Community Structure and Spatial Distribution of Phytoplankton in the Polar Front Region off the East Coast of Korea in Summer (여름철 한국 동해 극전선해역에서의 식물플랑크톤의 군집구조와 분포)

  • To characterize the community structure and spatial distribution of phytoplankton, observations on seawater temperature, salinity, nutrients, primary productivity and abundance and species composition of phytoplankton were made in the polar front region and its neighborhood off the east coast of Korea in summer 1990. Among the 96 taxa identified, Rhizosolenia setigera and Thalassionema nitzschioides were the most dominant species. The assemblage at the surface and 50 m depth was quite different in the northern inshore part of the study area but similar in the southern and offshore part. Principal component analysis by the species abundance showed that the phytoplankton consisted of the communities representing the surface of the northern inshore part with the neritic-warm dinoflagellates, the Northern Korean Cold Water with the cold water diatoms and the southern and offshore part, which seems to represent the Eastern Korean Warm Water, with the warm water diatoms. At the frontal region, diatoms were mixed with warm and cold water species. Primary productivity and phytoplankton standing crops were higher at the front than the neighboring waters. Nutrients were markedly high at the Northern Korean Cold Water. Horizontal advection of the Northern Korean Cold Water accompanied by nutrient supply seems to contribute to the high phytoplankton biomass at the front.

Expression of amino acid transport system L in the differentiation of periodontal ligament fibroblast cells (치주인대섬유모세포의 분화과정에서 아미노산 수송계 L의 발현)

  • The periodontium is a topographically complex organ consisting of epithelial tissue, soft and mineralized tissues. Structures comprising the periodontium include the gingiva, periodontal ligament (PDL) , cementum and the alveolar bone. The molecular mechanism of differentiation in PDL fibroblast cells remain unclear. Amino acid transporters play an important role in supplying nutrition to normal and cancer cells and for cell proliferation. Amino acid transport system L is a major nutrient transport system responsible for the Na+-independent transport of neutral amino acids including several essential amino acids. The system L is divided into two major subgroups, the L-type amino acid transporter 1 (LAT1) and the L-type amino acid transporter 2 (LAT2). In this study, the expression pattern of amino acid transport system L was, therefore, investigated in the differentiation of PDL fibroblast cells. To determine the expression level of amino acid transport system L participating in intracellular transport of amino acids in the differentiation of PDL fibroblast cells, it was examined by RT-PCR, observation of cell morphology, Alizaline red-S staining and uptake analysis after inducing experimental differentiation in PDL fibroblast cells isolated from mouse molar teeth. The results are as follows. 1. The LAT1 mRNA was expressed in the early stage of PDL fibroblast cell differentiation. This expression level was gradually reduced by differentiation- inducing time and it was not observed after the late stage. 2. The expression level of LAT2 mRNA was increased in time-dependent manner during differentiation induction of PDL fibroblast cells. 3. There was no changes in. the expression level of 4F2hc mRNA, the cofactor of LAT1 and LAT2, during differentiation of PDL fibroblast cells. 4. The expression level of ALP mRNA was gradually increased and the expression level of Col I mRNA was decreased during differentiation of PDL fibroblast cells. 5. The L-leucine transport was reduced by time from the early stage to the late stage in PDL fibroblast cell differentiation. As the results, it is considered that among neutral ammo acid transport system L in differentiation of PDL fibroblast cells, the LATl has a key role in cell proliferation in the early stage of cell differentiation and the LAT2 has an important role in the late stage of cell differentiation for providing cells with neutral amino acids including several essential amino acids.

Dynamic changes of yak (Bos grunniens) gut microbiota during growth revealed by polymerase chain reaction-denaturing gradient gel electrophoresis and metagenomics

  • Objective: To understand the dynamic structure, function, and influence on nutrient metabolism in hosts, it was crucial to assess the genetic potential of gut microbial community in yaks of different ages. Methods: The denaturing gradient gel electrophoresis (DGGE) profiles and Illumina-based metagenomic sequencing on colon contents of 15 semi-domestic yaks were investigated. Unweighted pairwise grouping method with mathematical averages (UPGMA) clustering and principal component analysis (PCA) were used to analyze the DGGE fingerprint. The Illumina sequences were assembled, predicted to genes and functionally annotated, and then classified by querying protein sequences of the genes against the Kyoto encyclopedia of genes and genomes (KEGG) database. Results: Metagenomic sequencing showed that more than 85% of ribosomal RNA (rRNA) gene sequences belonged to the phylum Firmicutes and Bacteroidetes, indicating that the family Ruminococcaceae (46.5%), Rikenellaceae (11.3%), Lachnospiraceae (10.0%), and Bacteroidaceae (6.3%) were dominant gut microbes. Over 50% of non-rRNA gene sequences represented the metabolic pathways of amino acids (14.4%), proteins (12.3%), sugars (11.9%), nucleotides (6.8%), lipids (1.7%), xenobiotics (1.4%), coenzymes, and vitamins (3.6%). Gene functional classification showed that most of enzyme-coding genes were related to cellulose digestion and amino acids metabolic pathways. Conclusion: Yaks' age had a substantial effect on gut microbial composition. Comparative metagenomics of gut microbiota in 0.5-, 1.5-, and 2.5-year-old yaks revealed that the abundance of the class Clostridia, Bacteroidia, and Lentisphaeria, as well as the phylum Firmicutes, Bacteroidetes, Lentisphaerae, Tenericutes, and Cyanobacteria, varied more greatly during yaks' growth, especially in young animals (0.5 and 1.5 years old). Gut microbes, including Bacteroides, Clostridium, and Lentisphaeria, make a contribution to the energy metabolism and synthesis of amino acid, which are essential to the normal growth of yaks.

