O/W형 유화상에서 pH변화에 따른 액정구조의 생성에 관한 연구 (A Study on the Formation of Liquid Crystalline Structure depend on pH Change in O/W Emulsion)

  • 이 연구는 국산화 한 액정형성 유화제에 대하여 선진사의 액정형성제를 사용하여 pH변화에 따른 안정성에 관한 것이다. 국산화한 액정유화제는 SugarCrystal-LC ((주)바이오뷰텍, 한국)를 사용하였고, 비교시료는 Nikkomulese-LC (니코캐미칼즈, 일본)와 Alacel-LC (Croda 캐미칼즈, 영국)를 사용하여 액정형성의 생성력을 실험하였다. 액정형성은 산성 (pH=4.2)에서, 중성 (pH=7.0) 그리고 알칼리성 (pH=11.7)에서 액정구조가 생성되는지 현미경으로 확인하였다. 또한 교반 속도에 따른 액정의 형성 입자를 편광현미경으로 관찰 하였다. 교반 시간은 모두 동일하게 3분 동안 Homo-mixer로 교반하였고, 교반속도는 2500rpm, 3500 rpm, 4500rpm으로 증가시켜 액정의 상태를 관찰하였다. 그 결과 한국산 계면활성제가 2종의 외국산 보다 안정하고, 선명한 액정구조가 형성되는 것을 알 수 있었고, 영국산의 경우 유화입경이 분균일하여 불안정한 상태를 보였다. 일본산의 경우, 한국산과 유사한 구조와 성능을 가지고 있음을 알 수 있었다. 한국산 계면활성제는 외국산과 비교하여 우수한 유화성능을 가지고 있음을 알 수 있었다. 응용분야로 국산화한 Sugar Crystal-LC 계면활성제를 사용하여 액정크림, 로션, 아이크림 등 다양한 제형개발이 가능하고, 화장품산업은 물론 의약품산업 및 제약산업에서 피부외용제의 유화기술로 폭넓게 응용이 가능할 것으로 기대한다.

맥문동 물 추출물을 첨가한 바게트 빵의 품질 특성 및 당뇨유발 흰쥐의 혈당, 혈청지질에 미친 효과 (Quality Characteristics and Dietary Effect of Baguette Bread Added with Water Extracts of Liriopis Tuber on the Blood Glucose and Serum Cholesterol in Diabetes Induced Rats)

  • The quality characteristics of baguette bread added with water extracts of Liriopis tuber(LT) and effect of the diets containing 20% dried and powered baguette with 10% LT on the level of blood sugar and lipids in the diabetes induced rat by streptozotocin were investigated. pH of the dough added with 5∼10% LT was 5.00∼5.03 which were lower than that of control(5.12). The volume of the dough with 10% LT after 2nd fermentation was the biggest. Loaf volume index of the bread with 10% LT was significantly higher than that of control. $L^{*}$ values of outside and inside of the baguette decreased in proportion to the increase of the LT, while the $a^{*}$ values increased. Color of the bread with LT showed light dark-red. There was no significant difference in hardness, but springiness and gumminess were higher in the bread with 10% LT. Sweet taste increased, color acceptability and overall taste were enhanced by addition of LT, but it did not affect the sour and astringent taste. The overeating phenomenon showed in the diabetes rat decreased in the group fed with baguette bread with LT, while feed efficiency increased by feeding of the bread with LT. The level of glucose, neutral lipid and total cholesterol in blood were 118.92,95.47 and 37.42mg/dL in the control group, 245.60, 121.54 and 102.43 mg/dL in the diabetes control, 148.40, 103.25 and 57.63 mg/dL in the baguette diet group, respectively. There was no significant difference of HTR between control group and baguette diet group, while atherogenic index was considerably decreased, which might represent improving and preventing effects of diabetes.s.s.

