Isolation and Characterization of Double Deletion Mutants of nsdD and veA or nsdC, the Genes Positive Regulating Sexual Development of Aspergillus nidulans (Aspergillus nidulans에서 유성분화 능동조절 유전자 nsdD와 veA 또는 nsdC의 이중결실돌연변이의 분리 및 분석)

  • Three genes, nsdD, nsdC, and veA are known to be necessary for positive regulating sexual development of Aspergillus nidulans. Since the mutants of those genes hardly form fruiting bodies in heterokaryons constructed by cross between two of them, it is difficult to isolate double mutants. In this work, double mutants of ${\Delta}nsdD$ ${\Delta}veA$ and ${\Delta}nsdD$ ${\Delta}nsdC$ were isolated using the characteristic of the nsdD deletion mutant that it could develop mature cleistothecia in hypoxic and low temperature culture condition. According to the phenotypes of double mutants, the nsdD gene controls the apical growth independently with veA or nsdC. Deletion of veA or nsdC was epistatic to nsdD deletion for pigment production. Conidia formation in submerged culture with lactose as sole carbon source was observed in ${\Delta}nsdD$ ${\Delta}nsdC$ double mutant implicating it to be unique phenotype of nsdC deletion.

  • The nsdD gene has been predicted to encode a GATA type transcription factor with the type IVb zinc finger DNA binding domain functions in activating sexual development of A. nidulans. In several allelic mutants of nsdD producing truncated NsdD polypeptides lacking the C-terminal zinc finger, the transcription level of nsdD gene was greatly increased. Also in an over-expressed mutant, the transcription under its own promoter was reduced. These results suggest that the expression of nsdD is negatively autoregulated. When the nsdD gene was over-expressed, cleistothecia were formed in excess amounts even in the presence of 0.6 M KC1 that inhibited sexual development of the wild type. Northern blot analysis revealed that the expression of nsdD was repressed by 0.6 M KC1. These results strongly suggest that the inhibition of sexual development by salts was carried out via the nsdD involved regulatory network.

  • Several mutants which never underwent to sexual development(NSD) of Aspergillus nidulans were analyzed genetically and physiologically. They were divided into two groups according to their characterisitics of asexual development after release from aeration block. The mutants in first group proceeded asexual development immediately after removal of aeration block, while those in second group did 10 hours or more later. The NSD mutants were separated into 4 complementation groups, nsdA, nsdB, nsdC and nsdD. The nsdA and nsdD genes were linked to AcrA1 on linkage groupⅡ and pabaA1 on linkage group I, respectively. The mutant alleles were all recessive to wild type allele in heterokayon state. The mutants did not developed cleistothecia on any of carbon sources, except NSD208 which developed cleistothecia on lactose.

  • A lot of mutants which cannot initiate sexual development were screened and several loci including nsdA, nsdB, nsdC, and nsdD were identified in homothallic ascomycetes Aspergillus nidulans. The NSD206, which has nsdC6 allele, showed typical phenotype of NSD (Never in sexual development) mutants. The nsdC gene was cloned by transforming NSDP697 ($nsdC^-$, $pryG^-$) with AMA1-NotI genomic library. The transforming library DNA recovered from several transformants showing wild phenotype carried about 10 kb genomic DNA insert. The DNA sequence of nsdC was analysed using GPS (Genome priming system). The nsdC gene has an open reading frame (ORF) of 1,929 bp encoding a putative polypeptide of 643 amino acids. The NsdC carries $C_2H_2C_2H_2C_2HC$ type zinc finger DNA binding domains in the middle of the polypeptide. A coiled-coil domain at its C terminus were also found. In nsdC6 allele, a single T insertion was occurred between 407-408 bp leading to the frameshift mutation and early termination of translation producing the truncated protein which has only 139 amino acids.

