Industrial Utilization and Function of Omega Fatty Acid and Their Content Variation in Perilla (들깨 오메가 지방산의 기능과 함량변이 및 이용)

  • The n-3 family fatty acids containing ${\alpha}$-linolenic acid(18:3, ALA) have been known as physiological activation materials such as inhibitory effects on the incidence of hyper-tension, coronary heart disease and cancers as well as the control of senilc dementia. Although a lot of ALA(about $63\%$) are contained in perilla oil, it has not been commercialized yet because the purification technique of the ALA has not been well established. The procedure of purification of ALA from perilla oil was saponified with 1 N-KOH /ethanol and then saturated and low level unsaturated fatty acids were removed by low-temperature crystallization method. The concentrated unsaturated fatty acids (containing about $75\%$ ALA) went down through the silver nitrate-impregnated silica column chromatography for separation of high purity of ALA. The results obtained we Fraction B, C and D contained ALA more than $85.5\%$(recovery, >$88.9\%,\;95.4\%$(recovery, >$54.4\%$) and $99.9\%$(recovery, >$31.5\%$) in purity, respectively. Seed oil content of the tested varieties were ranged from 34.8 to $54.1\%$ with $45.3\%$ of varietal means. The major omega fatty acids contained in the oil were oleic acid(n-9) $15.2\%$, linoleic acid(n-6) $13.9\%$ and linolenic acid(n-3) $63.1\%$ in the mean value. Varietal variation of n-9, 6 and 3 fatty acids ranged of $9.5\~21.4\%,\;9.1\~20.4\%$ and $50.6\~70.5\%$ respectively. Unsaturated fatty acid were averaged $92.2\%$ of seed oil in fatty acid composition. The ratios of n-6 to n-3 ranged of $0.13\~0.34\%$($0.22\%$ in mean value). The highest n-3 fatty acid variety was Yecheonjong being $70.5\%$. The lowest variety in ratios of n-6 to n-3 was Goseongjong being $0.13\%$. Oil content showed positive correlation with stearic acid and linolenic acid, while the negative correlation with oil content and linoleic acid. On the other hand, A significant negative correlation were showed between linolnic acid and the ratios n-6/n-3 fatty acid, saturated fatty acid. Saturated fatty acid was highly correlated with unsaturated fatty acid negatively being $r= -0.723^{**}$.

Changes in Nitrogen Mineralization as Affected by Soil Temperature and Moisture

  • Soil is the main nitrogen (N) provider for plants but N in soil is not all available to advanced plants. Mineralization is a critical biological process for transferring organic N to inorganic N that can be used by plants directly. To investigate the effect of different levels of soil temperature and water content to soil mineralization, a field experiment was established on three different sites (A, B and C). We measured soil temperature, moisture and electrical conductivity once daily after swine slurry application. Average soil moisture and temperature in site A is the highest among three sites (40.9% and $9.7^{\circ}C$, respectively). Following is in site C (37.3% and $9.6^{\circ}C$) and the lowest is in site B (28.0% and $9.0^{\circ}C$). Ammonium N (NH4+-N) and nitrate N (NO3--N) were determined on the first and fifth day after treatment. Compared with site B and C, site A always had the highest soil total N content (1.54 g N kg-1 on day one; 1.22 g N kg-1 on day five) and highest NO3-- N content (93.18 mg N kg-1 on day one; 16.22 mg N kg-1 on day five) and a significant decrease on day five. Content of NH4+-N in site B and C reduced while in site A, it increased by 6.7%. Results revealed that net N mineralization positively correlated with soil temperature (P<0.5, $r=0.675^*$) and moisture (P<0.01, $r=0.770^{**}$), suggesting that to some extent, higher soil moisture and temperature contribute more to inorganic N that can be used by plants.

Effect of Compost Application on Radish Quality and Changes of Soil Physico-chemical Properties in Organic Farming (유기농산물 생산을 위한 퇴비시용이 무의 품질과 토양의 이화학성에 미치는 영향)

  • Depending on compost treatment the changes of radish morphology and soil physico-chemical properties were investigated in the sandy loam soil, pH of the soil was decreased and the contents of organic matter, total nitrogen, phosphate and cation exchange capacity was increased with increase of compost. Root weight and length were the lowest, but deficit rate was the highest(79.3%) in the treatment of 120Mg/ha of compost. The contents of chlorophyll and sugars were increased with increasing the amount of compost treatment, but the inorganics were scarcely changed. The nitrate content in radish root ranged from 10 to 120mg/kg in various treatments.

