PAMAM Dendrimer Conjugated with N-terminal Oligopeptides of Mouse Fibroblast Growth Factor 3 as a Novel Gene Carrier

  • In this study, we introduced the RRLR oligopeptide sequences on the surface of polyamidoamine (PAMAM) dendrimer and characterized the physical properties and gene carrier activity of the novel polymer using HEK 293, NIH3T3, and HeLa cells. The RRLR peptide sequences were derived from a mouse fibroblast growth factor 3 (FGF3) protein containing a bipartite NLS motif. The entire sequence of FGF3 is RLRRDAGGRGGVYEHLGGAPRRRK and it has two functional sequences RLRR and RRRK at N-terminus and C-terminus, respectively. In particular, PAMAM G4-RRLR conferred enhanced transfection efficiency and lower cytotoxicity compared with those of PEI 25 kDa, PAMAM G4-R, and PAMAM G4 in various cell lines. These results suggest that the introduction of N-terminal oligopeptides of FGF3 on the surface of PAMAM holds promise as an effective non-viral gene delivery carrier for gene therapy.

  • The conformational transitions of oligopeptide -(HPPHPPP)$_n$- (H: hydrophobic amino acid, P: polar amino acid) which becomes cross linked-dimer are detected at various temperatures, pHs, ionic strengths, and the concentrations of denaturant. In this study, the transitions of oligopeptide due to denaturant, and those due to temperature are theoretically studied. Oligopeptide 20R as chain-dimer and guanidum-HCl as denaturant are used(20R, which contains 10 interchain and /or 10 intrachain electrostatic repulsions). Alpha helix-coil transitions by denaturant are very steep. This shows that the denaturations have transition states of, presumably all helical forms and random coils. The transitions by temperature are smoother than those by the concentration of denaturant. At low temperature the oligopetides which contain long helices exist more than those which contain short helices. As temperature rises, the mole fractions of the partially denatured oligopeptides increase. So the partially denatured oligopeptides are widely distributed at the transition temperature.

  • Molecular dynamics of nanosecond timescale could provide a significant result for the structure analysis of oligopeptides. The most surprising was the fact that Chou-Fasman parameters which have no direct relationship with nanosecond molecular dynamics could implicate the result to a certain extent. A novel parameter termed X %-stickiness, which is another measure of compactness of a molecule, was first introduced effectively.

Antihypertensive Angiotensin I-Converting Enzyme Inhibitory Activity and Antioxidant Activity of Vitis hybrid-Vitis coignetiae Red Wine Made with Saccharomyces cerevisiae

  • A Vitis hybrid-Vitis coignetiae red wine was vinified by fermentation of a mixture of a Vitis hybrid.Vitis coignetiae must with Saccharomyces cerevisiae KCTC 7904 at $25^{\circ}C$ for 10 days. The Vitis hybrid-Vitis coignetiae red wine showed high antihypertensive angiotensin I-converting enzyme (ACE) inhibitory activity (67.8%) and antioxidant activity (76.7%). The antihypertensive ACE inhibitor in the Vitis hybrid-Vitis coignetiae red wine was partially purified by solid phase extraction chromatography, and its ACE inhibitory activity yielded an $IC_{50}$ of 1.8 mg/mL. Six kinds of oligopeptides, including five new kinds, were contained in the partially purified ACE inhibitor fraction from the red wine after 10 days of fermentation. Antioxidant activity decreased significantly from 76.7% to 40.5% when the post-fermentation period was prolonged to 30 days.

  • Aster species has been used in traditional chinese medicine for treatment of a bruies and asthma. On reviewing the literatures of this species, monoterpene glycosides, diterpenoids, triterpene glycosides, cyclic pentapeptides, oligopeptides and flavonoids 1) were isolated and some pharmacological activites were investigated 2). Comparison of Extractive Nitrogenous Constituents in Cultured and Wild Olive Flounder (Paralichthys olivaceus) Muscle (양식 및 자연산 넙치의 함질소엑스성분 비교)

  • The nitrogenous compounds in the muscle extracts of cultured and wild olive flounder, Paralichthys olivaceous, were analyzed. The analyzed coumpounds were extractive nitrogen, free amino acids, oligopeptides, ATP and its related ompounds, quaternary ammonium bases, and guanidino compounds. The distribution pattern of these compounds in cultured and wild fish was found to be very similar. Although the ATP and its related compounds and creatine in the muscle of cultured fish were slightly abundant than those in the muscle of wild one, the extractive nitrogen, total free amino acid, oligopeptides, and TMAO were found to be slightly rich in the muscle of wild fish than those in the muscle of cultrued one. The moisture content of cultured fish was relatively lower but the protein and fat contents of cultured one were higher than those of wild fish. However the differences in the proximate composition, extractive nitrogen and nitrogenous compounds between two fishes were not significantly different.