표고버섯 헤미셀룰로즈의 식이가 쥐의 혈당과 콜레스테롤 함량에 미치는 영향 (Dietary Effect of Hemicellulose from Shiitake Mushroom(Lentinus edodes) on Blood Glucose and Cholesterol Content in Rats)

  • The effect of hemicellulose extracted from Shiitake mushroom(Lentinus edodes) on the level of blood sugar and cholesterol in the diabetes-induced rat by streptozotocin(STZ) was investigated. The yield of hemicellulose by extraction process of 5% salt extraction, preparation of alcohol insoluble substance, IN KOH extraction, acid precipitation(pH 3.0), and dialysis was 9.24%. The experimental plots divided to 1% cellulose group(control), 0.5% hemicellulose group(H-l) and 1% hemicellulose group(H-2). The groups were fed for 6 weeks, then continuously fed for 1 week after induction of diabetes by STZ. Feed intakes, weight gain and feed efficiency of the each groups were not significantly different, while water intakes and liver weight of H-2 group were lower than those of control and H-l group. Weight of liver in the H-2 group was significantly lower than those of control and H-l groups. The amounts of feces were 0.32 g/day in the control group, 0.43∼0.44 g/day in the H-l and H-2 groups, while the amounts of urine were 15.28 mL/day in the control group, 10.83∼11.20 mL/day in the H-l and H-2 groups. The content of blood glucose before diabetes induction(fed for 3∼5 weeks) was 111.2-132.6 mg/dL in the control group, not significantly different from others; After diabetes induction, however, the contents were 212.8 mg/dL in the control group, 140.0-144.0 mg/dL in the H-l and H-2 groups, which showed significant difference. Urine glucose contents of H-2 group before and after diabetes induction were lower than those of control and H-l groups. There was no significant difference in the content of neutral lipid between each groups. Total cholesterol contents were 101.6 mg/dL in the control group, 56.∼64.0 mg/dL in the hemicellulose groups. HDL-cholesterol content and atherogenic index of hemicellulose groups were lower than those of control group, respectively. In conclusion, the hemicellulose extracted from Shiitake mushroom represented improving and preventing effects for diabetes.

Comparison of Nutritional Compositions and Antioxidant Activities of Building Blocks in Shinseoncho and Kale Green Vegetable Juices

  • Shinseoncho and kale were divided into stem [shinseoncho stems (SS) and kale stems (KS)] and leaf parts [shinseoncho leaves (SL) and kale leaves (KL)] and made into green vegetable juices for analyses of nutritional compositions and antioxidant activities. Higher values of total acidity were observed in SL (0.736%) and KL (0.841%) than in SS (0.417%) and KS (0.335%) (p<0.05). Neutral sugar content showed higher values in SS (21.740 mg/mL) and SL (18.657 mg/mL) when compared with KS (1.497 mg/mL) and KL (1.452 mg/mL) (p<0.05). Protein content showed the highest value in SL (7.610 mg/mL) (p<0.05), while SS (0.403 mg/mL) and KS (0.403 mg/mL) showed similar lower values. Total polyphenol contents of SL (423.139 ${\mu}g/mL$) was significantly higher value (p<0.05) than those of other samples, which occurred in the following order: SL>KL (218.494 ${\mu}g/mL$) KS (107.269 ${\mu}g/mL$)>SS (75.894 ${\mu}g/mL$). KL exerted the highest DPPH radical scavenging activity (84.834%) (p<0.05), which occurred in the following order: KL>SL (63.473%)>KS (52.894%)>SS (35.443%). ABTS radical scavenging activity showed that SL (66.088%) and KL (38.511%) had higher scavenging activities, whereas SS (7.695%) and KS (9.609%) demonstrated to be lower activities (p<0.05). In general, leaf parts had much higher antioxidant activities as well as total polyphenol contents than those of the stem parts. In conclusion, shinseoncho and kale, particularly their leaf parts, offer antioxidant properties in green vegetable juices and the consumption of them may be beneficial as a nutrition source and in health protection.

