The Role of Autophagy in Apoptosis Induced by Water Extract of Platycodonis Radix in H460 Human Lung Cancer Cells (H460 인체 폐암세포에서 길경 물 추출물에 의해 유도된 세포사멸에서 자가포식의 역할)

  • Objectives : Recent studies have suggested that Platycodonis Radix has various pharmacological effects such as anti-cancer, antioxidant, anti-asthma, anti-diabetes, anti-obesity, hepatoprotective, and cardiovascular protection effects. The aim of this study was to investigate the role of water extract of Platycodonis Radix (WPR)-induced autophagy in H460 human lung cancer cells. Methods : H460 cells were treated with WPR and cell viability was calculated by an MTT assay. To evaluate changes in apoptosis- and autophagy-related genes, Western blotting was performed. Two kinds of autophagy inhibitors, 3-Methyladenine (3-MA) and bafilomycin A1, were pretreated to confirm the role of WPR-induced autophagy. Results : WPR reduced the viability of H460 cells in a treatment concentration-dependent manner, which was associated with induction of apoptosis. It was also confirmed that WPR induced autophagy based on the formation of specific intracellular vacuoles and changes in the expression of autophagy-related genes. Interestingly, pretreatment with 3-MA and bafilomycin A1 increased WPR-induced cytotoxicity and apoptosis. Conclusions : WPR induced autophagy at low concentrations and early stages of treatment, but promoted apoptosis at high concentrations and late stages. Moreover, WPR-induced autophagy had a cytoprotective role in H460 cells.

  • Objectives : This paper focuses on recent research related to the relationship between 'Pizhuweiwei(脾主爲衛)' and intestinal flora, and suggests Korean Medical treatment methods that can treat and prevent diseases related to Spleen Deficiency(脾虛) such as ulcerative colitis, atrophic gastritis, diabetes, and obesity that is prevalent today. Methods : This study summarizes recent research results based on various literature on the relationship between the spleen and intestinal flora. Results : Polysaccharides contained in Jianpi herbs(健脾藥) have the effect of increasing beneficial bacteria and maintaining the diversity of intestinal microorganisms to improve intestinal function, managing intestinal metabolites to improve the body's immune function, and regulating the intestinal immune defense system. Therefore, based on the theory of 'Pizhuweiwei(脾主爲衛)', if the symptoms are treated through the spleen with Spleen-strengthening herbal medicinals, it could provide a substantial starting point for improving immunity. Conclusions : Polysaccharides contained in Jianpi herbs(健脾藥) could be considered as potential probiotics based on research findings which show that polysaccharides can regulate the intestinal flora and strengthen weak spleen, playing an important role.

Prevalence and Risk Factors for falls of Older Adults with Dementia in Korea: Based on the Korean Longitudinal Study of Aging (우리나라 치매노인의 낙상 유병률과 위험요인: 고령화연구패널조사 결과를 이용하여)

  • This study is a data analysis study to identify the factors influencing the prevalence and risk factors for falls of older adults with dementia in Korea. Using the data of the 7th Aging Research Panel in 2018, 119 people were enrolled. We used response data on the dementia-related factors that is the duration of dementia and whether or not activity of daily living was restricted due to dementia. For comorbid diseases, data on hypertension, diabetes, and obesity were used. For statistical analysis of the collected data, logistic regression analysis was performed using SPSS statistics 22.0. Dementia-related factors and comorbidities of the analyzed subjects had a significant effect on the falling index. In particular, it was found that the influence was greatest in the order of obesity, diabetes, hypertension, daily life restrictions due to dementia, and the duration of dementia. This study is meant to identify factors that should be prioritized in the composition of a fall prevention program for the elderly with dementia. Based on the findings of this study, strategies for preventing falls due to the duration of dementia and limiting daily life, intensive management of high-risk groups for falls due to comorbid diseases, and training in the use of safety aids such as walking aids will be required in the care of the elderly with dementia,

Analysis of the Relationship between Abdominal Ultrasound Based Kidney Stones, Obesity and Metabolic Syndrome (복부 초음파 검사 기반 신장결석과 비만 및 대사증후군 관련성 분석)

  • Kidney stones are a common disease with an annual prevalence of about 30,000 people in Korea, and are deeply related to an increase in chronic kidney disease, high blood pressure, coronary artery disease, metabolic syndrome, type 2 diabetes, hyperlipidemia, and abdominal obesity. Therefore, in this study, 135 examinees who visited a general hospital in Dalseong-gun from May 2019 to June 2020 for a medical examination were examined. The relationship between kidney stones and factors related to obesity and metabolic syndrome were found in abdominal ultrasound. I tried to find out the relevance. As a result of the study, the risk of kidney stones in the abnormal group was increased by 4.255 times compared to the normal group in total cholesterol factor, and the risk of kidney stones in the abnormal group was increased by 2.072 times compared to the normal group in the low-density lipoprotein cholesterol factor. Total cholesterol factor and low-density lipoprotein cholesterol It was found that the factor affects the prevalence of kidney stones and metabolic syndrome. Since the risk of kidney stones is related to total cholesterol and low-density lipoprotein cholesterol factors, active attention should be paid to preventive purposes through health check-ups.

