Influence of Dietary Protein Source and Level on Lead Accumulation in Rat (단백질 급원과 수준을 달리한 식이가 흰쥐의 납축적에 미치는 영향)

  • In this experiment forty-eight Sprague Dawley male rats were chosen and used in order to measure the growth rates and to see the effects of lead acumulation in their organs resulting from variously controlled lead protein diet. Protein sources were casein and isolated soyprotein (ISP), and each source was divided into three groups : 7% low protein [LP], 20% standard protein (SP) and 40% high protein (HP) groups. The six experimental groups were given lead acetate(25 mg/kg B.W.) and six control groups were given sodium chloride by oral administration 6 times a week for weeks. The results from this experiment were summeraized as following ; 1) Food consumption, weight gain, organ weight and food efficiency ; Lead acetate administration with protein source had no effects on food consumption, weight gain and organ weight . By their different levels of protein, food consumption of LP group was less the that of SP and HP groups after 3 weeks, weight gain of LP group was less than that of SP and HO groups after 1 weeks. The organ weight in LP group was significantly lower than SP and HP groups except teeth and adrenal s. Effect of lead acetate administration on food efficiency have significantly lower in LP-ISP diet and HP -casein diet than other groups only first week. By their different levels LP group showed significantly lower than SP group until 3 weeks. 2) Hematopoietic effect ; The hematopoieteic effect was not influencec by lead acdtate administration and protein source. But the LP group showed a significantly lowe hematopoietic effect than the SP, HP, groups. 3) Accumulation of lead in the liver, kidney, teeth by protein source showed no significantly differences. Accumulation of lead in blood, heart of LP group, spleen of LP and HP groups. femur of SP and HP groups fed with casein diet groups were significantly higher than fed with ISP diet groups. By their different levels of group showed generally higher than SP and HP groups. But accumulation of lead in teeth of HP group was high also.

Glycolytic and oxidative muscles under acute glucose supplementation differ in their metabolic responses to fatty acyl-CoA synthetase gene suppression

  • Purpose: Skeletal muscles display significant heterogeneity in metabolic responses, owing to the composition of metabolically distinct fiber types. Recently, numerous studies have reported that in skeletal muscles, suppression of genes related to fatty acid channeling alters the triacylglycerol (TAG) synthesis and switches the energy substrates. However, such responses may differ, depending on the type of muscle fiber. Hence, we conducted in vitro and animal studies to compare the metabolic responses of different types of skeletal muscle fibers to the deficiency of fatty acyl-CoA synthetase (Acsl)6, one of the main fatty acid-activating enzymes. Methods: Differentiated skeletal myotubes were transfected with selected Acsl6 short interfering RNA (siRNA), and C57BL/6J mice were subjected to siRNA to induce Acsl6 deficiency. TAG accumulation and expression levels of insulin signaling proteins in response to acute glucose supplementation were measured in immortalized cell-based skeletal myotubes, oxidative muscles (OM), and glycolytic muscles (GM) derived from the animals. Results: Under conditions of high glucose supplementation, suppression of the Acsl6 gene resulted in decreased TAG and glycogen synthesis in the C2C12 skeletal myotubes. The expression of Glut4, a glucose transporter, was similarly downregulated. In the animal study, the level of TAG accumulation in OM was higher than levels determined in GM. However, a similar decrease in TAG accumulation was obtained in the two muscle types in response to Acsl6 suppression. Moreover, Acsl6 suppression enhanced the phosphorylation of insulin signaling proteins (Foxo-1, mTORc-1) only in GM, while no such changes were observed in OM. In addition, the induction ratio of phosphorylated proteins in response to glucose or Acsl6 suppression was significantly higher in GM than in OM. Conclusion: The results of this study demonstrate that Acsl6 differentially regulates the energy metabolism of skeletal muscles in response to glucose supplementation, thereby indicating that the fiber type or fiber composition of mixed muscles may skew the results of metabolic studies.