Purification and Anticoagulant Activity of a Fucoidan from Korean Undaria pinnatifida Sporophyll

  • Crude fucoidan was extracted from the sporophyll of Korean Undaria pinnatifida collected at a coastal area ofWando, Korea, mainly by dilute acid extraction, ethanol precipitation, CaCU Precipitation, with an yield of approxi-mately 3.9% in mass. It was further purified by DEAE-cellulose column chromatography and its chemical composi-don and in vitro anticoagulant activity was determined. The average molecular mass of the purified fucoidan wasestimated about 2.1 x 103 kDa by size-fractionation HPLC and it consisted of neutral sugar (52.34% in mass), uronicacid (26.2%), and sulfate esters (7.4%). From the HPAEC-PAD analysis, the monosaccharide composition of thepurified fucoidan was shown to be fucose, galactose, xylose, and mannose, with a molar ratio of 1, 0.2, 0.02, 0.15,respectively, demonstrating that major monosacd-iande was fucose (72.3% in mol percentage) and other sugars,xylose (1.5%), galactose (14.6%), and mannose (10.9%) were present as minor component. The results suggested thatthis fucoidan is a sulfated, U-type fucoidan. The activated partial thrombloplastin time (APTT) assay of the purifiedfucoidan showed that the purified fucoidan elicited anticoagulant activity in a dose-dependent manner. Five jUg ofsporophyll fucoidan delayed the blood clotting time up to 5 times than untreated control and also up to 1.5 timesthan the same amount of the commercial fucoidan, respectively. Although it is preliminary, these results suggestthat the fucoidan of Korean Undaria vinnatifida sporophyll would be promising candidates for the development ofan anticoaeulant.

Effects of Nitrogen Fertilization on Forage Production and Nutritive Nalue of Geukdong 6, Teosinte Hybrid [Zea mays L. subsp. mexicana (Schrad.) H. H. Iltis]

  • This study was conducted to evaluate the effects of different nitrogen levels on the yield and nutrient quality of a newly developed domestic hybrid of Teosinte, Geukdong 6〔Zea mays L. subsp. mexicana (Schrad.) H. H. Iltis〕. The field experiment was conducted in a randomized block design with three replicates and consisted of four nitrogen (N) application rates, T1 (200 kg/ha), T2 (300 kg/ha), T3 (400 kg/ha), and T4 (500 kg/ha). No differences were found in plant length, leaf length, leaf width, leaf number, dead leaves, stem hardness, tiller number, and fresh yield (p>0.05). The T3 showed significantly greater dry matter yield at harvest (heading stage) compared to the other treatments (p<0.05). The crude protein content of T4 (10.49%) was higher than those of T1 and T2 (p<0.05). However, there was no significant difference between T4 (10.49%) and T3 (9.63%). The effects on crude fat, crude ash, neutral detergent fiber (NDF), acid detergent fiber (ADF), and crude fiber were not significant (p>0.05). The sugar content was higher in the T2 treatment than the other treatments (p<0.05). For Ca, T3 showed significantly greater content (p<0.05). However, no significant effects were found in the contents of Cu, Fe, K, Mg, Mn, Mo, and Zn (p>0.05). Na content was higher in order of T2 > T4 > T3 > T1 (p<0.05). Total mineral contents were not significantly different among the treatments (p>0.05). Given these results, we recommend the amount of nitrogen fertilization necessary for "Geukdong 6" to be around 400 kg per ha (T3), when considering, high fresh yield, dry matter yield, number of leaves and content of crude protein.

공장산 고추장의 저장기간중 이화학적 특성의 변화 (Changes in Physicochemical Properties of Industry-type Kochujang during Storage)

  • 국내에서 생산되고 있는 공장산 고추장의 제조후 경과기간에 따른 품질특성치의 변화를 구명하기 위하여 시제된 공장산 고추장을 $37^{\circ}C$에서 105일간 저장하면서 고추장의 품질변화를 측정한 결과는 다음과 같았다. 환원당은 저장 15일까지는 비교적 큰폭으로 감소한 다음 완만하게 감소하였다. 또한 유리당으로는 glucose, fructose, maltose가 검출되었으며 이중에서는 glucose가 가장 많았다. 유리아미노산 함량은 저장기간이 길어짐에 따라 감소하는 경향이었다. 즉, 저장초기 유리아미노산 총량은 2092.50mg%에서 저장 60일후 22.3% 정도 감소하였으며 90일 후에는 1360.57mg%로 저장초기에 비해 35% 정도가 감소한 것으로 나타났다. 저장기간중 전반적으로 효소활성은 유의적인 변화를 보이지 않았으나, 증성 protease의 활성이 미약하나마 완만하게 증가되는 경향을 보였다. 수분함량 및 수분활성도는 각각 저장기간의 증가에 비례하여 감소하는 추세를 보였으며, 양자간의 상관계수는 r=0.964였다. 저장기간의 경과에 따른 겉보기점도는 증가하였으며, 수분함량보다는 수분 활성도에 의해 더 큰 영향을 받는 것으로 판단되었다.

