Changes in Non-cellulosic Neutral Sugar of Strawberry during Maturation (성숙중 딸기의 세포벽 비섬유성 중성당 변화)

  • The changes of non-cellulosic neutral sugar composition of strawberry during maturation were investigated. Arabinose, xylose, galactose and glucose were the main non-cellulosic neutral sugar of cell wall and increased until ripe stage. The main non-cellulosic neutral sugar of alkali soluble hemicellulose were arabinose, xylose, mammose, galactose. The contents of non-cellulosic neutral sugar of alkali soluble hemicellulose were increased during maturation.

  • This study was carried out to investigate the changes and composition of the non-cellulosic neutral sugars in cell wall of soybean sprouts during growth. The composition of non-cellulosic neutral sugars in cell of soybean sprouts was rhamnose, fucose, xylose, arabinose, mannose, galactose and glucose. The galactose content of cell wall was higher than other non-cellulosic neutral sugars, and was remarkably decreased during growth. The major non-cellulosic sugars of pectic substances were rhamnose, arabinose, and galactose. The arabinose content of pectic substance was increased in cotyledon and hypocotyl during growth. The contents of non-cellulosic neutral sugars were decreased in hypocotyl during growth. The galactose content of pectic substance was higher in cotyledon than those in hypocotyl, and was increased in cotyledon. The content of rhamnose was higher in ionically associated pectic substance than that in covalently bounded pectic substance. The major non-cellulosic neutral sugars of hemicellulose were glucose, rhamnose, arabinose and galactose. The galactose of hemicellulose was decreased remarkably during growth.

Changes in the Non-cellulosic Monosaccharides of Cell Wall Polysaccharides of Persimmon Fruits during Maturation and Postharvest (감과실의 성숙과 추숙중의 세포벽 다당류의 비섬유성 단당류의 변화)

  • Glucose, arabinose and total non-cellulosic neutral sugar contents of alcohol-insoluble substance were increased during maturation of persimmon, but arabinose, galactose and total non-cellulosic neutral sugar contents were decreased in soft persimmon. The main non-cellulosic neutral sugars of cell wall were galactose, arabinose and glucose. Arbinose and galactose contents were decreased during maturity and this tend was remarkable in soft persimmon. Pectic fraction contained $70{\sim}82%$ of uronic acid, and galactose, arabinose and uronic acid of pectic fraction were decreased. The main non-cellulosic neutral sugars of hemicelluloses were glucose, xylose, and galactose. Galactose was decreased during maturation and postharvest, and contents of non-cellulosic neutral sugar were decreased in soft persimmon.

  • This paper was carried out to investigate changes in chromatograms of polysacctatides and soluble pectins on Sephadex G-50 and non-cellulosic neutral sugars of polysaccharides isolated from cell wall of persimmon fruits treated with polygalacturonase and $\beta$-galactosidase in vitro. The chromatogram pattern of soluble pectins extracted from cell wall treated with $\beta$-galactosidase on Sephacryl S-500 column were similar to those of untreatment, but contents of soluble pectins treated with $\beta$-galactosidase were different from those of untreatment. The patterns of chromatograms In soluble pectins extracted from cell wall treated with polygalacturonase were more complex and lower molecular polymer than those of other cell wall-degrading enzyme treatments. Non-cellulosic neutral sugar of polysaccharides in fraction I of soluble material treated with polygalacturonase was rhamnose, those in fraction II were similar to those in fraction III and contents of arabinose, xylose and glucose were higher than contents of other non-cellulosic neutral sugars. Non-cellulosic neutral sugars of polysaccharides in fraction I in soluble material by $\beta$-galactosidase treatment were rhamnose, arabinose, galactose and mannose. Content of glucose of polysaccharides in fraction II was higher than that in fraction I . Non-cellulosic neutral sugars treated with mixed enzyme were rhamnose, fucose, arabinose, xylose, mannose, galactose and glucose. Compositions of non-cellulosic neutral sugars of polysaccharides in fraction I were similar to those in fraction II and III.

