국내 소 유산에 관한 임상학적 고찰 I. 경기지역을 중심으로한 임상학적 고찰 (Clinical Aspect of Bovine Abortion in Korea I. Clinical Aspect of Bovine Abortion in Kyunggi-do Area)

  • The objective of this study was to determine the cause and prevalence of bovine abortion and stillbirth in Kyungi-do area. Seventy three bovine fetuses were collected from farms and submitted to the College of Veterinary Medicine, Seoul National University. Submitted fetuses were evaluated during a 4-month period (July to November, 1999) for pathological lesion, tissue protozoa, bacteria and viral infection. The average proportion of abortions was decreased with parity in 73 abortion heifers and cows. Monthly incidence rate of bovine abortion was not different in this study. In fetuses from 90 to 282 days gestation, the majority were between 150 and 250 days gestation(58%). The cause of abortion or stillbirth was determined in 51% of the cases examined. In 15(21%) of the fetus, neosporosis were diagnosed by pathological findings. In three (4%) additional fetuses in three additional fetuses, suspected Neosporosis by pathological lesion, and in 3 (4%) fetuses examined Neopsorosis were diagnosed in 15 feturses and in 3 fetuses, Neosporosis was suspected by pathological legions. Neosporosis / viral infection were diagnosed in three additional fetuses). Miscellaneous bacterial infection, BVDV, iatrogenic cause, Neosporosis / IBRV / BVDV, miscellaneous viral, IBRV/BVDV and others were 3(4%), 3(4%), 2(3%), 2(3%), 1(1%), 1(%) and 9(12%) respectively. The cause and incidence of bovine abortion in different area in Kyungi-do was not different in this study.

Review of Neospora caninum and neosporosis in animals

  • Neospora caninum is a coccidian parasite of animals. It is a major pathogen for cattle and dogs and it occasionally causes clinical infections in horses, goats, sheep, and deer. Domestic dogs are the only known definitive hosts for N. caninum. It is one of the most efficiently transmitted parasite of cattle and up to 90% of cattle in some herds are infected. Transplacental transmission is considered the major route of transmission of N. caninum in cattle. Neospora caninum is a major cause of abortion in cattle in many countries. To elicit protective immunity against abortion in cows that already harbor a latent infection is a major problem. This paper reviews information on biology, diagnosis, epidemiology and control of neosporosis in animals.

  • 1. 질병명 : Neosporosis 2. 본 질병의 개요 역사 및 역학 : 1984년 노르웨이에서 부전마비로 폐사한 개를 검사하던 중 뇌척수염과 근염을 확인하고 T. gondii와 다른 새로운 원충성 질병을 최초로 보고한 이래 Dubey등이 미국에서 톡소플라즈마 감염으로 진단되었던 30두의 개조직에 대한 재검토를 실시하여 그 중 10 두는 새로운 원충성 질병으로 확인한 후 Neospora caninum을 명명하였다. 현재까지 본 원충은 개, 소, 염소, 말, 사슴 등에서 자연발생 예가 보고되었다. 특히 소에서는 유산의 한 주요한 원인체임이 한국을 비롯한 여러 나라에서 확인되었다.   • A case of neosporosis is described in a 6-month-old aborted bovine fetus. Grossly, numerous, well-demarcated, 2-3mm, yellowish white foci were scattered in the heart and skeletal muscle. Histopathologic changes were confined to the brain, heart, and skeletal muscle. Multifocal areas of necrotizing encephalitis were observed in the cerebrum as well as cerebellum. Severe multifocal to coalescing nonsuppurative inflammation was observed in the myocardium and skeletal muscle. The tachyzoites of Neospora caninum were demonstrated in the brain, myocardium and skeletal muscle with immunohistochemical method using Neospora caninum-specific antibody. Based on the histopathology and immunohistochemistry, this case was diagnosed as neosporosis in aborted Korean cattle. This is believed to be the first reported case of bovine neosporosis in Korea.

