  • Six previously unreported N-acylated-DL-1-amino alkyl phosphonic acids were prepared; N-Acetyl-DL-1-amino-3-methyl butyl phosphonic acidN-Benzoyl-DL-1-amino-2-methyl propyl phosphonic acidN-Benzoyl-DL-1-amino-3-methyl butyl phosphonic acidN-Benzoyl-DL-1-amino-2-methyl butyl phosphonic acidN-Acetyl-DL-1-amino-2-methyl propyl phosphonic acidN-Acetyl-DL-1-amino-2-methyl butyl phosphonic acidThe first four compounds were characterized, and the last two compounds were obtained in the crude oil state. The above three DL-1-amino-alkyl phosphonic acid were synthesized from iso-valeric acid, iso-caproic acid and ${\beta}$-methyl valeric acid using Hell-Volhard-Zelinsky reaction, the condensation reaction with triethyl-phosphite and the modified Curtius Reaction. Iso-caproic acid and ${\beta$-methyl valeric acid were prepared by the conventional methods.

The Effect of Salt Concentrations on the Production of Volatile Organic Acids by Zygosaccharomyces rouxii, a Soy Sauce Yeast (간장에서 분리한 Zygosaccharomyces rouxii의 휘발성 유기산 생성에 미치는 식염농도의 영향)

  • By using a synthetic medium simulated on the amino acid composition of soybeam, the effect of salt concentrations on the production of volatile organic acid by the strains of Zygosaccharomyces rouxii So-3101, a soy sauce yeast, was studied at the concentrations of 12.5, 18.0, 22.0 and 28.5% NaCl. The growth, consumption of glucose, and production of alcohol, total acid and volatile organic acid, showed the highest values at a concentration of 12.5% NaCl, and those values were decreased with an increase in the salt concentration. The ratio of volatile organic acid to total organic acid was remained at approximately the same level within the range of salt concentrations between 12.5~22.0%, whereas the ratio was decreased at a salt concentration of 28.5%. After incubation for 16 days, 8 volatile organic acids, i.e. acetic, propionic, n-butyric, isobutyric, isovaleric, isocaproic, n-caproic, and heptanoic acids, were detected by gas chromatography. Among the volatile organic acids, acetic acid was produced in the appreciable amiunt and its ratio to the other volatile acids was increased with an increase in the salt concentration.A small amount of isocaproic, propionic, isobutyric and isovaleric acids were produced, and n-caproic, n-butyric and heptanoic acids were detected only at the lower salt concentration.

  • This study was conducted to investigate the biochemical properties of isolated bacteria, low temperature tolerant methane-producing clostridia which were selected for using them as inoculum to anaerobic fermentation of agricultural and livestock wastes at low temperature. The results were; 1. Low temperature tolerant methane-producing clostridia were isolated from the samples which showed the high methanogenesis rate by enrichment culture at low temperature in cellulose medium. These clostridia, Clostridium botulinum SRC-64, Clostridium scatologens SRC-91 and Clostridium tyrobutyricum SRC-100, were isolated from swampy sediment at latitude $56.9^{\circ}N$, lake sediment IV at latitude $55.0^{\circ}N$, and tidal land soil II at latitude $37.0^{\circ}N$, respectively. The optimum growth temperature for these isolates was $37^{\circ}C$ and the minimum, around $10^{\circ}C$. They all had detectable amount of $F_{420}$, specific coenzyme of methanogens. 2. As anaerobic fermentation products of glucose SRC-64 produced $H_2$, acetic, isovaleric and caproic acid, SRC-91 produced $H_2$, propionic, butyric, valeric, and caproic acid, and SRC-100 produced only acetic and propionic acid. The isolates were produced $CH_4$ ranged from 2.6 to 8.68 n moles/ml for 2 days at $13^{\circ}C$.

Antifungal Properties of Some Short Chain Fatty Acids against Phytopathogenic Fungi (식물병원균에 대한 몇가지 저급지방산의 항균특성)

  • The five short-chain fatty acids such as isobutyric(C-4), butyric(C-4), isovaleric(C-5), valeric(C-5) and caproic (C-6) acids obtained from the extract of common purslane showed wide antifungal spectra against spore germination and mycelial growth of the twenty five phytopathogenic fungi tested in vitro, although there were differences in antifungal potency among them. The antifungal potency of each fatty acid varied significantly against different fungi in spore germination and mycelial growth. The seventeen fungi used for spore germination test and the sixteen fungi used for mycelial growth test can be divided into three groups depending upon differences in minimal inhibitory concentration of each fatty acid for them, respectively. Caproic acid was significantly more toxic to germination than to mycelial growth of the test fungi, while the other four fatty acids did not show such a significant differences in toxicity with a few of exceptions as shown in valerie acid. The longer the chain-length of fatty acid was, the higher the antifungal potency was shown. The normal fatty acids such as butyric and valerie acid were more toxic than their isomers to spore germination and mycelial growth of the test fungi. Each fatty acid was more toxic to spore germination of obligate parasites and some of facultative parasites, and mycelial growth of facultative parasites than to spore germination and mycelial growth of saprophytes, respectively.