Enhanced Production of Astaxanthin in Paracoccus haeundaensis Strain by Physical and Chemical Mutagenesis (물리·화학적 돌연변이 유도를 통한 Paracoccus haeundaensis의 astaxanthin 생산량 증대)

  • Carotenoids are natural lipid-soluble pigments, which are produced primarily by bacteria, algae, and plants. Many studies have focused on the identification, production, and utilization of natural sources of astaxanthin from algae, yeast, and crustacean byproducts as an alternative to the synthetic pigment, which is mostly used today. The aim of the present study was to identify a mutant of Paracoccus haeundaensis by exposure to UV and ethyl methanesulfonate (EMS). The mutant was then exposed to nutrient stress conditions to isolate an astaxanthin-hyperproducing strain, followed by characterization of the mutant. The survival rate decreased in accordance with an increase in the UV exposure time and an increase in the EMS concentration. A mutant of the original P. haeundaensis strain was identified that showed hyperproduction of astaxanthin following exposure to UV irradiation (20 min) and EMS treatment (0.4 M concentration). The optimal culture conditions for the PUE mutant were $25^{\circ}C$, pH 7-8, and 3% NaCl. The effects of various carbon and nitrogen sources on the growth and astaxanthin production of PUE were examined. The addition of 1% raffinose and 3% potassium nitrate influenced cell growth and astaxanthin production. The selected mutant exhibited an increase of 1.58 folds in astaxanthin content compared to initial wild type strain. A genetically stable mutant strain obtained using mutagen (UV irradiation and EMS treatment) may be a suitable candidate for further industrial scale production of astaxanthin.

The Formation of N-nitrosamine in Alaska Pollack during its Drying (명태의 건조중 N-nitrosamine의 생성)

  • Dried alaska pollack is one of the representatives among dried marine fish and shellfish products in our country. This study was performed to obtain the basic data about the effect of drying method on the formation of N-nitrosamine and its precursor to ensure the safety of dried alaska pollack. The contents of nitrate and nitrite were detected 1.5 and <1.0 mg/kg in raw samples, and $3.0\~4.2,\;1.4\~2.7mg/kg$ in dried products, respectively. There was no significant change of betaine contents during drying while TMAO decreased, TMA and DMA increased in alaska pollack during d교ing. N-nitrosodimethylamine (NDMA) was only detected in alaska pollack and its dried products, and recovery from above samples spiked with $10{\mu}/kg$ for N-nitrosodipropylamine was $87.2\~107.4\%$. The levels of NDMA were found to be $2.8{\mu}/kg$ on an average in raw samples, but the levels of NDMA increased remarkably during drying of alaska pollack and its content in dried products was $8.7\~51.4{\mu}g/kg$. Regardless of drying methods, NDMA tend to increased in dried products, and its contents were 15.5 times higher in hot-air dried than raw samples, 9.0 times in sun dried and 4.4 times in freeze dried products. less NDMA was produced in the freeze dried products, so it is believed that freeze drying method is effective to decrease the NDMA levels in the dried products of alaska pollack.

Removal Characteristic of Ammonia Nitrogen and Behavior of Nitrogen in Synthetic Wastewater Using Leclercia Adecarboxylata (Leclercia Adecarboxylata를 이용한 합성폐수의 암모니아성질소 제거특성 및 질소거동)

  • In this study, the removal characteristic of ammonia nitrogen and behavior of nitrogen was investigated using Leclercia adecarboxylata, which was derived from the culture contaminated by ammonia nitrogen of high concentration. The method of ammonia nitrogen removal was not biological nitrification and denitrification but elimination of nutrient salt with internal synthesis of microorganisms which use ammonia nitrogen as substrate. L. adecarboxylata(one of ammonia synthesis microorganisms) was highly activated and showed the most high removal efficiency in free salt condition but the removal efficiency decreased badly in salt concentration of more than 4%. About 80 mg/L of $NH_3-N$ was mostly removed within 20 hours and 500 mg/L of $NH_3-N$ showed less then removal efficiency of 50% because carbon source was not enough. However, ammonium nitrogen concentration was decreased again when the carbon source was inserted additionally thus, ammonium nitrogen removal efficiency by L. adecarboxylata, was related to amount of carbon source. pH decreased from 8.0 to 6.36 according to growth of L. adecarboxylata. Concentration of nitrite nitrogen and nitrate nitrogen did not increase and TKN concentration showed no variation while ammonia nitrogen was removed by L. adecarboxylata. In addition to, when content of protein in organic nitrogen was measured, protein was not detected at the beginning of microorganism synthesis but protein of 193.1 mg/L was detected after 48 hours. Hence, ammonium nitrogen was not decomposed as nitrate nitrogen and nitrite nitrogen but synthesized by L. adecarboxylata, which has excellent ability of nitrogen synthesis and can threat ammonia nitrogen of high concentration in wastewater.

