돼지고기에서 7종 mycotoxins 잔류실태 조사 (Monitoring of 7 mycotoxins in pork)

  • This study was conducted to determine the content of 7 mycotoxins (aflatoxin $B_1$, $B_2$, $G_1$, $G_2$, $M_1$, ochratoxin A and zearalenone) using LC-MS/MS in pork available on the Korean markets. The analysis was carried out using following conditions; C18 column ($2.1{\times}100mm$, $1.7{\mu}m$), mobile phase composed of $H_2O$ (0.1 mM $NH_4Ac$ 0.01% HCOOH) : Methanol (0.1 mM $NH_4Ac$ 0.01% HCOOH), binary pump at a flow rate of 0.5 mL/min and $2{\mu}L$ of injection volume, MS/MS detector with ESI positive and negative mode. The quantication of mycotoxins was based on matrix-matched calibration curves with a correlation coefficient in excess of 0.99 for the 7 mycotoxins. The dectection limits were ranged 0.74~2.13 ng/g, with mean recoveries between 73.10~97.46% except aflatoxin $B_1$ (61.31%). We also monitored mycotoxin residues in 208 pork samples. The test results, mycotoxins were not found except one sample. Ochratoxin A in one sample of the test samples was detected below the quantification limit.

  • Mycotoxin contamination is a global phenomenon and causes a wide array of negative effects and other complications. This study focused on commonly found mycotoxins in Africa and the possible means of prevention or reduction of their contaminating effects. Mycotoxins are secondary metabolites of mold and fungi; they are generally toxic to living organisms. Hundreds of mycotoxins have been identified thus far, with some, such as aflatoxins, ochratoxins, trichothecenes, zearalenone, fumonisins, and patulin, considered agro-economically important. Several factors contribute to the presence of mycotoxins in food, such as climatic conditions, pest infestation, and poor harvest and storage practices. Exposure to mycotoxins, which occurs mostly by ingestion, leads to various diseases, such as mycotoxicoses and mycoses that may eventually result in death. In light of this, this review of relevant literature focuses on mycotoxin contamination, as well as various methods for the prevention and control of their prevalence, to avert its debilitating consequences on human health. Clear evidence of mycotoxin contamination is present in Africa, and it was therefore recommended that adequate prevention and control of these toxic substances in our food system should be encouraged and that appropriate measures must be taken to ensure food safety as well as the enhanced or long-lifespan of the African populace. Governments, research institutions, and non-governmental organizations should tailor the limited resources available to tackle mycotoxin prevalence, as these will offer the best prospects for successful development of a sustainable food system in Africa.

Combined Genotoxic Effects of Aflatoxin B1, Ochratoxin A and Zearalenone in Rat Bone Marrow and Blood Leukocytes

  • Mycotoxins such as aflatoxin B1 (AFB1), ochratoxin A (OTA) and zearalenone (ZEA) are widespread contaminants of food and feedstuffs. It is very likely, that humans and animals are always exposed to mixtures of mycotoxins rather than to individual compounds. Therefore, risk assessments should consider mixture toxicity data. In the present study the combination of AFB1, OTA and ZEA was tested for genotoxicity in rat bone marrow and blood leukocytes after 15, 30 and 60 days treatment. The level of DNA damage was determined by the comet assay. The tail intensity and Olive tail moment in leukocytes and bone marrow cells were significantly higher than in controls. At the same time, the level of DNA damage in bone marrow cells was higher than in leukocytes. The data suggests that prolonged exposure to mycotoxins combination through food consumption can induce DNA damage contributing to the harmful effects in vivo.

