 돌연변이를 유도한 결과, 색소생성이 우수한 P-57 변이주를 분리하였다. 분리된 P-57 변이주는 고체 배양에서 현미를 기질로 했을 때 색소생성이 가장 우수한 것으로 나타났으며, 색소생성의 최적 배양조건은 배양온도 $30^{\circ}C,\;90\%$의 배양습도에서 30일간 배양이 가장 우수했다. 이상의 조건에서 배양한 적색색소, 오렌지색소와 황색색소의 양은 각각 160.0 unit, 193.6 unit와 141.6 units-나타났다.

반응표면 분석법을 이용한 바질 첨가한 홈메이드 파스타 제조의 최적화 (Optimization of Homemade Pasta with Addition of Basil using Response Surface Methodology)

  • The purpose of study was to determine the organoleptic characteristics of homemade basil-pasta made in various compounding ratio according to central composite design. The optimum mixing condition for basil-pasta was optimized by response surface methodology. The texture, including color, flavor, elasticity and overall quality, was measured as sensory evaluation. The optimum mixing rates of durum wheat semolina, basil and egg were 73.97g, 5.95g and 36.17g for color, 53.14g, 4.52g and 35.85g for flavor, 48.75g, 2.95g and 36.85g for elasticity. The organoleptic overall quality of basil-pasta showed the maximum score in the mixing condition of 55.52 g durum wheat semolina, 4.55g basil paste and 35.51g egg.

Bacillus subtilis IJ-31에서 Hydrocinnamic Acid 생산을 위한 최적배양조건 (Optimization of Culture Condition for the Hydrocinnamic Acid Production from Bacillus subtilis IJ-31)

  • 본 연구에서는 식물생장억제미생물 B. subtilis IJ-31이 생산하는 HCA의 최적생산조건을 검토하였다. HCA생산 최적조건으로 탄소원은 1%의 미강, 질소원은 0.5% tryptone, 무기염은 0.1% $ZnCl_2$, 배양 온도는 $37^{\circ}C$, 배양시간은 60시간, pH는 7.0에서 $90.5\;{\mu}g/ml$의 HCA를 생산하였으므로 이를 최적생산조건으로 확립하였다. 이를 바탕으로 통계프로그램(SAS) 중 반응표면 분석법(RSM)을 이용한 jar fermentor 배양에서의 HCA 최적생산 조건을 확립하였다. 즉 토양추출물 2.24% 첨가, 교반속도 290 ppm, tryptone 0.83% 첨가, 배양온도 $37^{\circ}C$, 배양시간 60시간, pH 7.0에서 $102.99\;{\mu}g/ml$의 HCA를 생산하는 최적생산 조건으로 예측되었다. 따라서 실제 상기 RSM 최적 조건에서 실험한 결과 HCA는 $102.5\;{\mu}g/ml$를 생산하였다. 이러한 결과는 플라스크에서 HCA는 $90.5\;{\mu}g/ml$로 생산되었으나 jar fermenter 배양에서는 플라스크 배양에 비하여 12%가량 HCA 생산량이 증가된 결과를 보여주는 것이다.
