본 연구에서는 방울토마토 라이코펜 품종의 식품학적 유용성을 알아보기 위해 유리아미노산, 아미노산 대사물질 및 폴리페놀 화합물의 조성을 분석하였다. 방울토마토 라이코펜 품종은 L-Cys과 L-Try을 제외한 18종의 유리아미노산을 함유하고 있었다. 방울토마토 라이코펜 품종의 유리아미노산 중 L-Glu이 건조 중량 100 g 당 2,499.02 mg이 함유되어 전체 아미노산 중 55.5%를 차지하고 있어 유리아미노산 중 가장 많은 함유량을 보였다. 그리고 L-Gln이 전체 아미노산 중 15.9%, L-Asp가 9.9% 함유되어 있어 L-Glu, L-Gln 및 L-Asp가 전체 아미노산 함량의 81% 이상을 차지하는 주요 구성 아미노산임을 알 수 있었다. 또한, 트립토판을 제외한 히스티딘, 이소류신, 류신, 라이신, 메티오닌, 페닐알라닌, 트레오닌 및 발린 등 필수아미노산이 고루 함유되어 있어 영양적인 측면에서 좋은 식품소재로 판단된다. 라이코펜 품종은 ${\gamma}$-aminobutyric acid(GABA), carnitine(L-Car), o-phosphoethanolamine(o-Pea), hydroxylysine(Hyl) phosphoserine(p-Ser), N-methyl-histidine(Me-His), ethanolamine($EtNH_2$) 등 아미노산 대사물질을 함유하고 있었으며 이 중 GABA가 건조 중량 100 g 당 305.99 mg으로 유리아미노산 대사물질 중 가장 많이 함유되어 있었다. LC/MS/MS 분석을 통해 라이코펜 품종으로부터 caffeic acid-hexose isomer I (CH I), caffeic acid-hexose isomer II (CHII), 3-caffeoylquinic acid(3-CQA), 5-caffeoylquinic acid(5-CQA), caffeoylquinic acid isomer(CQAI), quercetin-hexose-deoxyhexose-pentose(QTS), quercetin-3-rutinoside(Q-3-R), di-caffeoylquinic acid(di-CQA), tri-caffeoylquinic acid(tri-CQA), naringenin chalcone(NGC) 등 10종의 폴리페놀 화합물을 확인하였다. 특히, 항알러지효과, 2형 당뇨 억제 및 비만억제효과를 보이는 NGC이 건조 중량 100 g 당 67.6 mg으로 가장 많이 함유되어 있었고 Q-3-R이 50.9 mg으로 다량 함유되어 있었다. 결과적으로 방울토마토 라이코펜 품종은 18종의 유리아미노산과 트립토판을 제외한 8종의 필수아미노산이 고루 함유되어 있고 GABA, NGC, Q-3-R 등 생리활성 물질이 다량 함유되어 있어 영양이나 건강 측면에서 매우 유용한 식품 소재라 할 수 있다.
본 연구는 가토안에서 나린진과 키토산의 효능을 조사하였다. 나린진의 주성분은 자몽씨 추출물로서 이는 항산화, 항균, 항암제로서 널리 알려져 있다. 이는 식품보존제와 화장품에 널리 사용된다. 또다른 천연 보존제중 하나인 키토산은 식품보존제, 건강음료, 차에 사용되며, 갑각류의 외피나 곤충의 표피, 곰팡이 및 효모 등의 세포벽에 널리 분포되어 인체에 독성이 없으면서도 다양한 미생물에 대해 항균성을 보이는 천연항균제로 알려져 있다. 본 연구의 목적은 소프트콘택트렌즈를 장용함으로써 각 결막 독성과 감염을 일으키는 화합보존제를 대체할 천연 보존제의 적당한 농도를 찾는 것이다. 본 연구는 LM과 TEM을 사용하여 각막상피, 내피를 형태학적으로 비교하였다. In vivo 방법으로, 나린진과 키토산을 가토안의 안구에 직접 3-4방울을 일주일간 하루 4차례씩 점안하였다. 안구 적출 후, LM과 TEM을 통하여 각막의 손상상태를 관찰하였다. 손상된 세포는 대체적으로 키토산이 나린진보다 심하였다. 하지만 나린진도 1%의 농도에서는 세포에 손상을 주었다. 콘택트렌즈 보존제로서 가장 중요한 요건은 적당한 농도를 사용하는 것이다.