전통 침채류 유래 다당 생산균의 분리 특성 (Isolation and Characterization of Exopolysaccharide-Producing Bacteria from Korean Fermented Vegetables)

  • 탄닌 분해균 선발배지를 이용하여 탄닌 분해활성 보유 유산균을 발효 침채류로부터 분리하는 과정에서 오이소박이, 파김치, 총각김치로부터 다당류로 추정되는 점질물질을 생산하는 균주 4종을 선발하였다. 이들 균주의 165 rDNA 염기서열에 의한 계통유전학적 동정 결과, CK31은 Enterobacter cowanii, CK32는 Escherichia senegalensis, YK5는 Enterobacter asburiae, GK23은 Enterobacter ludwigii와 99% 이상의 매우 높은 상동성을 나타내었다. 가장 많은 양의 점질 물질을 세포 외로 생성하는 CK3l 균주는 rhamnose, fucose, arabinose, mannose, galactose, glucose의 중성당 조성을 가지고 있는 분자량 약 420 kDa heteropolysaccharide를 생산하는 것으로 나타났다.

  • 홍삼으로부터 염기성 수웅액 추출, 음이온교환 크로마토그래피 및 겔여과 크로마토그래피를 순차적으로 수행하여 항트롬빈 활성을 보이는 다당체를 분리하였다. 이 다당체는 셀룰로스 아세테이트막을 사용한 전기영동에서 하나의 띠로 나타나 비교적 순수하게 정제되었음을 알 수 있었고, 겔여과 크로마토그래피에서 결정된 평균 분자량은 약 177kDa이었다. $40.2\%$의 유론산, $9.2\%$의 황산기 및 $1.5\%$의 단백질을 포함하는 산성다당체였고, 구성 중성당으로 rhamnose, mannose, galactose, arabinose, glucose, fucose, xylose를 1.00 : 0.88 :0.86 : 0.78 : 0.70 : 0.33 : 0.22의 비율로 포함하고 있었다. 이 다당체는 트롬빈에 의한 피브리노겐의 응고를 농도에 비례하여 저해하였고, 혈장을 사용한 실험에서 내인성 경로를 통해 혈액응고를 저해하는 것으로 나타났다.

소나무 수피 페놀-유기설폰산 액화에 의하여 제조된 액화물의 특성 (Characterization of Liquefied Pine Bark Prepared from Phenol-Organic Sulfonic Acids Liquefaction.)

  • 소나무 수피를 유기 설폰산 촉매존재 하에서 페놀액화하고 얻어진 액화물에 대한 특성을 검토하였다. 수피의 페놀액화시 유기 설폰산은 염산보다 뛰어난 촉매임을 확인 할 수 있었으며, 본 촉매의 사용에 의하여 소나무 수피의 완전한 액화가 가능하였다. 이들 유기 설폰산을 촉매로하여 제조된 수피 액화물의 경우 액화용매로 사용된 페놀이 염산을 촉매로 한 경우에 비하여 2-3배 많이 결합해 있었으며, 유리 전이점(Tg)이 낮았다 (PTSA: 85℃, MSA: 169℃, HCI: 181℃). 또한 이들 액화물은 전수산기, 페놀성 수산기 및 지방족 수산기가 많이 존재하여 반응중 액화 용매인 페놀의 도입과 2차적인 축합억제를 예상할 수 있었다. 액화물의 IR 스펙트럼 및 중성당류 분석 결과로부터 수피 액화물 중의 탄수화물은 대부분 분해되었다는 것을 알 수 있었다. 액화물 및 그 잔사의 EDS 분석결과 유기 설폰산 촉매는 염산 촉매에 비하여 반응용기의 심각한 부식을 초래하지 않는다는 것을 알 수 있었다.