  • Congenital $Neospora$ $caninum$ infection was diagnosed in two Saanen goat kids from two distinct herds with a history of abortion and weak newborn goat kids in the Southern region of the State of Minas Gerais, Brazil. The first kid was weak at birth, had difficulty to rise and was unable to nurse. Gross lesions of porencephaly and hydrocephalus ex vacuo were seen. Multifocal necrosis, gliosis and non-supurative encephalitis were observed in the brain. Several parasitic cysts with a thick wall that reacted strongly only with polyclonal antiserum to $Neospora$ $caninum$ were seen in the cerebral cortex, brain stem and cerebellum. The second kid was born from a $Neospora$ $caninum$ seropositive mother that aborted in the last pregnancy. It was born without clinical signs. The diagnosis of neosporosis was based on antibody titer of 1:800 to $N.$ $caninum$ by indirect fluorescence antibody test obtained from blood collected before the goat kid ingested the colostrum and $Neospora$ $caninum$ DNA was detected by polymerase chain reaction and sequenced from placenta. This is the first report of neosporosis in goats in the southeast region of Brazil.

Generation and Immunity Testing of a Recombinant Adenovirus Expressing NcSRS2-NcGRA7 Fusion Protein of Bovine Neospora caninum

  • Neospora caninum is the etiologic agent of bovine neosporosis, which affects the reproductive performance of cattle worldwide. The transmembrane protein, NcSRS2, and dense-granule protein, NcGRA7, were identified as protective antigens based on their ability to induce significant protective immune responses in murine neosporosis models. In the current study, NcSRS2 and NcGRA7 genes were spliced by overlap-extension PCR in a recombinant adenovirus termed Ad5-NcSRS2-NcGRA 7, expressing the NcSRS2-NcGRA7 gene, and the efficacy was evaluated in mice. The results showed that the titer of the recombinant adenovirus was $10^9TCID_{50}/ml$. Three weeks post-boost immunization (w.p.b.i.), the IgG antibody titer in sera was as high as 1:4,096. IFN-${\gamma}$ and IL-4 levels were significantly different from the control group (P<0.01). This research established a solid foundation for the development of a recombinant adenovirus vaccine against bovine N. caninum.

울진군 한우 Neospora caninum 감염 실태 조사 (Investigation of Neospora caninum infection in Korean indigenous Cattle in Uljin)

  • This study was carried out to investigate an epidemiological state of neosporosis in Korean indigenous cattle in Uljin. Bloody samples were collected from 552 female cattle (337 farms), more than 5 years old, in 10 districts of Uljin, Antibody to N caninum were examined by enzyme linked immunosorbent assay (ELISA). Seroprevalence of individual and farm were 7.6% (42/552) and 8.6% (29/337), respectively. Positive rates by districts was variable $(0%{\sim}14,9%)$, and it was seemed to be a tendency that positive farms gathered with in a small zone. In seroprevalence by herd size farms having between 5-9 heads was top (37%), but there was no significant difference among herd size.

Detection of IgG antibody against Neospora caninum in cattle in Korea

  • A total of 492 cattle sera was screened by IgG-ELISA against Neospora caninum (Nc-1 strain and a Korean isolate, KBA-2) and Toxoplasna gondii. Out of 492, 113 sera (23.0%) reacted positively to either Nc-1 or KBA-2 strains of N. caninum. Among the 113 positive sera, 92 sera (81.4%) reacted with antigens of both strains, but 6 sera (5.3%) with Nc-1 and 15 sera (13.3%) with KBA-2 strain only And with T. gondii antigen, 6 sera (1.2%) were positive but all reacted with N. caninum antigen also. Western blot revealed typical binding pattern according to ELISA values, such that high OD group reacted specifically to the major surface proteins including 43 kDa protein. Seroprevalence of 23.0% indicates that neosporosis seemed to be one of major causes of abortion in cattle. It is suggested here to establish more epidemiological researches nationwide systematically.

ELISA detection of IgG antibody against a recombinant major surface antigen (Nc-p43) fragment of Neospora caninum in bovine sera

  • An ELISA was established to measure bovine IgG directed against the recombinant antigenic determinant of Nc-p43, a major surface antigen of Neospora caninum. In a previous study, two thirds of the C-terminal of the molecule was expressed as a $6{\;}{\times}{\;}His$ tagged protein (Ncp43p) for ELISA using 2/3 of the N-terminal of SAG1 from Toxoplasma gondii as a control (TgSAG1A). Among 852 cattle sera collected from stock farms scattered nation-wide, 103 sera (12.1%) were found to react with Ncp43p positively, but no positive reaction was observed with TgSAG1A. This study shows that Ncp43p could be available as an efficient antigen for the diagnosis of neosporosis in cattle. Furthermore, it together with TgSAG1A, could be useful for the differential diagnosis of N. caninum and T.gondii infections in other mammals.