감귤은 국내 및 동남아시아 지역에서 많이 즐기는 식품 중 하나이며, 이러한 감귤류의 껍질을 건조시킨 것을 진피라고 한다. 이러한 진피는 한방에서는 이뇨작용, 비장기능을 강화하는 것으로 알려져 있으나, 천연 플라보노이드를 다량 함유하고 있어 학계에서 여러 분야로 연구가 되고 있는 실정이다. 본 연구에서는 플라보노이드 배당체를 함유한 진피 추출물을 ${\beta}$-glucosidase 효소를 이용하여 아글리콘(aglycone)으로 변환 할 수 있도록 치마버섯 배양을 실시하였다. 진피 발효 배양액을 HPLC를 이용하여 분석하였으며, UVA에 의한 광손상 보호능을 섬유아세포를 이용하여 측정하였다. 진피 발효 배양액은 COX-2, LOX-5와 같은 항염증 관련 대사에도 관여하여 염증완화에도 도움을 줄 수 있음을 확인하였다. 또한 각질 형성세포를 이용한 interleukin-$1{\alpha}$를 측정한 결과 진피발효배양액 처리군의 저해활성이 더 높았다. 이러한 결과로 미루어 볼 때, 진피발효 배양액은 피부에 대한 항염 및 항산화 효과가 우수한 것으로 사료된다.
한국에 분포하고 있는 82종의 약용식물에 대하여 환경내성 및 기능성 소재의 개발을 위한 탐색을 위하여 항산화효소와 관련된 첫 번째 방어기작인 SOD 효소활성과 Phenol 화합물의 함량정도를 검정하여 본 결과 SOD 효소활성은 원지 (P. tenuifolia Willd.), 백편두 (D. lablab L.), 백개자 (S. alba L.), 대황 (R. palmatum L.), 감초 (G. uralensis Fisch), 연자육 (N. nucifera Gaertn), 지황 [R. glutinosa (Gaertner) Liboshitz], 현삼 (S. buergeriana Miq.), 인진 (A. capillaris Thb.)등에서 다른 약용작물보다 비교적 높은 SOD 효소 활성 능력을 나타냈다. Total phenol함량은 $(3{\sim}249.731\;{\mu}g/g)$의 범위를 보였으며 지유(S. officinalis L.), 대황 (R. palmatum L.), 복분자 (R. chingii Hu)에서 각각 $249.731\;{\mu}g/g,\;209.546\;{\mu}g/g,\;170.333\;{\mu}g/g$으로 가장 높은 함량을 나타냈다. 16종의 phenol 화합물의 경우에서는 82종의 약용식물이 각기 매우 상이한 농도의 분포를 나타내었다. 앞으로의 연구에서는, 항산화 효소 및 phenol 화합물이 많이 함유하여 우수한 기능성을 가지고 있는 것을 확인된 약용식물에 대해 그들의 이용성을 증대시킬 수 있는 구체적인 연구가 수행되어져야 할 것으로 사료된다. 또한 phenol성 물질을 많이 함유하는 약용식물 추출물을 이용한 기능성 식품, 화장품, 천연 보존재 및 의약품 등으로 개발 (Cook & Samman, 1996)할 수 있을 것으로 판단되어진다.
본 연구에서는 발아기간 및 고압처리 시간에 따른 항산화 성분 및 항산화 활성을 조사함으로써 발아벼의 항산화 활성에 미치는 고압처리공정의 효과에 대해 연구하였다. 발아기간은 6일까지로 하였고, 기간별로 발아된 벼는 30 MPa의 압력 하에서 24시간 및 48시간 동안 처리하였다. 총 폴리페놀함량은 48시간 고압처리를 실시한 6일차 발아벼가 5.15 mg/g으로 고압처리를 실시하지 않은 6일차 발아벼의 1.11에 비하여 증가하였으며, 15종의 페놀산 중 gallic acid 외 6종의 페놀산과 총 페놀산함량이 발아기간 및 고압처리 시간이 증가함에 따라 유의적으로 증가하였다. ABTS 라디칼 소거능과 DPPH 라디칼 소거능은 대조구의 경우 발아 4일차에서 최대 값을 나타내었으며, 고압처리 24시간 처리구와 48시간 처리구 모두 대조구에 비해 높았다. 환원력과 이온킬레이팅 효과는 총 폴리페놀, 페놀산 함량과 유사하게 발아기간 및 고압처리시간이 증가함에 따라 증가하였다. 본 연구결과 발아와 가압을 병행처리 하였을 때 일반벼 및 발아벼에 비해 항산화 성분 및 항산화 활성이 증가함을 알 수 있었으며, 발아벼의 기능성을 증대시키기 위하여 고압처리공정의 적용이 효과적이라고 판단된다.