Inorganic Nutrient Removal Efficiency of Aquatic Plants from Recirculating Aquaculture System (수생식물을 이용한 담수 순환여과식 양식용수내의 무기영양염 처리 효율)

  • Inorganic nutrients such as nitrogen and phosphate compounds accumulate in recirculating aquaculture systems. These nutrients must be removed from the system before they affect pH and fish health. For this purpose, aquatic plants are a simple and inexpensive method of removal. There are four commonly used aquatic plants: Eichhornia crassipes (water hyacinth), Pistia stratiotes (water lettuce), Hygrophila angustifolia, and Hydrocotyle leucocephala in freshwater recirculating aquaculture systems in Korea, but their efficiencies are not known. Therefore, removal efficiencies of inorganic nutrients from a freshwater recirculating aquaculture water with four commonly used aquatic plants were tested. Removing efficiencies of TAN, N $O_2$$^{[-10]}$ -N, and N $O_3$$^{[-10]}$ -N of the plants in 210 L aquaria for 48-hour period were tested. The removing efficiencies of TAN, N $O_3$$^{[-10]}$ -N, and P $O_4$$^{3-}$-P of the two most effective plants, water hyacinth and water lettuce, were also tested in 690 L (surface area of 1.55 $m^2$) tanks under 2 different initial stocking densities, 4 kg and 6 kg, for 22 days. Proximate analysis major nutrients and N and P contents of the all plants were analyzed for calculating net removal weight of N and P by the plants. Water lettuce was the most effective for removing TAN, N $O_2$$^{[-10]}$ -N, and N $O_3$$^{[-10]}$ -N from the water for 48-hour period tested followed by water hyacinth and Hygrophila angustifolia. Water lettuce reduced TAN, N $O_2$$^{[-10]}$ -N, and N $O_3$$^{[-10]}$ -N concentration from 2.3 mg/L, 0.197 mg/L, and 21.4 mg/L to 0.4 mg/L, 0.024 mg/L and 17.4 mg/L, respectively while water hyacinth reduced them down to 0.6 mg/L, 0.029 mg/L and 17.9 mg/L, respectively. The concentrations of TAN, N $O_2$$^{[-10]}$ -N, and N $O_3$$^{[-10]}$ -N in Hydrocotyle leucocephala group were rather increased up to 3.7 mg/L, 5.7 mg/L and 48.2 mg/L, respectively. This is because the creeping stem of Hydrocotyle leucocephala had to be cut to meet stocking weight resulting in decaying of the stem in the aquaria during experiment. The net growth in weight of water hycinth and water lettuce of 4 kg each in the 1.55 $m^2$ tanks for 22 days were 9.768 kg and 10.803 kg respectively, and those at initial weight of 6 kg each were 8.393 kg and 9.433 kg, respectively. The reason of lower net growth in the later group was restricted growth space. Nitrogen and phosphorus contents in water hyacinth were 2.89% and 0.27%, and those in water lettuce were 3.87% and 0.36%, respectively. Average quantities of removed N and P from 1.55 $m^2$ tanks by water hyacinth for 22 days were 18.9 g and 1.75 g, while those by water lettuce were 36.8 g and 3.5 g, respectively. Therefore water lettuce showed much higher efficiencies for removing both N and P from recirculating aquaculture water than water hyacinth.