  • Vegetables and fruits purchased from several markets in Seoul from July to October in 1977 were analyzed to know the level of nitrate-and nitrite-nitrogen accumulation in relation to a public health. Radishes and chinese cabbages utilized mainly as pickled vegetables in Korea resulted in the highest concentration of nitrate-nitrogen. Some of the levels for radishes and chinese cabbages were notably high and exceeded a recommended upper limit of 300 ppm $No_{3}-N$, and thus these levels would render these samples unsafe for use. The levels in some of vegetables other than radish and chinese cabbage, e.g., spinach, lettuce, green onion, cabbage were relatively high and considered to be unsafe for use in feeding infants, where as those of green pepper, bean sprouts and parsely were very low and safe. And also the levells in fruits were very low and safe. Nitrite-nitrogen contents in all tested vegetables and fruits ranged to trace and appeared not to be accumulated in fresh vegetables and fruits. Stems and roots of radishes and chinese cabbages accumulated approximately 2 fold more nitrate-nitrogen than leaves in 5 samples of each vegetable tested.

  • This study was carried out to evaluate the effect of Ganoderma lueuidum (GL) water extract on fat accumulation in the fatted rats Induced by the oral high fat administration for six weeks. Total cholesterol, neutral fat and epididymal fat pad of GL groups were lower than the control group. On the other hand, high density lipoprotein(HDL) -cholesterol was somewhat higher in GL than In the control group. In addition, the same result as the above case was obtained when examining adipose for liver and epididymal fat components. During the feeding experiment, level of low density lipoprotein(LDL) and very low density lipoprotein (VLDL) increased while HDL was decreased in the control group as compared with normal group. The decree of LDL and VLDL was decreased, but HDL was increased in GL group as compared with the control group. Experiment group showed the Increased excretion as compared with the control group, in the excretion. Based on the above results, It was shown that it is possible to improve fat accumulation induced by high fat dietary through using the oral administration of Ganoderma lueidum boiling extract solutions.

Association of daily carbohydrate intake with intermuscular adipose tissue in Korean individuals with obesity: a cross-sectional study

  • BACKGROUND/OBJECTIVES: The prevalence of obesity, a worldwide pandemic, has been increasing steadily in Korea. Reports have shown that increased intermuscular adipose tissue (IMAT) is associated with an increased risk of cardiovascular disease, independent of body mass index. However, the relationship between dietary intake and IMAT accumulation in the Korean population remains undetermined. The objective of this study was to evaluate regional fat compartments using advanced magnetic resonance imaging (MRI) techniques. We also aimed to investigate the association between IMAT amounts and dietary intake, including carbohydrate intake, among Korean individuals with obesity. SUBJECTS/METHODS: This cross-sectional study, performed at a medical center in South Korea, recruited 35 individuals with obesity (15 men and 20 women) and classified them into 2 groups according to sex. Anthropometry was performed, and body fat distribution was measured using MRI. Blood parameters, including glucose and lipid profiles, were analyzed using commercial kits. Linear regression analysis was used to test whether the IMAT was associated with daily carbohydrate intake. RESULTS: Carbohydrate intake was positively associated with IMAT in all individuals, with adjustments for age, sex, height, and weight. No significant differences in blood indicators were found between the sexes. CONCLUSIONS: Regardless of sex and age, higher carbohydrate intake was strongly correlated with greater IMAT accumulation. This suggests the need to better understand sex differences and high carbohydrate diet patterns in relation to the association between obesity and metabolic risk, which may help reduce obesity prevalence.

