  • The native forms of common globular proteins are in their most stable state but the native forms of plasma serpins (serine protease inhibitors) show high-energy state interactions. The high-energy state strain of a ${\alpha}$$_1$-antitrypsin, a prototype serpin, is distributed throughout the whole molecule, but the strain that regulates the function directly appears to be localized in the region where the reactive site loop is inserted during complex formation with a target protease.(omitted)

  • The purpose of this study was to develop and succeed local foods such as Yangte miyuck-gook, Getjango-tang, Boongjango-gook, Sook doenjang-gook, Mulmegi-gook and Mukjang-gook in the Namhae region. To derive standardized recipes; first, a test recipe was prepared based on the information obtained from literature material, personal interviews of Namhae natives, and surveys of restaurant recipes through restaurant owners and chefs in the Namhae area. After that, standardized recipes were made in accordance with the collected data. Then, CAN Program 2.0 was used for the nutritional evaluation. Most of these fish soups contained high percentages of protein, minerals and vitamins. Getjango-tang and boongjango-gook were higher than the other foods in most of nutrition contents, which were particularly good sources of calcium and iron.

Genetic Diversity of mtDNA D-loop Polymorphisms in Laotian Native Fowl Populations

  • Here, we studied the genetic diversity of native fowls in Laos by analyzing a mitochondrial DNA (mtDNA) sequence polymorphism. A 546-bp fragment of the mtDNA D-loop region was sequenced in 129 chickens from the areas of Vientiane, Luang Prabang and Pakse. In total, 29 haplotypes were identified and formed five clades. Haplotype diversity and nucleotide diversity of the native fowls in Laos were $0.85536{\pm}0.0172$ and $0.010158{\pm}0.005555$, respectively. Although the Laotian native fowls were distributed across five clades, most of them were clustered in two main clades (A and B), which were originated in China. The other haplotypes were contained in clades D, F, and I, which originated from continental southeast Asia. These results suggest that multiple maternal lineages were involved in the origin of domestic chicken in Laos. Moreover, there appear to be at least two maternal lineages, one from China and the other from the southeast Asian continent.

The Influence of the Annealing of Corn Starch on the formation and Characteristics of Enzyme-resistant Starch

  • The Physical properties of corn starch were investigated by scanning electron microscopy, X-ray diffractometry and differential scanning calorimetry during the formation of enzyme-resistant starch(RS). Samples were studied in their native states and after annealing at 50, 55, 60 and 65℃ in excess water(starch : water=1:3) for 48hr. Starch granules became smaller and more rounded after annealing than in their native state. Annealing did not change the X-ray profile of native corn starch. After autoclaving-cooling cycles, native starch lost most of its crystallinity but annealed ones showed some of their crystallinity left as diffuse or poor B-type, which didn't relate to increasing Rs yields. During formation of RS, however, both native and annealed starches changed their X-ray profile from A-type to poor B-type of retrograded amylose. Annealing caused an increase in gelatinization temperature and enthalpy, but a narrowing of gelatinization temperature range. Only starch annealed at 65℃, however, showed a decrease in enthalpy even though its gelatinization temperature increased, which appeared to be due to the partial gelatinization in the amorphous region during annealing. Peak height index(PHI), the ratio of ΔH to Ti-To, increased by annealing. PHI values, therefore, showed the possibility as an indicator to predict RS yield which cannot be differentiated by differential scanning calorimetry and X-ray diffraction data.

Genetic Diversity Analysis of the Cheju Horse Using Random Amplified Polymorphic DNAs (PCR-RAPD를 이용한 제주말의 유전적 다양성분석)

  • This experiment was carried out to analyze genetic characteristics and to develop the breed specific DNA marker for Cheju-native horse. If this marker contains high repetitive sequences, it is possible to convert a RAPD marker of interest into a single-locus PCR marker called a sequence characterized amplified region(SCAR). Twenty six Cheju-native horse and Fifty thoroughbred genomic DNA were pooled and PCR. were accomplished using 800 random primers. Comparing the pooled DNA from Cheju-native horse and thoroughbred, we found 9 primers which identified markers present in the pooled DNA from breed but absent in the other breed. Among 9 random primers, 6 primers were thoroughbred specific and 3 primers were Cheju-native horse specific. Testing individual horse revealed that 5 marker showed the similar band pattern between Cheju-native horse and Thoroughbred. However, 4 marker were wholly absent in breed while present in the other breed. UBC $126_{3500bp}$, UBC $162_{500bp}$, and UBC $244_{1200bp}$ was detected only Thoroughbred and UBC $562_{560bp}$was detected Cheju-native horse, respectively. After determining of the cloned breed-specific fragment sequence, we designed the SCAR-primers and carried out PCR. Compared to random primer, RAPD-SCAR primer didn't show significantly higher specific band. However, RAPD analysis is useful for genetic characterization of Cheju-native horse.