Effect of fermented soybean curd residue (FSCR; SCR-meju) by aspergillus oryzae on the anti-obesity and lipids improvement (Aspergillus Oryzae로 발효시킨 비지메주의 항비만 및 지질개선 효과)

  • In this study, we designed to confirm the dietary effect of anti-obesity of fermented soybean curd residue (FSCR; SCR-Meju; Biji-meju) by A. oryzae, which is well known as a Korean traditional meju microbe. We observed that body weight gain, serum and hepatic lipid profile, as well as the activity of ROS generating enzyme and ROS scavenging enzyme in high-fat diet induced obese mice fed experimental diet (SCR and SCR-meju). Body weight gain and epididymal fat weight of HC (high-fat diet control) was markedly higher than that of NC (Normal control). Conversely, body weight gain and epididymal fat weight of the SCR (Biji) and SCR-meju (Biji-meju) group was significantly lower than that of HC; these of the SCR-meju group was lower than that of the SCR group. Furthermore, serum TG and total-cholesterol and LDL-cholesterol contents of SCR and SCR-meju groups were lower than that of HC, and HDL-cholesterol level of the SCR-meju group was significantly higher than that of HC. In conclusion, although precise mechanisms of the antiobese effects of SCR-meju in this study are unknown, the present study provides an experimental evidence that SCR-meju may prevent obesity and obesity related metabolic syndromes, such as hyperlipidemia, hypertension and diabetes, and liver disease by high-fat diet. Nevertheless, further study in this filed will be needed.

Obesity as associated factors for Insulin, Fasting blood sugar, and Hemoglobin A1c in Korean adults (한국 성인의 비만이 인슐린, 공복혈당 및 당화혈색소에 미치는 영향)

  • The purpose of this study was to investigate the effects of body mass index(BMI) on insulin. fasting blood sugar(FBS), and hemoglobin A1c(HbA1c). This study analyzed the data of 2015, The Sixth KNHANES, and it was included to 4554 subject over 20 years old. Data were analyzed using descriptive statistics, t-test, ANOVA, Pearson correlation coefficient and stepwise multiple regression analysis(SPSS 25.0). The major findings, BMI was positively correlated with insulin(r=0.397, p<0.001), FBS(r=0.241, p<0.001), and HbA1c(r=0.237, p<0.001). Factors affecting insulin are overweight(${\beta}=.130$, t=8.193, p<0.001), obesity(${\beta}=.390$, t=24.390, p<0.001) and age. the explanatory power is 13.1% (F=53.584, p<0.001). Factors affecting FBS were overweight(${\beta}=.072$, t=4.478, p<0.001), obesity(${\beta}=.186$, t= 11.485, p<0.001), age, the explanatory power was 10.6% (F=42.351, p<0.001). Factors affecting HbA1c were overweight(${\beta}=.059$, t=3.712, p<0.001), obesity(${\beta}=.190$, t=11.895, p<0.001) age and sex, the explanatory power is 14.1% (F=58.134, p<0.001). Based on this study result, diabetes mellitus prevention and management programs should include interventions related to obesity.

Pathophysiological Functions of Deubiquitinating Enzymes in Obesity and Related Metabolic Diseases (탈유비퀴틴화 효소 DUBs의 비만 및 대사 관련 질환에서 병태생리학적 기능)

  • Ubiquitin signaling regulates virtually all aspects of eukaryotic biology and dynamic processes in which protein substrates are modified by ubiquitin. To regulate these processes, deubiquitinating enzymes (DUBs) cleave ubiquitin or ubiquitin-like proteins from these substrates. DUBs have been implicated in the pathogenesis of cancer, leading to the development of increasing numbers of small-molecule DUB inhibitors. On the other hand, recent studies have focused on the function of DUBs in metabolic diseases such as obesity, diabetes, and fatty liver diseases. DUBs play a positive or negative role in the progression and development of metabolic diseases. Their involvement in cell pathology and regulation of major transcription factors in metabolic syndrome has been examined in vitro and in animal and human biopsies. UCH, USP7, and USP19 were linked to adipocyte differentiation, body weight gain, and insulin resistance in genetic or diet-induced obesity. CYLD, USP4, and USP18 were found to be closely associated with fatty liver diseases. In addition, these liver diseases were accompanied by body weight change in certain cases. Collectively, in this review, we discuss the current understanding of DUBs in metabolic diseases with a particular focus on obesity. We also provide basic knowledge and regulatory mechanisms of DUBs and suggest these enzymes as therapeutic targets for metabolic diseases.