BACKGROUND/OBJECTIVES: Citrus and its peels have been used in Asian folk medicine due to abundant flavonoids and usage of citrus peels, which are byproducts from juice and/or jam processing, may be a good strategy. Therefore, the aim of this study was to examine antioxidant and anti-inflammatory effects of bioconversion of Jeju Hallabong tangor (Citrus kiyomi ${\times}$ ponkan; CKP) peels with cytolase (CKP-C) in RAW 264.7 cells. MATERIALS/METHODS: Glycosides of CKP were converted into aglycosides with cytolase treatment. RAW 264.7 cells were pre-treated with 0, 100, or $200{\mu}g/ml$ of citrus peel extracts for 4 h, followed by stimulation with $1{\mu}g/ml$ lipopolysaccharide (LPS) for 8 h. Cell viability, DPPH radical scavenging activity, nitric oxide (NO), and prostagladin $E_2$ ($PGE_2$) production were examined. Real time-PCR and western immunoblotting assay were performed for detection of mRNA and/or protein expression of pro-inflammatory mediators and cytokines, respectively. RESULTS: HPLC analysis showed that treatment of CKP with cytolase resulted in decreased flavanone rutinoside forms (narirutin and hesperidin) and increased flavanone aglycoside forms (naringenin and hesperetin). DPPH scavenging activities were observed in a dose-dependent manner for all of the citrus peel extracts and CKP-C was more potent than intact CKP. All of the citrus peel extracts decreased NO production by inducible nitric oxide synthase (iNOS) activity and $PGE_2$ production by COX-2. Higher dose of CKP and all CKP-C groups significantly decreased mRNA and protein expression of LPS-stimulated iNOS. Only $200{\mu}g/ml$ of CKP-C markedly decreased mRNA and protein expression of cyclooxygenase-2 in LPS-stimulated RAW 264.7 cells. Both 100 and $200{\mu}g/ml$ of CKP-C notably inhibited mRNA levels of $interleukin-1{\beta}$ ($IL-1{\beta}$) and IL-6, whereas $200{\mu}g/ml$ CKP-C significantly inhibited mRNA levels of $TNF-{\alpha}$. CONCLUSIONS: This result suggests that bioconversion of citrus peels with cytolase may enrich aglycoside flavanones of citrus peels and provide more potent functional food materials for prevention of chronic diseases attributable to oxidation and inflammation by increasing radical scavenging activity and suppressing pro-inflammatory mediators and cytokines.
BACKGROUND/OBJECTIVES: Citrus flavonoids have a variety of physiological properties such as anti-oxidant, anti-inflammation, anti-cancer, and anti-obesity. We investigated whether bioconversion of Citrus unshiu with cytolase (CU-C) ameliorates the anti-adipogenic effects by modulation of adipocyte differentiation and lipid metabolism in 3T3-L1 cells. MATERIALS/METHODS: Glycoside forms of Citrus unshiu (CU) were converted into aglycoside forms with cytolase treatment. Cell viability of CU and CU-C was measured at various concentrations in 3T3L-1 cells. The anti-adipogenic and lipolytic effects were examined using Oil red O staining and free glycerol assay, respectively. We performed real time-polymerase chain reaction and western immunoblotting assay to detect mRNA and protein expression of adipogenic transcription factors, respectively. RESULTS: Treatment with cytolase decreased flavanone rutinoside forms (narirutin and hesperidin) and instead, increased flavanone aglycoside forms (naringenin and hesperetin). During adipocyte differentiation, 3T3-L1 cells were treated with CU or CU-C at a dose of 0.5 mg/ml. Adipocyte differentiation was inhibited in CU-C group, but not in CU group. CU-C markedly suppressed the insulin-induced protein expression of CCAAT/enhancer-binding protein ${\alpha}$ ($C/EBP{\alpha}$) and peroxisome proliferator-activated receptor gamma ($PPAR{\gamma}$) as well as the mRNA levels of $CEBP{\alpha}$, $PPAR{\gamma}$, and sterol regulatory element binding protein 1c (SREBP1c). Both CU and CU-C groups significantly increased the adipolytic activity with the higher release of free glycerol than those of control group in differentiated 3T3-L1 adipocytes. CU-C is particularly superior in suppression of adipogenesis, whereas CU-C has similar effect to CU on stimulation of lipolysis. CONCLUSIONS: These results suggest that bioconversion of Citrus unshiu peel extracts with cytolase enhances aglycoside flavonoids and improves the anti-adipogenic metabolism via both inhibition of key adipogenic transcription factors and induction of adipolytic activity.
