Some Unrecorded Species of Oribatid Mites (Acari : Oribatida) from Korea (한국 산 날개응애류의 미기녹종(3))

  • In the course of taxonomical studies of oribatid mites, eight species, Tenuiala nuda Ewing, 1913, Cosmopirnodus angulatus Ichisawa and Aoki, 1998, Neoribates pallidus Aoki, 1988, Belba sellnicki Bulanova-Zachvatkina, 1962, Camisia horrida (Hermann 1804), Hermannia gibba (C. L. Koch 1839), Eueremaeus elongatus (Fujikawa 1972), and Licnodamaeus undulatus (Paoli 1908) are recorded from Korea for the first time.

A Novel Metalloprotease from the Wild Basidiomycete Mushroom Lepista nuda

  • A 20.9-kDa metalloprotease was isolated from dried fruiting bodies of the wild basidiomycete mushroom Lepista nuda. The N-terminal amino acid sequence of the protease was seen to be ATFVLTAATNTLFTA, thus displaying no similarity with the sequences of previously reported metalloproteases. The protease was purified using a procedure that entailed ion-exchange chromatography on CM-Cellulose, Q-Sepharose, and Mono S, and FPLC-gel filtration on Superdex 75. The protease functioned at an optimum pH of 7.0 and an optimum temperature of $50^{\circ}C$. It was also noted that the protease demonstrated a proteolytic activity of 1,756 U/mg toward casein. The $K_m$ of the purified protease toward casein was 6.36 mg/ml at a pH of 7.0 and with a temperature of $37^{\circ}C$, whereas the $V_{max}$ was 9.11 ${\mu}g\;ml^{-1}\;min^{-1}$. The activity of the protease was adversely affected by EDTA-2Na, suggesting that it is a metalloprotease. PMSF, EGTA, aprotinin, and leupeptin exerted no striking inhibitory effect. The activity of the protease was enhanced by $Fe^{2+}$, but was curtailed by $Cd^{2+}$, $Cu^{2+}$, $Hg^{2+}$, $Pb^{2+}$, $Zn^{2+}$, and $Fe^{2+}$ ions. The protease also exhibited inhibitory activity against HIV-1 reverse transcriptase with an $IC_{50}$ value of 4.00 ${\mu}M$. The $IC_{50}$ values toward hepatoma Hep G2 and leukemia L1210 cells in vitro were 4.99 ${\mu}M$ and 3.67 ${\mu}M$, respectively.

Study on the Anti-tumor Effects of Extracts from Lepista nuda Mushroom (민자주방망이버섯 추출물의 항암 효과에 대한 연구)

  • This study was carried to investigate the inhibition effects of Lepista nuda mushroom extracts on the growth of the human cancer (HepG2, KATOⅢ, AGS) cells. The fraction Ⅰ of Lepista nuda mushroom extracts strongly inhibited to 71.4-91.8% for the growth of cancer cells. The growth of cancer HepG2, KATOⅢ and AGS cell which treated with 0.5 and 1㎎/mL of hot water fraction I was inhibited to 71.4% and 85.5%, 45.0% and 86.6%, 71.6% and 90.7% respectively. The growth of cancer cell HepG2, KATOⅢ and AGS which treated with 0.5 and 1㎎/mL of microwave fraction I was inhibited to 79.8% and 85.2%, 78.6% and 88.9%, 85.8% and 91.8%, respectively. The inhibition ratios of cancer cell growth were higher by microwave extracts than hot water extracts. And the more fraction were insoluble for water, the more inhibition ratios of cancer cell growth were lower.

  • Twelve species of Taiwanese liverworts from recent collection were examined by GC/MS and some of the major components were isolated and identified spectroscopically. Interesting findings from the view point of chemotaxonomy are discussed in the paper.

The Screening of Fibrinolytic Activities of Extracts from Mushrooms in Mt. Chiak (치악산 버섯추출물로부터 Fibrin 분해활성의 검색)

  • Extracts from 65 species of mushrooms in mount Chiak were screened for their fibrinolytic activities. Extracts from Armillariella mellea, Calocybe sp., Lepista nuda and Trichaptum abietinum showed to have almost twice of activity of plasmin 1.5 U/ml, 193%, 213%, 198%, and 193% respectively. Collybia maculata showed 98% of activity, and Coprinus comatus and Lepiota sp. 56% and 58% activities, respectively. Other two, Agrocybe sp. and Stropharia rugosoannulata were less than 10% of activities.