Isolation of Pediococcus Strain from Nuruk and Anti-Lipid Accumulation Effect of Ornithine-Containing Makgeolli on 3T3-L1 Cells (누룩으로부터 오르니틴 생성능을 갖는 Pediococcus 속 균주의 분리 및 오르니틴 함유 막걸리의 3T3-L1 세포의 중성지질 축적 억제 효과)

  • To evaluate the functional effect of ornithine produced by isolated lactic acid bacteria, we examined the anti-lipid accumulation effect of ornithine produced by isolate lactic acid bacteria on 3T3-L1 cells. Lactic acid bacteria (Pediococcus strain) were isolated from nuruk, which is made from wheat, rice, and barley (whole grain, grits, or flour) by fermenting microorganisms (Aspergillus, Rhizopus, and yeasts). Pediococcus strain was identified by 16S rDNA sequencing analyses, and cells were collected by centrifugation and developed as an ornithine starter. makgeolli, an ornithine-containing Korean traditional alcoholic beverage, was made with isolated lactic acid bacteria and arginine. makgeolli was made with the help of ornithine starter using a makgeolli making kit. We evaluated the anti-proliferation effect of ornithine makgeolli on 3T3-L1 cells. To determine the anti-proliferation effect of ornithine makgeolli on preadipocytes, lipid droplets were quantified and stained with Oil Red O. makgeolli made with ornithine starter and arginine showed a 3-fold higher concentration of ornithine compared to makgeolli without starter and arginine. In the results of 3T3-L1 cell line experiment, lipid accumulation was significantly reduced by adding 0.05 mg/mL of ornithine makgeolli compare to the control (adipocyte without sample). In conclusion, ornithine makgeolli containing ornithine starter isolated from nuruk showed an anti-lipid accumulation effect with increased ornithine content without toxicity.

Effects of Fractions from Benincasa hispida on Inhibition of Adipogenesis in 3T3-L1 Preadipocytes (동과 분획물이 3T3-L1 지방세포 분화 억제에 미치는 영향)

  • The effects of three fractions, hexane (BHHH), chloroform (BHHC), and ethyl acetate (BHHE), from water extract of Benincasa hispida on the underlying mechanisms of adipogenesis were investigated in 3T3-L1 cells. Intracellular lipid droplets were stained with Oil Red O dye and quantified. Compared to control, lipid accumulation significantly decreased by 11% and 13% upon treatment with BHHC and BHHE, respectively at a concentration of 50 ${\mu}g/mL$. Intracellular triglyceride (TG) levels were also reduced by 21% and 16%, respectively, at the same concentration. To determine the mechanism behind the reductions in TG content and lipid accumulation, glycerol release and expression levels of adipogenic marker genes were measured. The levels of free glycerol released into culture medium increased by 13% and 17% upon treatment with BHHC and BHHE, respectively. In subsequent measurements using real-time polymerization chain reaction, the mRNA levels of $PPAR{\gamma}$, C/$EBP{\alpha}$, and leptin significantly decreased upon treatment with BHHE (45%, 67%, and 35%) in comparison with non-treated control. These results suggest that BHHE inhibits adipocyte differentiation by blocking $PPAR{\gamma}$, C/$EBP{\alpha}$, and leptin gene expression in 3T3-L1 cells, resulting in reduced lipid accumulation, increased glycerol release, and intracellular triglycerides.

Accumulation and Organ distribution of protein Bound Cadmium in Rats compared with CdCl2 (단백질에 결합된 카드뮴과 CdCl2를 섭취한 쥐에서 카드뮴의 체내축적 및 분포의 차이)

  • A low level exposure experiment was conducted on growing rats to investigate the accumulation and organ distribution of protein bound cadmium compared with cadmium chloride. Male Sprague-Dawley rats were fed for 21days with one of the semisynthetic diets, which contains cadmium as either bovine liver- or kidney meal bound cadmium, cadmium chloride with uncontaminated liver meal or cadmium chloride without organ meal, in the levels of ca. 0.5, 1 and 1.5mg/kg diet, respectively. After 21days of exposure cadmium was accumulated in liver, kidney and gastrointestinal tracts depending upon cadmium levels in diet. Inspite of very low cadmium accumulation in whole blood, it tends also to increase with dietary cadmium levels. The blood cadmium concentration of animals fed organ meal containing diets was about 4-7 fold higher than that without organ meal, regardless of cadmium was intrinsically bound to protein or not. However, significant effects of organ protein on cadmium accumulation in liver, kidney and digestive tracts were not detectable, when cadmium was supplemented as cadmium chloride. On the other hands, animals fed diet containing ca. 1.5mg Cd/kg as organ bound cadmium retained more cadmium in liver, kidney and digestive tracts compared to cadmium chloride with organ meal, whereby the increase of cadmium concentration in kidney was greater then in liver. However, when the concentration of protein bound cadmium was<1mg/kg diet, organ bound cadmium was not significantly different from cadmium chloride in bioavailability and organ distribution. From this result it is suggested that the intestinal absorption of protein bound cadmium is influenced of the amount of cadmium bound in protein. When cadmium concentration in protein is relatively low, protein bound cadmium seems to be absorbed in the same way as cadmium ions are absorbed. However, when the concentration is high, at least a small amount of intact protein bound cadmium could be absorbed and accumulated selectively in kidney.

