The Enhancement of Endotoxin-Induced Nitric Oxide Production by Elevation of Glucose Concentration in Macrophage

  • The production of nitric oxide (NO) and the expression of inducible nitric oxide synthase (iNOS) are known to be modulated by a variety of factors. Recent study showed that endotoxin-induced NO synthesis and iNOS expression were greatly enhanced by elevation of extracellular glucose concentration in murine macrophages. Although this was suggested to be due to the activation of protein kinase C (PKC) via sorbitol pathway, there was lack of evidence for this speculation. This study was performed to delineate the underlying intracellular mechanisms of glucose-enhancing effect on endotoxin-induced NO production in Raw264.7 macrophages. The levels of NO release induced by lipopolysaccharide (LPS) significantly increased by the treatment of glucose in a concentration dependent manner and also, this effect was observed in LPS-preprimed cells. Concurrent incubation of cells with PKC inhibitors, H-7 or chelerythrine, and LPS resulted in the diminution of NO production regardless of glucose concentration but this was not in the case of LPS-prepriming, that is, chelerythrine showed a minimal effect on the glucose- enhancing effect. PMA, a PKC activator, did not show any significant effect on glucose-associated NO production. Modulation of sorbitol pathway with zopolrestat, an aldose reductase inhibitor, did not affect LPS-induced NO production and iNOS expression under high glucose condition. And also, sodium pyruvate, which is expected to normalize cytosolic $NADH/NAD^+$ ratio, did not show any significant effect at concentrations of up to 10 mM. Glucosamine marginally increased the endotoxin-induced nitrite release in both control and high glucose treated group. 6-diazo-5-oxonorleucine (L-DON) and azaserine, glutamine: fructose- 6-phosphate amidotransferase (GFAT) inhibitors, significantly diminished the augmentation effect of high glucose on endotoxin-induced NO production. On the other hand, negative modulation of GFAT inhibitors was not reversed by the treatment of glucosamine, suggesting the minimal involvement, if any, of glucosamine pathway in glucose-enhancing effect. In summary, these results strongly suggest that the hexosamine biosynthesis pathway and the activation of PKC via sorbitol pathway do not contribute to the augmenting effect of high glucose on endotoxin induced NO production in macrophage-like Raw264.7 cells.

Empirical Analysis on Determinants of Air Pollution in China (중국의 대기오염 배출 결정요인에 대한 경험적 분석)

  • The rapid economic growth has brought tremendous pressure on the environment and caused severe air pollution in China. This study empirically examines causes of air pollution in China. Panel-corrected standard errors procedure (PCSE) was used to analyze major determinants of increasing or reducing emissions of sulfur dioxide (SO2) and nitrogen oxides (NOX) in 30 Chinese provinces. The estimation results show that SO2 emission is mitigated as per capita regional GDP increases, but the relation between emission of NOX and per capita regional GDP is found to have an inverse N-shaped curve, which implies that emission of NOX is ultimately expected to decline with economic growth. As for increasing factors of air pollutants, electricity consumption is a significant common source of SO2 and NOX emissions. Moreover, the results show that increment of coal consumption significantly affects emission of SO2 while increase of natural gas consumption reduce emission of SO2. On the other side, investment in energy industry, and investment on treatment of waste gases are determinants of mitigating emissions of SO2, but have no impact on NOX. Consumption of diesel, truck ratio and number of vehicles increase emission of NOX. Meanwhile, higher precipitation rate is a common determinant of mitigating emissions of SO2 and NOX. Policy implications are suggested in the conclusion.

The usability of Infraorbital Approach on the patients with Orbito-Zygomatic Fracture (안와-관골골절 환자에서 안와하 접근법의 유용성)

  • Purpose: There are various approaches for open reduction surgery for orbitozygomatic fractures. After the incisions are performed, patients might have various complications. In this point of view, we conducted a retrospective study of the usability of infraorbital incision on elderly patients, generally more vulnerable to scleral show and ectropion. Methods: In this study, 20 patients who are relatively more vulnerable to scleral show and ectropion through snap-back test having infraorbital incision were reviewed. We monitored the duration of the surgery, and complications resulted from the incision. We evaluated the scars with the researcher's observations and with the patient's level of satisfaction on a scale of 1 to 5. Results: On average, 5 minutes was spent between the skin incision and the fracture exposure. No case of scleral show and ectropion occurred. One year follow-up observation revealed 4 cases of visible scars, 9 cases of barely visible scars, and 7 cases of invisible scars. The subjective satisfaction level of the patients' scar is an average of 4.0. Conclusion: Infraorbital incision has many advantages. First, it can reduce the complications such as scleral show and ectropion. Second, it can shorten the operation time. Third, it can diminish incision-related scars.