We investigated the influence of germination and high hydrostatic pressure (HHP) treatment conditions on the conversion of functional compounds and antioxidant activity in adzuki bean. The adzuki bean germinated at $25^{\circ}C$ for three- or six-days, and was later subjected to HHP at 0.1, 50, 100, or 150 MPa for 24 h. The highest polyphenol content (5.36 mg gallic acid equivalents (GAE)/g) and flavonoid content (0.91 mg catechin equivalents (CE)/g) were observed after germination for six days and HHP treatment at 100 MPa for 24 h, respectively. The total phenolic acid contents increased with increasing applied pressure from 88.86 to $208.26{\mu}g/g$ (100MPa, 24h). Phenolic acids are divided into two categories; those that exhibit increased content upon HHP treatment, and those that exhibit decreased content. The increasing phenolic acids were gallic acid, chlorogenic acid, (+)-catechin, ${\rho}-coumaric$ acid, ferulic acid, heperidin, salicylic acid, protocatechuic acid, cinnamic acid, naringenin. The total anthocyanin content decreased with increasing applied pressure from 22.42 mg/100 g to 6.28 mg/100 g (150 MPa, 24 h). The highest ABTS radical scavenging activity (8.02 mg eq AA/g) and DPPH radical scavenging activity (1.22 eq Trolox/g) were observed after germination for six days and HHP treatment at 100MPa for 24h, respectively. These results suggested that the combination of HHP and germination can lead to improved functionality in adzuki bean.
Cytochrome P4501B1(CYP1B1) is known to be inducible by xenobiotic compounds such as policyclic aromatic hydrocarbon(PAH) and dioxins such as 2,3,7,8-tetrachloro-dibenzo-p-dioxin(TCDD). And these induction of CYP1B1 is also regulated by many categories of chemicals. In order to investigate the effects of several chemicals on CYP1B1 gene expression in Hepa-I and MCF-7 cells, 5' flanking DNA of human CYP1B1 was cloned into pGL3 basic vector containing luciferase gene, and then transfected into these cells. After treatment of chemicals, the luciferase activity was measured. CYP1B1 enzyme metabolize PAHs and estradiol. CYP1B1 metabolize estradiol to 4-hydrozyestradiol that is considered as carcinogenic metabolite. Recent industrialized industrialized society, human has been widely been exposed to widespread environmental contaminants such as PAHs(polycyclic aromatic hydrocarbon) that are originated from the imcomplete combustion of hydrocarbons. PAHs are known to be ligands of the AhR(aryl hydrocarbon receptor). Induction of cytochrome P4501B1(CYP1B1) in cell culture is widely used as a biomarker for PAHs. Therefore we have studied the effect of PAHs in the human breast cancer cells MCF-7 to evaluate bioactivity of PAHs. We have used the United State of America EPA selected 13 different PAHs, PAHs mixtures and extracts from environmental samples to evaluate the bioassay system. We examined effects of PAHs on the CYP1B1-luciferase reporter gene and CYP1B1 mRNA level. Benzo(k)fluoranthene and dibenzo(a, h)anthracene showed strong response to CYP1B1 promoter activity stimulation, and also CYP1B1 mRNAs increase in MCF-7 cells in a concentration-dependent manner. RT-PCR analysis indicated that PAHs significantly up-regulate the level of CYP1B1 mRNA. Some flavonoids such as genistein, daidzein, chrysin, naringenin and morin were also investigeted. These flavonoids decreased B(k)F infuced luciferase activity at low concentration. But, these flavonoids exhibited stimulatory effect at high concentration.
In this study, the optimum conditions of fermentation were determined by isolating the microorganisms with the ability to bioconvert the Citrus peel flavonoid, and the effect of the fermented Citrus peel extract which was bioconverted on the oxidative damage of HIT-T15 cell was investigated. The Aureobasidium pullulans Y-12 was isolated and identified with the strains having bioconversion activity. The fermentation conditions for bioconversion activity were confirmed to be optimal when culturing for three days at $25^{\circ}C$, 150 rpm in a culture medium containing 5% Citrus peel power and 0.8% casitone. As a result of bioconversion, 32.8 mg/g and 21.5 mg/g of naringenin and hesperetin, which were aglycone flavones, were produced respectively. Also in the flavonoid content, it was confirmed that FCP produced 154.8 mg/g while CP produced 33.7 mg/g, thus producing more by approximately 4.6 times. As a result of treating FCP and CP after inducing the oxidative damage for HIT-T15 cell by treating the deoxy-D-ribose with $IC_{50}$ (38 mM) concentration, the surviving rate was recovered to 90% for FCP treatments in the 0.01 mg/mL concentration and for CP treatments in the 0.025 mg/mL concentration. Also in the insulin secretion rate, FCP treatments increased by 206% and CP treatments by 132% when treated in the 0.1 mg/mL concentration. As the bioconverted FCP can inhibit the oxidative damage of HIT-T15 cell in the low concentration, it was considered its usability as the functional material for prevention of the type 2 diabetes.

제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에

제 5 장 계약 해지 및 이용 제한

일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
