A die casting mold releasing agent was prepared from aqueous mixture of powdery carbon black and water soluble natural polymeric materials such as xanthan gum(X-gum) and carboxymethyl cellulose(CMC), which were used as thickening agent as well as curing agent with aldehydes. The suitable concentration of natural polymers for stable dispersion of carbon black in water was 0.25 wt% of X-gum or 1.0 wt% of CMC. When CMC was used less than 1 wt%, the final carbon black disperion showed a rapid phase separation. The adhesion of carbon black releasing agent on glass plate was improved with the amount of crosslinking agent, glutaraldehyde and chain extender, oligosaccharide. However, the affinity of carbon black releasing agent prepared with X-gum was stronger than that with CMC on glass plate. The final carbon black mold releasing agents prepared under our mixing conditions can be applied to the production of castings of high quality with good workability and without worthening evironmental situations.
A strain of yeast which can convert starch directly to ethanol was developed by the intergeneric protoplast fusion between Schwanniomyces alluvius possessing $\alpha$ amylase as well as glucoamylase with debranching activity and FSC-14-75 which previously had been formed from a heterologous transformation and subsequent intergeneric protoplast fusion. Fusants were selected on minimal medium after protoplasts of auxotrophic mutant of S. alluvius fused with heat-treated protoplasts of FSC-14-75 in the presence of 30%(w/v) PEG and 20 mM $CaCl_2$. The fusion frequency was in the range of $10^{-6}$ order. All fusants tested were intermediate types of parental strains for carbon compound assimilation, and their cell volumes were approximately 1.1 times larger than FSC-14-75 and 1.8 times larger than S. alluvius. The fusants were unable to sporulate like FSC-14-75, while S. alluvius could sporulate. In flask scale the most promising fusant, FSCSa-R10-6, produced 7.83%(v/v) and 10.17%(v/v) ethanol from 15% and 20% of liquefied potato starch, respectively, indicating that the fermetation efficiency of each case increased 1.2 times and 1.6 times than that of FSC-14-75. The elution pattern on DEAE-cellulose chromatography showed that FSCSa-R10-6 has four distinct amylase peaks of which two peaks originated from S. alluvius and the other two from FSC-14-75. These results suggest that the enhanced fermentation efficiency of the fusant might be due to almost-complemented parental amylases.

Gambejaeseup-tang (GBJST) have recently been used as an anti-obesity herbal medicine but their effect and mechanism of action have not been studied. We modified ingredients of GBJST based on the previous experiments about exploring herbs to suppress triglyceride accumulation in 3T3-L1 adipocytes. We investigated the effects of modified GBJST on energy, glucose and lipid homeostasis using female rats with diet-induced obesity and their action mechanism was also determined. Rats fed a high-fat diet (HFD) were divided into 3 groups: rats in each group received 0.2 or 2 g water extracts of modified GBJST (L-GBJST or H-GBJST) or 2 g cellulose per kg body weight (a negative control) on a daily basis. A further group was fed a low-fat diet (LFD) as a positive control. We found that modified GBJST dose-dependently decreased body weight and mesenteric and retroperitoneal fat more than the control. This decrease was due to the reduction in energy intake and the increase of energy expenditure. HFD increased fat oxidation more than LFD and modified GBJST further increased fat oxidation as a major energy source more than the control in a dose-dependent manner. In addition, H-GBJST improved glucose tolerance without changing serum insulin levels during an oral glucose tolerance test. H-GBJST also suppressed the increase of serum total and LDL cholesterol and triglyceride levels by HFD. In conclusion, modified GBJST have a good anti-obesity effect by decreasing energy intake and increasing energy expenditure mainly as fat in female rats with diet-induced obesity. It also improves glucose tolerance and lipid metabolism.
