The Preventive Effects of Colon Cancer and Imflammatory Bowel Disease of Supercritical Heat-Treated Radish Extracts

  • There is a strong connection between the diet rich in antioxidants and the decreased incidence of inflammatory bowel disease and cancerous diseases. Diets that are rich in anti-oxidants particularly include fruits and vegetables containing the high amounts of vitamin A-E, carotenoids, and minerals. The supercritical heat-treated radish extracts of the research result had an inhibitory effect on the development of aberrant crypt foci (ACF), namely, preneoplastic lesions having a potential to become cancer cells and reduced the number of the aberrant crypt foci (ACF) consisting of four or more aberrant crypts (AC) having high risk to become tumors by about half. The supercritical heat-treated radish extracts can reduce the incidence of preneoplastic lesions having a high risk of developing cancer by about 28 %. DSS-treated mice developed symptoms similar to those of human UC, such as severe bloody diarrhea and weight loss. Supercritical heat-treated radish extracts, as well as sulfasalazine, suppressed colonic length and mucosal inflammatory infiltration. In addition, supercritical heat-treated radish extracts treatment significantly reduced the expression of pro-inflammatory signaling molecules through suppression both mitogen-activated protein kinases (MAPK) and nuclear factor-kappa B (NF-kB) signaling pathways, and prevented the apoptosis of colon. Moreover, supercritical heat-treated radish extracts administration significantly led to the up-regulation of anti-oxidant enzyme including SOD and Catalase.

Effects of Capsosiphon Fulvescens Extract on Contact Dermatitis in NC/Nga Mice (매생이(莓山苔) 추출물이 NC/Nga 마우스의 접촉성 피부염에 미치는 효과)

  • Objectives : The aim of this study is to report the effect of Capsosiphon fulvescens(Maesangi) extract(MSE) on contact dermatitis(CD). Methods : CD induced by Dermatophagoides farinae body(Dfb) in NC/Nga mice. 4% sodium dodecyl sulfate(SDS) $150{\mu}l$ and Dfb(10mg/ml) ointment 100mg were topically treated twice a week for four weeks. MSE 200 mg/kg was topically treated every 4weeks. Every week, we examined the skin lesions, weight of spleen and lymph node, epidermal thickness, production of Tumor necrosis factor-alpha($TNF-{\alpha}$) and interleukin-6(IL-6). Results : MSE reduced features of Dfb-induced skin lesions, weight of spleen and production of $TNF-{\alpha}$. MSE significantly reduced weight of lymph node, epidermal thickness, number of mast cells and production of IL-6. Conclusions : MSE may have a potential therapeutic effects for CD by suppressing allergic inflammation.

  • Pulmonary nodular lymphoid hyperplasia is a reactive lymphoproliferative disease. It is very rare, which means that many aspects of the disease are unknown or have not been proven. Pulmonary nodular lymphoid hyperplasia can be symptomatic or asymptomatic, progressive or not, and solitary or multiple, and a surgical approach is the current treatment of choice. We present a case of pulmonary nodular lymphoid hyperplasia that was visualized as multiple ground glass opacities on a computed tomography (C T) scan, and observed for 1 year because the patient was pregnant. Over this period, the number and extent of the opacities progressed, but no symptoms were reported. A surgical biopsy was done and some remaining lesions regressed on follow-up CT scans, while others progressed, without any appearance of symptoms.

Surgical Resection of Pulmonary Metastasis from Renal Cell Carcinoma

  • Background: Renal cell carcinoma has shown less response to systemic therapies including chemotherapy, radiation, and immunotherapy than other cancers. Surgery has therefore become an important treatment tool. The protocol for treatment is the same for pulmonary metastasis of renal cell carcinoma. We performed surgery for pulmonary metastatic renal cell carcinomas and analyzed the results. Materials and Methods: We retrospectively analyzed 15 patients who had undergone pulmonary metastasectomy from renal cell carcinoma at our hospital from January 2005 to December 2009. Results: No patients had extrathoracic metastatsis. The mean age was 60.2 years (range 35~73). There were 12 male and 3 female patients. The number of synchronous and metachronous patients were 8 and 7, respectively. The mean survival times of synchronous and metachronous patients were 32.6 and 42.9 months, respectively. 6 patients had single lesions and 9 patients had multiple (more than 3) lesions. The surgical procedures included wedge resection (10), lobectomy (2), wedge resection with segmentectomy (2), and segmentectomy (1). Median observation and survival time were 54.1 and 34.9 months. The 1-year and 3-year survival rates were 80% and 50%, respectively. Conclusion: Pulmonary resection for pulmonary metastatic renal cell carcinoma was found to be a safe and effective treatment modality when complete resection was performed.

