Effect of Dual Inoculation with Glomus mosseae and Bradyrhizobium sp. R938 on the Nitrogenase Activity of Arachis hypogeae L. (Glomus mosseae와 Bradyrhizobium sp. R938의 동시 접종에 의한 땅콩의 질소고정력 증진효과)

  • The effect of dual inoculation with vesicular-arbuscular (VA) mycorrhizal fungi, Glomus mosseae, and Bradyrhizobium sp. R938 on the nitrogenase activity of peanut plant (Arachis hypogeae L. cv. Youngho) was studied in pot experiment. As a result of the dual inoculation with Glomus mosseae and Bradyrhizobium on peanut plant, nitrogenase activity was increased by 1.5 times compared with single inoculation with Bradyrhizobium sp. R938. After 69 days growth of peanut plant, the size of nodules was also increased due to the mycorrhizal infection. The mean fresh weight per nodule was 6.7 mg in treatment with the dual inoculation, which is the 1.5 times increase compared with single inoculation with Bradyrhizobium sp. R938. These results suggested that there might be a hormonal effect of VA mycorrhizal fungi in addition to the effect of increase in nitrogen fixing ability through the enhancement of nutrient absorption.

  • We investigated the effects on various crops of inoculation with species of arbuscular mycorrhizal fungi (AMF) in soils from different sources and selected AMF species suitable for domestic environment-friendly farming. Effects on plants varied with the AMF species used. In carrot, Scutellospora heterogama, Acaulospora longula, and Funneliformis mosseae had a positive effect on growth of the host, whereas AMF had only weak effects on the growth of red pepper and leek. AMF inoculation had positive effects on the growth of carrot and sorghum. The results of this study indicate the nature of the relationship between soil, plants, and AMF; this study therefore has important implications for the future use of AMF in environment-friendly agriculture.

Dual Inoculation of Native Rhizobium spp. and Arbuscular Mycorrhizal Fungi: An Impact Study for Enhancement of Pulse Production

  • Fifteen Rhizobium spp. from nodules of 6 common pulses collected from 6 districts of Assam were studied for their infectivity, intrinsic antibiotic resistance, nitrogenase activity and effect of dual inoculation with two native Arbuscular Mycorrhizal Fungi viz. Glomus mosseae(GM) and Gigaspora gilmarie(GG). Out of the 15 isolates 9 were found nodulation positive and 6 of them(AR1, BR8, BR12, AR10, UR10 & GR21) were subjected to intrinsic antibiotic sensitivity test of which AR1 showed resistance against all the 9 test antibiotics. Isolates AR1 and GR21 showed the highest(4.25 mole, $gm^{-1}hour^{-1}$) and the lowest(1.05 mole, $gm^{-1}hour^{-1}$) nitrogenase activity respectively. In Most Probable Number count, the maximum Rhizobium population $5.8{\times}10^5$, was found in both Blackgram and Greengram variety of pulses. The maximum dry weight of nodules(3.14 g), dry weight of shoot(10.08 g), nitrogen content(7.68 mg, $plant^{-1}$), chlorophyll content(1.89 mg, $g^{-1}$), phosphorus content of shoot(6.17 mg, $g^{-1}$) and yield(535.67 kg, $Ha^{-1}$) were found when AR1 dually inoculated with GM in Blackgram.

Influence of Arbuscular Mycorrhizal Fungus and Kinetin on the Response of Mungbean Plants to Irrigation by Seawater

  • An experiment was carried out to investigate the effects of pre-inoculation with the mycorrhizal fungus Glomus clarum and foliar application of kinetin on the growth of mungbean plant irrigated wht different dilution of seawater. Arbuscular-mycorrhizal(AM) infection significantly increased dry weight, height, chlorophyll, sugar and protein content, nitrogen and phosphorus-use efficiencies, leaf conductivity, transpiration rate, nitrogenase, acid and alkaline phosphates activities of all salinized mungbean plants in comparison with control and non-mycorrhizal plants irrespective of the presence or absence of kinetin. Mycorrrhizal plants showed higher concentrations of N, P, K, Ca and Mg and lower Na/N, Na/P, Na/K, Na/Ca and Na/Mg ratios than non-mycorrhizal plants when irrigated with certain dilution of seawater. Mungbean plants showed 597% and 770% dependency on AM fungus G. clarum in absence and presence of kinetin, respectively, for biomass production under a level of 30% of seawater. The average value of tolerance index for mycorrhizal plants accounted 267% and 364% in absence and presence kinetin respectively. This study provides evidence for the benefits of kinetin which are actually known for mycorrhizal than non-mycorrhzal plants. AM fungus and kinetin protected the host plants against the detrimental effects of salt. However, mycorrhizal infection was much more effective than kinetin applications. Thus management applications of this arbuscular mycorrhizal endophyte(G. clarum) with kinetin could be of importance in using seawater in certain dilution for irrigation in agriculture.

