Evaluation of Antioxidant Activity of Solvent Fractions of Angelica gigas Root Using TOSC Assay (TOSC 방법을 이용한 참당귀 뿌리 용매분획(Angelica gigas)의 항산화 활성 평가)

  • Objectives: The aim of this study was to elucidate the antioxidant effect of solvent fractions of Angelica gigas root. Methods: The ethanol extract of Angelica gigas root was suspended in water and then partitioned with dichloromethane (MC Fr.), ethyl acetate (EA Fr.) and butanol (BuOH Fr.), sequentially. The antioxidant activities of solvent fractions of Angelica gigas root were evaluated for radical scavenging activity against stable free radicals (2,2-diphenyl-1-picrylhydrazyl) DPPH, hydrogen peroxide and superoxide anion radicals. In addition the antioxidant activities of solvent fractions of Angelica gigas root against peroxyl radicals, hydroxyl radicals and peroxynitrites were determined by the total oxy-radical scavenging capacity (TOSC) assay. Results: Among the solvent fractions of MC Fr., EA Fr., and BuOH Fr., BuOH Fr. was found to have stronger antioxidant activity with $IC_{50}$ values of 59.72, 14.36, 30.96 and $44.75\;{\mu}g/m{\ell}$ on the DPPH radical, nitrite, superoxide anion and hydrogen peroxide, respectively, than BHA used as a positive control. Moreover, specific TOSC values(564.8, 276.4 and 405.5 TOSC/mM) of BuOH fr. against peroxyl radicals, hydroxyl radical and peroxynitrite were 4 times higher than GSH (136.5, 67.4 102.6 TOSC/mM) used as a positive control. Conclusions: These results suggest that the BuOH fr. of Angelica gigas root has a high antioxidant activity and can be useful to develop functional food against oxidative stress conditions.

  • Background: Ganoderma lucidum cultured on hulled barley was investigated as a potential natural source of antioxidants and anti-inflammatory agents. Methods and Results: The yields from Ganoderma lucidum cultured on hulled barley water and ethanol extract were 17.69% and 25.77%, respectively. The antioxidant activity of Ganoderma lucidum cultured on hulled barley extracts was confirmed by various methods including assayss of 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2-azino-bis-(3-ethylbenzo thiazoline-6-sulfonic acid) (ABTS), nitrite radical scavenging, and $Fe^{3+}$ to $Fe^{2+}$ reducing power activity. The ethanol extract of Ganoderma lucidum cultured on hulled barley showed improved DPPH, ABTS and nitrite radical scavenging activity compared with the water extract. After treatment of RAW264.7 cells with Ganoderma lucidum cultured on hulled barley ethanol extracts, the cell viability compared with the control was 92.82%, even at a concentration of $3,000{\mu}g/m{\ell}$. The ethanol extract inhibited reactive oxygen species (ROS) generation in RAW264.7 cells stimulated with $H_2O_2$, even at low concentrations. In addition, the ethanol extract showed an inhibitory effects on the production of lipopolysaccharide-induced nitric oxide (NO) in RAW264.7 cells. Conclusions: This study suggests that the extract of Ganoderma lucidum cultured on hulled barley is a potential source of natural antioxidants and anti-inflammatory agents.

Evaluation of the Antioxidant Activity and Anti-Inflammatory Effect of Hericium erinaceus Water Extracts (노루궁뎅이 버섯 열수추출물의 항산화 활성과 항염증 효능 평가)

  • This study was conducted to determine the antioxidative activity of Hericium erinaceus (HE) and we investigated that HE extract contributes to cell proliferation and to anti-LPS-induced inflammation. HE extract showed high DPPH free radical scavenging activity. However, the reducing power activity slightly increased in compare with control. Nitrite scavenging activity, ABTS radical scavenging activity, SOD-like activity of HE were elevated in dose-dependent. The level of total phenolic and flavonoid showed 30.06 mg/100 g and 33.86 mg/100 g, respectively. Linoleic acid oxidation inhibition had reached a maximum level on the fourth day and started to drop from the fifth day. HE extract contributes to cell proliferation on the RAW 264.7 cell. Our finding demonstrated that water extracts of HE possess significant antioxidant activities and may be suggested a new potential source of anti-inflammatory medicines.