A xylanase (xylanase I) was purified 11.9-fold from the culture filtrate of Trichoderma koningii ATCC 26113 by the column chromatography on Sephadex G-75, SP-Sephadex C-50, DEAE-Sephadex A-50 and Sephadex G-50 with an overall yield of 8.2%. The molecular mass determined by gel filtration and sodium dodecyl sulfate polyacrylamide gel electrophoresis was found to be a monomeric polypeptide of ca. 35 kDa. The isoelectric point of the enzyme was estimated to be 9.3. The optimal reaction pH and temperature are 5.8 and 55.deg.C, respectively. The enzyme is stable up to 60.deg.C, while 78% of its activity is lost after the incubation for 10 min at 70.deg.C. The enzyme hydrolyzes sylan with relatively high activity, as well as carboxymethyl cellulose and avicel. The $K_{m}$ values of the enzyme for oat-spelf sylan, larchwood xylan and Avicel were 3.5, 1.6 and 10. 1 mg/ml, respectively. The enzyme hydrolyzed oat-spelt sylan to sylose, sylobiose, sylotriose and arabionoxylobiose, while it degraded larchwood xylan to xylose, xylobiose, xylotriose and arabionoxylobiose, while it degraded larchwood xylan to xylose, xylobiose and xylotriose as the major products. The hydrolysis patterns indicate that xylanase I is endo-enzyme.e.
The purpose of this study was conducted to develop the slow-released N fertilizer(SRNF) using of waste paper cellulose. Properties of trial product was investigated. Contents of nitrogen, phosphorus, and potassium in trial product were showed 26%, 0.04 and 0.01%, respectively. The contents of Cr, Cu, Pb and Zn were showed 17.4ppm. 259ppm, 12.2ppm and 60.0ppm in the trial product, respectively. However, As and Cd was not detected. Nitrogen of SRNF could be released 60.4% within 12hr after dissolution in water. However, the releasing velocity was thereafter remarkably delayed, showing 75% after dissolution for 72hr.
In this study, natural dyeing using Polygonum tinctoria was performed with linen, ramie, and hemp, which are biodegradable cellulose fibers, considering environmental aspects. In particular, the impacts of alkali NaOH and reducing agent Na2S2O4 were examined, and the possibilities of minimizing the use and reusing the dye were explored. The surface dye concentrations were found to be in the following order: hemp>linen>ramie. With the increase in all additives, the L* value decreased, and the ⊿E and K/S values increased gradually. When Na2S2O4 was 1g/L, the surface color of the dye appeared uniformly from the NaOH concentration of 0.4g/L (pH 10.84). When NaOH was 0.4g/L, the K/S values of linen and ramie increased rapidly after 0.4g/L of Na2S2O4, and hemp maintained a stable color from 0.6g/L of Na2S2O4. With the increase in the dye concentration from 1 to 6g/L, all the fibers were dyed uniformly. The K/S value increased or higher doubled upon repeated dyeing six times for 5 min than when dyed only once for 30 min. Therefore, the linen, ramie, and hemp fibers dyed repeatedly exhibited good washing, rubbing, and colorfastness to perspiration, which was rated between 4 and 4-5, and that to light was rated as 5. Moreover, no discoloration due to sunlight was observed. Finally, linen exhibited a bacterial reduction of 99.9%, thereby indicating its excellent antibacterial property.

Microbial production of cellulolytic enzymes by Aspergillus niger in solid state fermentation (SSF) and submerged state fermentation (SF) in laboratory scale was compared. Czapek Dox liquid broth amended with cellulose (0.5%) was used for cultivation in SF, whereas rice bran was used as a solid support, moistened with cellulose, amended Czapek Dox broth for growth in SSF. The production of Carboxymethyl cellulase, Filter paperase and ${\beta}$-Glucosidase was monitored at regular intervals. The peak production of the enzymes occurred within 3 days of incubation in SSF as against $\geq$ 7 days in SF. SSF gave higher yields of enzymes in comparison to SF. Maximum titres of 0.40, 0.62 and 0.013 U/ml in respect of FPase, CMCase and ${\beta}$-glucosidase in SSF were recovered as against 0.13, 0.06 and 0.0013 U/ml in SF respectively, at their respective peak time intervals. Hence, SSF appeared to be a better choice for production of cellulolytic enzymes by Aspergillus niger.
