Synthesis and Microphase Separation of Biodegradable Poly($\varepsilon$-caprolactone)-Poly(ethylene glycol)-Poly($\varepsilon$-caprolactone) Multiblock Copolymer Films

  • Poly($\varepsilon$-caprolactone)-poly(ethylene glycol)-poly($\varepsilon$-caprolactone) (PCL-PEG-PCL) multiblock copolymers at various hydrophobic-hydrophilic ratios were successfully synthesized by the chain extension of triblock copolymers through isocyanate (hexamethylene diisocyanate). Biodegradable films were prepared from the resulting multiblock copolymers using the casting method. The mechanical properties of the films were improved by chain extension of the triblock copolymers, whereas the films prepared by the triblock copolymers were weak and brittle. Atomic force microscopy (AFM) of the multiblock copolymer film showed that the hydrophilic PEG had segregated on the film surface. This is consistent with the observed contact angle of the films.

Preparation of the Dexamethasone-incorporated Lipid Nanosphere: Characteristics of Lipid Nanosphere by Varying Species and Ratio of Lipid (덱사메타손이 봉입된 지질나노입자의 제조: 지질의 종류와 함량 변화에 따른 지질나노입자의 특성)

  • the coricosteroid drug dexamethasone is an efficacious antiinflammatory drug, it is difficult to formulate in an injectable formulation due to its poor aqueous solubility. A lipid-based nanosphere formulation containing dexamethasone was designed for solubilization of the drug in aqueous solution and sustained release of the drug from the nanosphere. The lipid nanospheres, composed of phospholipid, cholesterol and cationic lipid, were prepared by self emulsification-solvent diffusion method followed by diafiltration. Physicochemical characteristics such as mean particle diameter, zeta potential and drug loading efficiency of the lipid nanospheres were investigated according to the variation of either the kind of lipid or the lipid composition. The lipid nanospheres had a mean diameter 80-120 nm and dexamethasone loading efficiency of greater than 80%. The drug loading efficiency increased with the increase of the length of aliphatic chain attached to the phospholipid. However, the drug loading efficiency was inversely proportional to the increase of cholesterol content in the lipid composition. The lipid nanosphere could not be prepared without the use of cationic lipid and the drug loading efficiency was proportional to the increase of cationic lipid content. The lipid nanospheres containing dexamethasone are a promising novel drug carrier for an injectable formulation of the poorly water-soluble drugs.

  • 광결정(photonic crystal)은 주기적인 유전율 차이를 가지고 있는 인공적인 광학 물질이다. 그래서 이러한 광결정을 지나가는 전자기파는 마치 반도체와 같은 밴드갭을 가지게 된다. 우리는 silica nanosphere를 사용해서 자연의 보석 중, opal과 유사한 3차원 fcc(face-centered cubic) structure를 가지는 광결정을 만들었다. 평평한 유리기판을 용액에 수직으로 담근 후, 용액을 상온에서 증발시키면 자기 조립 방법(self-assembly method)으로 광결정이 만들어진다.   • Pressure sensitive adhesive (PSA) was prepared by semi-batch emulsion polymerization of acryl monomers in the presence of reactive and non-reactive carboxylated polyurethane resin (PUR). Effects of the PUR type, its content, and crosslinker feeding method on the adhesive properties of the PSA was investigated. In this experiment, the PSA prepared with the reactive PUR showed better adhesive property then the PSA with the non-reactive PUR. Especially, peel strength of the PSA, where acetoacetoxy ethyl methacrylate and 1,6-hexane diamine as crosslinkers were introduced, was dramatically enhanced in severe humidity condition due to the interfacial crosslinking.

  • We have successfully synthesized relatively monodisperse and cross-linked melamine-formaldehyde (M-F) microspheres by dispersed polycondensation and subsequent pH adjustment with serum replacement cleaning. The average particle sizes (equation omitted): weight-average and (equation omitted) : number-average), the polydispersity index (equation omitted), the number of particles N$\_$p/ and the gel content of the M-F microspheres were observed by varying the pH, the surfactant concentration, and the polymerization temperature. We observed that both the pH and the polymerization temperature were predominant factors in determining (equation omitted) and N$\_$p/, but the effect that the temperature and pH had on the gel content ( > 94% for all samples) was negligible. The exponents of the slopes of plots of N$\_$p/ versus pH and surfactant concentration were -10 and 0.6, respectively. Particle nucleation and growth were achieved within short periods; the incessant coagulation occurred even in the presence of surfactants.

