Analysis of Distances for MRI Scan to Maintain Pptimal Signal Intensity in a Surface Coil (MRI 검사 시 코일내 최적의 신호강도를 유지할 수 있는 거리의 기준 분석)

  • The purpose of this study is to statistically analyze the signal intensity pattern according distance from the surface coil center and to maintain optimal signal intensity under clinical circumstances where the surface coil centers are not correctly positioned due to various causes. The cylindrical fluid phantom was placed and moved from the coil center in vertical direction with 1 cm increments. The signal intensities were measured and compared. As a result, the signal intensity showed no significant difference within 4 cm and 1 cm from in T1 weighted images while in T2 weighted images the signal intensity was maintained up to 5 cm and lower 3 cm in the upper and lower direction from the coil center. In conclusion, to maintain the optimal signal intensity the target region should be located within the reference distances proposed in this study.

  • We demonstrated positioning accuracy of iRTK system, which has been developed by Korea Astronomy Observatory, using a low-price GPS receiver. Each four different baseline positioning tests using the iRTK system, we certified 1-3 meters level positioning accuracy of the iRTK system. While the iRTK is similar to conventional RTK at coverage and accuracy, the iRTK positioning carried out from data processing center. And also, the iRTK system has no limit of data communication coverage because of using wire/wireless Internet. But the iRTK system has a weakness of short available coverage within 5km. Therefore we discussed a plan to adopt VRS (Virtual Reference System) as completion of the iRTK system preparing nation-wide iRTK service in near future.

Manufacturing a Functional Bolus Using a 3D printer in Radiation Therapy (방사선치료에서 3D 프린터를 이용한 기능적 조직보상체의 제작)

  • Commercial plate bolus is generally used for treatment of surface tumor and required surface dose. We fabricated 3D-printed bolus by using 3D printing technology and usability of 3D-printed bolus was evaluated. RT-structure of contoured plate bolus in the TPS was exported to DICOM files and converted to STL file by using converting program. The 3D-printed bolus was manufactured with rubber-like translucent materials using a 3D printer. The dose distribution calculated in the TPS and compared the characteristics of the plate bolus and the 3D printed bolus. The absolute dose was measured inserting an ion chamber to the depth of 5 cm and 10 cm from the surface of the blue water phantom. HU and ED were measured to compare the material characteristics. 100% dose was distributed at Dmax of 1.5 cm below the surface when was applied without bolus. When the plate bolus and 3D-plate bolus were applied, dose distributed at 0.9 cm and 0.8 cm below the surface of the bolus. After the comparative analysis of the radiation dose at the reference depth, differences in radiation dose of 0.1 ~ 0.3% were found, but there was no difference dose. The usability of the 3D-printed bolus was thus confirmed and it is considered that the 3D-printed bolus can be applied in radiation therapy.

  • Due to the rise of international oil prices, with the continued increase of vessel operating costs, profitability has gradually deteriorated in the some case of South Korea ferry shipping. Therefore, it is necessary to improve the resistance performance of passenger ship. Goose neck bulb can be one of the methods to improve the resistance performance of passenger ship. Goose neck bulb has been applied to passenger ships operated in Europe and large cruise line. But there is no application example in the passenger ship to be operated on a regular basis in the sea near Korea. It is needed to provide reference data that can be applied efficiently goose neck bulb on the medium-sized passenger ships. This study, intended for the medium-sized passenger ship operated short international routes, presents the design of the hull form that goose neck bulb has been applied. And the resistance performance of the designed hull confirmed by numerical simulation. The numerical simulation is performed while changing the local shape of the goose neck depending on the bulb parameters. This study finds bulb parameters and their range that can affect the resistance performance. Thus, it is possible to provide a foundation to develop the optimal design technique and regression analysis on the resistance performance and goose neck bulb.

  • A 13-month old, intact female, miniature Schnauzer dog was presented with history of dermatitis, pruritus and pain of ear pinnae with delayed puberty. The basal serum growth hormone level was 0.01 ng/ml, which was much lower than the reference range (1.80-2.00 ng/ml). On the xylazine stimulation test, the dog was no responsive to administration of xylazine. The dog was diagnosed as isolated type pituitary dwarfism. Treatment was aimed to improve alopecia with oral melatonin. After initiation of treatment, alopecia of the ear pinnae has been improved.

Application of next generation sequencing (NGS) system for whole-genome sequencing of porcine reproductive and respiratory syndrome virus (PRRSV) (돼지생식기호흡기증후군바이러스(PRRSV)의 전장 유전체 염기서열(whole-genome sequencing) 분석을 위한 차세대 염기서열 분석법의 활용)

  • In the present study, fast and robust methods for the next generation sequencing (NGS) were developed for analysis of PRRSV full genome sequences, which is a positive sensed RNA virus with a high degree of genetic variability among isolates. Two strains of PRRSVs (VR2332 and VR2332-R) which have been maintained in our laboratory were used to validate our methods and to compare with the sequence registered in GenBank (GenBank accession no. EF536003). The results suggested that both of strains had 100% coverage with the reference; the VR2332 had the coverage depth from minimum 3 to maximum 23,012, for the VR2332-R from minimum 3 to maximum 41,348, and 22,712 as an average depth. Genomic data produced from the massive sequencing capacities of the NGS have enabled the study of PRRSV at an unprecedented rate and details. Unlike conventional sequence methods which require the knowledge of conserved regions, the NGS allows de novo assembly of the full viral genomes. Therefore, our results suggested that these methods using the NGS massively facilitate the generation of more full genome PRRSV sequences locally as well as nationally in regard of saving time and cost.