The Effects of Demographic Characteristics and Psychological Characteristic on Make-up Behavior of Women (성인여성의 인구통계적 변인과 심리적 특성이 화장행동에 미치는 영향)

  • This study intends to examine the influencing factors in personal intention and show-off of make-up behavior. For the study, a questionnaire survey was conducted of 436 women in between their twenties to fifties residing in the Kyeongnam region. Obtained data were analyzed pearson's correlation coefficient and multiple regression analysis. The result is summarized as follows.; The influence on personal intention and show-off of make-up behavior can be described into 4 factors that are included in the decision making process of personal intention and show-off, in other words, the demographical characteristic, the body cathexis, the need and the clothing value. The demographic characteristics, the marriage status, the occupation status and the media contact had a direct influence on personal intention, and the educational background had a direct influence on show-off. The body cathexis did not have a direct influence on any subordinate factors of personal intention and show-off, but had indirect influence having the need as a parameter or had no influence having the clothing value as a parameter.

Catalytic Removal of Nitric Oxide in Oxygen-Rich Exhaust with Methane over Metal Ion-Exchanged Zeolites (금속이온교환 제올라이트 촉매상에서 메탄을 이용한 산소과잉 배출가스중의 NO 제거)

  • Selective catalytic reduction of nitric oxide by methane in the presence of excess oxygen was investigated over copper and cobalt ion-exchanged ZSM-5 zeolites. Copper ion-exchanged ZSM-5(Cu-ZSM-5) has the limitations for commercial applications to lean-bum gasoline and diesel engines due to low thermal stability and resistance to water vapor and sulfur dioxide. But cobalt ion-exchanged ESM-5(Co-ZSM-5) is more active at high temperatures and also stable to water vapor and sulfur dioxide for catalytic reduction of nitric oxide by methane. The catalytic activity of Cu-ZSM-5 for NO reduction increases with increasing temperatures, reaches the maximum conversion of 23.0% at 350\"C. and then decreases with higher temperatures. In the meantime catalytic activities of Co-ZSM-5 show the maximum conversion of 25.8% at $500^{\circ}C$ Therefore Co-ZSM-5 catalysts have higher thermal stability at high temperatures. Catalytic activities of both zeolites were remarkably enhanced with the existence of oxygen in the exhaust. It is noted that the catalytic activity of Cu-ZSM-5 decreases with the increasing concentration of methane while the catalytic activity of Co-ZSM-5 decreases with increasing contents of methane in the exhaust. This may imply the existence of different paths of NO reduction by methane in the presence of excess oxygen fur Cu-ZSM-5 and Co-ZSM-5 catalysts. For binary metal ionexchanged ZSM-5, the primary ion-exchanged metal may be masked by secondary ion-exchanged component, which plays the important role for catalytic activities of binary metal ion-exchanged ZSM-5, Therefore CuCo-ZSM-5 catalysts show the similar volcano-shaped curves to Cu-ZSM-5 catalysts between the activity and temperature. It Is interesting that the activities of CoCu-ZSM-5 catalysts indicate almost no dependence on the concentration of methane in the exhaust.aust.

Different Effects of Sound Stimuli on Performing Left-and Right-Hemispheric Tasks (뇌의 좌반구와 우반구의 작용에 미치는 음의 서로 다른 영향)

  • In order to examine the different effects of noise and music on mental tasks, an addition of figures as a left-hemispheric task and a pattern search as a right-hemispheric task were given to elementary school pupils under conditions of jet noise stimulus, music stimulus, and no-sound stimulus, respectively. Results showed that effects of music and noise stimuli during these tasks were significantly different. The subjects under music stimulus tended, when performing additions, to show occasional short periods in which they produced substantially less than their own average rate of work. This is due to interaction between calculation and hearing music in the left hemisphere, whereas there was no detrimental effects on the task of addition under noise stimulus as well as no-sound stimulus because the addition and the noise may be separately processed in different cerebral hemispheres. As effects of noise on performing search task, the subjects tended to show instantaneous agitations in their working curves. Since noise with no-meaning is processed in the right hemisphere, it may be explained as an interacting effect in this hemisphere.

