토양환경내 오염물질의 고정화 현상은 오염물질의 거동을 결정하는 주요 과정중 하나이다. 특히 생물화학적 반응에 대해 비가역적이며, 이로부터 오염물질의 독성도 동시에 제거되는 산화-공유결합반응에 의한 고정화 반응은 오염물질의 주요한 자연정화 메커니즘중 하나일 뿐만 아니라, 이를 공학적으로 응용함으로써 기존의 분해에 의존해 오던 정화 방법에 비해 보다 효과적으로 오염토양 및 지하수의 복원에 적용될 수 있다. 특히 이러한 산화-공유결합반응을 일으키는 촉매로서의 역할을 하는 망간산화물은 미생물 자체, 혹은 미생물을 포함한 균류, 식물체 등으로부터 추출한 산화-환원 효소를 이용하는 것에 비해 실용적인 측면에서 많은 장점을 가지고 있다. 이에 본고에서는 망간산화물을 이용한 유기오염물질의 정화 기작에 대한 최근의 다양한 연구 결과들을 정리하였다. 특히 망간산화물에 대해 반응성을 가지지 않는 안정한 유기오염물의 처리를 위한 관련 연구로서 반응매개체를 적용한 사례와, PAHs 처리기법, $Fe^0$를 이용한 환원 전처리 등 적절한 전처리 기법과의 조합에 의한 처리방법 등에 대한 연구결과를 소개하였으며, 이로부터 보다 광범위한 적용 가능성을 제시하고자 하였다. 자연계 내에서 일어나는 탄소의 순환과정을 고려할 때 산화-공유결합 반응에 의한 고정화 및 안정화 반응은 특히 분해에 대해 높은 내성을 가지는 방향족 유기오염물질의 제거에 보다 효과적으로 적용될 수 있는 친환경적 기법으로 사용될 수 있을 것이다.
Nasal-type extranodal natural killer (NK)/T-cell lymphoma (ENKL) is a highly invasive cancer with a poor prognosis. More effective and safer treatment regimens for ENKL are needed. Pegaspargase (PEG-Asp) has a similar mechanism of action to L-asparaginase (L-Asp), but presents lower antigenicity. The aim of the present research was to evaluate the safety profile and the latent efficacy of a PEG-Asp-based treatment regimen in patients with ENKL. Data collected from 20 patients with histologically confirmed ENKL, admitted to the Third Affiliated Hospital of Sun Yat-Sen University from January 2009 to August 2013, were included in the study. All patients received $2500IU/m^2$/IM PEG-Asp on day 1 of every 21-day treatment cycle. Patients received combination chemotherapy with CHOP (n=5), EPOCH (n=7), GEMOX (n=7) or CHOP with bleomycin (n=1). After 2-5 treatment cycles (median, 4 cycles) of PEG-Asp-based chemotherapy, five patients (25%) showed a complete response (CR), and the overall response rate (ORR) was 60%. Grade 3/4 neutropenia occurred in fourteen patients (70%). Grade 3 alanine aminotransferase (ALT) elevation was observed in two. Grade 1-2 non-hematological toxicity consisted of activated partial thromboplastin time (APTT) elongation (n=9), hypofibrinogenemia (n=6), hypoproteinemia (n=17), hyperglycemia (n=3), and nausea (n=6). No allergic reactions were detected. No treatment related death was reported. Our results suggested that PEG-Asp-based chemotherapy presented an acceptable tolerance and a potential short-term outcome in patients with nasal-type ENKL.

