Lymphoma - clinical questions

  • Lymphoma is a group of neoplastic disease of lymphoid tissues, which can be classified into categories of Hodgkin's disease and non-Hodgkin's lymphoma(NHL). Prognosis of lymphoma depends on the extent of disease(staging) especially in Hodgkin's disease, but also depends on the histologic make up in non-Hodgkin's lymphoma. Although non-Hodgkin's lymphoma is a neoplastic transformation of lymphoid cell it is a collection of disease with merphologically and immunologically diverse make up. Consequently the classification of NHL has changed frequently and evolved according to the progress of immunologic and molecular knowledge added to the original morphologic classification. Lymphoma is a disorder sensitive to chemotherapy which often leads to cure of the disease even in advanced stage, while many other patients die from the progression of disease. Therefore, better understanding in newer classification and sensitive imaging technique, such as PET, in lymphoma will likely lead to the improvement of survival rate.

Cell Behavior of Human Papillomavirus-immortalized and Tumorigenic Human Oral Keratinocytes Does Not Depend on the Integrin Expression

  • Cell behavior of the transformed cells is known to affect by interaction with extracellular matrix (ECM) proteins and integrin. To investigate the alterations of both integrin expression and cell-matrix interaction during neoplastic conversion of human oral kerationcytes, we studied expression levels of integrin subunits by flow cytometry and cellular responses to the ECM proteins in normal human oral keratinocytes (NHOKs), HPV-immortalized HOK-16B line, and three oral cancer cell lines established from HOK-16B line, CTHOK-16B-BaP, CTHOK-16B-DMBA, and CTHOK-16B-Dexa lines. The expression levels of ${\alpha}\;and\;{\beta}$ integrin subunits were shown decreased tendency in human oral keratinocytes undergoing immortalization and tumorigenic transformation except CTHOK-16B-DMBA line tested. Although ${\alpha}v{\beta}6$ integrin is known to be highly expressed in squamous cell carcinomas, and the altered integrin expression is suspected to be associated with cellular carcinogenesis, ${\alpha}v$ integrin subunit and ${\alpha}v{\beta}6$ integrin did not express in oral cancer cell lines tested. Cell behavior to the ECM proteins in HOK-16B line was generally similar to that of exponentially proliferating NHOKs. The adhesion activity profiles of type I collagen were very similar to that of its laminin counterparts, but fibronectin showed minimal adhesion activity under our conditions compared to the BSA control. The ability of the CTHOK-16B-BaP line to spread upon type I collagen and laminin markedly decreased, but migration was notably increased on type I collagen. In contrast, CTHOK-16B-DMBA and CTHOK-16B-Dexa lines spread less but migrated more upon type I collagen than immortalized HOK-16B line. These data indicate that downregulation of integrin subunits causes the changes of cellular responses to the ECM proteins during neoplastic conversion of human oral keratinocytes, and that cellular responses to the ECM proteins in oral cancer cell lines established by exposing different carcinogens are variable according to chemical carcinogens treatment.

Squamous cell carcinoma arising within a maxillary odontogenic keratocyst: A rare occurrence

  • Squamous cell carcinoma (SCC) arising within the lining of an odontogenic keratocyst (OKC) is a rare occurrence. Although potentially locally destructive, OKC is a benign odontogenic process that typically presents with clinical and radiographic features characteristic of a benign intraosseous neoplasm. We present the clinical and radiographic features of a maxillary mass that demonstrated SCC arising from the lining of an OKC. Although the initial clinical and radiographic presentation suggested an infection or malignant neoplasm, biopsies revealed an infiltrative well-differentiated SCC contiguous with and arising from the focus of a pre-existing OKC. The patient subsequently underwent a type II hemi-maxillectomy with neoadjuvant chemoradiation. This report discusses the clinical and radiographic features associated with intraosseous malignancies, especially those arising from an otherwise benign odontogenic lesion. While the majority of OKCs are benign, the current report illustrates the potential for carcinomatous transformation within the lining of an OKC.

  • Mazabraud 증후군은 다골성 섬유성 이형성과 근육 내 점액종이 동반되는 매우 드문 양성 질환이다. 섬유성 이형성에서 악성 변환이 일어나는 경우는 약 1%로 알려져 있고, Mazabraud 증후군의 섬유성 이형성에서 발생한 골육종으로 악성 변환에 대한 보고는 아직까지 전 세계에 8예에 불과하다. 저자들은 Mazabraud 증후군 환자에서 섬유성 이형성이 있었던 경골에 골육종이 발생한 증례를 경험하였기에 영상 소견을 중심으로 대한민국에서 처음으로 보고한다.

3,4-Dihydroxytoluene의 Raf-1 신호전달체계 억제를 통한 암예방 효능 (3,4-Dihydroxytoluene Inhibits Epidermal Growth Factor-induced Cell Transformation in JB6 P+ Mouse Epidermal Cells by Suppressing Raf-1)

  • 이전 연구들에서 rutin과 quercetin을 포함한 여러 flavonoids의 암예방 활성이 보고되었으나, rutin의 경우 섭취 시 체내에서 HVA, HPAA, DHT라는 대사체로 변형되어 흡수된다. 그러나, 이들 대사체와 관련한 암예방 효능 및 그 분자생물학적 작용기작에 대한 연구 결과는 보고된 바가 없어, 본 연구에서 이를 규명하였다. DHT는 EGF로 유도된 세포 변형을 억제하였으며, AP-1 전사인자의 활성 또한 억제하였다. DHT는 Raf-1 효소 활성을 효과적으로 저해하므로서 MEK 및 ERK의 인산화를 억제하였으며, Raf-1과 ATP는 비경쟁적으로 직접 결합하여 Raf-1 효소 활성을 저해한다는 사실을 밝혀내었다. 이와는 대조적으로, rutin은 EGF로 유도된 세포 변형, AP-1 활성, ERK 신호전달체계, Raf-1 효소 활성을 억제하지 못하였다. 이상의 연구결과는 DHT의 암예방 활성이 발암과정과 밀접한 연관이 있는 Raf-1 효소 활성을 억제하여 세포 변형을 억제하는 것과 관련되어 있다는 것을 제시한다.