A Study on the Preventive Attitudes and Health Behavior of Life-style Related Diseases in College Students (일부 대학생들의 생활습관병에 대한 예방태도와 건강행동)

  • Objectives: An investigative effort was made to provide basic data upon which effective public health education can be constructed for schools. Methods: The research was made by delivering self-administered questionnaires to 536 men and women with unknown names from a national university during 1-month period on September of 2004. The questionnaire items included preventive attitudes, interests, anxiety, health behavior, consciousness, etc. centering upon life-style related diseases. Results and Conclusions: The preventive attitudes against life-style related diseases was of a higher level in female than male students, The smaller number of students had knowledge of the definition of life-style related diseases, willingness to learn, and gathered information on them. The number of students who had anxiety about them was also small and as low as 54.3% of students were willing to undertake periodic health examination. A great variation ranging from 4.7% to 81.7% was shown in the accuracy rate of answers on the questions asking about three leading causes of death, overconsumption of dietary salt, obesity, diabetes, hypercholesterolemia and colon cancer. With regard to health behaviors, a smaller number of students were involved in health behaviors. On the other hand, $25.7{\sim}35.1%$ of students had everyday exercises and breakfasts, 65.5% had no tobacco use. The study results showed that the survey subjects had deficient knowledge as well as a low level of preventive attitudes and interests against life-style related diseases. Moreover, their health behavior and health consciousness were undesirable apart from their low rates of taking periodic health examinations. Accordingly, the college students should be encouraged to take health education and to make more effective the existing health education in the university.

A Survey on the Consumer Attitude Toward Health Food in Korea (I) -Consumer Perception on Health and Food Habit- (건강식품에 대한 소비자 인식 연구 (I) -건강과 식습관에 관한 소비자 의식구조-)

  • The consumer perception on health and food habit, the experience of health food use and the discrimination between health food and drug of Korean consumer were surveyed by using a questionnaire containing 15 items in order to obtain the basic data for the assessment of the benefit and risk of health foods in Korea. A total of 1,000 people over 20 years of age living in Seoul and the vicinities were interviewed and asked to fill out the questionnaire during the period from the October 1995 to the February 1996. Among the 882 answers collected, 23 was incomplete data, and 859 answers were used for the statistical analysis by using SAS program. The perception of Korean consumer on health and food habit indicated that food habit was considered the most important factor for the maintenance of health, as appeared in 39.8% of the subjects, among which 93.9 % believed that food habit could cause disease, and 97.1% believed that disease could be cured by changing food habit. The most worried disease was cancer (30.6%), degenerative diseases (14.1%), diseases by accident (12.6%) and obesity (10.0%). The disease which likely to be caused by food habit was diabetes (35.6%), obesity (22.4%), high blood pressure (12.8%), constipation (12.7%) and cancer (7.9%). The disease which was believed to be cured by changing food habit was diabetes (40.1%), obesity (25.9%), constipation (16.5%), high blood pressure (7.4%) and cancer (3.3%). It appeared that the people had a perception that food habit was highly related with diabetes and obesity, but less with cancer which was mostly worried.

Nutritional Status and Health Habit of College Students by Body Mass Index (수도권 남녀대학신입생의 체질량지수에 따른 건강행동과 영양상태)

  • It is important for young adults to establish good dietary behavior for healthy life. The prevalence of obesity in college students has increased gradually. The obesity is one of the most prevalent health problems in S. Korea. Moreover, obesity is closely related with the disease such as cardiovascular disease, diabetes mellitus. The purpose of this study was to evaluate health practice, serum components and nutrient intake of the college students according to Body Mass Index (BMI) level. 400 male and female freshmen students in 4 year university located near to Seoul metropolitan area were surveyed respectively through the health check-up procedure for college entrance in February. Survey was carried out for them by questionnaires, including a 24-hour dietary recall and anthropometric measurements. Blood samples were obtained while fasting. Nutrient intake was expressed by DDS(Dietary Diversity Score by 5 food groups) and DVS(Dietary Variety Score). Nutrition level was analyzed by Can-Pro (Korea Nutrition Association). And for the quantity intake, percentage ratio against daily requirement(by KDRIs) and MAR(Mean Adequacy Ratio) were calculated. Underweight group($BMI<18.5kg/m^2$), normal weight group($18.5) and overweight group($BMI<23.0 kg/m^2$) were 18.5%, 61.2% and 19.3% respectively. Nutrient intake levels and food intake status were not significantly different among three groups. The mean nutrient adequacy ratio (MAR) was not significantly different by BMI status. Nutrient intakes of calcium, iron, and riboflavin were especially low for all the students. Overweight group showed high blood pressure(120.9/79.3 mmHg) compared with the underweight group (116.4/ 75.8mmHg) but the difference was not significant. The serum triglyceride level in overweight group (94,0mg%) was higher than that in normal (70.9mg%) or underweight group(70.8mg%)(p<0.05). HDL-cholesterol level in overweight (43.8 mg%) group was not significantly different from that in underweight group (45.9mg%). BMI was positively correlated with blood pressure, hemoglobin, and the intake of total fat and cholesterol. But it was negatively correlated with HDL-cholesterol level. With these results overweight group is closely related with the cardiovascular disease risks, such as high blood pressure and high triglycerides and cholesterol. Thus, a health intervention program including weight control is required for young adults.