Comparison of Seasonal and Regional Variation in Extractive Nitrogenous. Constituents of the Raw Anchovy (Engraulis japonica) (생 멸치의 함질소 엑스성분조성 및 지역과 계절에 따른 변동)

  • In order to investigate the composition, seasonal and regional variation of extractive nitrogenous components in the raw anchovy (Engraulis japonica) harvested from June 1991 to June 1992 at Nam-hae and Ki-jang in the southern coast of Korea, the contents of extractive nitrogen (EN), free amino acids (FAA), oligopeptides (OP), ATP and its related compounds (ARC), quaternary ammonium bases, and guanidino compounds were measured. The EN contents of raw anchovy were $444{\~}773\;mg/100\;g$. Thirty-four kinds of FAA were found and their total amount were $1,049{\~}2,079\;mg/100\;g$. Histidine, taurine, alanine, leucine and glutamic acid were the major free amino acids in the anchovy extracts. The amount of ARC was average $5.02\;{\mu}mol/g$, and IMP was the major compound among them, who and creatine contents in the extracts or raw anchovy were average 164 and 229 mg/100 g, respectively. The contents of glycinebetaine,${\brta}-alaninebetaine,\;{\gamma}-burobetaine$, homarine, TMA, and creatinine were extremely small. As for the regional and seasonal disparities, the Nam-hae samples contained more EN, FAA, OP, betaines and creatine than the Ki-jang samples. On the other hand the Ki-jang samples contained more ARC and TMAO. The spring samples contained more EN, FAA, OP, ARC and betaines than the autumn samples. On the other side the autumn samples contained more TAMO and creatine.

Extractive Nitrogenous Constituents of Toha (Caridina denticulata denticulata, Freshwater Shrimp) and Jeotsaeu (Acetes japonicus , Seawater Shrimp) (토하(土蝦) 및 젓새우의 함질소(含窒素) 엑스성분(成分)에 관(關)한 연구(硏究))

  • The extracts of toha, a kind of freshwater shrimp (Caridina denticulata denticulata) being cultivated in Naju district, Chonnam of Korea and jeotsaeu, a kind of seawater shrimp (Acetes japonicus) being caught in Shinan district, Chonnam of Korea, were analyzed for the taste constituents-extractive nitrogen, free amino acids, oligopeptides, nucleotides and related compounds, quaternary ammonium bases, and guanidino compounds-using specimens collected from December in 1994 to November in 1995. The content of extractive nitrogen. free amino acids, oligopeptides, nucleotides and related compounds in toha extract was higher than that in jeotsaeu extract. On the contrary, the amount of homarine, trimethylamine oxide and trimethlamine in toha extract was lower than that in jeotsaeu extract. But in case of creatine and creatine both species showed almost the same level. Arginine, lysine, glutamine, glutamic acid, phenylalanine, leucine, alanine, asparagine and glycine were the major amino acids in toha extract, while arginine, glutamic acid, proline, glutamine, lysine, leucine, glycine, alanine and taurine were the major amino acids in jeotsaeu extract. Methionine, glutamine, valine were almost the same level between the two species. As for nucleotides and related compound, AMP was the principal constituent in toha extract, and inosine was the principal constituent in jeotsaeu extract. In conclusion, the content of major nitrogenous constituents in toha extract was more abundant than that in jeotsaeu extract. So toha was more excellent raw material than jeotsaeu.

Physiological Activities and Amino Acid Compositions of Korean Dried Laver Porphyra Products (국내산 마른 김(Porphyra)의 생리활성 및 아미노산 조성 비교)

  • The physiological activities and amino acid compositions of dried lavers of Porphyra from the Jangheung, Seochen and Haenam coasts were determined. The ethanol extract of the Haenam dried laver exhibited the greatest 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity (56.1%), ferric-reducing antioxidant potential (FRAP) value(86.5 mM) and total polyphenol content (3.51 ppm) of these dried laver products. In addition, there was a strong positive correlation between the antioxidative activity and total polyphenol content of the laver products, suggesting that polyphenol compounds contribute to the antioxidant capacity in the ethanol extract of dried laver. However, there was no difference among the extracts in terms of cell proliferation activity. Fourteen combined amino acids were identified in oligopeptides from the dried laver products. Of the three extracts, that of the Haenam dried laver contained the highest levels of both free and total amino acids including alanine, glutamic acid, taurine, phosphoserine, and aspartic acid.

Inhibitory Effects of Ricinus communis on HIV-1 Essential Enzymes in vitro and Prediction of Inhibitory Factor Using QSAR in silico (구조활성상관(QSAR)에 의한 피마엽 추출물의 HIV-1 효소억제활성인자 예측)

  • Objectives : For the purpose of developing new anti-HIV agents from natural sources, the extracts of Ricinus communis were tested for their inhibitory effects on essential enzymes reverse transcriptase (RT), protease and alpha-glucosidase. Inhibition activity of major compounds of Ricinus communis were predicted from quantitative structure activity relationships (QSAR) in silico. Methods and Results : In the anti-HIV-1 RT using enzyme-linked oligonucleotide sorbent assay (ELOSA) method, water and methanol extracts (100ug/ml) of Ricinus communis showed strong activity of 94.2% and 82.7%, respectively. In the HIV-1 protease and alpha-glucosidase inhibition assay, neither water nor methanol extracts of Ricinus communis inhibited the activity of the enzyme to cleave any substrates as oligopeptides and oligosaccharides. Conclusions : We found that for these samples it is possible that the inhibition of the RT in vitro is due to the secondary metabolites of Ricinus communis such as ricinine and quercetin. It would beof great interest to identify the compounds which are responsible for this inhibition, since all therapeutically useful agents up to date are RT inhibitors.