Changes in Biochemical Components of the Ovary and the Trunk Tissues Including the Digestive Organ Associated with Gonadal Development of the Female Octopus ocellatus (주꾸미 (Octopus ocellatus) 의 생식소 발달에 따른 난소와 소화기관을 포함한 몸통부위 조직의 생화학적 성분변화)

  • Gonadosomatic index, condition index and reproductive cycle with the gonadal development of the female Octopus ocellatus were investigated by histological observations and morphometric data, from January to December, 2000. And changes in biochemical components of the ovary and the trunk tissues including the digestive organ associated with gonadal development were studied by biochemical analysis from January to October, 2001. The specimens were collected at the coastal waters of Buan, Jeollabuk-do, Korea, from January 2000 to October 2001. O. ocellatus is a dioecious organism. The gonad of O. ocellatus locates medially in posterior region of the body. Morphology of the ovary shows round and oval in shape, the average diameter and external colour of ripe ovary was 32 mm and semitransparent light brown in colour. As the ovary was getting mature, transparent elongated eggs covered with chorion were present in the ovarian cavity. Monthly changes in the gonadosomatic index (GSI) showed a similar pattern with those of the condition index. The GSI and condition index began to increase in March and reached the maximum in April. And then, their values decreased from May and reached the minimum in September. Reproductive cycle of O. ocellatus can be categorized into five successive stages: early developing stage (September to December), late developing stage (November to March), ripe stage (March to May), partially spawned stage (April to June), and degenerative/resting stage (June to October). Follicle cells attached to an oocyte were involved in vitellogenesis in the cytoplasm of the vitellogeneic oocyte and formation of chorion (secondary egg membrane) of the ovarian eggs. Spawning occurred between April and June. The spawning period was once a year and the peak took place between May and June. This species belongs to semelparity. According to changes in biochemical contents of the ovary and the digestive organ, monthly variations of moisture, total protein, total lipid and glycogen contents (%) in the ovary showed a negative correlationship with those of the trunk tissues including the digestive organ. Accordingly, it is assumed that the ovary only may be received nutrient supply (total lipid content) for gonadal development from the trunk tissues including the digestive organ (r = -0.55, p < 0.05).

Seasonal Changes in Biochemical Components of the Adductor Muscle and Visceral Mass Tissues in the Female Cyclina sinensis, in Relation to Gonad Developmental Phases (암컷 가무락조개, Cyclina sinensis의 난소 발달단계에 따른 폐각근 조직과 내장낭 조직의 생화학적 성분의 계절적 변화)

  • We investigated the reproductive cycle with gonadal development of the female Cyclina sinensis by histological observations and seasonal changes in biochemical components of the adductor muscle and visceral mass tissues were studied by biochemical analysis, from January to December, 2001. The reproductive cycle of this species can be classified into five successive stages: early active stage (February to April), late active stage (March to June), ripe stage (May to August), partially spawned stage (July to October) and spent/inactive stage (September to February). Total protein contents in the adductor muscle tissues reached the maximum in February (early active stage) and appeared the minimum in June (ripe stage), while their contents in the visceral mass tissues reached the maximum in the late active and ripe stages (June) and gradually decreased from July (partially spawned stage) to November (spent/inactive stage). Changes in total protein contents showed a negative correlationship between the adductor muscle and visceral mass tissues (r = -0.499, p = 0.099). Total lipid contents in the adductor muscle tissues reached the maximum in January (the inactive stages) and their contents gradually decreased from February. Their contents in the visceral mass tissues, however, reached the maximum in June (late active and ripe stage) and gradually decreased from July (the partially spawned stage). On the whole, total lipid contents showed a negative correlationship between the adductor muscle and visceral mass tissues (r = -0.631, p < 0.05). Therefore, These results indicate that the nutrient contents of the adductor muscle and visceral muscle tissues change in response to gonadal energy needs. Glycogen contents in the adductor muscle tissue reached the maximum in March (early and late active stages) and decreased from July to September (partially spawned stage). while their contents in the visceral mass tissues reached the maximum in June (late active and ripe stages) and gradually decreased from July (partially spawned stage). Thereafter, their levels gradually increased in November (spent/inactive stage). On the whole, changes in glycogen contents appeared negative correlationship between the adductor muscle and visceral mass tissues. However, they showed no significant different (r = -0.307, p = 0.331).

The Antibacterial Activity Against Fish Pathogen of Paenibacillus sp. MK-11 Isolated from Jeju Coast (제주연안으로부터 분리한 Paenibacillus sp. MK-11의 어류 질병 세균에 대한 항균활성 탐색)

