Identification of Root Age by Histochemical Staining of Secretory Duct Layers in Ginseng (인삼 분비도관의 조직화학적 염색에 의한 연근판별)

  • Identification of the age of ginseng root is very important in commercial market as well as in research field. However, any criterion abut it has not been clearly established yet. We studied to find a clear morphological key for identification of ginseng root\\`s age using the histochemical staining method. Fresh sections of 3, 4, 5, and 6 year old roots were stained with 1% nile blue, observed under the light microscopy, and compared each other. The number of secretory duct layers(SDL) is a useful key to confirm the age of ginseng root as follow; three-year-old root has two, four-year-old one has three, fie-year-old one has four, and six-year-old one has five resin duct layers on each cortical region of roots. Secretory ducts are thought to be formed by the vascular cambium every year. Unlike the surrounding parenchyma cells, secretory epithelial cells lack starch grains in the cytoplasm.

  • The culture of makeup has early developed in China. Especially, beautiful eyebrows were the symbol of a beauty. This study is related to fashionable Cheong-Dae in Han Dynasty. In Han Dynasty, women painted thier eyebrows with blue color in a different color from former periods. This fact has close connection with the bordering countries on Western China. Together with Hu fashion having been in fashion during trading directive with Western China women has seemed to use Cheong-Dae for their makeup in Han Dynasty. In Hand Dynasty, Ferghana women had very often used it for their makeup. And it had produced in Ferghana, Jaguda, Kashkar. In Tang Dynasty, it has been said that Persia was the best. Let's take a look at the producing method. Soak indigo plants in water add lime and stir it up, then bubbles are formed. They are called it as Cheong-Dae. Nearly, they are produced in the course of indigo dying. So we can see inflowing course of Cheong-Dae into China through the history of Indigo dying. We can guess the use of Cheong-Dae in Egypt. Indeed, Nile Blue is used for the Protection their access from insects as well as effect of coolness visually. This guess is very reasonable because the fact is considered that some people dye with indigo color by the way of sedimentation in Peniasula of Sinai. Also, the PH of Cheong-Dae is about 7. So, it does no harm in skin and its color is dark blue. Cheong-Dae has effects of insecticide and removal of fever. Cheong-Dae that had been used in Peninsula of Egypt seems to have been carried into China through Silk Route. And by trial to make Cheong-Dae it is proved that its ingredient is plant.

  • One most feasible way to measure the concentration field in the micro-channel is using micro-LIF(Laser Induced Fluorescence) method. However, an accurate concentration field at a given cross plane in a micro-channel has not been successfully achieved so far due to various limitations in the light illumination and fluorescence signal detection. The present study demonstrates a novel method to provide an ultra thin laser sheet beam having five(5) microns thickness by use of a micro focus laser line generator. The laser sheet beam illuminates an exact plane of concentration measurement field to increase the signal to noise ratio and considerably reduce the depth uncertainty. Nile Blue A was used as fluorescent dye for the present LIF measurement. The enhancement of the fluorescent intensity signals was performed by a solvent mixture of water $(95\%)$ and ethanol (EtOH)/methanol (MeOH) $(5\%)$ mixture. To reduce the rms errors resulted from the CCD electronic noise and other sources, an expansion of grid size was attempted from $1\times1$ to 3(3 or 5(5 pixel data windows and the pertinent signal-to-noise level has been noticeably increased accordingly.

  • Mixing between two or more reagents is one of important processes in biochemical microfluidics. In efficient micromixer design, it is essential to analyze flow pattern and evaluate mixing efficiency with good precision. In this work, mixing efficiency for Y-channel micromixer is measured by fluorescence intensity using LIF(Laser Induced Fluorescence) Confocal Microscope. The Y-channel micromixers are fabricated with polydimethylsiloxane(PDMS) and those are bonded to glass plate through Plasma bonding. Nile Blue A is injected into the micromixer as a fluorescence dye for measuring of fluorescence intensity by He/Ne laser. For visualization of the flow pattern, dynamic image capturing is carried out using CAM scope. For the comparison with computer simulation, modified SIMPLE algorithm for incompressible flow equation is solved for the same geometry as in the experiment. Throughout the experiments and computer simulation, accurate mixing efficiency evaluation process for a PDMS Y-channel micromixer is established.