Genome-Wide Transcriptomic Analysis of n-Caproic Acid Production in Ruminococcaceae Bacterium CPB6 with Lactate Supplementation

  • n-Caproic acid (CA) is gaining increased attention due to its high value as a chemical feedstock. Ruminococcaceae bacterium strain CPB6 is an anaerobic mesophilic bacterium that is highly prolific in its ability to perform chain elongation of lactate to CA. However, little is known about the genome-wide transcriptional analysis of strain CPB6 for CA production triggered by the supplementation of exogenous lactate. In this study, cultivation of strain CPB6 was carried out in the absence and presence of lactate. Transcriptional profiles were analyzed using RNA-seq, and differentially expressed genes (DEGs) between the lactate-supplemented cells and control cells without lactate were analyzed. The results showed that lactate supplementation led to earlier CA p,roduction, and higher final CA titer and productivity. 295 genes were substrate and/or growth dependent, and these genes cover crucial functional categories. Specifically, 5 genes responsible for the reverse β-oxidation pathway, 11 genes encoding ATP-binding cassette (ABC) transporters, 6 genes encoding substrate-binding protein (SBP), and 4 genes encoding phosphotransferase system (PTS) transporters were strikingly upregulated in response to the addition of lactate. These genes would be candidates for future studies aiming at understanding the regulatory mechanism of lactate conversion into CA, as well as for the improvement of CA production in strain CPB6. The findings presented herein reveal unique insights into the biomolecular effect of lactate on CA production at the transcriptional level.

  • DL-1-Amino-pentyl phosphonic acid, DL-1-Aminopropyl phosphonic acid were synthesized with good yield from caproic acid and butyric acid using the modified Curtius reaction and N-derivatives, N-acetyl-DL-1-amino propyl phosphonic acid, N-acetyl-DL-1-aminopentyl phosphonic acid, N-Benzoyl-DL-1-amino-propyl phosphonic acid, were also prepared by a variety of methods including the Schotten-Baumann reaction. In addition, the chemical and physical properties of the products were investigated. All the products were also identified by means of elemental analysis, Infrared spectrophotometry, Ninhydrin test, and the determination of the neuralization equivalent.

Alcohols and Volatile Organic Acids as Stimulants of Rhizomorph Production by Armillaria mellea (알코올 및 휘발성 유기산류가 뽕나무버섯의 균사속 생산에 미치는 영향)

  • To investigate symbiotic relationship of 'Chunma (Gastrodia elata) and the rhizomorph of Armillaria mellea, volatile organic acids and alcoholic compounds which were considered to be contained in Gastrodia elata were tested to determine stimulatory effects on rhizomorph growth on a chemically defined medium. Also, volatile organic acids were isolated from Gastrodia elata and analyzed by gas chromatography. The growth of rhizomorph was stimulated by the presence of alcohols and volatile organic acids, but acetic acid and methanol were ineffective. In the presence of valeric acid and ethanol, Armillaria mellea produced abundant rhizomorph at concentrations of 0.1 and 1%, respectively. Ethanol and valeric acid supplemented at regular intervals of 3 days as lower concentrations in the medium stimulated the growth of Armillaria mellea. The concentrations of ethanol and valeric acid as low at 0.01% added 3 days intervals for 15 days were more effective than initial concentrations of 0.1 and 1% in stimulating rhizomorph development of Armillaria mellea. Eight kinds of volatile organic acids were identified and quantified by gas chromatography. The major compounds were n-propionic, valeric, iso-carproic and caproic acids, and the minor compounds were iso-butyric, butyric, iso-valeric and hepatanoic acids. Valeric acid was the most abundant among them.

  • We quantitated the major chemical components of ramie leaves (Boehmeria nivea L.) powder. The proximate compositions (all w/w) was 5.42% moisture, 28.15% crude protein, 6.95% crude fat, 15.27% crude ash, and 54.79% carbohydrate, respectively. The total, insoluble, and soluble dietary fiber contents were $39.66{\pm}1.84g/100g$, $20.32{\pm}2.02g/100g$, and $19.34{\pm}2.84g/100g$, respectively. The major free sugars were glucose, galactose and lactose. Seventeen amino acids were isolated. Essential amino acids constituted 44.65% of the total. When free amino acid levels were evaluated, 25 kinds of components were detected, of which 18.15% were essential. Only caproic acid methyl ester and pentadecanoic acid were detected when fatty acid examined.   • The changes of the content of organic acids, carbon dioxide, alcohols and carbonyl compounds of the various Kimchis which were made of cabbage with green onion, garlic, ginger or red pepper and fermented at $12^{\circ}C-16^{\circ}C$ were investigated. Nonvolatile organic acids identified were lactic, succinic, fumaric and malic acid. Volatile organic acids identified were acetic, formic, propionic, butyric, valeric, n-caproic and n-heptanoic acid. Carbonyl compounds identified were acetaldehyde and acetone. The content of lactic acid was increased with fermentation, and higher in Kimchis containing red pepper, garlic and green onion. The content of acetic acid was increased with fermentation, especially in Kimchi containing garlic. The content of carbon dioxide was higher in Kimchi containing garlic. Alcohols identified in all Kimchis was only ethanol. Carbonyl compounds had no direct effect on off-flavor of Kimchi.

  • A simple and modified method is developed to determine a lipase activity. A linear relationship exists between the color intensity and the free fatty acid liberated by enzyme action. The range of determination is from 0.05 to $1.5\;{\mu}moles$ of long chain fatty acid (oleic acid) and 0.2 to $2.0\;{\mu}moles$ of short chain fatty acid (caproic acid). The cumbersome procedure of the removal of the upper aqueous phase which was required in the previous copper soap extraction method was eliminated by the movement of solvent phase to upper phase in the respective biphasic system with a mixture solvent (chloroform: n-hexane:ethanol = 49:49:2) and copper reagent saturated with sodium chloride.