Background: Phytochemical compounds are emerging as a new generation of anticancer agents with limited toxicity in cancer patients. The purpose of this study was to investigate the potential effcts of thymoquinone, caffeic acid phenylester (CAPE) and resveratrol on inflammatory markers, oxidative stress parameters, mRNA expression levels of proteins and survival of lung cancer cells in Vitro. Materials and Methods: The A549 cell line was treated with benzo(a)pyrene, benzo(a)pyrene plus caffeic acid phenylester (CAPE), benzo(a)pyrene plus resveratrol (RES), and benzo(a)pyrene plus thymoquinone (TQ). Inflammatory markers, oxidative stress parameters, mRNA expression levels of apoptotic and anti-apoptotic proteins and cell viability were assessed and results were compared among study groups. Results: TQ treatment up-regulated Bax and down-regulated Bcl2 proteins and increased the Bax/Bcl2 ratio. CAPE and TQ also up-regulated Bax expression. RES and TQ down-regulated the expression of Bcl-2. All three agents decreased the expression of cyclin D and increased the expression of p21. However, the most significant up-regulation of p21 expression was observed in TQ treated cells. CAPE, RES and TQ up-regulated TRAIL receptor 1 and 2 expression. RES and TQ down-regulated the expression of NF-kappa B and IKK1. Viability of CAPE, RES and TQ treated cells was found to be significantly decreased when compared with the control group (p=0.004). Conclusions: Our results revealed up-regulation of the key upstream signaling factors, which ultimately cause increase in their regulatory p53 levels affecting the induction of G2/M cell cycle arrest and apoptosis. Overall these results provide mechanistic insights for understanding the molecular basis and utility of the anti-tumor activity of TQ, RES and CAPE.
Background: The objective of this research is the computed axial tomography (CT) imaging grading of radiation induced pneumonitis (RP) and its correlation with clinical and radiotherapeutic parameters. Materials and Methods: The chest CT films of 20 patients with non-small cell lung cancer who have undergone threedimensional conformal radiation therapy were reviewed. The proposed CT grading of RP is supported on solely radiological diagnosis criteria and distinguishes five grades. The manifestation of RP was also correlated with any positive pre-existing chronic obstructive pulmonary disease (COPD) history, smoking history, the FEV1 value, and the dosimetric variable V20. Results: The CT grading of RP was as follows: 3 patients (15%) presented with ground glass opacity (grade 1), 9 patients (45%) were classified as grade 2, 7 patients (35%) presented with focal consolidation, with or without elements of fibrosis (grade 3), and only one patient (5%) presented with opacity with accompanying atelectasis and loss of pulmonary volume (grade 4). Both univariate and multivariate analysis revealed as prognostic factors for the radiological grading of RP the reduction of FEV1 and the V20 (P=0.026 and P=0.003, respectively). There was also a significant (P<0.001) correlation of radiological grading of RP with FEV1 and V20 (spearman rho 0.92 and 0.93, respectively). Conclusions: The high correlation of the proposed radiological grading with the FEV1 and the V20 is giving a satisfactory clinical validity. Although the proposed grading scale seems relevant to clinical practice, further studies are needed for the confirmation of its validity and reliability.
Objective: The antiplatelet agent aspirin has been widely used for treating atherosclerosis in western medicine, and its efficacy has been proven in cardiac and extracardiac vascular diseases. On the other hand, Hyeolbuchukeo-tang has been widely used for treating blood stasis syndrome in traditional medicine. Therefore we investigated whether Hyeolbuchukeo-tang could have a synergic effect along with aspirin. Methods & Materials: Male $ApoE^{(-/-)}$ mice were randomly divided into three different experimental groups: a non-treated group(Control group), an aspirin-treated group(AP group), and an aspirin with Hyeolbuchukeo-tang-treated group(APH group). The control group was fed only an atherogenic diet, the AP group an atherogenic diet plus Aspirin 5 mg/kg, and the APH group an atherogenic diet plus Aspirin 5 mg/kg with Hyeolbuchukeo-tang 100 mg/kg. We investigated plasma lipid with liver function test, and performed the histological investigation of liver and abdominal aorta. Results: 1. We investigated photomicrographic changes of liver and abdominal aorta tissue. They showed that histological injury of aorta and lipid accumulations of the liver were lower in the AP and APH groups than in the control group. 2. In the APH group, plasma triglyceride levels were significantly lower than those in the control and AP groups. 3. There were no differences in aspartate aminotransferase and alanine aminotransferase levels among the control, AP and APH groups. Conclusion: The above results show that a combined treatment of Hyeolbuchukeo-tang and aspirin has a somewhat synergic effect in terms of inhibiting vessel injury and decreasing lipid deposits on liver cells without liver toxicity.
