Effects of Ginseng Total Saponin on Caffeine-induced Stimulation of Locomotor Activity and the Related Brain Catecholamine Contents in Mice (인삼사포닌이 생쥐에 있어서 카페인으로 유도된 보행활동과 뇌 카테콜아민 함량에 미치는 영향)

  • This study was undertaken to investigate the effect of ginseng total sapoin (GTS) on locomotor activity that had been increased by caffeine. Catecholamines, noradrenaline and dopamine, possible mediators for the locomotor activity, were measllred in the mouse whole brain, cortex and the re- mainder. The locomotor activity was measured in circlllar activity cages equipped with six light sources and photocells. The catecholamine contents in the mollse brain were determined by HPLC-fluorescence detection. GTS (50 and 100 mg/kg) reduced the increased locomotor activity by caffeine (25 mg/kg) dose-devendently. Caffeine increased the norevinephrint and dopamine in mouse whole brain and cortex dose-dependently. GTS reduced the norevinevhrine in the remainder, and reduced the dopamine in the cortex which had been increased by caffeine.

Effect of an herbal formulation on DOCA-salt and fructose induced models of hypertension in rats

  • The present study was carried out to investigate the antihypertensive effect of a folklore herbal formulation (HF) (300mg/kg/day; p.o.) in deoxycorticosterone acetate (DOCA)-salt induced and fructose induced hypertensive rats. In DOCA model, DOCA (15 mg/kg, s.c., twice a week) was administered to unilateral nephrectomized rats for 4 weeks. In fructose model, drinking water was replaced with 10% fructose solution for 6 weeks to induce hypertension. Systolic blood pressure (BP) was measured once every week during the treatment schedule. After completion of treatment schedule, BP and vascular reactivity to various agonists like Noradrenaline, Adrenaline, Phenylephrine and Serotonin (5-hydroxytrptamine; 5-HT) were recorded in rats of both models. A cumulative concentration response curve of 5-HT was carried out in isolated rat fundus strip of the DOCA-salt induced and fructose induced hypertensive rats. The results tend to suggest that HF possesses antihypertensive activity.

  • Abnormality in the central noradrenergic system may be related to the seizure prone state in the genetically epilepsy prone rats (GEPR). The present work deals with the characterization of the deficit in noradrenergic system if susceptitibility and intensity of seizure are dependent on central noradrenregic activities by comparing the activities of dopamine $\beta$-hydroxylase (DBH) which hydroxylates dopamine into noradrenaline. DBH activities were measured in 5 areas of brain of normal rats, native GEPR and severe GEPR. The results suggest that lower DBH activities in the midbrain of GEPRs may positively be coupled to the susceptibility to seizure, whereas the same characteristics of the native or severe GEPR are not neccessarily in parallel with the intensity of seizure.

  • The vocal fold has three major function-phonation, respiration and protection, and is richly innervated. The vocal 1314 its autonomic innervation-adrenergic and cholinergic from superior cervical ganglion and the vagus nerve, respectively. The action of both system account for vasoregulation and glandular activity. In e vocal fold several kin of neuropeptides, including SP, CGRP, VIP, TH, NPY, ENK have been reported at the animal including cat or dog. But information regarding the distribution of autonomic nerve fibers containing neuropeptides in the human vocal fold is lacking. Two neuropeptides are of special interest : 1) vasoactive intestinal polypeptide(VIP)that is known to be contained in the parasympathetic(cholinergic) neuron. 2) tyrosine hydroxylase(TH)is located in the cytoplasm of noradrenergic neuron and is the rate-limiting enzyme in noradrenaline synthesis. To understand specific autonomic function of vocal fold we did immunohistochemical examination of VIP and TH in the human vocal fold.

The Effects Liquid Stick Packs of Dongshingihyeolyangsubang on Stress and Sleep-Related Substance of Rats Induced by Chronic Mild Stress (동신기혈양수방(東新氣血養睡方) 액상 스틱 파우치가 Chronic Mild Stress 유발 흰쥐의 스트레스 및 수면 관련 호르몬에 미치는 영향)

