Folic acid supplementation prevents high fructose-induced non-alcoholic fatty liver disease by activating the AMPK and LKB1 signaling pathways

  • BACKGROUND/OBJECTIVES: The present study aimed to evaluate the effects of folic acid supplementation in high-fructose-induced hepatic steatosis and clarify the underlying mechanism of folic acid supplementation. MATERIALS/METHODS: Male SD rats were fed control, 64% high-fructose diet, or 64% high-fructose diet with folic acid for eight weeks. Plasma glutamate-pyruvate transaminase, glutamate-oxaloacetate transaminase, lipid profiles, hepatic lipid content, S-adenosylmethionine (SAM), and S-adenosylhomocysteine (SAH) were measured. RESULTS: The HF diet significantly increased hepatic total lipid and triglyceride (TG) and decreased hepatic SAM, SAH, and SAM:SAH ratio. In rats fed a high fructose diet, folic acid supplementation significantly reduced hepatic TG, increased hepatic SAM, and alleviated hepatic steatosis. Moreover, folic acid supplementation in rats fed high fructose enhanced the levels of phosphorylated AMP-activated protein kinase (AMPK) and liver kinase B (LKB1) and inhibited phosphorylation of acetyl coenzyme A carboxylase (ACC) in the liver. CONCLUSIONS: These results suggest that the protective effect of folic acid supplementation in rats fed high fructose may include the activation of LKB1/AMPK/ACC and increased SAM in the liver, which inhibit hepatic lipogenesis, thus ameliorating hepatic steatosis. The present study may provide evidence for the beneficial effects of folic acid supplementation in the treatment of non-alcoholic fatty liver disease.

포도주스의 보충섭취가 흡연성인의 혈압, 혈장지질 및 자유 라디칼 생성에 미치는 영향 (The Effects of Purple Grape Juice Supplementation on Blood Pressure, Plasma Lipid Profile and Free Radical Levels in Korean Smokers)

  • Flavonoids contained in grapes are potent antioxidants that may protect against oxidative stress and reduce the risk of chronic diseases related with free radical damage. In this study we investigated the effect of daily grape juice supplementation on blood pressure (BP), plasma lipid profiles and the generation of free radicals in 67 healthy volunteers (29 smoker, 38 nonsmokers). The daily 480 ml of grape juice supplementation for 8 weeks resulted in a significant decrease in diastolic BP by 6.5% in smokers and systolic and diastolic BP by 11.2 and 3.7% in non-smokers. Plasma total cholesterol, HDL- and LDL-cholesterollevels in smokers and total cholesterol in non-smokers were significantly increased after the intervention. Plasma triglycerides and conjugated dienes were not affected by grape juice supplementation. Levels of free radical determined by reading the lucigenin-perborate ROS generating sources, decreased significantly by 18% compared to the beginning of the study. The results indicated that the consumption of grape juice may reduce BP and free radical generation in smokers, which was possibly exerted by flavonoids. Our findings suggested that the grape juice has protective effect on chronic disease due to the overproduction of free radical in smokers.

손바닥선인장 복합물이 Streptozotocin으로 유발된 당뇨 쥐의 지질대사에 미치는 효과 (Effects of Opuntia ficus-indica Complex on Lipid Metabolism in Streptozotocin-induced Diabetic Rats)

