Recent Advances in Biotechnology Applications to Aquaculture

  • Biotechnological research and development are moving at a very fast rate. The subject has assumed greatest importance in recent years in the development of agriculture and human health. The science of biotechnology has endowed us with new tools and tremendous power to create novel genes and genotypes of plants, animals and fish. The application of biotechnology in the fisheries sector is a relatively recent practice. Nevertheless, it is a promising area to enhance fish production. The increased application of biotechnological tools can certainly revolutionise our fish farming besides its role in biodiversity conservation. The paper briefly reports the current progress and thrust areas in the use of synthetic hormones in fish breeding, production of monosex, uniparental and polyploid individuals, molecular biology and transgenesis, biotechnology in aquaculture nutrition and health management, gene banking and the marine natural products.

Detection of Taurine in Basidiomycetes

  • Taurine is one of the essential amino acids for humans and many of mammals. It is produced and contained in fleshes, shells, plants and algae, but has never been found in fungi. We examined six mushrooms for taurine and detected taurine in five of them. Taurine was determined by an automated amino acid analyzer using ion-exchange chromatography, being eluted between phosphoserine as a distinct peak on the chromatogram. Fruit bodies of Flammulina velutipes contained $83\mu$moles/100g fresh wt. the highest level among them, Agaricus bisporos $65\mu$moles, Lentinus edodes $49\mu$moles, Pleurotus ostreatus $9\mu$moles, and Auricularia auricula-judae $20\mu$moles. Taurine was not detected in Ganoderma lucidum. As far as fungi are concerned, this is the first report of the detection of taurine in Basidiomyces.

Analysis of Nitrate Contents of Some Vegetables Grown in Korea (국내 채소류의 질산염 함량 분석)

  • Nitrate is taken up from the soil by plants for protein synthesis and present in vegetables as a natural component and/or contaminant. The objective of this study was to estimate nitrate(NO3-) contents of some vegetables(Chinese cabbage, radish, lettuce, spinach) which were produced in Korea and to provide a scientific basis for the evaluation of risk to public health arising from dietary exposure to nitrate. A total of 400 samples were analysed for nitrate contents using our ion chromatography. From the results, in general, nitrate levels in vegetables produced by 2 harvest seasons were not different. The minimum, maximum and mean values of nitrate were 311, 5522 and 2788 for spinach; 542, 4484 and 2287 for lettuce; 273, 4151 and 1551 for radish; 362, 3015 and 1498(mg/kg) for Chinese cabbage. Nitrate contents of vegetables grown in Korea were similar to those of vegetables grown in other countries.

Detection of Irradiated Astragalus membranaeus Bunge and Havenia duzcis Thumb Using DNA Comet Assay

  • Ionizing radiation can be used to sanitize herbs contaminated by various microorganisms. However, health concerns related to irradiation damage to complex molecules in plants necessitate that methods be developed to monitor such damage. To elucidate DNA damage of herbs caused by irradiation, the DNA comet assay was used for Astragalus membranaceus Bunge and Havenia dulcis Thumb, irradiated at 1, 5, 7, and 10 kGy. With increasing irradiation doses, the tails of comets became longer with average tail length increasing from 17 (non-irradiated) to 124 (10 kGy) $\mu$m in Astragalus membranaceus Bunge. Above 7 kGy, some of the tails were separated from the heads of comets. Distribution patterns of the tail length of In comets selected randomly in the irradiated herbs were analyzed to quantify the DNA damage. These results clearly suggest that the DNA comet assay is an effective and inexpensive tool for the detection of irradiation damage to DNA in herbs.

  • By the 505 chromotest which utilized Escherichia bolt PQ 37, Korean medicinal plants had been screened to Investigate the antimutagenic effect to aflatoxin B1(AFBl). Ikmocho(IMC, Leonurus sibiricus L.) was extracted with hot water. The extract was not found to be mutagenic in the Salmonella mutation test with or without metabolic activation, and the extract was showed to possess the antimutagenic properties towards AFB1-induced metation. The mutagenicity of AFB1 was inhibited by methanol soluble fracstion (IMC-MS) in dose-dependent. However, water-soluble fraction exhibited comutagenic activity. The greatest inhibitory effect of IMC-MS on AFB1 mutagenicity occurred when IMC-MS was first incubated, AFB1 followed by a second incubation with the cells and 59 mixture. Also lower inhibition was occurred when S9 mixtures were first incubated, with IMC-MS followed by a second incubation with AFBI. The results of the sequential incubation study support the probability that one mechanism of inhibition could involve the formation of chemical complex between IMC-MS and AFB1 rather than deactivation of S9 enzyme.

  • The effects of Artemisia princeps var orientalis(mugwort), Circium japonicum var ussuriense(Unggungqui) on serum lipid componets in rats was evaluated. Thirty rats divided into 5 experimental groups were fed with the diet containing 1% cholesterol, 0.25% sodium cholate, 10% coconut oil and 5% lard(control diet), supplemetned with 5% plant powder or its water soluble extract(experimental diet) for 4 weeks. Proximate analysis data and compositions of ash, dietary fiber and fatty acids in plants were presented. The concentrations of the total cholesterol was significantly lower in Ungungqui powder, Ungungqui extract and mugwort powder diet groups than the control. Serum HDL-cholesterol level was significantly higher in Unggunqui powder and Unggunqui extract diet group than the control. The concentration of LDL was significantly lower in Unggungqui power or extract diet group compared to the control. The values of VLDL and chylomicron were comparatively lower in Unggungqui power and mugwort powder diet group than in the other diet groups. The concentration of seum triglyceride and phospholipid was lower in Unggungqui powder diet group than in the control.

Fatty Acid Composition of Domestic Seed Lipids (한국산 종자 지방질의 지방산 조성)

  • The seeds of 16 domestic plants were studied for their moisture, total lipids and fatty acid composition. Of the 16 seeds, chestnut, corn, mungbean and ginko nut yielded less than 9% by weight of total lipids compared to others that gave 20-73%. The identified fatty acids from the seed lipids ranged from lauric acid (12:0) to lignoceric acid(24:0). It was intended in this study to classify the seed lipids according to their major fatty acids: Group t-Oleic acid; Group 2-0leic acid and linoleic acid; Group 3-linoleic acid; Group 4-linolenic acid ; Group 5-erucic acid ; Group 6-ricinoleic acid. The saturated fatty acid content of mungbean (33%) was the highest among the seed lipids studied. The highest value for the P/S fatty acid ratio(10) was in perilla.

  • Five flavonoids isolated from the ethyl acetate fraction of Cedrela sinensis A. Juss. were identified by high performance liquid chromatography. Separation was achieved by reversed phase chromatography on ${\mu}-bondapak$ C18 column with isocratic elution method. The content of the major flavonoid, quercitrin was about 9.48%(w/w) and 37.06%(w/w) for the methanol extract and ethyl acetate fraction, respectively.

  • To develop a new anti-wrinkle agent from medicinal plants, this study investigated the optimal conditions for extracting elastase inhibitor from Rheum undulatum L. Maximal extraction occurred by using 70% methanol at $50^{\circ}C$ for 24 hr; the elastase inhibitory activity was 60.4%($IC_{50}:6.7{\times}10^3{\mu}g/m{\ell}$). Systematic solvent extraction, thin layer chromatography, silica gel column chromatography, Sephadex LH-20 column chromatography, and reverse-phase high performance liquid chromatography were used in the partial purification of the elastase inhibitor. The compound was soluble in dimethylsulfoxide, methanol, and ethanol, and had maximum absorption spectra at 231.5 nm and 275.5 nm.
