Some species of the Trichoderma genus are reported as the major problem in oak wood mushroom production in Korea. In spite of economic loss by the fungi, scientific information on airborne Trichoderma species is not much available. To generate information for disease management development we analyzed airborne Trichoderma. A total of 1,063 fungal isolates were purely obtained from indoor air sampling of cultivation houses used for oak wood mushroom using sawdust media. Among the obtained isolates, 248 isolates were identified as Trichoderma fungi including T. harzianum, T. atroviride, T. citrinoviride, and T. pseudokoningii, by morphological and molecular analysis. T. harzianum was dominant among the four identified species. All the four Trichoderma species grew fast on solid nutrient media tested (potato dextrose agar [PDA], malt extract agar [MEA], Czapek's Dox + yeast extract agar [CYA] and cornmeal dextrose agar). Compact mycelia growth and mass spore production were better on PDA and CYA. In addition, T. harzianum and T. citrinoviride formed greenish and yellowish mycelium and spores on PDA and CYA. Greenish and yellowish pigment was saturated into PDA only by T. pseudokoningii. These four Trichoderma species could produce extracellular enzymes of sawdust substrate degradation such as β-glucosidase, avicelase, CM-cellulase, amylase, pectinase, xylanase, and protease. Their mycelia inhibited the growth of oak wood mushroom mycelia of two tested cultivars on dual culture assay. Among of eleven antifungal agents tested, benomyl was the best to inhibit the growth of the four Trichoderma species. Our results demonstrate that the airborne Trichoderma fungi need to be properly managed in the cultivation houses for safe mushroom production.
Recently, one of the most interesting topic in the field of wastewater treatment is the disposal of excess sludge. The new concept of excess sludge reduction with recirculation of solubilized sludge via effective microorganisms for cell disruption within the wastewater treatment process has been developed in this study. The alkalophiles for degradation of sludge cell wall were isolated as Exiguobacterium sp., which could be more effectively solubilized sludge in the anaerobic condition. The SCOD of solubilized excess sludge by Exiguobacterium sp. was up to about 2,000mg/L and average TN and TP concentration of solubilized component were 117mg/L and 58mg/L, respectively and C/N ratio was more than 17. To investigate the effects of solubilized sludge by alkalophiles on excess sludge reduction and nutrient removal efficiency, the pilot plant of $DF^{(S)}-MBR$ process, combined with membrane bioreactor and sludge solubilization tank, was operated. In the control run(without sludge solubilization), the daily sludge production was about 4.54 kgMLVSS/day. However, in the $DF^{(S)}-MBR$(with sludge solubilization), the daily sludge production was decreased to 1.39kgMLVSS/day. The effluent quality satisfied the effluent regulation in both cases. Furthermore, the $DF^{(S)}-MBR$ showed relatively better TN removal efficiency in spite of high influent loading. So we concluded that the solubilized excess sludge by alkalophiles was effectively degraded in the MBR process as the carbon source and 70% of sludge reduction efficiency can be achieved.
Background: Previous studies on the biological integrity on habitat and landuse patterns demonstrated ecological stream health in the view of regional or macrohabitat scale, thus ignored the mesoscale habitat patterns of pool, riffle, and runs in the stream health analysis. The objective of this study was to analyze influences on the mesohabitat structures of pool, riffle, and run reaches on the fish guilds and biological integrity in Geum-River Watershed. Results: The mesohabitat structures of pool, riffle, and run reaches influenced the ecological stream health along with some close relations on the fish trophic and tolerance guilds. The mesoscale components altered chemical water quality such as nutrients (TN, TP) and BOD and these, then, determined the primary productions, based on the sestonic chlorophyll-a. The riffle-reach had good chemical conditions, but the pool-reach had nutrient enrichments. The riffle-reach had a predominance of insectivores, while the pool-reach has a predominance of omnivores. Also, the riffle-reach had high proportions of sensitive fish and insectivore fish, and the pool-reach had high proportions of tolerant species in the community composition. The intermediate fish species in tolerance and omnivorous fish species in the food linkage dominated the community in the watershed, and the sensitive and insectivorous fishes decreased rapidly with a degradation of the water quality. All the habitat patterns were largely determined by the land-use patterns in the watershed. Conclusions: Trophic guilds and tolerance guilds of fish were determined by land-use pattern and these determined the stream health, based on the Index of Biological Integrity. This study remarks the necessity to include additional variables to consider information provided by mesohabitats and land-use distributions within the selected stream stretch. Overall, our data suggest that land-use pattern and mesohabitat distribution are important factors to be considered for the trophic and tolerance fish compositions and chemical gradients as well as ecological stream health in the watershed.