Antioxidant Activity of Solvent Fraction from Black Garlic (흑마늘 용매 분획물의 항산화 활성)

  • To confirm antioxidant activity of black garlic, methanol extract of black garlic was fractioned by hexane, chloroform, ethyl acetate, buthanol and water. Antioxidant activities of solvent fractions were assayed in 100, 250, 500 and $1,000\;{\mu}g/mL$ concentrations. The contents of total phenol and flavonoids were significantly higher 5.5~11.6 times in chloroform, ethyl acetate and hexane fraction than other fractions. Antioxidant activities of solvent fractions were increased by higher sample concentrations and their activities were significantly higher in chloroform and ethyl acetate fractions than others. DPPH radical scavenging activity was over 50% in $1,000\;{\mu}g/mL$ concentration, except butanol and water fraction. In the same concentration, reducing power was also significantly lower in butanol and water fraction. ABTS radical scavenging activity was higher in hexane, chloroform and ethyl acetate fractions and was over 70% at $1,000\;{\mu}g/mL$ concentration. In $1,000\;{\mu}g/mL$ concentration, the range of hydroxy radical scavenging activity was 50.27~81.02% and SOD-like ability was 26.73~47.64%. Antioxidant activity in linoleic acid reaction system was significantly higher when storage time was longer and sample concentration was higher in non-polar solvent fractions. Nitrite scavenging activity was relatively higher than antioxidant activity and the activity in $100\;{\mu}g/mL$ concentration was over than 50%, except butanol fraction.

Chemical Composition and Antioxidant Activities of Prunus salicina Formosa Produced in Gimcheon (김천산 자두 후무사의 화학적 성분과 항산화 활성)

  • The present study was designed to investigate the chemical composition and antioxidant activities of Prunus salicina Formosa produced in Gimcheon. The free sugar amounts in Prunus salicina Formosa were fructose>glucose>sucrose>maltose, and malic acid contents in Prunus salicina Formosa were the highest among all the organic acids found in the present study. Each Prunus salicina Formosa extract was obtained by treating Prunus salicina Formosa with distilled water, 80% ethanol, 60% acetone, and 80% methanol at 25 and $50^{\circ}C$. The highest contents of total polyphenol and flavonoid were observed in the 60% acetone extract and 80% ethanol extract, respectively. The 60% acetone extract exhibited the greatest DPPH radical scavenging ability, reducing power, and nitrite scavenging ability. However, SOD-like activity was not considerably different for all the extracts studied. The results from the present study could be helpful for developing antioxidant products using Prunus salicina Formosa produced in Gimcheon.

Antioxidant Activities of Pleurotus cornucopiae Extracts by Extraction Conditions (추출조건에 따른 노랑느타리버섯 추출물의 항산화 활성)

  • Physiological activities of extracts form Pleurotus cornucopiae were examined. DPPH radical scavenging activity of water extract was 70.22 percent at 4 mg/mL, which was the highest among all extracts. Total polyphenol content of water extract was 26.34 mg/g, which was the highest value for all extraction conditions. Superoxide anion radical scavenging activities ranged from 50.17 to 80.56 percent at 1, 2, and 4 mg/mL, which were higher than the ascorbic acid activity (56.19% at 10 mg/mL) (P<0.05). ACE inhibitory activities were higher in ethanolic extracts than in water extracts. Nitrite-scavenging abilities under acidic conditions (pH 1.2 and pH 3.0) were the most effective among all the extracts. These results will be useful for understanding the physiological activities of Pleurotus cornucopiae.

Amino Acid Contents and Various Physiological Activities of Allium victorialis (산마늘의 아미노산 함량과 생리활성 효과)

  • This study was conducted to gather the basic data on the alpine leek (Allium victorialis) for the expand of consumption and the production of its manufactured goods. Amino acid content in alpine leek leaves and various physiological activities were examined. Seventeen component amino acids and 38 free amino acids from alpine leek leaves were analyzed, and the total contents were 2,693.28 mg/100g for component amino acids and 535.39 mg/100g for free amino acids. Total phenolic compounds in the leaves of alpine leek showed the highest level from the methanol extract (37.7 mg/l), and followed by ethanol extract (31.9 mg/l) and hot water extract (25.4 mg/l). Total flavonoid contents in 1,000 mg/l extract was the highest in the methanol extract (22.2 mg/l). DPPH radical scavenging activity at 1,000 mg/l extract was high in the order of ethanol extract (51.6%), methanol extract (47.3%) and hot water extract (37.2%). nitrite radical scavenging activity Methanol extract from Allium victorialis leaves was the highest nitrite radical scavenging activity (79.5%). Hyperplasia suppression of lung cancer cells (Calu-6) and gastric cancer cells (SNU-601) by the methanol extract from the bulb of alpine leek were 99.9% in the extracting concentration of over 200 mg/l. No significant difference in antimicrobial activity among the 3 different solvents and extract concentrations was observed, and the inhibition zones against the gram-positive and negative microorganisms were ranged from 8.23 to 10.15 mm. It was concluded that physiological activities in a human body could be improved by the intake of alpine leek as a pharmaceutical material, and that it would be useful for the prevention of health risk such as lung and gastric cancers.