  • Objectives: This study evaluates anti-stress and sleep-inductive effects of Dongshingihyeolyangsubang (DSGYSB) on rats induced by chronic mild stress (CMS). Methods: Twenty-five healthy rats were randomly divided into five groups: normal, CMS (Control), DSGYSB50, DSGYSB100, and DSGYSBS200. All rats except the normal group were exposed to unpredictable stress conditions such as water deprivation, empty bottles, and forced treadmill. A week after starting the experiment, rats in DSGYSB50, DSGYSB100, and DSGYSB200 groups were fed orally with water once a day for two weeks. Then blood samples were taken from the rats for analysis of complete blood count, AST, ALT, and glucose. Noradrenaline, corticosterone, serotonin, GABA and melatonin were measured by ELISA kit. BDNF, CREB, TrkB and $TNF-{\alpha}$ were measured by RT-PCT. Results: In Noradrenaline contents, the DSGYSB200 group revealed significant decrease compared to the control group. In corticosterone contents, the DSGYSB200 group revealed significant decrease compared to the control group. In serotonin contents, the DSGYSB200 group revealed significant increase compared to the control group. In GABA contents, the DSGYSB50, DSGYSB100, and DSGYSB200 groups revealed significant increase compared to the control group. In the activity of BDNF, the DSGYSB50, DSGYSB100 and DSGYSB200 groups revealed significant increase compared to the control group. In the activity of CREB, the DSGYSB100 and DSGYSB200 groups revealed a significant decrease compared to the control group. In the activity of TrkB, the DSGYSB100 and DSGYSB200 groups revealed significant decrease compared to the control group. In the activity of $TNF-{\alpha}$, DSGYSB100 and DSGYSB200 groups revealed significant decrease compared to the control group. In glucose contents, the DSGYSB100 and DSGYSB200 groups revealed significant decrease compared to the control group. In the leukocyte changes, white blood cells, neutrophil, lymphocytes, and monocyte significantly increased in the DSGYSB100 and DSGYSB200 groups than the control group. In the erythrocyte changes, hemoglobin significantly increased in the DSGYSB200 group than the control group. Conclusions: Results suggest that Dongshingihyeolyangsubang has anti-stress and sleep-inductive effects on rats induced by CMS.

Altered Functions of Adrenoceptors in Splanchnic Vascular Beds in Portal Hypertensive Rat Model: Effect of Propranolol (문맥 고혈압 흰쥐에 있어서 내장혈관의 아드레나린성 수용체의 기능변동과 이에 대한 Propranolol의 효과)

  • Alterations in splanchnic circulatory hemodynamics along with reactivities to the alpha adrenoceptor agonists were assessed in association with the preventive effects of propranolol 10 days after portal ligation. Decreases in precapillary resistance (Ra) and postcapillary resistance (Rv) along with increases in mesenteric blood flow (MBF) and capillary pressure (CP) were observed in conjunction with an increment of splenic pulp pressure (SPP). Dose-dependent increase in Rv in response to noradrenaline, increases in Ra and RV to adrenaline, and increases in superior mesenteric arterial pressure (SMAP), Ra and Rv to phenylephrine observed in sham group were significantly attenuated by portal vein stenosis. In PPL-3 group (propranolol 3 mg/kg, i.p. three times daily for 10 days), MBF was significantly decreased in association with decrease in mesenteric venous pressure (MVP) when compared with those of protal ligated (PL) group, and decreased Ra and Rv in PL group were recovered toward the values of sham group. Likewise, in PPL-1 group (propranolol 5 mg/kg, i.p. once daily for 10 days), the pressor response of Rv to adrenaline was recovered up to the level of sham group. Thus, it is suggested that decreases in Ra and Rv in association with increases in MBF and CP may have a close relevance to the increased SPP, and the changes in circulatory hemodynamics and vascular reactivities were effectively reversed by longterm propranolol treatment. Based on these results, it is concluded that these changes observed in portal hypertension are closely related with the altered functions of the adrenoceptors in the splanchnic vascular beds.

The Optimum Conditions for the Simultaneous Determination of Neurotransmitters in Rat Brain Striatum by High Performance Liquid Chromatography with Electrochemical Detection (HPLC-ECD를 이용한 흰쥐 뇌의 선조체 중 신경전달물질의 동시분석시 최적 조건)

  • A simple, efficient and sensitive method was described for the simultaneous determination of catecholamine, indoleamine and related metabolites from the homogenates of the rat brain striatum by HPLC-ECD. The optimum mobile phase on a reverse phase $C_{18}$ column was 35mM sodium acetate buffer(included 10mM citric acid, 0.13mM $Na_4EDTA$, 0.58mM SOS, pH3-4):MeOH=85:15. The column temperature was $30^{\circ}C$. Dopamine(DA), 3, 4-dihydroxyphenyl acetic acid(DOPAC), homovanilic acid(HVA), 5-hydroxyindole acetic acid(5-HIAA), serotonin(5-HT) and noradrenaline(NA) could be separated and analysed to very small amount. The detection limits of this method were 2~10pg per injection for all components. The effects of age and sex of rat on the contents of the catecholamines and their metabolites in rat brain striatum were studied. The levels of DA and 5-HT contents of the 7 week old female rats were higher than those of the 7 week old male rats. As the age of rat increases, the contents of DOPAC increased significantly.