In order to develope interspecific fusion of the genus Cellulomonas capable of assimilation cellulose, the optimun conditions for the protoplast formation was investigated to examine the susceptibility of cell wall, between different species of the same genus using scanning electron microscope. The variation in the susceptibilities of Cellulomonas sp. CS 1-1 and C. flavigena to lysozyme treatment were considerably remarkable, although they belong to the same genus. The rate of protoplast formation of CS1-1 was 99.9% being treated with lysozyme $(100{\mu}g/ml)$ for 30 minute and that of C. flavigena was about 80% being treated at the concentration of $600{\mu}g/ml$ of lysozyme for 6 hours. The susceptibility of cell wall to the lysozyme treatment on protoplast formation of the strain, CS1-1 seems not to be depend on the cultural periods of cells. On the contrary, that of C. flavigena was considerably depend on the periods. Cells of C. flavigena at mid exponential phase could be more efficiently converted to protoplast cells than those at late exponential phase be done. The rate of the protoplast formation was 95%, when cells of C. flavigena at mid exponential phase were treated with lysozyme $600{\mu}g/ml$ for 6 hours and observed by SEM. In the evalution of protoplast formation of the CS1-1 results of counting method in plate after osmotic shock treatment were similar to the results of the direct observation method by means of SEM. But in the case of C. flavigena the latter method was much more reliable than the former, because the differences between the number of spheroplasts and protoplasts were not able to figure out on conuting the number of protoplast after osmotic shock tretment.
The Aphyllophorales is a large order containing about 2,000 known species. Many of these are the bracket and coral fungi. The vast majority of these fungi are saprophytic on the plant debris. Many species are significant in decomposing plant remains, as they are able to digest cellulose or lignin that occurs in plant cell walls. Many of these fungi have been involved in everyday human affairs. A few were used medicinally by the Greeks and Romans as a remedy for many complaints, including colic, fractured limbs and bruises. Other bracket fungi have been used as curry combs for horses, as snuff, as razor strops and as a source of dye for clothing. The texture of the basidiocarp may be similar to that of cork, wood, leather, paper, or cartilage. Unlike the basidiocarps of the Order Agaricales, the basidiocarps of the Aphyllophorales are not fleshly and moist. Division of the members of the Aphyllophorales into genera was originally made on the basis of gross morphology of the basidiocarp and hymenium and Donk(1964) recognizes 22 families in this order. The species and genus whose typical in Aphylloporales were listed in Table. with related information. The ITS region sequence of some genus were found by BLAST search. Sequences retrieved from GenBank were visually aligned by the program CLUSTAL G. As a result, the medicinal mushroom was separated in four groups. In this multiple alignment, the sequence analysis among Fomes group, Inonotus group and Phellinus group showed high genetic similarity except Hericium group and Sparassis group.
Fertilized hen eggs are rich in a variety of bioactive ingredients. In this study, we aimed to obtain an antioxidant protein from fertilized eggs and the radical scavenging abilities on 1, 1-diphenyl-2-picrylhydrazyl (DPPH), hydroxyl radical (${\bullet}OH$), superoxide anion ($O^{2-}{\bullet}$) were used to evaluate the antioxidant activity of the purified protein. During 20 d of incubation, the radical scavenging ability of protein extracted from fertilized eggs exhibited significantly differences and the protein on day 16 showed higher antioxidant capacity. Based on this, the antioxidant protein of the samples on day 16 were isolated for the follow-up study. With a molecular weight 43.22 kDa, the antioxidant protein was purified by Diethylaminoethyl cellulose -52 (DEAE-52) column and Sephadex G-100. The LC-MS analysis showed that the purified protein molecular weight was 43.22 kDa, named D2-S. The sequence of amino acids was highly similar to ovalbumin and the coverage reached to 84%. The purified protein showed a radical scavenging rate of $52.34{\pm}3.27%$ on DPPH and $63.49{\pm}0.25%$ on ${\bullet}OH$, respectively. Furthermore, the C-terminal amino acid sequence was NAVLFFGRCVSP, which was consistent with the sequence of ovabumin. These results here indicated that purified protein may be a potential resource as a natural antioxidant.