A chitinase-producing bacterium was isolated from seashore mud around Beobseongpo in Chunnam province by selective enrichment culture, and among it, one isolate which was the best in producing of chitinase was selected. Nutrient or MacConkey medium was confirmed with secreting of prodigiosin pigment by Serratia sp. JM, and it was performed by the production of clear zone on medium containing chitin. Serratia sp. JM was almost same compared with Serratia marcescens ATCC 27117 in respect of its morphological, physiological and biochemical characteristics except succinic, urea and pyruvic acid. Serratia sp. JM was resistant to tetracycline but was not resistant to kanamycin and chloramphenicol. The optimal temperature and pH for the production of chitinase from Serratia sp. JM were $30^{\circ}C$ and 7.5, respectively. Production of chitinase and pH in the medium increased until the cultivation of 120 hours, but after 120 hours, they were decreased due to the acetic acid accumulated from degradation of chitin by Serratia sp. JM.
A digestion trial in vitro was conducted to study effects of supplementation of NSP (non-starch polysaccharides) degrading enzyme (feed grade) on cell wall degradation and digestibility of nutrients in barley. The slices of barley were soaked in distilled water with or without 0.15% non-starch polysaccharides degrading enzyme. Microscopic examination of the slices showed that the endosperm cell wall of barley was completely degraded by the non-starch polysaccharides degrading enzyme. The residues and supernatant of digesta in vitro were separated by filtration with 0.1 mm nylon fabric. The residues were used for measurement of crude protein, crude fat, crude fiber, and moisture. The supernatant was used for determination of viscosity, as well as amino-nitrogen and glucose content. The results showed that compared with the control, the amino-nitrogen and glucose content of the supernatant increased by 17.58% (p<0.05) and 10.26% (p<0.05), respectively, while viscosity did not change. Enzyme supplementation increased the digestibilities of dry matter, crude protein, nitrogen-free extract, crude fat and crude fiber of barley by 18.1% (p<0.05), 20.3% (p<0.05), 16.4% (p<0.05), 26.9% (p<0.05) and 30.0% (p<0.05), respectively. The present study suggests that cell wall hydrolysis may contribute to improved nutrient digestion in vivo when non-starch polysaccharides degrading enzymes are fed to swine.

The purpose of this study is to investigation of the decomposition characteristics in a pilot-scale burial site of livestock in three kinds of typical soils in Korea: sandy loam soil, clay loam soil, and sandy soil. In this study, we confirmed that most of the animals in the condition were decomposed within three years as mentioned in the "Livestock burial regional environmental research guidelines." We also determined that the decomposition rate of dead cows was higher than that of dead pigs, and that the biodegradation rate depends on the soil types in the following order: sandy soil > clay loam soil > sandy loam soil. The various external environment factors, such as temperature, moisture, pH, earthiness, nutrient, and the burial depth, should be managed properly for appropriate decomposition of dead animals.

In this study, degradation of carbohydrates and composition of fermentative products were investigated in semi-continuous acid fermenter varying hydraulic retention time (HRT). Rice soup was used as a sole substrate for the acid fermentation. Solubilization efficiency of the substrate was higher than 70% for all HRT, however the gas conversion was ignorable implying that most of organic contents in the influent remained in the form of volatile fatty acids (VFAs) and ethanol after acid fermentation. The VFAs were the predominant product and the VFAs conversion increased as the HRT decreased. The VFAs conversion reached the maximum value at 12 hr HRT accounting for 70% of the influent COD. Similar to VFAs, ethanol conversion was increased with the decrease of HRT and the maximum ethanol conversion efficiency was 8% at the HRT of 12 hr. Composition of VFAs was markedly dependent on HRT. As HRT increased, the composition of acetic acid was increased as a product of acetogenesis from butyrate, valerate and ethanol. This study demonstrated that HRT affected acid fermentation of a carbohydrate containing organic wastes producing VFAs and ethanol which could be effectively used to compensate the lack of carbon in wastewater for biological nutrient removal.