  • This study was conducted to examine the effects of Opuntia ficus-indica complex (OF) on the lipid metabolism, bile acid in feces, alanine aminotransferase (ALT) activity, aspatate aminotransferase (AST) activity, composition of urine and expression of cholesterol related mRNA in streptozotocin (STZ)-induced diabetic rats. Thirty two male Sprague-Dawley rats were randomly divided into non-diabetic control (NC), diabetic control (DC), diabetic OF of 2% (OF-2) and diabetic OF of 5% (OF-5), then each group was fed for 3 weeks. Plasma total cholesterol, non-esterified fatty acids (NEFA), total cholesterol, low density lipoprotein (LDL), very low density lipoprotein (VLDL) were decreased significantly (p<0.05) in OF-5 group compared to DC, but high density lipoprotein (HDL) was not changed. AST and ALT were also reduced and bile acid excretion was improved. Composition of urine in OF-5 was almost same in NC. The expression of cholesterol $7{\alpha}$-hydroxylase (CYP7A1), 3-hydroxy-3-methylglutaryl-coenzyme A reductase (HMG-CoA-R), Low density lipoprotein receptor (LDL-R) mRNA indicated that feeding OF have the effects of cholesterol decreation in plasma by synthesis of bile acid from cholesterol. These results provide experimental evidence about improved lipid metabolism of the OF feeding in the STZ-induced diabetic rats.

Effect of Aerva lanata against oxalate mediated free radical toxicity in urolithiasis

  • This study was undertaken to evaluate the antioxidant potential of A. lanata on oxalate mediated free radical toxicity in ethylene glycol induced calcium oxalate urolithic rats. Calcium oxalate (CaOX) stone was induced by 0.75% ethylene glycol in drinking water for 28 days. From $29^{th}$ day onwards, the CaOX urolithic rats were treated with A. lanata aqueous suspension (2,000 mg/kg body weight/dose/day) orally for another 28 days. At the end of experimental periods the animals were sacrificed, samples were collected and analyzed the lipid peroxidation product, protein oxidation product, enzymatic and non-enzymatic antioxidants in normal and experimental groups. Lipid peroxidation and protein oxidation products were significantly elevated while enzymatic and non-enzymatic antioxidant levels were significantly decreased in ethylene glycol induced CaOX urolithic rats when compared with control rats. The above alterations were reverted to near control in rats treated with aqueous suspension of A. lanata. This study suggests that A. lanata could prevent the free radical formation from calcium oxalate urolithiasis in rats and protecting the renal cells from oxidative injury.

  • The purpose of this study to examine lipid profiles and lipoprotein (a) concentrations and identify the risk factors of type 2 diabetes mellitus patients with macrovascular complications. The subjects were consisted of 618 out patients with type 2 diabetes mellitus from beginning of March through the end of April in 2001, who visited at the endocrinology department at Kangnam St. Mary's Hospital of Catholic University in Seoul. The patient's clinical laboratory data and the occurrence of chronic complications of diabetes were assessed at medical record review. The data were analyzed using for t-test. chi-square test and logistic regression. The results were as follows : 1. There were no significant differences in age. duration of diabetes. body mass index, sex, fasting blood glucose, 2-hour postprandial blood glucose and $HbA_{1}c$ level between macrovascular complication group and non macrovascular complication group. 2. There were significant differences in the level of total cholesterol. triglyceride. HDL(High density lipoprotein) cholesterol, LDL(Low density lipoprotein) cholesterol and lipoprotein(a) between macrovascular complication group and non macrovascular complication group. 3. Significant factor associated with macrovascular complication in the logistic regression best gut model was HDL cholesterol.

체육전공과 체육비전공 남자대학생의 식이섭취 실태조사 및 혈청 지질 함량 비교분석 (A Comparative Study on the Serum Lipid Levels and the Dietary Intake of Physical Education Major and Non-Major Male Students)