Studies on Mycological Status of Salted Fish "Moloha" in Upper Egypt

  • Chemical analysis of salted fish was analyzed in 60 samples collected from various moloha markets in Sohag, Qena and Aswan Governorates, Upper Egypt. Moloha contained 52.9% water content, while organic matter content represented 71.79% of dry weight and 33.81%($338.12{\pm}8.64mg\;g^{-1}$) of fresh weight. Total salts and soluble salts represented 13.29% and 10.19%($132.88{\pm}7.65\;and\;101.93{\pm}5.76mg\;g^{-1}$ of fresh weight), respectively. pH values were more or less neutral. Mycological investigation of examined samples revealed that fifty-five fungal species and one variety belonging to 11 genera were identified. The fungal genera of highest occurrence and their respective number of species were Aspergillus(A. flavus, A. niger, A. fumigatus, A. montevidensis, A. ficuum, A. parasiticus and A. mangini) and Penicillium(P. citrinum, P. puberulum, P. aurantiogriseum and P. roquefortii). On the other hand, yeast represented 18.2% and 3.0% of total counts of fungi on Czapeks-dextrose agar and 15%NaCl-Czapeks-dextrose agar media, respectively. Samples were assayed for potential presence of mycotoxins. Ten out of 60 samples(16.7%) were proved to be toxic. It is the first record of mycotoxins contamination of salted fish in Egypt. The ability of 340 isolates of recovered fungi was screened for production of mycotoxins and extracellular enzymes.

오염된 곡물류(밀, 콩, 옥수수)에서 주요 진균독소 검출 (Detection of Major Mycotoxins from Contaminated Cereals (Wheat, Soybean and Corn))

  • Penicillium, Aspergillus 및 Fusarium에 오염된 밀(Triticum aestivum L.), 콩(Glycine max Merr), 옥수수(Zea mays L.)에서 HPLC를 이용하여 주요 독소들을 검정하였다. Penicillium 독소인 brefeldin A는 밀에서 3.1~270 ppm, 콩에서 45~230 ppm, 옥수수에서 1030~1240 ppm 다량 검출되었다. Citreoviridin과 griseofulvin은 밀, 콩, 옥수수에서 각각 40~80 ppm과 3.6~26.0 ppm이 검출되었다. 그리고 citrinin과 patulin은 밀, 콩, 옥수수에서 각각 0.3~4.0 ppm과 420~3800 ppm로 검출되었고, 특히 옥수수에서 다량 검출되었다. Aspergillus 독소로 ochratoxin A는 밀에서 730 ppm, 콩에서 12.4 ppm, 옥수수는 310 ppm이 검출되었다.

Studies on mycotoxins using LC-MS/MS for the forage produced in Incheon

  • The purpose of this study was to investigate the contamination level of representative mycotoxins that have adverse effects on livestock by using LC-MS/MS method and to utilize the results as basic data for the establishment of quality control system for feed, and to provide information on production and storage. A total of nine mycotoxins, including aflatoxin $B_1$, aflatoxin $B_2$, aflatoxin $G_1$, aflatoxin $G_2$, ochratoxin A, fumonisin $B_1$, fumonisin $B_2$, deoxynivalenol (DON), zearalenone (ZEN) were simultaneously analyzed in LC-MS/MS under ESI positive mode. Fumonisin $B_1$ and fumonisin $B_2$ were detected from 3 cases of 75 forage produced in Incheon area, the detection rate was 4.0%. The detection concentration was 0.01~0.02 mg/kg, which was lower than the domestic recommended limit. Fumonisins were detected in a slightly different manner from the results of mycotoxin studies reported in Korea, which is attributed to the high temperature and dry summer weather of the year. The result of LC-MS/MS method performance of 9 mycotoxins, the recovery of DON was quite low as $41.53{\pm}3.91%$ that is not suitable for simultaneous analysis. This is probably due to that the extract solution used in this study was not suitable for the extraction of DON, along with the characteristics of a very dry forage. For the study of mycotoxins in Incheon area forage for the first time, further investigation is needed for the safe supply of livestock products.

양돈장 사료의 곰팡이독소 오염률 조사 (Prevalence of mycotoxin contamination in pig feedstuffs)

  • To analyze prevalence of mycotoxins, a total of 74 feedstuff samples were collected from silos (n=37) and hoppers (n=37) in nine pig farms. Six mycotoxins were tested with commercialized ELISA kits. All samples were contaminated with four or more mycotoxins. Zearalenone was detected in all of the tested samples. Ochratoxin, deoxynivalenol and H-2/HT-2 toxin were detected in more than 90% of the samples. And also, fumonisin was positive in 89.2% of the samples from the silos, 75.2% from the hoppers, respectively. On the other hand, aflatoxin was detected in about 40% of the samples. When the behavior of lactating sows was observed, possible mycotoxicosis was suspected. It was confirmed that their feedstuffs were contaminated with high levels of mycotoxins such as ochratoxin and T-2/HT-2 toxin. After cleaning the feedline, the clinical symptoms in sows suspected with mycotoxicosis were disappeared. Although mycotoxin concentration in most of the feedstuffs was below the acceptance level, these data indicate that what are required is more monitoring and continuous management for mycotoxins in pig feedstuffs.