  • This paper was performed to investigate the changes of non-cellulosic neutral sugars composition in cell wall of persimmon fruit by treatment of cell wall degrading enzyme in vitro. Rhamnose, xylose and galactose in cell wall by polygalacturonase treatment, arabinose, galactose and rhamnose in cell wall by mixed enzyme treatment and arabinose and galactose in cell wall by ${\beta}-galactosidase$ treatment decreased, respectively. Noncellulosic neutral sugars of pectins extracted cell wall by enzyme treatments decreased and those by polygalacturonase treatment decreased remarkably. Rhamnose, arabinose and xylose in hemicellulose I of cell wall by polygalacturonase treatment were higher than those of untreated, and rhamnose and xylose in that by ${\beta}-galactosidase$ treatment were higher but arabinose, mnnose and galactose decreased. Xylose, mannose and glucose in that by mixed enzyme treatment were higher than those of untreatment and arabinose and galactose decreased. Contents of total non-cellulosic neutral sugars in hemicellulose of untreatment, and contents xylose, and glucose in hemicellulose II of cell wall by polygalacturonase treatmet decreased but those of other treatments were not changed.

  • This study was carried out to investigate changes in cell wall components and solubilization and depolymerization of pectin and neutral sugar polymers during softening of 'Tsugaru' apples. Pectic polysaccharides were solubilized in different solvents, distilled-water, 0.05 M CDTA, 0.05 M $Na_2CO_3$, and 8 M KOH, from cell wall materials during fruit softening. The uronic acid contents in distilled-water fraction rapidly increased along with fruit softening at 4 weeks after ambient storage. In the change of non-cellulosic neutral sugars in the cell wall of ‘Tsugaru’ fruits, the major sugar was galactose and arabinose in distilled-water, 0.05 M CDTA and 0.05 M $Na_2CO_3$ soluble fractions, and it was glucose, galactose and xylose in 8 M KOH fraction. Especially the change of galactose contents in distilled-water fraction was increased greatly along with fruit softening. When uronic acid polymers (UAP) and carbohydrate polymers (CP) in distilled-water fraction were filtered and separated using Sepharose CL-2B column, the high molecular UAP and CP were degraded to the low molecular ones from at harvest to softening fruit. Thus, the amount of high molecular polymers were greatly decreased along with fruit softening.

Changes in the Cell Wall Components and Glycosidases Activity during Development of Peach Fruits (복숭아 과실의 발육 중 세포벽성분 및 Glycosidase 활성의 변화)

  • This study was carried out to know whether $\beta$-galactosidase is directly important or not on fruit softening during the development of peach fruits compared to those in the stage stage. It was investigated that the flesh firmness, cell wall components, and the glycosidase activities of the peach fruits with a fast softening cultivar, 'Mibeakdo', a slow softening cultivar,'Yumyung'and a middle softening cultivar, 'Okubo$\beta$, at different developmental stages, on 13 May, 16 June, 16 July, and 5 August and on 28 August which harvested only 'Yumyung' fruits. In order to investigate the amounts of total sugar and non-cellulosic neutral sugar, the cell wall materials of each fruit were solubilized in distilled water, 0.05M CDTA, 0.05M Na$_2$CO$_3$, 4% KOH, and 24% KOH sequentially. During the fruit development, the fruit firmness of three cultivars decreased and the fruit firmness of 'Yumyung' was higher than that fo 'Mibeakdo' and 'Okubo' in the overall period. During the fruit development, the changes of total sugar amounts of each measured fractions were similar among peach cultivars. Arabinose and galactose were the predominant non-cellulosic neutral sugars in all the fractions including cell wall material of the three cultivars. There was an active relationship between the changes of flesh firmness in three cultivars and the mol % changes of rhamnose on 5 August which was the harvest date of 'Mibeakdo' and 'Okubo' fruits. The activity of soluble $\beta$-galactosidase was high at the early developmental stage and then dropped to a very low activity level in all cultivars. The activity of cell wall-bound $\beta$-galactosidase was high at the early developmental stage and then decreased continuously through the harvest date. In addition the changes of other glycosidase activities were similar among cultivars.