KRG and its major ginsenosides do not show distinct steroidogenic activities examined by the OECD test guideline 440 and 456 assays

  • Background: Ginseng has been used as a traditional medicine for treatment of many diseases and for general health maintenance. Previously, we showed that ginseng did not demonstrate estrogenic property in ovariectomized mouse model. However, it is still possible that disruption of steroidogenesis leading to indirect hormonal activity. Methods: The hormonal activities were examined in compliance with OECD guidelines for detecting endocrine disrupting chemicals: test guideline (TG) No. 456 (an in vitro assay method for detecting steroidogenesis property) and TG No. 440 (an in vivo short-term screening method for chemicals with uterotrophic property). Results: Korean Red Ginseng (KRG) and ginsenosides Rb1, Rg1, and Rg3 did not interfere with estrogen and testosterone hormone synthesis as examined in H295 cells according to TG 456. KRG treatment to ovariectomized mice did not show a significant change in uterine weight. In addition, serum estrogen and testosterone levels were not change by KRG intake. Conclusion: These results clearly demonstrate that there is no steroidogenic activity associated with KRG and no disruption of the hypothalamic-pituitary-gonadal axis by KRG. Additional tests will be performed in pursuit of cellular molecular targets of ginseng to manifest mode of action.

Modulatory Effects of 21 kinds of Medicinal Herbs Including Herba Pogostemi (Agastache rugosa) on Nitric Oxide Production in Macrophage Cell line RAW 264.7 cells (곽향(Agastache rugosa)을 포함한 21종의 한약재가 대식세포주 RAW 264.7 세포의 nitric oxide(NO) 생산 조절에 미치는 효과)

  • Aqueous extracts were prepared from 21 medicinal herbs including Herba Pogostemi (Agastache rugosa) to examine their modulatory effects on NO production in mouse macrophage cell line RAW264.7 cells. While almost all medicinal herb extracts failed to show marked scavenging activities to NO produced by LPS stimulation, only Herba Pogostemi showed a rather strong induction of NO production in RAW264.7 cells without stimulation with LPS. When we treated the cell with $200{\mu}M\;of\;N^G-monomethyl-L-arginine\;(N^GMMA)$, a NOS2 inhibitor, a significant reduction in NO production could be observed. Moreover, a treatment of $100{\mu}M$ pyrrolidine dithiocarbamate (PDTC) led to about a 79% reduction of NO production. These results demonstrated that the aqueous extract of Herba Pogostemi might provide a second signal for the expression of NOS2 in RAW264.7 cells, and suggested that Herba Pogostemi induces NO production through L-argininedependent pathway.

  • In this paper, we show two ways of the time reparametrization of piecewise Pythagorean-hodograph $C^1$ Hermite interpolants. One is the time reparametrization with no shape change, and the other is that with shape change. We show that the first reparametrization does not depend on the boundary data and that it is uniquely determined by the size of parameter domain, up to the general cases. We empirically show that the second parametrization can cause the change of the shape of interpolant.

REGULAR INJECTIVITY AND EXPONENTIABILITY IN THE SLICE CATEGORIES OF ACTIONS OF POMONOIDS ON POSETS

  • For a pomonoid S, let us denote Pos-S the category of S-posets and S-poset maps. In this paper, we consider the slice category Pos-S/B for an S-poset B, and study some categorical ingredients. We first show that there is no non-trivial injective object in Pos-S/B. Then we investigate injective objects with respect to the class of regular monomorphisms in this category and show that Pos-S/B has enough regular injective objects. We also prove that regular injective objects are retracts of exponentiable objects in this category. One of the main aims of the paper is to draw attention to characterizing injectivity in the category Pos-S/B under a particular case where B has trivial action. Among other things, we also prove that the necessary condition for a map (an object) here to be regular injective is being convex and present an example to show that the converse is not true, in general.