  • In this study, we isolate and identify bacteria from seawater collected from Jeju coast, to evaluate the antimicrobial activity against the fish pathogenic bacteria. 14 bacterial strains were isolated and identified using physiological, biochemical and molecular tools. Antibacterial activity of all the 14 isolates were screened against four major fish pathogens namely, two Gram-positive: Streptococcus iniae, Streptococcus parauberis and two Gram-negative: Vibrio anguillarum, Edwardsiella tarda. Results revealed that among the 14 isolates, MK-11 was found to have antibacterial activity against S. iniae, S. parauberis, V. anguillarum Particularly, S. iniae was susceptibility with the MIC value of $250{\mu}g/ml$. The biochemical and physio-chemical results reveal that MK-11 had the sugar-alcohol disassemble ability of the D-sorbitol and D-mannitol. Also the utilization of the yeast extract, sorbitol and di-potassium phosphate were noted to be high. The optimum culture condition such as pH and temperature was recorded as pH 6.0, $25^{\circ}C$ and along with 1% NaCl which differs from the previous reports particularly in nutrient resolutions. As results of the analysis of 16S rDNA sequences, MK-11 show the high similarity with Paenibacillus polymyxa, P. jamilae, P. brasilensis 99.78%, 99.43%, 99.39%, repectively. Hence, in the present study, the isolated Paemibacillus sp. MK-11 from Jeju seawater possesses the antibacterial activity against fish pathogens and it could be used as a new antibiotic agents against the gram positive fish pathogens.

Bacterial Community Structure of Food Wastewater Treatment System Combined with Rotating Biological Contactor and Tapered Aeration Reactor (회전접촉장치와 점감포기 반응조를 이용한 식품폐수 처리시설의 세균군집 구조)

  • A pilot-scale wastewater treatment plant combined with rotating biological contactor and tapered aeration reactors was operated with the wastewater discharged from a food factory for 5 months. The bacterial communities of this plant were investigated by terminal restriction fragment length polymorphism (T-RFLP) and phylogenetic analysis of 16S rRNA genes. In spite of high concentration of nitrogen and phosphorus as well as organic carbon, removal efficiency of chemical oxygen demand, total nitrogen, and total phosphorus was 98%, 93%, and 95%, respectively. Bacterial community at the initial operation stage was clearly distinguished from that of the stable operation stage. The most predominant phylum in the sample of stable stage was Bacteroidetes. Major population of operation period was Haliscomenobacter, Sphaerotilus, and candidate division TM7, which were classified as filamentous bacteria. However, sludge bulking caused by these bacteria was not observed. The population that has a close relationship with Haliscomenobacter increased during the stable operation stage, emerging as the most predominant group. These results suggest that the filamentous bacteria participated in nutrient removal when using rotating biological contactor and tapered aeration reactor.

Evaluation of Field Application for the Developed Retrofitting Process and Analysis of Bacterial Community Structure in Pilot Plant (하수처리장 Retrofit 공정의 현장적용성 평가 및 세균 군집 분포 연구)

  • In this study, a retrofitting BNR process that was modified for the economical applicability was proposed and evaluated in the pilot plant($50m^3/d$). At the same time the bacterial community structure was investigated in the pilot plant by using FISH(fluorescent in situ hybridization) method. Economically 16% of the initial construction cost for the proposed process(introduction of a biological nutrient removal process of $60,000m^3/d$ scale basis) was reduced due to the absence of a bioreactor. Water treatment efficiencies and maintenance facilities of the modified process were satisfied with the strengthened discharge permits in Korea throughout a long term pilot plant operating including a winter season. Bacterial populations in the pilot plant and in the control plant(A2/O process, B SIP(Sewage Treatment Plant)) were remained uniformly during the test period, but bacterial structure in the bioreactor was changed drastically. Proportions of ${\beta}$-proteobacteria group including soil bacteria which play a important role in wastewater treatment increased $25{\sim}607%$ in population.

Water Quality of a Rural Stream, the Hwapocheon Stream, and Its Analysis of Influence Factors (보와 습지가 있는 화포천의 수질 영향인자 분석)

  • This study was aimed to analyze the water quality characteristics of the Hwapocheon Stream and to be utilized in the further related research. Water in the upper stream became a dammed pool due to the existence of 14 weirs, and pollutants such as both sewage and irrigation water were introduced into the mainstream passing through farming settlements and agricultural land. For these reasons, filamentous cyanobacteria (Oscillatoria sp.) bloomed at the bottom of the dammed pool. Also in the midstream and downstream, tributaries with high pollutant concentrations [e.g., Comocheon (T3) and Yongdeokcheon (T8)] were inflowed, and had a negative impact on water quality of the mainstream, especially nitrogen and phosphorus. In the Hwapocheon Stream, dissolved oxygen (DO) decreased, and suspended solid (SS) increased toward the downstream. The result showed that hydraulic retention time, SS, COD, and concentration of $NH_4{^+}$ were important water quality factors of the Hwapocheon Stream. The high concentration of benthic organic matter and rich in attached algae in the core of Hwapo-wetland were expected to give impact on the water quality of the mainstream. In the spatial manner, water quality showed increasing trend in the weir zone, and it was constant or decreased trend in wetland. In the seasonal manner, the nutrient concentrations were high in the winter dry season, however, the organic matter concentrations were high in spring and summer. Generally, the concentrations of phytoplankton value were $40{\mu}g\;chl-{\alpha}/L$ or less in all reaches except for the high concentrations in the weir and wetland area in June.
