여름느타리버섯과 느타리버섯의 균계(菌系) 생장(生長)에 영향(影響)을 미치는 몇가지 요인(要因)에 관한 시험(試驗) (Effect of Temperature, pH, Carbon and Nitrogen Nutritions on Mycelial Growth of Pleurotus sajor-caju (Fr.) Sing. and Pleurotus ostreatus (Fr.) $Qu{\acute{e}}l$.)

  • 여름느타리버섯 및 느타리버섯의 균사생장(菌絲生長) 적온(適溫)은$25{\sim}30^{\circ}C$이며 배지(培地)의 최적(最適) pH는 5{\sim}6이였다. 여름느타리버섯은 이당류(二糖類) 및 다당류(多糖類)가, 느타리버섯은 다당류(多糖類)가 균사생장(菌絲生長)에 가장 좋은 탄소급원(炭素給源)이었으며 질소급원(窒素給源)으로는 yeast extract등(等)의 복합(複合) 유기태질소(有機態窒素)가 단일(單一) amino산(酸) 및 무기태질소(無機態窒素)보다 좋았다. 단일(單一) amino산(酸) 중(中) asparagine이 가장 좋았으며 무기태질소급원(無機態窒素給源) 중(中) ammonium태(態) 질소(窒素)가 Nitrate태(態) 질소(窒素)보다 균사생장량(菌絲生長量)이 많았다. 균사생장(菌絲生長)에 알맞는 glucose와 ammonium tartrate의 비(比)는 여름느타리버섯이 각각(各各) 6% 및 0.6%, 느타리버섯이 4% 및 0.4%였다.

효모세포벽(酵母細胞壁) 용해효소(溶解酵素)의 미생물 생산(生産) (Microbial Production of Yeast Cell Wall Lytic Enzymes)

  • 1) 효모세포벽(酵母細胞壁) 용해활성(溶解活性)이 큰 미생물을 얻기 위하여 baker's yeast-peptone-bouillon agar 평판(平板)배지에서 투명하게 용해된 부위를 형성하는 균주(菌株)를 서울 및 경기지방의 토양 및 하수(下水)시료에서 156개 분리하였고 이들중 활성(活性)이 가장 큰 균주(菌株)로 K-42를 선발하여 Bacillus circulans로 동정(同定)하였다. 2) 균주(菌株) K-42에 의한 효모세포벽 용해효소의 생산을 보면 당류 첨가의 경우 배양 2일째에는 maltose>glucan>xylose>control의 순으로, 3일째에는 lactose>galactose>glucan>control의 순으로 나타났다. 무기 질소원의 경우는 배양2일째 ammonium acetat>sodium nitrate>control의 순으로, 3일째는 ammonium chloride>ammonium oxalate>control의 순으로 나타났으며 milk casein을 제외한 거의 모든 유기질소원은 배양 2일째 활성(活性)의 증가를 보였으나 3일째는 모두 감소하였다. 당류와 질소원의 배합첨가는 상승효과가 없었다. 3) 효소생산에 미치는 당류와 질소원의 첨가 효과는 배양기간 중 pH의 변화와 깊은 관계가 있었는 바 배양중 $pH\;7{\sim}8$을 계속 유지시켜 주면서 당류 또는 질소원을 첨가하면 높은 활성(活性)을 상당기간 계속 유지할 수 있었다. 4) K-42균주(菌株)가 분비(分泌)하는 세포벽(細胞壁) 용해효소의 작용최적(作用最適) 조건은 $pH\;7{\sim}8$, $60^{\circ}C$이었고 열처리(熱處理) 효모세포벽의 용해율은 65%이었다.

Growth and Amino Acid Contents of Spirulina platensis with Different Nitrogen Sources

  • The growth and amino acid contents of the cyanobacterium, Spirulina platensis strain NIES 46, were investigated using ammonium, nitrate, nitrite, or urea as the sole nitrogen source in a batch culture. Chlorophyll a concentration was highest at 2,096$\mu\textrm{g}$/L in the nitrate group after 10days of cultivation, while the dry weight of S. platensis was highest at 4.5g/L in the ammonium group after 30days of cultivation. The total amino acid content was highest at 174mg/g dry weight of S. platensis in the urea group at the end of the cultivation period, yet the amino acid patterns for S. platensis were similar for all the experimental groups. Therefore, it seemed that the growth and amino acid composition of S. platensis varied depending on the type of nitrogen sources, while the amino acid patterns were not changed. Also, the most efficient harvesting time for S. platensis seemed to be approximately 10 days after cultivation.

