Production of Tropane Alkaloids by Two-stage Culture of Scopolia parviflora Nakai Adventitious Root

  • Scopolia parviflora Nakai, a rare and endangered species, is the sole plant producing tropane alkaloids (TA) among the Korean native species. In order to enhance TA productivity the SP72 root line was selected by screening 100 of root line, and the optimal culture media for root growth and TA production were investigated with the SP72 roots. Based on the several media, SH and 2B5 medium were determined as growth medium and White and NN medium as production medium. Among the four combinations of two-stage culture, 2BN (2B5 as growth medium plus NN as production medium) showed more enhanced root growth and TA production as compared with production media of White and NN medium and growth media of SH and 2B5 medium, respectively. However, bubble column bioreactor (BCB) cultures applying two-stage culture did not reveal the effective results despite of the each successful operation of two-stage culture in conical flasks and BCB cultures.

Shape Optimal Design to Minimize Stress Concentration in the Journal of the Heating Drum for a Hot-Rolling Press (열간압연 프레스용 가열드럼 저어널부의 응력집중 최소화를 위한 형상 최적설계)

  • The present research deals with structural analysis and optimal design of the heating drum of a hot-rolling press for medium density fiberboard. Stress concentration in the journal of the heating drum is analyzed by the submodel technique of the finite element method. The fatigue life under operating conditions is analyzed and evaluated by the stress-life theory. Shape optimal design problems, to minimize the maximum stress occurring in the journal, are formulated and shape parameters of the corner fillets of the journal are defined as the design variables. The problems are solved by the numerical optimization method and optimal shapes are found. The optimal designs are shown to be reliable in terms of the maximum stress and the fatigue life.

The Medium Development for Entomopathogenic Fungi by Using Silkworm Powder (누에분말을 이용한 곤충기생균의 최적생장배지 개발)

  • This study was carried out to investigate the optimal medium and optimal dextrose concentration of 12 entomopathogenic fungi. The colonies of Cordyceps gracilioides, C. sp.(J23), Paecilomyces farinosus(J5), P tenuipes, Paecilomyces sp.(J4), Beauveria bassiana and B. brongniartii on SLA(silkworm larva agar) medium are similar to those on PDA(Potato dextrose agar) medium. On the contrary, the colonies of 5 fungi are very various according to its medium. The optimal medium for 1 1 fungi was SLA-A medium, which was better than PDA medium with regard to the mycelial growth. The optimal concentration on mycelial growth was superior to 6% for 6 fungi. When silkworm larvae was injected with P. tenuipes on SLA medium for fruiting body production. Infection rate, fruiting body formation rate, synnemata number and synnemata length were 91.3 $\pm$ 1.5%, 88.6 $\pm$ 2.3%, 56.6 $\pm$ 3.2 and 33.4 $\pm$ 2.88 ㎜ respectively. In economical analysis, the prototype medium can reduce 61.9% of production cost per 1 liter compared with PDA medium.

Optimization of Culture Conditions for Erythritol Production by Torula sp.