Ultrasonographic assessment of experimentally induced gastric perforation in beagle dogs

  • The goals of this study were, first, to evaluate the feasibility of inducing gastric perforation with 99% alcohol injection after electrocautery (EA-method), and, second, to observe "enhanced peritoneal stripe sign (EPSS)" and other lesions upon induction of gastric perforation. Six clinically normal beagle dogs were prepared for gastric perforation using endoscopy. After gastric perforation, EPSS and other lesions on ultrasonography were observed eventually (at 0 h, 3 h, day 1, day 2, day 3, day 4, day 5, and day 6). We graded the EPSS depending on its width and number. EPSS was observed until day 4 of the examination in all the 6 dogs. The grades of EPSS were the highest at 3 h and declined gradually. Peritoneal effusion was observed in all dogs at 3 h and on day 1. Regional bright mesenteric fat was confirmed in all dogs on days 3 and 4. In conclusion, gastric perforation can be induced by EA-method. EPSS and peritoneal effusion appear at a very early stage, and regional bright mesenteric fat was identified on days 3 and 4 in almost all dogs with gastric perforation.

Transmembrane protein 64 modulates prostate tumor progression by regulating Wnt3a secretion

  • Wnt3a is a glycosylated ligand that activates the β-catenin-dependent signaling pathway. Wnt signaling is also important in the prostate tumor microenvironment, and Wnt proteins secreted by the tumor stroma promote resistance to therapy. Bioactive Wnt3a production requires a number of dedicated factors in the secretory cell, but their coordinated functions are not fully understood. We previously reported transmembrane protein 64 (Tmem64) as a novel regulator of the Wnt/β-catenin signaling pathway, which is correlated with β-catenin regulation. In the present study, the role of Tmem64 in prostate cancer cells was investigated by modulating Wnt3a secretion. Overexpression of Tmem64 inhibited Wnt3a secretion and Lef/Tcf-sensitive transcription. By contrast, a Tmem64 mutation deleting the protein's transmembrane region restored Wnt3a secretion. Notably, Tmem64 protein and mRNA in PC3 cells were significantly overexpressed compared with that observed in LNCaP and DU145 cells. In a mouse metastasis model intracardially injected with PC3 cells, Tmem64 expression was downregulated in the metastatic spine and mandible lesions compared with in the primary injection regions. However, Wnt3a was strongly expressed in the metastatic spine and mandible lesions. Collectively, these findings suggest that Tmem64 is involved in the metastatic progression of prostate cancer cells by regulating Wnt3a secretion.

  • Palmoplantar pustulosis may be associated with peculiar bone and joint conditions. Initially restricted to the anterior chest wall, these conditions were later found to involve peripheral bones and joints. The same aseptic pseudoinfectious bone and joint lesions were also found associated with severe acne. In 1987, Kahn et al suggested this acronym to describe a number of reports of peculiar bone, joint, and skin lesions published over the last 25 years. SAPHO sydrome stands for synovitis, acne, pustulosis, hyperostosis, and osteitis syndrome. We report one case of SAPHO syndrome: A 27-year-old female presented left clavicular and right sacroiliac joint pain with hyperostosis and pustulosis palmaris.