Core-shell Poly(D,L-lactide-co-glycolide )/Poly(ethyl 2-cyanoacrylate) Microparticles with Doxorubicin to Reduce Initial Burst Release

  • Monodispersed microparticles with a poly(D,L-lactide-co-glycolide) (PLGA) core and a poly(ethyl 2-cyanoacrylate) (PE2CA) shell were prepared by Shirasu porous glass (SPG) membrane emulsification to reduce the initial burst release of doxorubicin (DOX). Solution mixtures with different weight ratios of PLGA polymer and E2CA monomer were permeated under pressure through an SPG membrane with $1.9\;{\mu}m$ pore size into a continuous water phase with sodium lauryl sulfate as a surfactant. Core-shell structured microparticles were formed by the mechanism of anionic interfacial polymerization of E2CA and precipitation of both polymers. The average diameter of the resulting microparticles with various PLGA:E2CA ratios ranged from 1.42 to $2.73\;{\mu}m$. The morphology and core-shell structure of the microparticles were observed by scanning electron microscopy (SEM) and transmission electron microscopy (TEM). The DOX release profiles revealed that the microparticles with an equivalent PLGA:E2CA weight ratio of 1:1 exhibited the optimal condition to reduce the initial burst of DOX. The initial release rate of DOX was dependent on the PLGA:E2CA ratio, and was minimized at a 1:1 ratio.

Size Tunable Nano Patterns Using Nanosphere Lithography with Ashing and Annealing Effect (나노 구체 리소그라피법에 Ashing과 Annealing 효과를 적용하여 크기조절 가능한 나노패턴의 제조)

  • This work presents a fabrication procedure to make large-area, size-tunable, periodically different shape metal arrays using nanosphere lithography (NSL) combined with ashing and annealing. A polystyrene (PS, 580 ${\mu}m$) monolayer, which was used as a mask, was obtained with a mixed solution of PS in methanol by multi-step spin coating. The mask morphology was changed by oxygen RIE (Reactive Ion Etching) ashing and temperature processing by microwave heating. The Au or Pt deposition resulted in size tunable nano patterns with different morphologies such as hole and dots. These processes allow outstanding control of the size and morphology of the particles. Various sizes of hole patterns were obtained by reducing the size of the PS sphere through the ashing process, and by increasing the size of the PS sphere through annealing treatment, which resulted in tcontrolling the size of the metallic nanoparticles from 30 nm to 230 nm.

Nanosphere Form of Astaxanthin Restores the Mucin Depletion Induced by V. vulnificus

  • Astaxanthin, a natural carotenoid component of shrimp, has been used as a food additive for the treatment of various diseases, but a functional role of Astaxanthin Nanosphere (AN) in the regulation of intestinal mucin (Muc) 2 production during bacterial infection has not described yet. In this study, we have investigated the effect of AN prepared from astaxanthin during Muc2 repression elicited by the Gram-negative bacterium V. vulnificus in human gastrointestinal epithelial (HT-29) cells. AN significantly inhibited the level of ROS production and PKC activation in recombinant protein (r) VvpE-stimulated HT-29 cells. Moreover, AN inhibited the PKC-mediated phosphorylation of extracellular signal-regulated kinase and nuclear factor-kappa B responsible for region-specific hypermethylation in the Muc2 promoter in rVvpE-treated HT-29 cells. In the mouse models of V. vulnificus infection, treatment with AN maintained the level of Muc2 expression in the intestine. On the basis of these results, we suggest that AN blocks the hypermethylation of the Muc2 promoter to restore the level of Muc2 production in HT-29 cells infected with V. vulnificus.

Recyclable Porphyrin Catalyst with Core-shell Nanostructure

  • In the search for a simple preparation method of heterogeneous catalyst, the iron porphyrins were coordinated bonded to the surface of a polymeric core-shell nanosphere. The heterogeneous catalyst was characterized by FT-IR, scanning electron microscope, and UV-vis spectrophotometer. The iron porphyrin bound core-shell nanospheres was about 470 nm in diameter and their catalytic activity for cyclohexene oxidation was similar to a homogeneous iron porphyrin in a solvent composition range of 25-75% acetonitrile/water (v/v). In addition, they could be recovered by simple centrifugation and their catalytic activity was maintained more than the third cycle.

  • 나노스피어 리소그래피는 기존의 리소그래피 방법에 비해 나노 크기 패턴을 제작하는데 공정이 간단하며 재현성있게 대면적에 패터닝이 가능하다는 장점이 있다. 본 연구에서는 Vertical LED(V-LED)의 External quantum efficiency 향상을 위하여 나노스피어 리소그래 피를 이용하여 V-LED의 n-GaN 표면을 패터닝을 하였다. n-GaN 위에 Sputter를 이용하여 $SiO_2$를 증착 후 나노스피어를 스핀 코팅을 이용하여 단일막을 형성하였다. 그 후, 반응성 이온 식각 장치를 이용하여 나노스피어의 크기를 조절하고 $SiO_2$층을 식각하였다. 다음과 같은 공정 후 $SiO_2$층을 Mask층으로 하여 n-GaN 표면을 식각하였다. 실험 결과 나노스피어 리소그래피를 이용하여 V-LED의 External quantum efficiency 향상을 위한 n-GaN 표면의 패턴 제작이 가능함을 확인할 수 있었다.