  • The medium for erythritol production by Torula sp. in a 500-ml baffled flask was optimized to be 300 g/I sucrose, 10 g/I yeast extract, 3 g/I $KH_2PO_4$, and 10 mg/I $CuSO_4{\cdot}5H_2O{\;}at{\;}34^{\circ}C$ with initial pH of 5.5. Using this optimal medium, erythritol of 166 g/I was obtained after 140 h of cultivation, corresponding to 55.3% of the erythritol yield from sucrose with a productivity of 1.11 g/I/h. Optimal concentrations of carbbon and nitrogen sources in a fermentor were higher than that in a flask due to the higher oxygen supply of the fermentor. Employing the medium containing 300 g/I or 400 g/I sucrose for the determination of optimal C/N ratio, the C/N ratio was found to be more important than the nitrogen concentration for effective erythritol production, The optimal ratio of yeast extract to sucrose (g/g) was 20. The yield and productivity of erythritol were maximal in the medium containing 400 g/I sucrose and 20 g/I yeast extract. when dissolved oxygen in the culture was increased, the cell mass increased but the erythritol production was manimal in the range of 5 to 10% of dissolved oxygen. Under the optimal the rane of 5 to 10% of dissolved oxygen. Under the optimal culture condition of the fermentor, a final erythritol concentration of 200 gI was obtained after 120 h with a yield of 50% and the productivity was 1.67 g/I/h.   • An Immunostimulating polysaccharide was produced from the suspension culture of Angelica gigas H4, plant cells. In order to enhance the polysaccharide production by the A. gigas cell culture, medium composition and physical conditions were optimized. Schenk and Hildebrandt (SH) medium was selected as an optimal basal medium for the growth of A. gigas. The maximum cell and polysaccharide concentration obtained in SH medium were 15.8 g DCW/l and 0.85 g polysaccharide/l, respectively, at $25^{\circ}C$ under dark condition. For the enhanced polysaccharide production, a polysaccharide production medium (PPM) was established by modifying Gamborg B5 medium with optimized carbon sources, growth regulators, organic and inorganic elements. Optimal initial pH and temperature were 6.0-6.6 and $20^{\circ}C$, respectively, and the dark condition was better than the light condition. The maximum polysaccharide concentration of 1.5 g/l could be obtained through the optimization of the medium composition and physical conditions.

Mutational and Nutritional IMprovement of Tylosin Production (Tylosin 생성 우수 균주선별과 Tylosin 발효)

  • A tylosin-hyperproductive mutant of S. fradiae MNU20 was isolated among 3500 strains obtaincd from either MNNG- or UV-treated Streptomyces fradiae NRRL2702. The composition of optimal medium for tylosin production was formulated as followed as: 4 g soluble starch, 1 g glucose, 1 g corn steep liquor, 7.5 ml soy bean oil, 0.2 g KH$_{2}$PO$_{4}$, 1 g Na$_{2}$S$_{2}$O$_{3}$$\cdot $5H$_{2}$O, 2 g CaCO$_{3}$, 2 g NaCl, 0.001 g CoCl$_{2}$$\cdot $6H$_{2}$O in 1 liter of distilled water. With the optimal medium, S. fradiae MNU20 was able to produce 159 mg tylosin (g biomass)$^{-1}$, indicating that tylosin productivity of Streptomyces fradiae NRRL2702 was increased 14 times higher by mutation. When the effect of valine, succinate, and natural zeolite on tylosin production was investigated by using the optimal medium, these substances essentially enhanced tylosin production and their addition time during culture period appeared to be critical for the increase of tylosin production.

Proliferation of Embryogenic Callus of Kalopanax pictus through Suspension Culture System (현탁배양을 통한 음나무(Kalopanax pictus) 배발생 캘러스의 증식)

  • Kalopanax pictus was cultured in vitro to find out optimal condition for embryogenic cells proliferation in liquid media rapidly. Embryogenic cells were induced from leaves and petiols of Kalopanax pictus. Optimum culture medium appeared to be a 1/2MS medium supplemented with 2.0mg/L 2,4-D and 0.1mg/L BA. To find out optimal conditions, embryogenic cells were cultured some condition as different concentrations of 2,4-D, medium and sucrose. There was cultured on 1/2MS liquid medium containing different concentration of 2,4-D. When embryogenic cells were cultured on 1/2MS liquid medium supplemented with 1.0mg/L 2,4-D, cell propagation rate was higher than other concentration of 2,4-D. When embryogenic cells were cultured on different media that MS, Gambols B5, N6, White, SH medium, observed the highest multiplication rate among Gambols B5 and White medium. To find out of effect of sucrose to embryogenic cells propagation, we tested cells under different concentrations. Optimal concentration of sucrose appeared to be a basal medium added 3% sucrose. Above results suggest that optimal conditions for proliferation of embryogenic cells were established Gambols B5 and White medium added 1.0mg/L 2,4-D and 3% sucrose. There is every possibility achieving embryogenic cells proliferation via bioreactor culture system in Kalopanax pictus.