Ethnobotany of Wild Baobab (Adansonia digitata L.): A Way Forward for Species Domestication and Conservation in Sudan

  • Selection of superior phenotypes of fruit trees and products based on established criteria by local people is a prerequisite for future species domestication and conservation. Thus the study objective was to identify the local people's perceptions and preferences on baobab trees and products. A sample of 142 respondents was randomly selected using structured interviews in Blue Nile and North Kordofan, Sudan in 2013. Descriptive analysis was employed using SPSS and Excel programs. The study results indicated that local people use the morphological characteristics of the tree (leaves, fruits, seeds, kernels and bark) to differentiate individual trees. Based on the perceptions, local people recorded trees with delicious leaves, white pulp color, big fruit size and mature capsule size, and high pulp yield as criteria for differentiating between baobab trees in the study areas. In contrast, the undesirable traits were connected to trees with acidic pulp, slimy pulp, bitter leaves, and low pulp yield. The study concluded that the ethnobotanical knowledge of the baobab tree and its products may play an important role in tree domestication and improvement in Sudan. However, further research on tree genetics is needed to complement the ethnobotanical knowledge for baobab resources domestication and conservation.

Polyhydroxyalkanoate (PHA) Production Using Waste Vegetable Oil by Pseudomonas sp. Strain DR2

  • To produce polyhydroxyalkanoate (PHA) from inexpensive substrates by bacteria, vegetable-oil-degrading bacteria were isolated from a rice field using enrichment cultivation. The isolated Pseudomonas sp. strain DR2 showed clear orange or red spots of accumulated PHA granules when grown on phosphate and nitrogen limited medium containing vegetable oil as the sole carbon source and stained with Nile blue A. Up to 37.34% (w/w) of intracellular PHA was produced from corn oil, which consisted of three major 3-hydroxyalkanoates; octanoic (C8:0, 37.75% of the total 3-hydroxyalkanoate content of PHA), decanoic (C10:0, 36.74%), and dodecanoic (C12:0, 11.36%). Pseudomonas sp. strain DR2 accumulated up to 23.52% (w/w) of $PHA_{MCL}$ from waste vegetable oil. The proportion of 3-hydroxyalkanoate of the waste vegetable-oil-derived PHA [hexanoic (5.86%), octanoic (45.67%), decanoic (34.88%), tetradecanoic (8.35%), and hexadecanoic (5.24%)] showed a composition ratio different from that of the corn-oil-derived PHA. Strain DR2 used three major fatty acids in the same ratio, and linoleic acid was the major source of PHA production. Interestingly, the production of PHA in Pseudomonas sp. strain DR2 could not occur in either acetate- or butyrate-amended media. Pseudomonas sp. strain DR2 accumulated a greater amount of PHA than other well-studied strains (Chromobacterium violaceum and Ralstonia eutropha H16) when grown on vegetable oil. The data showed that Pseudomonas sp. strain DR2 was capable of producing PHA from waste vegetable oil.

  • The present investigation was focussed on histochemical and electron microscopical observations of utero-vaginal(U-V) glands in U-V junctions of domestic hens. In histochemical observations, fine granules by PAS technique for mucopolysaccharides and nile blue stain for acidic lipids were slightly stained on the cytoplasm of U-V glandular epithelium. Larger granules by Sudan black B stain for neutral fat and phospholipids and Sudan III stain for neutral fat were heavily stained on the perinuclear region of the U-V gland epithelial cells. These positive materials were heavily stained on the U-V glandular epithelium of lowfecundity hens and non-laying hens. In scanning electron microscopic findings of the U-V junction surface, the orifices of U-V glands are seen as the crater-like invagination. The neck of the U-V gland and the epithelium of U-V iunction were covered by ciliated epithelial cells. Aggregates of spermatozoa are observed often to be on the necks of the U-V gland. These spermatozoa heads are embedded in the glandular tubules and many spermatozoa tails are free on the epithelium of uterine surface. In transmission electron microscopic findings, the epithelial cells of the U-V glandular orifices were ciliated, columnar cells. The apical regions of these cells contained numerous electrondense, round secretory granules of uniformly size. The epithelial cells of the U-V glandular tubule were columnar or pyramid shape with round or oval nuclei. These epithelial cells have numerous microvilli and also contained electron-dense, round secretary granules of uniformly size and electron-lucent vesicles of various size. Spermatozoa are seen as the cross-sections of various regions of heads and tails in glandular tubules. Also spermatozoa arranged longitudinally parallel within the glandular tubules.