LC-MS/MS를 이용한 닭 간과 신장 중 곰팡이 독소 6종 동시분석법 개발 (Method Development for Determination of Multi-Mycotoxins in Chicken Liver and Kidney Tissues by LC-MS/MS)

  • 본 연구에서는 곰팡이 독소에 대한 노출을 확인하기 위한 목적으로 닭의 간, 신장 조직에서 곰팡이 독소 분석법을 확립하였다. 곰팡이 독소의 경우 닭에서 독성이 강하며, 본 실험에서는 가축의 사료에서 주로 확인되는 곰팡이 독소 6종(아플라톡신 $B_1{\cdot}M_1$, 오크라톡신 A, 푸모니신 $B_1$, 데옥시니발레놀, 제랄레논)을 선별하여 추출, 정제조건을 확립하고 LC-MS/MS를 이용하여 분석하였다. 확립된 분석조건에서 검량선은 $R^2$값이 0.99 이상으로 우수한 직선성을 나타내었다. QUECHERS법을 응용하여 닭 간, 신장 시료에서 곰팡이 독소를 추출, 정제하여 분석하였을 때 곰팡이 독소 4종(아플라톡신 $B_1$, 오크라톡신 A, 데옥시니발레놀, 제랄레논)의 평균 회수율은 80.94~98.10%이고, 상대표준편차도 14% 미만으로 조사되어 높은 정확도와 정밀도를 확인할 수 있었다. 검량선에 근거하였을 때 곰팡이 독소 6종에 대하여 닭 간 시료의 경우 검출한계는 $7.6{\sim}145.79{\mu}g/kg$, 정량한계는 $23.04{\sim}441.78{\mu}g/kg$이었다. 닭 신장의 경우 검출한계는 $6.07{\sim}197.20{\mu}g/kg$, 정량한계는 $18.40{\sim}597.59{\mu}g/kg$으로 나타났다. 본 연구의 결과 LC-MS/MS를 이용하여 닭의 간, 신장에서 곰팡이 독소 6종 동시 분석법을 확립하였으며, 이는 생체시료에서 효율적인 곰팡이 독소 동시 분석법으로 활용이 가능할 것으로 기대된다.

  • Of the 200, 000 known species of molds, only 50 or 60 are known to be harmful to humans or livestock. Certain fungi that grow on grains and grasses can produce chemical substances called mycotoxins that adversely affect performance in poultry. There are several methods of preserving feed ingredients. The list includes: drying, antioxidants, mold inhibitors, organic acids, phosphates, cooking or toasting, fat extraction, blending and fermenting. Mold inhibitors are manufactured to inhibit mold growth and prevent the production of toxic substances. They are fungistats and not fungicides, that is, they only stop the growth of molds. Practical and cost-effective methods to detoxify mycotoxin containing feedstuffs are in great demand. 0.5% hydrated sodium calcium aluminosilicate added to the diet protected chicks from the deleterious effects of aflatoxin-contaminated feed. The dietary addition of antioxidants and methionine also significantly diminished the negative effects on body weight in chicks toxicated with 3.0 ppm aflatoxin B1.

Microbe-Mediated Control of Mycotoxigenic Grain Fungi in Stored Rice with Focus on Aflatoxin Biodegradation and Biosynthesis Inhibition

  • Rice contaminated with fungal species during storage is not only of poor quality and low economic value, but may also have harmful effects on human and animal health. The predominant fungal species isolated from rice grains during storage belong to the genera Aspergillus and Penicillium. Some of these fungal species produce mycotoxins; they are responsible for adverse health effects in humans and animals, particularly Aspergillus flavus, which produces the extremely carcinogenic aflatoxins. Not surprisingly, there have been numerous attempts to devise safety procedure for the control of such harmful fungi and production of mycotoxins, including aflatoxins. This review provides information about fungal and mycotoxin contamination of stored rice grains, and microbe-based (biological) strategies to control grain fungi and mycotoxins. The latter will include information regarding attempts undertaken for mycotoxin (especially aflatoxin) bio-detoxification and microbial interference with the aflatoxin-biosynthetic pathway in the toxin-producing fungi.
