The Strategic Plan for Preparedness and Response to Bioterrorism in Korea (우리나라의 생물테러 대비 및 대응방안)

  • Following the Anthrax bioterrorism attacks in the US in 2001, the Korean government established comprehensive countermeasures against bioterrorism. These measures included the government assuming management of all infectious agents that cause diseases, including smallpox, anthrax, plaque, botulism, and the causative agents of viral hemorrhagic fevers (ebola fever, marburg fever, and lassa fever) for national security. In addition, the Korean government is reinforcing the ability to prepare and respond to bioterrorism. Some of the measures being implemented include revising the laws and guidelines that apply to the use of infectious agents, the construction and operation of dual surveillance systems for bioterrorism, stockpiling and managing products necessary to respond to an emergency (smallpox vaccine, antibiotics, etc.) and vigorously training emergency room staff and heath workers to ensure they can respond appropriately. In addition, the government's measures include improved public relations, building and maintaining international cooperation, and developing new vaccines and drugs for treatments of infectious agents used to create bioweapons.

  • Segniliparus species is a novel genus that is reported to be the new emerging respiratory pathogens. Here, we report a very rare case of S. rugosus pulmonary infection in an immunocompetent patient with non-cystic fibrosis. The organism was identified by 16S rRNA gene sequencing. The patient was successfully treated with antibiotics.

  • To establish a competent method of microbiological assay for bacampicillin, one of new semi-synthetic derivatives of ${\beta}-lactam$ antibiotics, a comparison was carried out between different conditions for hydrolysis of bacampicillin into ampicillin which was then subjected to cylinder plate method. The results showed that the use of carboxylic ester hydrolase in vitro as a pretreatment of it in either pH value 6 or 8 was feasible and that the cylinder plate method with Sarcina lutea was adequate for potency estimation.

  • The absorption rate of lysino-methylene-ampicillin from the rat small intestine, compared with ampicillin, was determined in vitro and in situ to establish the biopharmaceutical properties of lysino-methylene-ampicillin which is one of the new penicillinase-resistance antibiotics. The half of administered dose was absorbed rapidly within thirty minutes. The water-oil distribution coefficient of lysino-methylene-ampicillin was 0.03 in chloroform versus buffer system with $Na_{2}HPO_{4}-citric$ acid of pH5.2 at $37^{\circ}C$, and its dissolution rate reached the plateau in an hour and then represented 0.6 percent of equilibrium solubility

  • Twelve strains of Bifidobacterium spp. were isolated from the feces of healthy Korean 20∼30 years. The identification of genera from isolates were performed by the microscopic observation and fructose-6-phosphate phosphoketolase (F6PPK) activity which is the key enzyme to distinguish the Bifidobacterium spp. from other anaerobic bacteria. To determine the antibacterial resistance patterns, minimum inhibitory concentration (MIC) of several antibiotics (including anti-tuberculosis agents) was determined. Five of the isolate!, showed the high degree of resistance to vancomycin. To investigate the genetic diversity between isolates and type strain of Bifidobacterium spp. from KCTC, we peformed the RAPD-fingerprinting. Using a total set of four primers, it is possible to distinguish the isolates and Bifidobacterium spp. from KCTC. Thus, Bifidobacterium strains isolated from our samples may be a new species or strains of Bifidobacteriurn genera, and have the potential as a probiotics.

  • To develop new cephalosporin antibiotics with improved antibacterial activities, a series of 7$\beta$-[2-(2-aminothiazol-4-y)-(Z)-2-(1-carboxy-1-methylethoxyimino)acetamido] -3-[5-(heterocycle)thiomethylpy-rrolidin-3-ylthio]methyl-3-cephem-4-carboxylic acid (14~18) having aminothiazol carboxymethylethoxy-imino group on the C-7 position and (heterocycle) thiomethyl pyrrolidinthiomethyl group on the C-3 position of the cephem ring were synthesized. These compounds were tested for antimicrobial activity in vitro against Gram(+) and Gram(-) bacteria. Compounds 15 and 16 showed remarkable antibacterial activity against Salmonella typhimurium TV119 and Alcalienes faecalis KCTC1004, but most of compounds showed lower activity than cefotaxime.

  • 6$\beta$-(trans-3-Aryl-5-oxo-pyrrolidin-2-yl)acetamidopenicillanic acid(7a~7c) and 7$\beta$-(trans-3-Aryl-5-oxo-pyrrolidin-2-yl) acetamidocephalosporanic acid(8a~8c) were synthesized and tested in vitro antibacterial activity. Of these new penicillins exhibited good antibacterial activity against Gram-positive bacteria whereas none of the compounds possessed the activity against Gram-negative bacteria at the concentration tested.

Antibacterial and Antioxidant Activities of the Essential Oil from the Roots of Anthriscus sylvestris (전호(Anthriscus sylvestris) 뿌리 정유의 항균 및 항산화 작용)

  • To develop a new effective and safe natural antibiotics and antioxidant the essential oil was extracted from the roots of Anthriscus sylvestris by steam distillation. Its composition was analyzed by GC-MS. The activities of the essential oil fraction and its main components were evaluated against antibiotic-susceptible and -resistant strains of some food-born bacteria. In addition the synergism was examined with this oil combined with antibiotic by checkerboard titer test. The antioxidant activities were determined by in 1,1-diphenyl-2-picryl-hydrazil (DPPH) free radical scavenging activity test and reducing power assay. The essential oil fraction of A. sylvestris revealed significant inhibiting activities against antibiotic-susceptible and -resistant species of Vibrio and Shigella with MICs ranged from 1.00~4.00 mg/ml. It showed synergistic or additive effects when it was combined with amphicillin or trimethoprim/sulfamethoxazole (1 : 9). Additionally, the essential oil fraction of A. sylvestris exhibited significant DPPH free radical scavenging activity and the reducing power.

  • Antimicrobial peptides are widely used in various organisms as a defense system against infection. The peptides are lethal towards bacteria and fungi, however have minimal toxicity in mammalian and plant cells. In this aspect, it is considered that antimicrobial peptides are new alternative materials for defensing against microbial infection. Here, we describe overall characteristics of antimicrobial peptides based on the mechanism of action, classification of the peptides, report detection/screening methods and chemical/biological production. It is expected that understanding of innate immune system based on antimicrobial peptides tends to develop novel natural antimicrobial agents, which might be applied for defensing pathogenic microorganisms resistant to conventional antibiotics.

  • Tetracycline is one of the mostly used antibiotics around the world. The purpose of this study was to evaluate the fate of two different types of tetracycline resistant bacteria in biological wastewater treatment plants. Tetracycline resistant enterics and heterotrophic bacteria were monitored under two different lab-scale experimental conditions. Tetracycline resistant enteric bacteria showed the lower percentages of total enteric bacteria and net specific growth rate in the monitored activated sludge system as compared to tetracycline resistant heterotrophic bacteria. Therefore, total enterics, potentially E.coli, might not be the best indicator microorganism for evaluating the antibiotic resistant bacteria in biological wastewater treatment plant.