Identification of Novel SNPs with Effect on Economic Traits in Uncoupling Protein Gene of Korean Native Chicken

  • The avian uncoupling protein (avUCP) is a member of the mitochondrial transporter superfamily that uncouples proton entry in the mitochondrial matrix from ATP synthesis. The sequencing analysis method was used to identify nucleotide polymorphisms within the avUCP gene in Korean native chicken (KNC). This study identified ten single nucleotide polymorphisms (SNPs) in the avUCP gene. We analyzed the SNPs of the avUCP gene to investigate whether polymorphism in the gene might be responsible for quantitative variations in economic traits in KNC. Three significant polymorphic sites for economic traits were avUCP C+282T (mean body weight, p<0.05), avUCP C+433T (daily percent lay, p<0.05), and avUCP T+1316C (daily percent lay, p<0.05). The frequency of each SNP was 0.125 (C+282T in avUCP gene exon 1 region), 0.150 (C+433T in avUCP gene intron 1 region), and 0.15 (T+1316C in avUCP gene exon 3 region), respectively. Among the identified SNPs, one pair of SNPs (genotype CC, C+282T and TT, avUCP C+433T) showed the highest daily percent lay (p<0.05) and mean body weight (p<0.05) and the frequency was 0.067. This study of the avUCP gene could be useful for genetic studies of this gene and selection on economic traits for KNC.

The histological studies on the development of cartilage canal in calcaneus of the Korean native cattle fetus (한우태자 뒷발꿈치골에서 연골관에 관한 조직학적 연구)

  • The development and change of the cartilage canals ralated to chondrocyte growth and the major histological pattern were investigated in the calcaneus of the Korean native cattle fetuses from CRL 28mm to 175mm. The results as follows; 1. The cartilage canal first appeared in the caudal subperichondral region of the sustentaculum tali of the CRL 28mm fetus, and well developed after the period. 2. The three major cartilage canals, one entered from the head of calcaneous into where achilles tendon insert and the others from the caudal region of sustentaculum tali and the calcaneal sulcus, growth distinctively. The latters participated in the ossification center prior to the former. 3. The number of cells in this canals and the size increased synchronously with chondrocytic hypertrophy. In the intermediate and deep canals, the chondrogenic change showed distinctinvely. Our results demonstrated that the cartilage canals from the lower part were important the formation of the ossification center, and the growth and development of canals associated with the hypertropic changes of chondrocytes in the calcaneus of the Korean native cattle fetus.

Institutionalization of Korean Native Beef Production and the Characteristics of Commodity Chain: the Case of Hampyong-gun in Jeonnam Province (한우 생산 제도화에 따른 한우 상품사슬의 특징: 전남 함평군을 중심으로)

  • Since the last decades, livestock ranching has been fast industrialized and the global trade of meat products is sheer increasing. And the Korean government opened its domestic beef market to meet increasing domestic demand for beef. In this context, domestic beef production took its way to specialization and scale economies, and subsequently the commodity chain of beef became different from the conventional one. Such institutions as beef quality grade, HACCP, and brand marketing have strong influence on current beef production systems. Furthermore, along with the macro-scale change, regional and local systems and actors also transform the beef commodity chain. Hampyong-gun, which had once been a declining rural and under-served region in the past, is now actively reviving its economy by producing industrialized, specialized, and localized beef. Such native beef production is both impacted by and associated with multi-scaled actors, their institutions and supporting discourses. These heterogeneous actors and institutions form a newer form of native beef system in the region.

An immunohistochemical study of the endocrine cells in gastrointestinal tract of the Korean native goat (한국 재래산양의 위장관에 출현하는 내분비세포의 면역조직화학적 연구)

  • The gastrointestinal endocrine cells of the Korean native goat were studied immunohistochemically, and 5-HT-, somatostatin-, Gas/CCK-, glucagon-, chromogranin- and PP- immunoreactive cells were revealed. The characteristic findings of the regional distribution and relative frequency of these immunoreactive cells in the gastrointestinal tract of the Korean native goat were as follows. 5-HT-immunoreactive cells were more numerous in the small and large intestine than in the abdomen stomach. Somatostatin-immunoreactive cell were more numerous in the abdomen stomach than in the small and large intestine. Gas/CCK-immunoreactive cells were concentrated very numerously in the pyloric region with a few in the other regions. Moderate numbers of glucagon-immunoreactive cells were found in the small and large intestine, but a few of them were found in the abdomen stomach. Very numerous chromogranin-immunoreactive cells were detected throughout the gastrointestinal tract. PP-immunoreactive cells were observed moderate numbers in the large intestine with few in the ileum. No insulin-immunoreactive cell was found in the gastrointestinal tract.