Loads of Nitrogen and Phosphorus from the Agricultural Watershed in Central Korea

  • Water quality monitoring network was established at the agricultural watershed located at the Namdae-chon watershed of Seolchon-myon, Muju-gun, Chollabuk-do, Korea which is 22,560 ha in size. Based on total amount of stream flow loads of nitrogen and phosphorus from the agricultural watershed were estimated. About 4.48 (1,011 ha), 7.02 (1,585 ha), and 86.82% (19,609 ha) of the site were used for paddy fields, upland fields, and forests, respectively. During the period of 6 months from May 1 to October 31, 1999, the total amounts of precipitation and stream flow were 993.2 mm and $148,533,000m^3$ respectively. The loads of agricultural non-point sources accrued by land use were 83,526 kg, 24,508 kg, 49,705 kg, and 215 kg for total-N, ammonia-N, nitrate-N, and total-P, respectively. Results showed that 23.4 and 0.1 % of the applied nitrogen and phosphorus fertilizers, respectively, were estimated to load into the streams as agricultural non-point sources.

질소 및 가리 급원이 들잔디(Zoysia japonica Steud.)의 생육 및 품질에 미치는 영향 (Effects of Nitrogen and Potassium Sources on the Growth and Quality of Zoysia japonica Steud.)

  • The purpose of the experiment was to determine the effect of nitrogen sources (urea, ammonium sulphate, ammonium nitrate) and potassium sources (potassium chloride, potassium sulphate) on the growth and quality of Zoysia japonica Steud. This experiment was conducted at Seoul City University turf field from 1988 to 1989. The results of this experiment were summarized as follows; 1. Urea and Ammonium sulphate resulted in superior clipping yield compared to ammonium nitrate. The growth of rhizome and stolon increased significantly with urea forms, but ammonium sulphate treated plots exhibited the highest the growth rate of root. 2. Ammonium sulphate showed best turf color rating while ammonium nitrate resulted in the poorest. Prolongation of the green period showed longer in ammonium sulphate and urea treated plots than ammoium nitrate. 3. Urea and ammonium sulphate exhibited superior visible quality and shoot density compared to ammonium nitrate. 4. The uptake of mineral nutrient showed the highest concentration with urea plots. Surface soil pH was allowed to become slightly acid with the ammonium sulphate and potassium sulphate treatments, while the application of N and K sources did not cause significant differences in mineral element content in soil.

영산강 하구역 퇴적물의 유기물 분포와 기원 (Organic Matter in the Sediments of Youngsan River Estuary : Distribution and Sources)

  • Total organic carbon(TOC), Total nitrogen(TN), and carbon and nitrogen stable isotopes were measured in the sediment and suspended parties in fresh lake water and saline estuarine water to determine the sources of Particulate organic matter(POM) in the sediments of the Youngsan river estuary. POM in the freshwater discharge water was mostly phytoplankton origin with little trace of terrestrial plants. POM from phytoplankton blooms formed in estuarine water in response to the nutrient enriched freshwater discharges was the most important sources of POM in the sediment near the dike, comprising more than 40% of the total organic matter. POM from freshwater phytoplankton and oceanic phytoplankton were also important sources of the sediment POM, and their contributions varied with the distances from the dike. Contribution of freshwater phytoplankton to sediment POM decreased from the dike to the outside of the estuary.