Effect of Dietary Iron Levels on the Cadmium Accumulation in Cadmium Poisoned Rats (카드뮴의 장기 중독시 철분의 섭취 수준이 흰쥐의 체내 카드뮴 축적에 미치는 효과)

  • This study was undertaken to investigate the effect of dietary Fe levels on Cd accumulation in Cd poisoned rats. Forty male weaning Sprague Dawley rats weighing 80-90g were divided into 4 groups(LFe : low Fe, LFeCd : low Fe and Cd, AFe : adequate Fe, AFeCd : adequate Fe and Cd) according to Cd administration(Cd : 0 or 50ppm in drinking water) and Fe levels (Fe : 6 or 40ppm in diet) for 12 weeks. The food intake and weight gain of LFe and LFeCd were significantly lower than those of AFe and AFeCd(p<0.01, p<0.001). The water intake was not affected significantly by Cd and Fe, therefore Cd intake was no significant difference between groups. The Cd accumulation of kidney in LFeCd was significantly higher than those of AFeCd(p<0.001). But the Cd accumulations of brain, liver and spleen were not significantly different between Cd groups and without Cd groups. The serum Cd content and urinary Cd excretion of LFeCd was significantly higher than those of AFeCd(p<0.01, p<0.01). But the fecal Cd excretion of LFeCd was significantly lower than that of AFeCd(p<0.001). The Cd retention, Cd retention rate, and apparent Cd digestibility of LFeCd were significantly higher than those of AFeCd(p<0.001, p<0.001, p<0.001). It was concluded that adequate Fe supplementation have protective effects on the long term Cd poisoning in rats.

The Effect of Dietary Cu and Fe on the Cd Accumulation in Long-Term Cd Poisoned Rats (장기간 카드뮴에 중독된 흰쥐에서 카드뮴 축적에 대한 식이 구리와 철분의 효과)

  • This study was undertaken to investigate the effect on the Cd accumulation in long-term Cd poisoned rats. 40 male weaning Sprague Dawley rats weighting 80-90g were divided into 4 groups (LCuLFeCd : low Cu, Fe and Cd group, ACuLFeCd : adequate Cu, low Fe and Cd group, ACuAFeCd : adequate Cu, adequate Fe and Cd group) according to Cu and Fe levels (Cu 0.5ppm, 8.5ppm : Fe 6ppm, 40ppm) for 12 weeks. There were no significant difference in water intake, feed intake, and body weight gain according to dietary Cu and Fe consumption. But the mean food intake and body weight gain of adequate Fe groups(LCuAFeCd, ACuAFeCd) were higher than those of deficient Fe groups (ACuLFeCd, LCuFeCd)in long-term Cd poisoned rats. The mean Cd levels of serum, liver, kidney, and urine in ACuAFeCd group were lower than those of Cu and /or Fe deficient groups. But the mean fecal Cd excretion of ACuAFeCd group was higher than that of Cu and/or Fe deficient groups. And the mean Cd retention amount of ACuAFeCd group was lower than those of Cu and/or Fe deficinet groups. In conclusion, these results provide an evidence that adequate Cu and Fe intakes can decrease Cd accumulation in rats. Therefore, in the point of increasing environmental Cd contamination, adequate Cu and Fe intakes must be suggested to prevent Cd accumulations.
