Occurrence of a Natural Interspecific Hybrid between Rhodeus pseudosericeus and R. notatus in Sangcheon Stream of the Han River, Korea (한강수계 상천천에서 한강납줄개 Rhodeus pseudosericeus와 떡납줄갱이 R. notatus의 종간 자연잡종 출현)

  • Two specimens presumed to be hybrids of Rhodeus pseudosericeus and R. notatus were collected from the Sangcheon Stream of the Han River, Korea. The body color of natural hybrid individuals was yellowish brown, showing the intermediate characteristics of R. pseudosericeus and R. notatus, but overall, the characteristics of R. notatus were similar. Meristic and morphometric characters, the number of dorsal fin rays, anal fin rays and longitudinal row scales showed a hybrid index (HI) of 0, indicating the characters of R. notatus. The hybrid index of predorsal length (HI=74.6), preanal length (HI=75.3), and preventral length (HI=77.6) were similar to the characters of the R. pseudosericeus. Also four characters were appeared to have intermediate characters between R. pseudosericeus and R. notatus; number of gill rakers (HI=55.3), body depth (HI=67.9), snout length (HI=43.4), and inter orbital width (HI=44.8). The rest of 14 characters deviated between 0 and 100, showing unique characteristics of hybrid individuals. Recombination activating gene 1 (RAG1) analysis, hybrids were identified as natural hybrids due to the overlapping peaks of their parent species. Also, as a result of analysis using the cytochrome b gene (COB), one individual was derived from R. pseudosericeus, while the other was derived from R. notatus.

  • The retroviruses carrying ${\nu}-myc$ and ${\nu}-raf$ oncogenes were infected into fetal thymic organ culture (FTOC) to study the molecular mechanisms involved in T cell development. T cell lymphomas in the different stages of T cell development were obtained from this culture system. Interestingly, a few cell lines obtained from this system have a lack of transfected oncogenes, however these cells have the characteristics of transformed cells. In spite of the discrete phenotype of these transformed cell lines, the same pattern of recombination of endogenous c-raf genes was detected from Southern blot analysis. We suggest in this regard that the translocation event of thymocytes, or abnormal promoter activity, can cause lymphomagenesis by way of c-raf.

  • Allozyme study on a small pitch pine stand originating and expanded rather rapidly from a few founder trees indicated that the colonization of the pitch pine population was made progressively from the place where the founder trees located to another by moving in cohorts of seeds from a limited number of family or genetically closely related family groups in line with the succeeding generations. This pattern of migration and colonization resulted marked differences in allelic and genotypic frequencies at many of the allozyme loci between the initially colonized subpopulation on the south-facing slope and the lately colonized subpopulation on north-facing slope of a hill. It appeared that gene fixation due to inbreeding and genetic drift occurred at some loci in the pitch pine population or subpopulations. However, even in t 1e inbreeding small pitch pine population or subpopulations, a comparatively large amount of genetic diversity or heterozygosity was maintained due to the high levels of gene recombination at many of the gene loci and natural selections favoring for heterozygotes.

  • Currently, hydrogen has been produced by Steam Reforming or partial oxidation reforming processes mainly from oil, coal, and natural gas and results in the production of $CO_2$. However, these are influenced greatly on the green house effect of the earth. so it is important to find the new way to produce hydrogen utilizing water without producing any environmentally harmful by-products. In our research, we use microwave water plasma and photocatalyst to improve dissociation rate of water. At low pressure plasma, electron have high energy but density is low, so temperature of reactor is low. This may cause of recombination in the generated hydrogen and oxygen from splitting water. If it want to high dissociation rate of water, it is necessary to control of recombination of the hydrogen and oxygen using photocatalyst. We utilize the photocatalytic material($TiO_2$, ZnO) coated plasma reactor to use UV in the plasma. The quantity of hydrogen generated was measured by a Residual Gas Analyzer.

  • Monogenic diseases are resulted from modifications in a single gene of human cells. Because their treatment with pharmacological medicine have a temporary effect, continuous nursing care and retreatment are required. Gene therapy, gene targeting and induced pluripotent stem cell (iPSC) are considered permanent treatment methods of them. In gene therapy, however, retroviral vectors that have potential toxicity caused by random insertion of harmful virus are used as vehicles for transferring genetic materials. On the other hand, gene targeting could replace and remove the modified gene though homologous recombination (HR) induced by site-specific endonucleases. This short review provides a brief overview on the recently tailored endonucleses with high selectivity for HR.