본 연구에선 바이오부탄올 생산을 위한 추출발효에 적용하고자 부탄올 추출에 효율적이고 미생물에 독성을 주지 않는 적합한 추출용매를 선정하고자 하였다. 추출 용매를 선정하기 위하여 부틸 부틸레이트, oleyl alcohol, 친환경 용매인 소수성 이온성 액체 2 종류를 이용하여 분배계수를 구한 결과, 부틸 부틸레이트와 oleyl alcohol이 높은 부탄올 분배계수를 나타내었고 부탄올 농도 1-20 g/L와 pH 4-5.5 범위에서 80% 이상의 우수하고 안정적인 추출효율을 나타내었다. 미생물에 독성이 없는 oleyl alcohol를 사용한 회분식 추출발효에서는 추출을 하지 않은 회분식 배양보다 11% 향상된 부탄올 생산을 보였다 (11.2 g/L vs. 12.4 g/L). 이는 배양액 내의 부탄올이 추출되어 배양액내의 부탄올 농도가 낮게 유지됨으로써 부탄올 독성 영향이 감소되었고, 이로 인해 향상된 부탄올 생산이 이루어진 것으로 판단된다. Oleyl alcohol:부틸부티레이트 부피비를 9:1로 혼합한 용매를 사용했을 경우는 미생물 생장에는 저해 영향이 미미했으나 부탄올 생산은 추출발효를 하지 않은 회분식 배양에 비해 60%에 그쳤다. 부틸부티레이트를 20% 이상 미생물 생장에 심각한 저해를 주는 것으로 관찰되어졌다. 본 연구를 통해, 실험에 사용된 4가지 추출 용매 중에서 oleyl alcohol이 미생물에게 독성영향을 끼치지 않으면서 부탄올 추출을 효율적으로 할 수 있음을 알 수 있었고, oleyl alcohol을 바이오 부탄올 연속 추출발효에 적용한다면 향상된 부탄올 생산성을 얻을 수 있을 것으로 기대된다.
내분비계 장애 추정물질로 알려진 alachlor의 어류 생체내 호르몬에 미치는 영향을 검색하고자 송사리를 이용한 급성독성, 수중 분해성 및 단기번식독성시험을 수행하였다. 시험결과 96시간 $LC_{50}$은 2.37(1.994~2.805) mg/L 이었으며, 수중 분해성이 15일에 초기 처리농도에 비해 20% 정도 분해되었으므로 단기번식독성시험의 시험용수 교체는 매 7일로 하였다. 송사리를 이용한 21일 단기번식시험의 노출농도는 용매대조군, alachlor(0.02, 0.04, 0.11, 0.27, 0.68ppm)처리군, 양성 대조군($17{\beta}$ estradiol, 0.01, 0.1, 0.5ppb)을 두어 수행한 결과 산란 수는 대조군에 비해 최고투여 농도에서 47.9% 감소하였고, 유정란 율은 47.8% 감소하였다. 기형율도 부분적으로 농도 의존성 경향이 관찰되었다. 또한 내분비계 장애성 중 에스트로겐 영향을 검색할 목적으로 어체내 vitellogenin 농도 변화를 조사한 결과 유의성이 나타나지 않았으므로 alachlor는 vitellogenin성 에스트로겐은 아닌 것으로 판단되었다.