Primary Productivity and Nitrogenous Nutrient Dynamics in the East Sea of Korea (한국 동해의 기초생산력과 질소계 영양염의 동적관계)

  • The daily net primary production by phytoplankton and ammonium excretion by macrozooplankton (> $350{\mu}m$) were measured to understand the nitrogenous nutrient dynamics in the southern part of the East Sea of Korea. At most of the staions, water columns were well stratified and strongly developed pycnoclines and matching nutriclines could be found near the 20-60m. Total chlorophyll ranged between $1.22-3.24{\mu}g$ ChI/l and nano-fractions of chlorophyll ranged from 43.2 to 99.6% in the surface layer. The daily net primary production by phytoplankton ranged from 0.75 to 2.04 gC/$m^2$/d and averaged to be 1.5 gC/$m^2$/d. 1t is evidenced that the primary production and chlorophyll content are relatively high in frontal waters where the North Korean Cold Water meets with the East Korean Warm Water. The turnover time of nitrate in the euphotic zone ranged from 0.2 day to 1.6 day and averaged to be 0.8 day. The N:P ratio of the study area shows on the average 13.4 which indicates nitrogenous nutrient to be the limiting factor for phytoplankton growth. Ammonium excretion by macrowoplankton averaged out to 1.3mg at-N/$m^2$/d, and contributed 7.3% of daily total nitrogen requirement by phytoplankton in this area. Calculation of upward flux of nitrate to the surface mixed layer from the lower layer approximates 7% of nitrogen requirement by phytoplankton.

Effect of Chemical Fertilizer and Liquid Manure Application on the Growth Characteristics and Feed Value of Corn for Silage and NO3 of Soil (화학비료와 액상분뇨 시용이 사일리지용 옥수수의 생육특성, 사료가치 및 토양의 NO3에 미치는 영향)

  • This experiment was carried out to determine the growth characteristics, dry matter and TDN yield of corn for silage (Zea mays L.), soil nitrate in infiltration water by chemical fertilizer and different manure application(C; chemical fertilizer + chemical fertilizer, T1; Lime +chemical fertilizer + chemical fertilizer, T2; swine manure + swine manure, T3; Lime + swine manure + swine manure, T4; swine manure +chemical fertilizer). The results obtained are summarizes as follows; The mean growth characteristics of T3 treatment(soil treatment: lime + base fertilizer: liquid manure + additional fertilizer: chemical fertilizer) was higher than those of other treatments. The highest yield of dry matter and TDN was obtained in T3 treatment with 18,611 and 13,746 kg/ha, respectively(P<0.05), and the lowest in T1 treatment with 13,529 and 9,541 kg/ha, respectively(P<0.05). The contents of NDF and ADF were not significant different among treatments. However, NDF and ADF content among treatments were the highest with T1 treatment and the lowest with T3 treatment. $NO_3$ level of soil was hardly influenced by the different manure application treatment, and not affected during the experiment time.

Refinement of the manganese nitrate solution prepared by leaching the reduced Ferromanganeses dust with nitric acid. (용해도 차이를 이용한 질산망간 용액의 정제)

  • Mn was extracted by using a nitric acid from the reduced ferromanganese dust and the basic experiments were taken to refine the manganese nitrate solution by means of precipitation of Ca, Mg oxalate. The dust was generated in AOD process producing a medium-low carbon ferromanganese and collected in the bag filter. Manganese oxide content in the dust was about 90% and its phase was confirmed as $Mn_3$$O_4$. $Mn_3$$O_4$ in the dust was reduced to MnO by roasting with activated charcoal. The main impurities in the extracted solution prepared by leaching the reduced dust with nitric acid were Na, K, Fe, Si, Ca, Mg etc. Among them, Fe was removed by controlling pH of the solution more than 4 and precipitating $Fe(OH)_3$, simultaneously silicious material solved in the solution was removed by co-precipitation with the ferric hydroxide. Addition of 150 g reduced dust into 4N HNO3 solution 1$\ell$ was appropriate to control the pH of the solution to pH 4. To differ greatly the solubilities of manganese oxalate and calcium or magnesium oxalate in a solution containing a high concentration of Mn, pH of 4 or less and addition of ($NH_4$)$_2$$C_2$$O_4$ in equivalent with Ca and Mg are recommended. At this time, the higher temperature was the shorter the precipitation reaction time was needed.

  • These studies were carried out to investigate the effects of rice straw on microflora in relation to nitrogen metabolism in submerged soil. Rice plants were cultured in submerged soil to which rice straw was applied. In the submerged soil applied with rice straw the value of Eh lowered. pH was higher in the upper layer than in the lower. The content of iron(II) in submerged soil increased, while that of ammonium nitrogen decreased when rice straw was applied and nitrate-nitrogen was hardly detected during the rice cultivation period Under application of rice straw the number of denitrifying bacteria observed to increase at the early growing stage of rice plant and to decrease thereafter, and that of nitrate reducing bacteria increased at the late growing stage. The number of ammonium oxidizing bacteria and that of nitrite oxidizing bacteria decreased continually but the latter were rather sharply decreased.