Effect of High CO2 Concentration on Activation of Sexual Development in Aspergillus nidulans (고농도 CO2 노출에 의한 Aspergillus nidulans의 유성생식 촉진효과)

  • Fungal development is largely affected by many environmental factors. In a model filamentous fungus Aspergillus nidulans, asexual development is promoted by exposure of light, presence of salt and non-fermentable sugars. In other hand, sexual development is largely induced by absence of light, fermentable sugars and hypoxic condition. Also, some important genes including veA and nsdD play positive roles in activating sexual development. Here, we reported that the effect of high concentration of $CO_2$ on developmental decision in A. nidulans. When wild-type $veA^+$ strain was cultured in normal condition, sexual and asexual development occurred in balanced manner. However, high concentration of $CO_2$ (~5%) strongly activated sexual development and inhibited asexual development. Furthermore, this $CO_2$ effect was controlled by the veA or nsdD gene. High $CO_2$ culture of $veA^-$ or $nsdD^-$ mutant didn't activate sexual development, suggesting that the activation of sexual development induced by high $CO_2$ cannot overcome the genetic requirement of sexual development such as veA or nsdD. Since 5% $CO_2$ is an important condition for human pathogenic fungi for surviving and adapting in human body, this developmental pattern of A. nidulans affected by $CO_2$ concentration may provide interesting clues for comparative study with human fungal pathogens including Aspergillus fumigatus.

  • A No Spin Differential (NSD) design has been improved from evaluation of two NSD models utilizing the axiomatic approach. New design parameters of the second level are developed to satisfy the independence axiom. The design matrices are determined to decouple the relationship between design parameters and process parameters. The values of process parameters are then determined to optimize and improve the NSD design. Consequently a unique and evolutionary NSD design is achieved with the aid of the axiomatic approach.

  • A GAL4 type transcription factor gene (formally annotated as AN3912) locating downstream of sndA (AN3911) was characterized. The putative transcription factor carries both Zn(II)2Cys6 binuclear cluster DNA-binding domain and transcription activator domain. The gene named gtfA (gal4 type transcription factor) had an open reading frame which consisted of 762 amino acids and was disrupted by three introns. The deletion mutant produced reduced amount of conidia but increased amount of fruiting bodies, suggesting that the GtfA make function in decision of asexual preferential to sexual development. The forced over expression of gtfA caused the retardation of fruiting body formation on high glucose concentration. The transcript level of gtfA was kept constant through the life cycle except late vegetative stage and early sexual development stage during which slight increase was found. The expression of gtfA was not significantly affected by sexual or asexual development regulators, such as VeA, NsdD or FluG, FadA, and SfaD. The GtfA repressed the nsdC transcription, which suggested that GftA control sexual development negatively via negative regulation of nsdC expression.

Effects of steaming and drying processing on Korean rice wine (Makgeolli) with deodeok (Codonopsis lanceolate)

  • The physicochemical properties of deodeok (Codonopsis lanceolate) was investigated in relation to the different steaming time and cycles of steaming and drying (S/D). Additionally, the quality characteristics of Makgeolli with different amount (0-0.45%) of steaming and drying deodeok (SD) were measured comparison to non-steaming and drying deodeok (NSD). L⁎ values of deodeok tended to decrease as the number of S/D cycles and steaming times increased, while BI showed the opposite trend for L⁎ values. Reducing sugar increased significantly from 1 to 3 S/D cycles and decreased thereafter (p<0.05). Also, processed with steaming for 4 h and 5 S/D cycles had the highest antioxidant properties. Principal component analysis (PCA) revealed that the S/D process notably influenced the properties of deodeok. Quality characteristics of Makgeolli showed that 0.45% SD resulted in higher antioxidant properties than control or NSD.

  • Two No Spin Differential(NSD) models were benchmarked for a project of Dual-Use Technology. The Axiomatic approach is utilized to evaluate the designs of the models. The Independence Axiom is satisfied at the top level of design but not at the second level, which implies the design exhibits coupling and will admit design improvements. The detailed process of design evaluation is described. It is shown that it is possible to develop a unique and evolutionary NSD design by solving the problems that cause coupling within two models.