Performance of Newly Evolved Bivoltine Silkworm Hybrids of Bombyx mori with Reference to Hybrid Vigour

  • In the present study, hybrid vigour in terms of heterosis and over dominance of newly evolved bivoltine hybrids and their reciprocals raised from the promising newly evolved bivoltine breeds were calculated for the important parameters like pupation, cocoon yield, cocoon weight, shell weight, shell ratio, raw silk percentage, filament length and neatness. Through there was no significant difference among the hybrids for the characters pupation, cocoon yield and cocoon weight, the magnitude of heterosis and over dominance were varied significantly for these characters. However, significant differences were observed between new hybrids and control hybrid for the characters like cocoon shell weight, cocoon shell ratio, filament length and raw silk percentage. The results indicate that majority of new hybrids have shown higher heterosis and over dominance than control hybrid. The data also indicate that highest mean heterosis of all new hybrids was recorded for cocoon yield followed by filament length. Whereas highest mean over dominance was recorded for cocoon yield followed by cocoon weight. Negative mean heterosis and over dominance were recorded for the trait cocoon shell ratio. Based on the results, the importance of three hybrids viz., CSR2 ${\times}$ CSR4, CSR2 ${\times}$ CSR5 and CSR3 ${\times}$ CSR6 and their reciprocals (CSR4 ${\times}$ CSR2, CSR5 ${\times}$ CSR2 and CSR6 ${\times}$ CSR3) which exhibited significant heterosis and over dominance, for commercial exploitation was discussed.

Comparative Study of Gene Expression Profiles in Posterior Silk Glands of the Silkworm, Bombyx mori L.

  • We used serial analysis of gene expression (SAGE) approach to derive a profile of expressed genes of the posterior silk glands (PSG) and to create a reference for understanding gene cluster related to the mechanism of silk protein synthesis in the silkworm, Bombyx mori. We constructed a 3' SAGE library from the PSG of the fifth instar larvae of the silkworm. In total we obtained 2,406 SAGE tags, of which 682 were unique tags. Sorted by tag count number, 27 (4%) unique tags were significantly more abundant genes (ten or more times), whereas 445 (65%) unique tags were detected as single copies. The annotation of 682 unique SAGE tags revealed that 462 (68%) of the SAGE tag sequences represented known genes, whereas 220 (32%) of the tag sequences had no matches in SAGE map and silkworm EST databases. Of the 682 SAGE tags, the most abundant tag sequences were that of the fibroin light chain gene and the silk protein P25. In addition, we compared two relative abundance results of the SAGE and the EST approaches to verify whether their transcript quantitative aspects are significant or not. The comparative results of relative abundances of the fibroin H-, L- chain and P25 glycoprotein genes indicated that the quantitative approach based on SAGE tags is effective for quantitative cataloging and comparison of expressed genes in same organs. The SAGE tag information reported in this study would be useful for researchers in the field to analyze genes associated with silk processing mechanisms of insects.

Ureolytic Vibrio parahaemolyticus Isolated from the Kamak Bay of Yeosu, in 2002 and 2003

  • Five urease-positive Vibrio parahaemolyticus strains were isolated from Kamak Bay in Yeosu in 2002 and 2003. V. parahaemolyticus YKB4 and YKB14 were isolated from seawater, YFB20 from black rockfish (Sebastes schlegeli), and YFO2l and YFO22 from olive flounder (Paralichthys olivaceus). The five urease-positive strains (YKB4, YKB14, YFB20, YFO21, and YFO22) did not show hemolysin and protease activity, while they did alter in color (to red) as the bacteria grew in the urea broth medium. All samples showed identical biochemical characteristics as a reference strain, V. parahaemolyticus KCTC2471, except in urease production. The five urease-positive strains showed urease activities at a mid stationary phase, and their activity was maximal in the late stationary phase of their culture supernatant. The addition of urea to the Luria-Bertani (LB) broth medium significantly affected the initial production of urease of V. parahaemolyticus isolates. Mortality by urease-positive V. parahaemolyticus YKB4, YKB14, YFO2l, and YFO22 was significantly high, being$60-80\%$, while YFB20 only reflected a rate of $20\%$. Protease-positive V. parahaemolyticus FM39 and FM50 showed a $40\%$ and $60\%$ mortality rate, respectively. However, hemolysin-positive V. parahaemolyticus had no mortality, like the non-pathogenic V. parahaemolyticus KCTC2471, while V. vulnificus resulted in a $40\%$ mortality rate. Injection with urease-positive V. parahaemolyticus strains showed mortality within 12 hrs in mice, and the strains could be isolated from the dead mice.

Integration of in-situ load experiments and numerical modeling in a long-term bridge monitoring system on a newly-constructed widened section of freeway in Taiwan

  • The widening project on Freeway No.1 in Taiwan has a total length of roughly 14 kilometers, and includes three special bridges, namely a 216 m long-span bridge crossing the original freeway, an F-bent double decked bridge in a co-constructed section, and a steel and prestressed concrete composite bridge. This study employed in-situ monitoring in conjunction with numerical modeling to establish a real-time monitoring system for the three bridges. In order to determine the initial static and dynamic behavior of the real bridges, forced vibration experiments, in-situ static load experiments, and dynamic load experiments were first carried out on the newly-constructed bridges before they went into use. Structural models of the bridges were then established using the finite element method, and in-situ vehicle load weight, arrangement, and speed were taken into consideration when performing comparisons employing data obtained from experimental measurements. The results showed consistency between the analytical simulations and experimental data. After determining a bridge's initial state, the proposed in-situ monitoring system, which is employed in conjunction with the established finite element model, can be utilized to assess the safety of a bridge's members, providing useful reference information to bridge management agencies.