Purpose: The Gated cardiac blood pool scan is non-invasive method that a quantitative evaluation of left ventricular function. Also this scan have shown the value of radionuclide ejection fraction measurements during the course of chemotherapy as a predictor of cardiac toxicity. Therefore a reliable method of monitoring its cardiotoxic effects is necessary. the purpose of this study is to minimize the overestimate of left ventricular ejection fraction (LVEF) by modified body position to reduce the influence of scattered rays from surrounding organs of the heart in the background region of interest. Materials and Methods: Gated cardiac blood pool scan using in vivo $^{99m}Tc$-red blood cell (RBC) was carried out in 20 patients (mean $44.8{\pm}8.6$ yr) with chemotherapy for a breast carcinoma. Data acquisition requires about 600 seconds and 24 frames of one heart cycle by the multigated acquisition mode, Synchronization deteriorates toward the end of the cycle and with the distance from the trigger signal (R-wave) by ECG gating. Gated cardiac blood pool scan was studied with conventional method (supine position and the detector head in $30-45^{\circ}$ left anterior oblique position and caudal $10-20^{\circ}$ tilt) and compared with modified method (left lateral flexion position with 360 mL of drinking water). LVEF analysis was performed by using the automatically computer mode. Results: The ROI counts of modified scan method were lower than LV conventional method ($1429{\pm}251$ versus $1853{\pm}243$, <0.01). And LVEF of modified method was also decrease compared with conventional method ($58.3{\pm}5.6%$ versus $65.3{\pm}6.1%$, <0.01). Imaging analysis indicated that stomach was expanded because of water and spleen position was changed to lateral inferior compared with conventional method. Conclusion: This study shows that the modified method in MUGA reduce the influence of scattered rays from surrounding organs. Because after change the body position to left lateral flexion and drinking water, the location of spleen, left lobe of liver and stomach had changed and they could escaped from background ROI. Therefore, modified method could help to minimize the overestimate LVEF (%).
본 연구는 섬오갈피 뿌리 추출물을 이용하여 돌연변이 유발을 관찰하기 위해 S. typhimurium TA100, TA98, TA1535, TA1537과 E. coli WP2 uvr A를 이용해 Ames test을 하였고 또한 S. typhimurium TA100, TA98을 이용한 항돌연변이원 억제 시험을 시행하였다. Ames test에 필요한 시험물질들은 최고농도 결정을 통해 $5000\;{\mu}g$/plate, $2500\;{\mu}g$/plate, $600\;{\mu}g$/plate의 시험물질을 양성대조군, 실험군, 음성대조군을 비대사활성계와 대사활성계로 나누어 시험을 시행하였다. 시험 결과 모든 농도에서는 집락군의 일관성 있는 증가는 보이지 않았고 이 점으로 미루어 보아 복귀돌연변이는 일어나지 않았고 음성으로 판정하였다. 항돌연변이원 시험에서는 양성물질의 농도결정과 시험물질의 최고농도 결을 통해 양성대조군, 실험군, 음성대조군을 비대사활성계와 대사활성계로 시험을 하였고 시험결과 S. typhimurium TA100, TA98 두 균주 돌연변이 억제를 보였으며 TA98에서 더 높은 억제율을 보였다. 시험결과로 섬오갈피의 뿌리는 항돌연변이 억제효과에 탁월한 효과가 있음을 시사하였다.
이 연구는 ${\gamma}$선 및 x선 조사 시 AuNPs 입자의 방사선감수성의 향상을 생물물리학적으로 평가하기 위해 두 가지 실험을 시행하였다. 생물학적 방사선 감수성의 평가에 앞서 6시간, 12시간, 18시간, 24시간 incubation한 후 세포생존율(surviving Fraction, SF)값을 비교하여 AuNPs 입자의 독성 여부와 세포질에 입자의 균일 된 분포 여부를 확인 해 보았다. incubation time에 따라 세포생존율(surviving Fraction, SF)이 낮아지지 않음을 확인한 다음 ${\gamma}$선 및 x선 조사 후 Clonogenic assay 실시하고 세포생존율(surviving Fraction, SF)을 구하여 방사선 감수성을 비교 평가하였다. 방사선량 증가 시 세포생존율(surviving Fraction, SF)이 계속해서 낮아졌고 8Gy 조사 시 최대치로 약 30%의 차이가 있음을 확인하였다. 또한 Gate v6.1을 사용하여 몬테카를로 시뮬레이션 후 percent depth dose(PDD)를 구한 뒤 선량 평가하여 약 40% Dose Enhancement가 있음을 확인하였다. 두 가지 실험에 따라 생물물리학적으로 AuNPs 입자는 방사선 감수성이 향상되도록 하며 이는 nanoparticle을 이용한 방사선 병합 치료 시 치료 성적 향상에 큰 도움이 될 것으로 보인다.