  • The paper examines the effect of physical exercise on blood lipid with young male students. Our analysis is based on the body measurement, food intake and blood lipid component for 70 male students of Kongju National University who either major in physical exercise (group A) or do not major in physical exercise (group B). The average weight of group A is 71.97 $\pm$ 10.79 kg while that of group B is 67.57 $\pm$ 8.66 kg. The former is significantly higher (p<0.05) than the latter. The SBP of group A (128.20 $\pm$ 11.40 mmHg) is significantly (p<0.001) higher than that of group B (136.27 $\pm$ 13.18 mmHg). In terms of the intake of total kcal, group A marks significantly higher than group B; group A goes with 2190.26 $\pm$ 581.96 kcal and group B goes with 2392.94 $\pm$ 769.03 kcal. The comparative analysis of the three nutrient intake shows that the intake proportions for group A are carbohydrate 55.08 $\pm$ 6.03%, protein 15.58 $\pm$ 2.75%, and lipid 29.34 $\pm$ 5.16% while the intake proportions for group B are carbohydrate 57.29 $\pm$ 8.09%, protein 15.62 $\pm$ 3.26%, and lipid 27.10 $\pm$ 6.90%. For group A, the vitamin B$_1$ intake amounts to 1.50 $\pm$ 0.5 mg which fulfills 100% of RDA, and the niacin intake amounts to 16.57 $\pm$ 5.54 mg N.E which is less than RDA. In contrast, both intakes for group B are more than the RDA. In the case with minerals except calcium, both group A and B mark more intake than RDA. Group A's calcium intake is 517.12 $\pm$ 200.63 mg and group B's is 409.56 mg. The total cholesterol intake for group A is 447.00 $\pm$ 245.08 mg which is significantly (p<0.05) higher than for group B with 352.35 $\pm$ 200.25 mg. The total cholesterol in the lipid of serum is 151.30 $\pm$ 12.92 mg/dl for group A and 182.30 mg/dl for group B, where the dominance of group B over group A is true at the level of significance 0.01. In the case with triglyceride, group A is 107.57 $\pm$ 31.60 mg/dl and group B is 108.07 $\pm$ 33.93 mg/dl so that the group difference is non-significant. The HDL-cholesterol for group A is 54.67 $\pm$ 6.49 mg/dl which is significantly higher (p<0.001) than for group B with 36.13 $\pm$ 4.64 mg/dl. We conclude that regular exercise may have an effect on blood lipid metabolism among young males.

마황과 의이인 혼합추출물이 비알콜성 지방간 모델에서 지질 축적 및 포도당 흡수에 미치는 효과 및 기전 연구 (A Study on the Effects and Mechanisms of the Combination Extract of Ephedrae Herba and Coicis Semen on Lipid Accumulation and Glucose Absorption in Non-Alcoholic Fatty Liver Disease)

  • Objectives: Ephedrae herba (EH) and Coicis semen (CS) has been frequently prescribed for the treatment of obesity. However, effects of combinational extracts of these two herbs on non-alcoholic fatty liver disease are unknown. The aim of the present study was to investigate the effects of EH and CS on lipid accumulation and glucose absorption in free fatty acids (FFAs) or palmitic acid (PA)-treated HepG2 cells. Methods: Five samples of EH and CS were extracted by combination ratios (S1=0:100, S2=25:75, S3=50:50, S4=75:25, S5=100:0). Oil Red O staining was used to measure lipid accumulation in FFAs-induced steatosis cells. Intracellular triglycerides and total cholesterol levels were measured in FFAs-induced steatotic HepG2 cells. In PA-treated cells, intracellular 2-NBDG was detected using a fluorescence microplate reader and flow cytometry. Phosphorylation of key metabolism-related factors of AMP-activated protein kinase and acetyl-CoA carboxylase, expression of key lipid synthesis-related factors carnitine palmitoyltransferase 1 alpha (CPT1α), sterol regulatory element-binding protein 1 (SREBP1), peroxisome proliferator-activated receptor gamma (PPARγ) and CCAAT enhancer binding protein alpha (C/EBPα) were confirmed by western blot. Results: Treatment of EH-CS combination in the FFAs-induced steatotic HepG2 cells significantly reduced lipid accumulation. As the relative ratio of Ephedrae herba increased, the lipid-lowering effects of the combination were increased. However, S1 and S5 of Ephedrae herba and Coicis semen did not significantly reduce triglycerides and total cholesterol induced by FFAs. However, the combination of Ephedrae herba and Coicis semen restored glucose absorption in PA-induced HepG2 cells. Major makers of SREBP1, PPARγ, C/EBPα, and CPT1α expression tended to decrease with EH ratio. Conclusions: The EH-CS combination has advantages over sole EH and CS extracts in improving lipid and glucose metabolism in liver steatosis models.