Antimicrobial and Antioxidant Activities of Extracts of Marine Green-algae Enteromorpha intestinalis (거대녹조류 창자파래 추출물의 항균, 항산화 활성)

  • In this study, we investigate the antimicrobial and antioxidant activities of extracts of Enteromorpha intestinalis. To evaluate the antimicrobial and antioxidant activities, three solvents (hexane, chloroform and methanol) were applied to obtain extracts. The extraction yields are hexane (A) 1.11%, chloroform (B) 0.94%, and methanol (C) 8.2%. Also, the contents of total phenolic compounds of extract A, B, C are 4.03%, 8.15%, and 2.33%, respectively. In the results of antimicrobial activity, extract A and B have higher activity than that of extract C. Especially, Vibrio vulificus, Pseudomonas aeruginosa, Bacillus subtilis, Escherichia coli are more sensitive than others. In antioxidant activity, extract A and B present the higher DPPH activity than that of control (BHA and ascorbic acid). The DPPH radical scavenging activity and SOD-like activity are ordered as B>C>A. However, extract C show high value in ferric reducing antioxidant power assay. In nitrite scavenging activity, extract A is relatively higher than others. However, the antioxidant activities are lower than that of controls (ascorbic acid and BHA). The antioxidant activities are presented the increasing pattern of increasing by the increasing of extract concentration.

Antioxidative and Anticancer Activities of Various Solvent Fractions from the Leaf of Camellia japonica L. (동백나무 잎 용매분획물의 항산화 및 항암 활성)

  • To obtain basic information on the potential use of Camellia japonica leaf as a raw material in functional food, leaf antioxidant and anticancer activities were investigated. The radical-scavenging activity of various solvent fractions from the leaf, as shown by the DPPH radical test, increased in a dose-dependent manner, with the water fraction showing the highest activity. The reducing power of various solvent fractions from the leaf was also dose-dependent, and, again, the water fraction showed the highest reducing power. The water fraction showed strong antioxidant activity in the linoleic acid test and was also capable of scavenging nitrite in a dose-dependent manner. Proportions of 92.15% and 95.61% of available nitrite were scavenged by the water and butanol fractions, respectively, at levels of $1,000{\mu}g/mL$. Both butanol and water fractions exhibited strong inhibitory effects on the growth of human lung and colon cancer cells. The total phenolic contents of the butanol and water fractions were 216.26 mg/g and 220.68 mg/g, respectively. High-performance liquid chromatography (HPLC) showed that quercetin and epicatechin were the predominant phenolic compounds in the water fraction. The activities of this fraction are attributable to the presence of these phenolic compounds, particularly quercetin and epicatechin.

Antioxidant Activities of Hot Water and Ethanol Extracts from Portulacae Herba (마치현 열수 및 에탄올 추출물의 항산화 활성)

  • Objectives : This study aims to provide basic data about Portulacae Herba (PH) extracts as natural antioxidants by considering diverse antioxidant activities of PH depending on solvents. Methods : The samples of PH were pulverized, and A hot water and a 70% EtOH were stir-extracted for two hours three times repeatedly in a water bath with a temperature of 95 degrees and at room temperature respectively to measure 7 kinds of antioxidant activities. Results : There were significant differences in total phenol content, because the total phenol content of the 70% EtOH extract was higher than the hot water extract's, and the total flavonoid content of the 70% EtOH extract($4.40{\mu}g/mg$) was nearly 3.8 times higher than the hot water extract's($1.16{\mu}g/mg$). DPPH and ABTS radical scavenging activities were 70% EtOH extract showed a little higher activity than the hot water extract, and at a concentration of $500{\mu}g/mg$, the highest scavenging activity was found in the 70% EtOH extract, not in the control group. Hydroxyl radical and $Fe^{2+}$ chelatingactivities were slightly higher in hot water extract than in 70% EtOH extract, and increased in a dose-dependent manner. Nitrite scavenging activities increased dose-dependently in the hot water and the 70% EtOH extract, regardless of the pH level, and scavenging activity of the 70% EtOH extract was higher at pH 1.2 than at pH 3.0. Conclusions : In conclusion, it is thought that PH hot water and 70% EtOH extract have antioxidant activities, and can be used as natural antioxidants in future.