Alternative Splicing and Its Impact as a Cancer Diagnostic Marker

  • Most genes are processed by alternative splicing for gene expression, resulting in the complexity of the transcriptome in eukaryotes. It allows a limited number of genes to encode various proteins with intricate functions. Alternative splicing is regulated by genetic mutations in cis-regulatory factors and epigenetic events. Furthermore, splicing events occur differently according to cell type, developmental stage, and various diseases, including cancer. Genome instability and flexible proteomes by alternative splicing could affect cancer cells to grow and survive, leading to metastasis. Cancer cells that are transformed by aberrant and uncontrolled mechanisms could produce alternative splicing to maintain and spread them continuously. Splicing variants in various cancers represent crucial roles for tumorigenesis. Taken together, the identification of alternative spliced variants as biomarkers to distinguish between normal and cancer cells could cast light on tumorigenesis.

줄기세포의 분화 결손으로 인한 노화와 암화 (Mal-differentiation of Stem Cells: Cancer and Ageing)

  • Adult stem cells, which have characteristic of self-renewal and multipotency, are specialized cell types, responsible for the tissue regeneration of the damaged tissue. Recent studies suggest that stem cells senescence (or stem cells' ageing) is closely associated with the variety of ageing-related phenotypes such as tissue atrophy, degenerative diseases and onset of cancers. During ageing, declining of stem cells function and subsequently occurring mal-differentiation of stem cells would be important to understand the biological process of development of ageing-related phenotypes such as tissue degenerations and cancers. This review focuses on the DNA damage stress as a cause of senescence of stem cells and their mal differentiation, which is closely link to defect of regeneration potentials and neoplastic transformation. Understanding of molecular mechanisms governingsuch events is likely to have important implications for developing novel avenues for balancing tissue homeostasis longer period of time, further leading to 'Healthy ageing'.

Expression of Matrix Metalloproteinase-10 at Invasive Front of Squamous Cell Carcinoma and Verrucous Carcinoma in the Oral Cavity

  • Background: Matrix metalloproteinases (MMPs) are a family of zinc metalloproteinases capable of degrading components of connective tissues. MMP-10 is frequently expressed in human cancers. The aim of this study was to immunohistochemically evaluate its expression in oral squamous cell carcinoma (OSCC) and verrucous carcinoma (OVC). Materials and Methods: A retrospective analysis of 73 samples (31 OSCC, 22 OVC and 20 non-neoplastic epithelium) was performed. All samples were immunohistochemically stained with monoclonal MMP-10 antibody and expression levels and staining intensity were evaluated with respect to microscopic features. Data were analyzed by SPSS (V.21), Mann-Whitney and Kruskal Wallis tests. Results: MMP-10 was detected in all OSCC and OVC cases. The expression of MMP-10 in OSCC was intensive (score 3) and in OVC was low and moderate (score 1 and score 2) more frequently. Non- neoplastic epithelium did not show MMP-10 expression. Differences between groups was statistically significant (p<0.05). However, the expression of MMP-10 was not obviously different between various grades of OSCC. Conclusions: According to our study, MMP-10 protein can be important possible factor in the transformation of normal oral epithelium to OVC and OSCC, also the level of MMP-10 expression at invasion front of the lesions can be helpful in the differentiation of OVC and OSCC.

Roles of Galectin-7 in Cancer

  • Galectins are ${\beta}$-galactoside binding lectins that contain one or more carbohydrate recognition domains. As a consequence of sugar-binding properties, galectins exhibit a variety of interactions with glycoproteins, thus playing important roles in various pathological processes. A number of studies have shown roles of galectins in cancer. Galectin-7 is a prototype member of the galectin family implicated in epithelial stratification and cell migration. It can act as a potent dual regulator in different types of cancer. Galectin-7 may contribute either to neoplastic transformation and tumour progression through regulation of cell growth, cell cycle, angiogenesis, apoptosis and cell migration or may have a protective effect in cancer depending on the tissue type. A perusal of the literature indicates particular roles of galectin-7 in carcinomas and melanomas, while contributions await greater exploration in other types of cancers including sarcomas and leukemia. This review collectively summarizes available literature on expression and roles of galectin-7 in different cancers.

Xenografted Tumorigenesis in the oral vestibule of nude mice by Snail transfection: Histological and immunohistochemical study

  • Purpose: The purpose of this study is to investigate the epithelial-mesenchymal transition (EMT) induced by Snail transcription factor and Snail-transfected in vivo tumors with histopathological features. Materials and methods: We induced in vivo xenografted tumorigenesis in the oral vestibules of nude mice by a Snail transfected HaCaT cell line and investigated morphological and immunohistochemical features in Snail expressive tumors. Results: We identified tumor masses in 14 out of 15 nude mice in the HaCaT-Snail cell inoculation group, but no tumors were present in any of the HaCaT cell inoculation group. Induced tumors showed features of poorly differentiated carcinoma with invasion to neighboring muscles and bones. The HaCaT-Snail tumors showed decreased expressions of E-cadherin and cytokeratin, but showed increased expressions of vimentin and N-cadherin. Discussion: The Snail transfected xenograft can improve productivity of malignant tumors, show various histopathological features including invasive growth, and aid in the investigation of tumor progression and the interaction with surrounding tissues.