Fermented Kochujang Supplement Shows Anti-obesity Effects by Controlling Lipid Metabolism in C57BL/6J Mice Fed High Fat Diet

  • The aim of the present study was to assess the anti-obesity effects of fermented kochujang supplement in C57BL/6J mice. Thirty mice were divided into 3 groups; normal diet control group (ND), high fat diet control group (HD), and high fat diet plus kochujang supplemented group (HDK). Results were as follows: 1. Fennented kochujang supplement in high fat diet decreased body weight and epidydimal and back fat weight compared to non-supplement in HD group. 2. Lipid content and blood glucose level were lower in HDK group than HD group. 3. Fermented kochujang supplement increased mRNA level of lipolytic genes such as acyl-CoA synthetase (ACS), carnitine palmitoyltransferase-1 (CPT-1), and uncoupling proteins-1 (UCP-1) expression, whereas decreased mRNA level of adipogenic genes such as acetyl CoA carboxylase (ACC) expression. These findings suggest that fermented kochujang supplement in high fat diet normalized body weight, epididymal and back fat weight, lipid content, and blood glucose levels through controlling lipid metabolism and provides basic information on the control of obesity.

쥐의 간암화 과정에서 타우린의 공급이 지질과산화물 함량, 생체방어 효소 및 세포막 안정도에 미치는 영향 (Effect of Taurine Supplementation on Lipid Peroxidation, Activities of Defense Enzymes and Membrane Stability During Rat Hepatocarcinogenesis)

  • The purpose of this study was to determine the effects of taurine supplementation on the hepatic lipid peroxidation, activiteis of defense enzymes and membrane stability during rat hepatocarcinogenesis. Hepatocarcinogenesis was induced by Solt & Farber modification. Lipid peroxide contents of carcinogen treated group which was not supplemented with taurine were lower than those of control group. This might be that peroxide is decreased because of the activation of detoxifing enzyme. Glutathione S-transferase(GST) activites of carcinogen treated groups were significantly (p<0.05) increased compared to those of control groups. The GST activities of group supplemented with taurine before treatment of carcinogen and during the all period of experiment were only less increased. In carcinogen treated groups, glutathione peroxidase(GPx) activites of groups supplemented with taurine were higher than those of non supplemented group. By carcinogen treatemtn, glucose 6-phosphatase(G6Pase) activites, index of membrane stability were decreased, but in carcinogen treated groups supplemented with taurine, they were less decreased. These results suggest that taurine supplementation seems to inhibit lipid peroxidation, to change the activities of defense enzymes and to prevent to membrane disintegration during chemically induced hepatocarcinogenesis.

The role of lipids in the pathogenesis and treatment of type 2 diabetes and associated co-morbidities

  • In the past decade, the incidence of type 2 diabetes (T2D) has rapidly increased, along with the associated cardiovascular complications. Therefore, understanding the pathophysiology underlying T2D, the associated complications and the impact of therapeutics on the T2D development has critical importance for current and future therapeutics. The prevailing feature of T2D is hyperglycemia due to excessive hepatic glucose production, insulin resistance, and insufficient secretion of insulin by the pancreas. These contribute to increased fatty acid influx into the liver and muscle causing accumulation of lipid metabolites. These lipid metabolites cause dyslipidemia and non-alcoholic fatty liver disease, which ultimately contributes to the increased cardiovascular risk in T2D. Therefore, understanding the mechanisms of hepatic insulin resistance and the specific role of liver lipids is critical in selecting and designing the most effective therapeutics for T2D and the associated co-morbidities, including dyslipidemia and cardiovascular disease. Herein, we review the effects and molecular mechanisms of conventional anti-hyperglycemic and lipid-lowering drugs on glucose and lipid metabolism.