The Type Analysis for Safety Accident of Water Sports Happening in River

  • Purpose: Recently, as the number of people who enjoy water sports life in river or lake easy to accessible are increasing, that of the patients who are injured in water sports also does gradually. We intend to investigate the type of the injured patients of water sports and the riskiness of the sports in this study. Methods: We retrospectively looked into the medical records of the patients who were injured in water sports and visited a general hospital in Gangwondo-province from 2010 to 2015. Results: Total 146 patients came to the hospital during six years. Patients mostly occurred at younger ages, in summer, and on holidays, rather than weekdays. The most common lesions of injuries were faces (53 patients). The most common types of injuries were contusions (62 patients), followed by fractures (32 patients) and lacerations (26 patients). The most frequent fracture sites were the upper extremities (11 patients). Most of the trauma patients were mild, but a small number of patients with aspiration pneumonia occurred and their severity was higher than trauma patients. Conclusions: In this study, facial injuries were most frequent in water sports injuries. In the fractures, upper extremities were the most common fractured lesions. In addition, even if there is no direct trauma, aspiration pneumonia is serious, so caution should be taken with protective equipment suitable for water sports.

Inhibitory Effects of Lactic Acid Bacteria (LAB) on the Azoxymethance-induced Colonic Preneoplastic Lesions

  • Epidemiological and experimental studies provide evidences that diet and intestinal microflora play an important role in colon carcinogenesis. In recent years, it has been suggested that lactic acid bacteria (LAB) used to ferment dairy products have an inhibitory effect on the colon cancer. This study was designed to determine the effect of Bifidobacterium longum HY8001 (Bif) and Lactobacillus acidophilus HY2104(Lac) of Korean origin on azoxymethane (AOM)-induced colonic preneoplastic lesions such as aberrant crypt foci(ACF) formation and cecal pH. At five weeks of age, Spraque-Dawley rats were divided at random into four (AOM alone, Bif, ,Lac, and Bif+Lac) groups. Animals were weighed weekly and oral administration of LAB cultures were performed daily until the termination of the study. Two weeks later, all animals were given a subcutaneous injection of AOM dissolved in normal saline at a dose of 15 mg/kg of body weight once per week for 2 weeks. All rats were necropsied 7 weeks after the last AOM injection , and the ACF were visualize under light microscopy in the formalin-fixed, unsectioned methylene blue-stained colons. The total number of aberrant crypt in Bif, Lac, and Bif+Lac groups were significantly lower than that of the AOM alone group and the percentage of inhibitions weas 35.0, 45.6%, respectively. Significant inhibition (p<0.001) in the total number of ACF was also observed in LAB treated groups (Bif , Lac, and Bif+Lac group by 3003, 38.6, and 41.2%, respectively). Furthermore, cecal pH appeared to significantly decrease by LAB administration. The results of present study provide some evidences for potential colon tumor-inhibitory properties of lactic cultures and fermented dairy products.

Effect of Yanghyeoljeseuptang (YHJST) Composition on Atopic Dermatitis NC/Nga Mice Induced by DNCB(dinitrochlorobenzene) (양혈제습탕(凉血除濕湯)이 DNCB로 유도된 NC/Nga mice의 아토피 피부염에 미치는 영향)

  • Objectives The purpose of this study is to investigate the effect of YHJST on atopic dermatitis in an experiment using an NC/Nga mice induced by DNCB, which has histological and clinical similarities to the condition in humans. Methods To investigate the effect of YHJST on atopic dermatitis(AD), we evaluated atopic dermatitis-like skin lesions by clinical skin index and analyzed immunological parameters in peripheral blood mononuclear cells(PBMCs) and performed skin histology in ears and dorsal skin of NC/Nga ato-mouse. Results YHJST medicines decreased Serum level of IgE, IL-6, TNF-$\alpha$. Also total number of $CD69^+$, $CD3^+$ in PBMCs, absolute cell number of $CCR3^+CD3^+$, $CD11b^+Gr-1^+$ in Dorsal skin tissue, Serum IgG1, IgM, IgG2a and IgG2b decreased significantly. Furthermore YHJST is extremely effective to histological symptoms; dermal and epidermal thickening, hyperkeratosis and inflammatory cell infiltration and suppressed histologic infiltration of $CD4^+$ & $CCR3^+$ in ear and dorsal skin lesions significantly. YHJST decreased gene-expression of IL-6, TNF-$\alpha$, CCR3, Eotaxin mRNA than that of control group. Conclusions YHJST on atopic dermatitis to atopic dermatitis NC/Nga mouse induced DNCB was incredibly effective.