A STUDY OF MORPHOGENESIS OF DIGITAL MALFORMATION ON RAT EMBRYO BY X-IRRADIATION (방사선조사시 태내백서의 지지기형성과정에 관한 실험적 연구)

  • The author studied on the effects of x-irradiation to the development of digital malformation in gestation rats. The time-matings occured between 6 p.m. and 8 a.m. and females with copulation. plugs at 8 a.m. were isolated and properly marked for evidence of copulation. The lower abdomen of mothers were exposed to x-irradiation on the 11½th day of gestation, the critical period developing digital malformation, respectively 100, 150, 200, 250, 300 and 350 rads. At 18½th day of post-conception total 50 pregnant females were dissected and the incidence of digital malformations were obtained. Rat embryos on the 12, 13, 14, 15, 16th day of gestation irradiated by 250 rads were examined for morphogenesis of digital malformation. Digital radiating lines were examined in water and histologically by H-E stain. Supra vital stain samples by Nile-blue sulfate in 37℃ normal saline were prepared for the observation of cell necrosis regions and morphogenesis of digits. The results obtained were as follows; 1. By x-irradiation on 11th day of gestation, digital malformations of Ectrodactylia, Syndactylia Polydactylia and Hematodactylia were developed. Ectrodactylia showed the effective relationship to the amount of irradiation, however Syndactylia ans Polydactylia did not. 2. By x-irradiation, cell necrosis of digital germ was appeared markedly, but in 48 hours after irradiation was depressed to the periphery of digital germ and in 72hours after irradiation was disappeared. Digital radiating line showed marked stage of malformation in 48hours after irradiation and continued to show the same amount of physiological cell necrosis as the compared control group in 72hours. after irradiaion. But in the Syndactylia, physiological cell necrosis was not able to be recognized. 3. Ectrodactylia induced by x-irradiation was considered as the direct resoult of cell necrosis of digital origin, however, Polydactylia and Syndactylia were considered as the resoult of some effect in repair process of x-irradiation damages.

DNA Dynamics: a Fluorescence Resonance Energy Transfer Study Using a Long-Lifetime Metal-Ligand Complex

  • Fluorescent probes bound to DNA typically display nanosecond decay times and reveal only nanosecond motions. We extend the time range of measurable DNA dynamics using $[Ru(pby)_2(dppz)]^{2+}$ (bpy=2.2'-bipyridine, dppz=dipyrido[3,2-a2',3'-c]phenazine) (RuBD) which displays a mean lifetime near 90 ns. To test the usefulness of RuBD as a probe for diffusive processes in calf thymus DNA, we compared the efficiencies of fluorescence resonance energy transfer (FRET) using three donors which display lifetimes near 5 ns for acridine orange (AO), 22 ns for ethidum bromide (EB) and 92 ns for RuBD, with nile blue (NB) as the acceptor. The F rster distances for AO-NB, EB-NB and RuBD-NB donor-acceptor pairs were 42.3, 52.3, and $30.6{\;}{\AA}$, respectively. All three donors showed dramatic decreases in fluorescence intensities and more rapid intensity decays with increasing NB concentrations. The intensity decays of AO and EB in the presence of varying concentrations of NB were satisfactorily described by the one-dimensional FRET model without diffusion (Blumen and Manz, 1979). In the case of the long-lifetime donor RuBD, the experimental phase and modulation somewhat deviated from the recovered values computed from this model. The recovered NB concentrations and FRET efficiencies from the model were slightly larger than the expected values, however, the recovered and expected values did not show a significant difference. Thus, it is suggested that the lifetime of RuBD is too short to measure diffusive processes in calf thymus DNA.

Lipid Production Characteristics of the Basophilic Blue-Green Algae Arthrospira platensis Depending on pH for Alkaline Wastewater Treatment (알칼리성 폐수처리를 위한 호염기성 남조류 Arthrospira platensis의 pH에 따른 지질생성 특성)

  • This study investigated the growth and metabolic characteristics of Arthrospira platensis (A. platensis) according to pH, which has an alkaline optimal pH. The intake of inorganic carbon was expected to be the highest at the optimum pH 9, but it was different from the expectation, so the cause of the excessive intake of inorganic carbon at pH 7 was investigated. We tried to verify the triacylglycerol (TAG) synthesis metabolic mechanism because it was assumedthat the inorganic carbon intake of A. platensis according to pH is closely related to lipid production inside the cell. To verify this, the effects of pH on inorganic carbon intake were examined through lipid analysis inthe cell of A. platensis according to pH. As a result, in the case of the effect of inorganic carbon intake of A. platensis according to pH on TAG content, pH 9 and pH 11 showed no significant difference in TAG content, but at pH 7, it was two times higher compared to pH 9 and pH 11. It was assumed that the reason why A. platensis excessively consumed inorganic carbon at pH 7 was because itincreased the TAG content in proportion to the intake of inorganic carbon to protect cells from external pH stress. In addition, it is considered that the TAG content produced in proportion to the intake of inorganic carbon is because acetyl-CoA produced in the Calvin cycle is required for the synthesis of TAG.