Assessment of Organic Matter Sources in the Singil Stream Flowing into Lake Shihwa, South Korea

  • The Singil Stream, flowing into an artificial lake, Lake Shihwa (South Korea), experiences a strong anthropogenic pressure with continuous organic matter (OM) inputs from rural, urban, and industrial areas. In this study, we investigated suspended particulate matter (SPM) and streambed sediments collected along the Singil Stream in 2014 and 2016, by applying a dual element approach (δ13C and δ15N) to identify OM sources. The SPM and streambed sediment samples from the indusrial area showed higher organic carbon and nitrogen concentrations (or contents) than those from the other areas, with distinctively lower δ15N values. Accordingly, our dual element approach indicates that the industrial area was the predominant OM source influencing OM quality and thus water quality of the Singil Stream flowing into Lake Shihwa during the study periods. However, further studies are necessary to better constrain OM sources in the Singil Stream since OM sources from the industrial area appear to be complex.

Effects of Nitrogen and Sodium on Growth in Phaeodactylum tricornutum (Bacillariophyceae)

  • Phaeodactylum tricornutum (Bacillariophyceae) is a marine diatom which has been supplied as a food of bivalves. In this study, growth responses of P. tricornutum to some nitrogen sources and sodium were investigated by measuring cell number and contents of chlorophyll a in culture. In medium with nitrogen and sodium, brisk cell division occurred and maximum growth rate was respectively found in the medium with 150 mg/l of nitrate and 10 mg/l of ammonium and urea. At 10-500 mg/l ammonium and urea and 200-500 mg/l nitrate, specific growth rate decreased slightly. However, no cell division observed in sodium-deficient medium, regardless of presence or absence of nitrogen. This suggests that sodium is required for the nitrogen uptake of P. tricornutum, resulting nitrogen uptake leading to cell division. Also the upper limits of ammonium and nitrate for the growth of P. tricornutum seem to be 10 mg/l and 500 mg/l, respectively.

소화성 궤양 흰쥐에서 체내 질소이용율 증진을 위한 체내 질소원에 관한 연구 - 단백질과 단백질 가수분해물의 비율을 중심으로 - (A Study on the Nitrogen Sources for the Enhancement of the Nitrogen Bioavailability in Rats with Peptic Ulcer -The Ratio of Casein and Casein Hydrolysate-)

  • This study aimed to verify the nutritional and curative effects of protein hydroysate and optimal ratio between protein and protein hydroysate as nitrogen source in rats with cysteamine-induced duodenal ulcer. Duodenal ulcer rat model was established by intraperitoneal injections of cysteamine. Sprague-Dawley, female rats weighing approximately 200g were intrapertionealy injected twice cysteamine(13mg/100g BW) at intervals of 3hours per day. This procedure was repeated 3 times at intervals of 3 days. Animals fed on 10% casein diet for injection periods. After last injection, 5 kinds of kiets (the ratio of casein and casein hydrolysate was 100 : 0(C100), 75 : 25(CH 25), 50 : 50(CH 50), 25 : 75(CH 75), 0 : 100(CH 100)) were given. The rate were sacrificed after feeding diet, 1, 3, 5 days. Ulcer index, hexosamine content of stomach and duodeum, gastric motility, trypsin activity, blood glutathione, plasma total protein, albumin, amino-N, urinalry urea nitrogen, creatinine, hydroxyproline and retention rate of nitrogen were analyzed for nutritional effects of diet treatments. There were no differences among diet groups in the view of the growth and diet treatments. There difference of ulcer curation by diet was appeared after 3 days. The ulcer indexes of C100 and CH 25 of 3, 5 days were significantly higher than those of CH 50, CH 75 and CH 100. This result was the same as hexosamine content of stomach, plasma protein, albumin concentration and nitrogen retention rate. The more casein hydrolysate diet had, the lower trypsin activity was. The more casein gydroysate diet had, the higher excretion of hydroxyproline was. These results show that protein hydrolysate can be applied in diet therapy for the patients with gastronitestinal ulcer. It suggests that it has curative effect of diet when nitogen sources include at least over than 50% of protein hydrolysate.

Effect of Nitrogen Sources on the Production of Polyols by Aureobasidium pullulans

  • Aureobasidium pullulans produced three kinds of extracellular polyols e.g. glycerol, mannitol, and sorbitol from either sucrose, glucose, fructose or mannose. Sorbitol was selectively produced when urea was used as a sole nitrogen source, and the amounts of sorbitol produced rapidly reached a plateau after 50 h where its maximum quantity was about 20 g/l with sucrose.