The Effects of Dietary $MgSO_4$ Supplement on Serum Stress Hormones Concentrations and Pork Quality in Late Finishing Pigs (출하 전 마그네슘 단기급여가 돼지의 혈중 스트레스 관련 호르몬 함량 및 육질에 미치는 영향)

  • This study was conducted to determine the effects of MgSO₄ supplement on meat quality and serum stress hormones concentrations in late finishing pigs. Sixty castrated Landrace pigs (average weight±S.D., 106.5±8.9 kg) were assigned to a control diet (without MgSO₄ supplement) and diet with MgSO₄(10 g/kg diet) supplemented. Pigs had free access to diets and water during five-day feeding period. At the end of feeding experiment, pigs were fasted for 16 hours and transported to commercial slaughter plant (30-minute transportation distance). After 2-hour lairage time, pigs were slaughtered, and blood samples were collected at bleeding procedure of slaughter. Pigs fed the diet with MgSO₄ supplement had higher (p<0.05) serum magnesium concentrations than those in pigs fed a control diet. Serum adrenaline, noradrenaline and cortisol concentrations determined at slaughter were not significantly different between dietary groups. Carcass temperature, pH at 24h post- slaughter, drip loss and color characteristics were not influenced (p>0.05) by short-term supplement of MgSO₄. Pigs fed the diet supplemented with MgSO₄ diet had lower PSE incidence (23.3%) compared to pigs fed the control diet (33%). These results indicate that short-term supplement of dietary MgSO₄ in late finishing pigs may not be beneficial in improving pork quality, and further study needs to assess feeding regime of magnesium supplement.

Effects of Pinelliae Rhizoma Extract on Sociopsychological Stress (반하(半夏) 추출물이 생쥐의 사회.심리적 스트레스에 미치는 영향)

  • Objectives : The effects of Pinelliae Rhizoma extract(PRe) were tested for the anti-stress action. Methods : PRe was fed to ICR male mice($20{\pm}2g$) orally with the dose of 100 mg/kg/day for five days. Mice were exposed to sociopsychological stress by restraining and seeing foot shock stressed mice for one hour for five days. Results : PRe administration had the effect of decreasing serum level of lipid peroxidation. The elevated plus-maze test is designed to detect the effect of anxiolytic drugs, and PRe administration group showed a significant increase of latency time. From Microarray, common features between mind-stimulus related genes and body-stimulus related genes were not so abundant When PRe was administered, there were some changes in distributions of mind-stimulus related genes but the distributions were not recovered to normal status. Conclusions : These results suggest that PRe can effectively rid the sociopsychological stress and stress concrened diseases.

Effects of Cold and Hot Drugs on the Activity of Monoamine Oxidase (한성 및 열성한약재가 모노아민 산화효소의 활성에 미치는 영향)

  • To explain the theory of KIMI which is the theory of therapeutics in oriental medicine, monoamine oxidase(MAO) activities were measured in the brain and liver of mice which were orally administered oriental medicinal herbs which were classified into cold and hot drugs. Rheum palmatum, Anemarrhena asphodeloides, Gardenia jasminoides, Scutellaria baicalensis and Coptis japonica were considered as the cold drugs and Zingiber officinale, Aconitum carmichaeli, Asiasarum sieboldi, Evodia officinalis and Cinnamomum cassia were included in the hot drugs. The effects of cold and hot drugs on in vitro enzyme activities were measured and compared with the in vivo effects. Serotonin is important neurotransmetter involved in the control of body temperature. The MAO plays a central role in the metabolism of many neurotransmetter monoamines including serotonin. MAO is a flavoprotein found exclusively in the mitochondrial outer membrane, occuring in the MAO-A and MAO-B subtypes. MAO-A deaminates serotonin and noradrenaline, whereas MAO-B prefers phenylethylamine and benzylamine as substrates. Coptis japonica and Aconitum carmichaeli elevated the in vivo MAO activities and especialy, in vivo MAO-B activities were significantly increased. In vitro MAO-A activities were increased by hot drugs, whereas the in vitro MAO-B activities were inhibited. Cold drugs inhibited both enzyme activities in vitro.