Effect of Aminoethoxyvinylglycine Dipping Treatment on Ethylene Production and Cell Wall Composition of 'Tsugaru' Apple Fruits during Cold Storage (Aminoethoxyvinylglycine 침지처리가 '쓰가루' 사과의 저온저장중 에틸렌발생과 세포벽성분들의 변화에 미치는 영향)

  • This study was conducted to determine the influence of postharvest dipping treatment with aminoethoxyvinylglycine (AVG) on ethylene production and composition of non-cellulosic neutral sugars in cell walls of 'Tsugaru' apple fruits during storage. Fruits were harvested on August 20, soaked in AVG 50 and 75 $mg L^{-1}$ solution for 5 minutes, and stored in cold storage chamber at $0{\pm}1^{\circ}C$ for 60 days. Fruit quality factor, ethylene productions, and cell wall component changes were investigated at 20 days interval. As a result, the fruit firmness and acid content were much higher in AVG treated fruits than those of untreated one during 60 days of cold storage. Ethylene production of AVG treated fruits was reduced to the level of 1/10 compared with untreated one. As to the change of non-cellulosic neutral sugars in the cell walls of 'Tsu- garu' fruits, the major sugar was arabinose and galactose in water, CDTA and $Na_2CO_3$ soluble fractions. The content of arabinose and galactose in untreated fruits increased as the softening of fruits was in progress, but the fruits treated with AVG showed a little change during storage, so it is predicted that these two cell wall compositional sugars were not solubilized by the treatment of AVG. Accordingly, the marketability of 'Tsu- garu' fruits could remarkably increase when soaking the fruits in AVG solution after harvest.

  • This paper was investigated to changes of cell components during drying for studies on the softening of jujube fruits. The contents of alcohol-insoluble material, cell wall and water-soluble material were not changed at 6 days of drying, but alcohol-insoluble materials and cell wall were decreased at 9 days of drying, however water-soluble materials were increased. Pectin and hemicellulose were not changed at 6 days of drying. Pectin and alkali-soluble hemicellulose were remarkable decreased at 9 days of drying, but acid-soluble hemicellulose was increased. Water-soluble pectin was not changed at 6 days of drying, but increased at 9 days of drying. EDTA-soluble and insoluble pectin were decreased after 6 days of drying. The non-cellulosic neutral sugars were not changed at 6 days of drying. The contents of arabinose, galactose and total neutral suger were decreased at 9 days of drying.

Phenolic Compounds and Antioxidant Activity in Cell Wall Materials from Deodeok (Codonopsis lanceolata) (더덕 부위별 세포벽 물질의 페놀성화합물과 항산화 활성)

  • This study measured the amounts of uronic acid, total sugars, non-cellulosic neutral sugars, phenolic compounds as well as antioxidants activity in cell wall materials (CWM) derived from different parts of deodeok (Codonopsis lanceolata). The values of the uronic acid (UA): neutral sugars (NS) ratio in polymers extracted from the CWM of the flesh and skin were 4 and 6, respectively. The total sugar contents of the flesh and skin were 788.6 and 824.9 ${mu}g/mg$ of CWM, respectively. Galactose and arabinose were the main noncellulosic neutral sugars. The chemical structure of five phenolic compounds from the CWM were analyzed and identified as vanillic acid, p-OH-benzaldehyde, vanillin, ferulic acid, and 8-O-4' diferulic acid by HPLC spectral data. Among them, p-OH-benzaldehyde, vanillin, and 8-O-4' diferulic acid were the first compounds identified from the deodeok. The content of 8-O-4' diferulic acid in the skin CWM was 56.1 ${mu}g/g$ AIR (alcohol insoluble residue). The ethanol-NaOH fractions from CWM had the highest oxygen radical absorbance capacity (ORAC) activities, followed by the AIR fractions and ethanol fractions.