Distribution of the Korean Barbatula Species Reviewed by the Morphological Traits of Nostrils (비공의 형태적 특징으로 재검토한 한국산 종개속 어류의 분포 특성)

  • According to the character states of nostrils in the Barbatula species comprising 1,366 specimens collected from the major river system in Korea, distribution pattern of the two Korean Barbatula species was reviewed. The taxonomic character related to nostrils in Barbatula was very stable and two character states (widely separated and closely set) were observed in the present materials. B. toni (Dybowski, 1869) with widely separated nostrils distributes in the streams flowing into the East Sea north of Sokcho, Gangwon-do Province; Eogokcheon Stream, a tributary of Namhangang River; and Anseongcheon Stream, Gyeonggi-do Province, whereas B. nuda (Bleeker, 1864) with closely set occurs in most rivers flowing into the Yellow Sea including Nakdonggang River as well as in the Yangyangnamdaecheon Stream, Gangwon-do Province south to Bangyulcheon Stream, Gyeongbukdo Province, flowing into the East Sea as previously reported with some exception. Especially, both Barbatula species were co-occurred in the Yeongokcheon Stream, Gangwon-do Province and Eogokcheon Stream. The present results will provide a useful guide to study the regional fish fauna or fish community analysis as well as to clarify the taxonomical status of the Korean Barbatula species.

  • Mycelial growth and survival ratio of ectomycorrhizal fungi were determined after storage at $-70^{\circ}C$ for 1, 3, or 6 mon. Seventeen of 23 ectomycorrhizal fungi did not survive after storage for more than 6 mon, whereas Cenococcum geophilum, Lepista nuda, and some species of Rhizopogon and Suillus did survive.

  • Present experiments were conducted to determine the possibility of cultivation of 9 edible wild species selected among the higher fungi growing in Korea. In the investigation on the mycelial growth according t6o the different media, the mycelial growth of Coprinus comatus was fast on the CSA medium, when malt extract was added to the basal medium PSA, the mycelial of Lapista nuda and Auricularia auricula-judae was fast in growth and density. In the spawning, the mycelial growth of Pholiota squarrosa on the oak tree's sawdust, Pleurotus cornucopiae on the broad-leaves' sawdust, and Coprinus comatus on the compost was respectably fast and also it shown to be possibility of artificial cultivation owing to their carporphore budding when Coprinus comatus and Lepiota alborubescens cultivated on the rice straw, Auricularia auriculajudae and Pleurotus cornucopiae on the sawdust of the popla and Pholiota squarrosaon the sawdust of the oak tree.

Analysis of Nutritional Components of Lepista nuda (민자주방망이버섯(L. nuda)의 영양성분 분석)

  • This study was carried out to analyze the nutritional component of Lepista nuda in order to estimate its nutritional and functional values. The content of moisture were $90.90{\pm}0.09%$ carbohydrate $4.34{\pm}0.07%$, crude protein $3.70{\pm}0.03%$, ash $0.55{\pm}0.04%$, and crude fat $0.47{\pm}0.01%$, respectively The reducing sugar was $636.17{\pm}7.50mg$. Total amount of free sugar was 97.32 mg, which were mainly composed of galactose (51.98 mg) and trehalose (22.48 mg). In the results of mined analysis, the content of K was the highest as $103.10{\pm}0.18mg$, followed by $56.01{\pm}0.20mg$ of Mg. The total content of hydrolyzed amino acid was 310.39 mg which consist of essential amino acids including valine (25.40 mg), leucine (22.52 mg), lysine (21.34 mg) and 198.89 mg of non-essential amino acids including glutamic acid (60.72 mg), aspartic acid (30.17 mg), and alanine (29.72 mg), as the major amino acids. free amino acids were 220.86 mg which were composed of essential amino acids (36.21 mg) inclusive of isoleucine (21.57 mg) and threonine (11.20 mg) and non-essential amino acids (184.65 mg) including glutamic acid (42.78 mg) and aspartic acid (32.44 mg) as major amino acids. Total content of amino acid derivatives was 187.85 me and that of hydroxyproline was highest as 121.50 mg.