Unidirectional Photo-induced Charge Separation and Thermal Charge Recombination of Cofacially Aligned Donor-Acceptor System Probed by Ultrafast Visible-Pump/Mid-IR-Probe Spectroscopy

  • A new ${\pi}$-stacked donor-acceptor (D-A) system, [Ru(1-([2,2'-bipyridine]-6-yl-methyl)-3-(2-cyclohexa-2',5'-diene-1,4-dionyl)-1H-imidazole)(2,2':6',2"-terpyridine)]$[PF_6]_2$ (ImQ_T), has been synthesized and characterized. Similar to its precedent, [Ru(6-(2-cyclohexa-2',5'-diene-1,4-dione)-2,2':6',2"-terpyridine)(2,2':6',2"-terpyridine)]$[PF_6]_2$ (TQ_T), this system has a cofacial alignment of terpyridine (tpy) ligand and quinonyl (Q) group, which facilitates an electron transfer through ${\pi}$-stacked manifold. Despite the presence of lowest-energy charge transfer transition from the Ru-based-HOMO-to-Q-based-LUMO (MQCT) predicted by theoretical calculations by using time-dependent density functional theory (TD-DFT), the experimental steady-state absorption spectrum does not exhibit such a band. The selective excitation to the Ru-based occupied orbitals-to-tpy-based virtual orbital MLCT state was thus possible, from which charge separation (CS) reaction occurred. The photo-induced CS and thermal charge recombination (CR) reactions were probed by using ultrafast visible-pump/mid-IR-probe (TrIR) spectroscopic method. Analysis of decay kinetics of Q and $Q^-$ state CO stretching modes as well as aromatic C=C stretching mode of tpy ligand gave time constants of <1 ps for CS, 1-3 ps for CR, and 10-20 ps for vibrational cooling processes. The electron transfer pathway was revealed to be Ru-tpy-Q rather than Ru-bpy-imidazol-Q.

  • Nonactin, known as an ionophore antibiotic, was antagonized by the actibity of quercetin, an oxygen radical scavenger. This compound generated superoxide radicals in Bacillus subtilis lysates. A recombination-deficient mutant strain of B. subtilis was more sensitive than a wild strain, and this hypersensitivity was reduced in the presence of dithiothreitol as an antioxidant. These results suggest that superoxide radical is important in the antibacterial action of this agent.

  • In Order to investigate the physiological role of thiol peroxidase in Bacillus subtilis, a thiol peroxidase (btpx) knock-out mutant was generated by homologous recombination. The growth of btpx knock-out mutant in aerobic condition showed a similar pattern with that of wild type of Bacillus subtilis 168/ But btpx knock-out mutant showed a retarded growht in response to oxidative stress such as $H_2O_2$, cumene hydroperoxide (CHP) treatments.

  • Proliferating cell nuclear antigen (PCNA) is a critical eukaryotic replication accessory factor that supports DNA binding in DNA processing, such as DNA replication, repair, and recombination. PCNA consists of three toroidal-shaped monomers that encircle double-stranded DNA. The diverse functions of PCNA may be regulated by its interactions with partner proteins. Many of the PCNA partner proteins generally have a conserved PCNA-interacting peptide (PIP) motif, located at the N- or C- terminal region. The PIP motif forms a 310 helix that enters into the hydrophobic groove produced by an interdomain-connecting loop, a central loop, and a C-terminal tail in the PCNA. Post-translational modification of PCNA also plays a critical role in regulation of its function and binding partner proteins. Structural and biochemical studies of PCNA-protein will be useful in designing therapeutic agents, as well as estimating the outcome of anticancer drug development. This review summarizes the characterization of eukaryotic PCNA in relation to the protein structures, functions, and modifications, and interaction with proteins.

Studies on Combustion Characteristics and Reduced Kinetic Mechanisms of Natural Gas Premixed Flames (천연가스 예혼합화염의 연소특성 및 축소반응메커니즘에 관한 연구)

  • Combustion characteristics of natural gas premixed flames is studied experimently and numerically by adopting a counterflow as a flamelet model in turbulent flames. Flame speeds are measured by employing LDV, and the results show that flame speed increases linearly with strain rate, which agrees well with numerical results. Parametric dependences of extinction strain rates are studied numerically with detailed kinetic mechanism to show that the addition of ethand to a methane premixed flame makes the flame more resistant to strain rate. The effect of pressure on the extinction strain rate is that the extinction strain rate increases up to 10 atm and them decreases, which is explained by competition of chain branching H+O2=OH+O and recombination reaction H+O2+M=HO2+M. Detailed mechanism having seventy-four step is systematically reduced to a nine-step and a five-step thermal NOx chemistry is reduced to two-step. Comparison between the results of the detailed and the reduced mechanisms demonstrates that the reduced mechanism successfully describes the essential features of natural gas premixed flames including extinction strain rate and NOx production.