The objective of this research was to develop ecological multi-metric models using natural fish assemblages for a diagnosis of current stream health condition, and apply the model to nationwide lotic ecosystems of the Geum River, the Youngsan River, and the Sumjin River. The ecological stream health model was based on the index of biological integrity (IBI), which was originally developed in North American streams by Karr (1981), and the Rapid Bioassessment Protocol (RBP), which was scientifically established by the US EPA (1999). The metric numbers and metric attributes were partially changed for the regional applications, so the scoring criteria was modified for the assessment. Overall, metric values, based on the IBI calculations, reflected conventional water quality characteristics, based on nutrient regime, and agreed with results of staticeco-toxicity tests. Some stations impaired in terms of stream health were identified by the IBI approach, and also major key stressors affecting the stream health were identified by additional evaluations of physical habitats. Our preliminary results suggested that biological integrity in stream ecosystems was largely disturbed by habitat degradation as well as chemical pollutions. This new approach would be used as a key tool for ecological restorations and species conservations in the degraded aquatic ecosystems in Korea and applied for elucidating major causes of ecological disturbances. Ultimately, this approach provides us an effective management strategy of stream ecosystems through establishments of ecological networks in various watersheds.
Isolation and identification of alga-lytic bacteria were carried out. Fifteen isolates of alga-lytic bacteria were screened by the double layer method using A. cylindrica NIES-19 as a sole nutrient and four isolates among them were compared with their alga-lytic activity. The isolate AK-05 exhibiting the highest alga-lytic activity was identified as Acidovorax temperans base on its 16S rDNA sequence. The culture supernatant of the isolate AK-05 was reliable for the alga-lytic. Alga-lytic activity assays of culture supernatant revealed that the major substances for alga-lytic activity were non-proteins and heat stable. The highest alga-Iytic activity was practical under alkaline conditions and at 25${\sim}$$30^{\circ}C$. It is indicating an advantage for the application of water blooms by cyanobacteria in eutrophic lakes where the pH is generally in alkaline region.
In this study, we analyzed fish tolerance guilds in mainstems and tributaries of 65 streams and rivers arid their relations to water quality using dataset sampled from April to November, 2009. For the study, water quality parameters including biochemical oxygen demand (BOD), electric conductivity (EC), total nitrogen (TN), total phosphorus (TP), ammonia nitrogen ($NH_3$-N), nitrate nitrogen ($NO_3$-N) and phosphate phosphorus ($PO_4$-P) were analyzed in the laboratory and also tolerance ranges in 3 category fishes of sensitive, intermediate, and tolerant species with high abundance were analyzed. According to fish guild analysis, tolerant species was 58% of the total community and the proportion of omnivore species was 63% of the total, indicating a degradation of habitats and water quality. Water quality was shown typical longitudinal gradients from the headwater to the down-river; TN and TP increased toward the down-rivers except for the big point-source area and ionic contents, based on, electric conductivity showed same pattern. Tolerance guild analysis of 9 major species with high abundance indicated that sensitive groups had narrower tolerance range in the water quality than the groups of intermediate and tolerant species. In contrast, tolerant groups including Zacco platypus, Carassius auratus, and Opsarichthys uncirostris amurensis had wider tolerance ranges than the groups of sensitive and intermediate species. Thus, each group was evidently segregated from the tolerance levels. Principal Component Analysis (PCA) employed for the relations of water quality to fish species in each groups suggests that water quality had highest eigenvalues with fish species in the 1st axis of the PCA and nitrogen (TN, $NH_3$-N, $NO_3$-N) and phosphorus (TP) were key components differentiating three groups of sensitive, intermediate and tolerance guilds.