Studies on Mycelial Growth and Artificial Cultivation of Pleurotus eryngii (큰느타리버섯(Pleurotus eryngii)의 균사배양 및 인공재배에 관한 연구)

  • This study was carried out to investigate the optimal mycelial growth and an artificial cultivation conditions of Pleurotus eryngii. The optimal medium for the mycelial growth and density was MYPA medium. The optimal temperature and pH for the mycelial growth were $25^{\circ}C$ and 6.0, respectively. The modified optimal medium composition were obtained to be soluble starch 3%(w/v), malt extract 0.25%(w/v), yeast extract 0.25%(w/v) and $CaCl_2{\cdot}2H_2O$ 0.05% (w/v). From the results of experiment on the nutritional requirements, the modified optimal medium was higher than MYP medium in mycelial production and growth yield (Yx/s) of Pleurotus eryngii. The optimal sawdust species of solid culture for the mycelial growth and density was Quercus spp. The optimal concentration of additives (rice bran and wheat bran) and moisture content for the mycelial growth were about 30%(v/v) and 70%(v/v), respectively. On the other hand, the optimal concentration of additives for the production of fruiting body was 20%(v/v) of rice bran.

Statistical Optimization of Medium Components for Milk-Clotting Enzyme Production by Bacillus amyloliquefaciens D4 Using Wheat Bran-an Agro-Industry Waste

  • In this paper, two statistical methods were applied to optimize medium components to improve the production of the milk-clotting enzyme by Bacillus amyloliquefaciens D4. First, wheat bran juice, skim milk powder, and $Na_2HPO_4$ were shown to have significant effects on D4 enzyme production using the Plackett-Burman experimental design. Subsequently, an optimal medium was obtained using the Box-Behnken method, which consisted of 3.31 g/l of skim milk powder, 5.0 g/l of sucrose, 0.1 g/l of $FeSO_4{\cdot}7H_2O$, 0.1 g/l of $MgSO_4{\cdot}7H_2O$, 0.1 g/l of $MnSO_4{\cdot}2H_2O$, 0.1 g/l of $ZnSO_4{\cdot}7H_2O$, 1.52 g/l of $Na_2HPO_4$, and 172.45 g/l of wheat bran juice. With this optimal medium, the milk-clotting enzyme production was remarkably enhanced. The milk-clotting enzyme activity reached 3,326.7 SU/ml after incubation of 48 h, which was 1.76-fold higher than that of the basic medium, showing that the Plackett-Burman design and Box-Behnken response surface method are effective to optimize medium components, and B. amyloliquefaciens D4 possessed a high rennet-producing capacity in the optimal medium.

High Production of L-Ornithine by L-Citrulline Auxotroph of Breviabcterium ketoglutamicum : PART II : Production of L-Ornithine by Controlled Feeding of L-Arginine (Brevibacterium ketoglutamicum을 이용한 L-Ornithine 생산 연구 PART II : L-Arginine 제한공급에 의한 :-Ornithine 유가식 발효생산)

  • A highly productive fed-batch fermentation process was developed for the production of L-ornithine by using a new stabilized strain, Breviabcterium ketoglutamicum BK52. Fed-batch cultures with a continuous feeding of the complex medium were conducted on various operating conditions. The optimal concentration of phosphate in the complex medium was 2.1g/L. The optimal feeding rate of L-arginine was 0.028g/L/hr. The optimal feeding point of the complex medium was determined to be at 40 OD of the cell mass. The final L-ornithine concentrations within 64hrs of cultivation in 5 and 50 liter fermenters were 73g/L and 71g/L, respectively. The maximum overall L-ornithine productivity was 1.14g/L/hr which was about 2 times higher than that of the conventional fed-batch culture with intermittent feeding. The overall productivity of the fermentation system is remarkably improved by employing the optimized conditions, and it offers a significant potential for industrial application.