  • Effects of the indigenous Vesicular-arbuscular mycurrhizal fungi(VAMF) on early growth response of greenhouse grown crops were experimented. This study was done to evaluate the benefit of indigenous VAMF inoculation on the early growth and the subsequent growth after transplanting of some crops such as cucumber, tomato, hot pepper, eggplant, and melon. Leaf area, shoot dry weight, and plant length of mycorrhizal greenhouse crops showed the tendency of significant or no significant increase over control plants receiving no inoculation. The levels of VA mycorrhizal colonization were increased with plant growth, and infection rates of horticultural crop except hot pepper around one week after transplanting were decreased, while that of 8 weeks after emergence of mycorrhizal seedlings were increased again and infected by around 50% at harvesting time. In spore densities in the rhizosphere soil of craps experimented, the number of spore ranged from $72.7{\pm}26.3$ to $100{\pm}10.3g^1$ on dried soil basis and high density showed in both cucumber and tomato. Total nitrogen contents in shoots were lower in the mycorrhizal plants than non-mycorrhizal one, whereas P uptake in mycorrhizal hot pepper and tomato were highly ramarkable. The K contents in the shoots of mycorrhizal cucumber and eggplent were highly enhanced. Inoculation of the indigenous VAMF enhanced shoot Ca and Mg in both tomoto and melon. The contents of Fe, Zn, Mn and Cu in shoots of mycorrhizal crops were higher than non-mycorrhizal plants and vice versa in case of eggplent. Inoculation of the indigenous VAMF to horticultural crops were effective for alleviation of transplanting shock, and pretransplanting infection improved subsequent growth by reducing the time required for establishment of a functional mycorrhizal symbiosis following transplanting.

  • The objectives of this study were to understand the characteristics of Cd and Pb accumulation in various tissues of poplar species and the effects of ectomycorrhizal inoculation on the accumulation of above two heavy metals in the tissues. The mycelial inoculum of ectomycorrizal fungus, Pisolithus tinctorius was produced in peatmoss and vermiculite mixtures, and inoculated into potted soil with fresh cuttings of four species of Populus, P. alba ${\times}$ glandulosa (Pag). P. koreana ${\times}$ nigra var. italica (Pkn), P. nigra ${\times}$ maximowiczii(Pnm), and P. euramericana(Pe). The potted soils were added with 0, 30, and 80 ppm Cd, and 0, 50, and 300 ppm Pb. The cuttings were grown outdoors for about five months until the plants were harvested for measurement of growth, mycorrhizal infection, and metal contents in leaves, stems, and roots. The total dry weight of Pe treated with Cd and Pb was increased by mycorrhizal inoculation, while that of three other species was not affected by the inoculation. Cd was accumulated in the highest concentration in Pag and its concentration was increased by four times by mycorrhizal inoculation. The Pag accumulated Cd in the highest concentration in the leaves, while three other species accumulated Cd most in the roots. Pb was accumulated in the highest concentration in the roots of all the four species, while Pkn accumulated Pb in the leaves as much as in the roots. Without mycorrhizal inoculation Pe accumulated Pb most among the four species, while Pkn with mycorrhizal inoculation accumulated Pb two times more than in the same species without mycorrhizal inoculation. It was concluded that Pag was the most effective species among the four poplar species in Cd absorption from contaminated soil, and that Pe instead effectively absorbed Pb. Mycorrhizal inoculation increased the Cd accumulation in the tissues by four times in Pag and also increased Pb accumulation by two times in Pkn, with leaves being the major sites of metal accumulation. It may be possible to use these two poplar species in remedying the metal in the soil through the raking and removing the litter out of the contaminate site.

  • For the application of ectomycorrhizal seedlings on damaged slope lands, studies on cultural characteristics of Lyophyllum shimeji and ectomycorrhizal associations of Lespedeza cyrtobotrya seedlings were carried out by artificial inoculation of L. shimeji. Mycelial growth of L. shimeji was best on MP (1% malt extract, 0.1% peptone, 1% glucose and 1.5% agar) medium. An optimum temperature and pH for the mycelial growth were $25^{\circ}C$ and pH6, respectively. Mycorrhizal root of L. cyrtobotrya seedlings inoculated with L. shimeji showed characteristics of ectomycorrhizas with Hartig net. Growth rate of the mycorrhizal seedlings's roots was higher than that of non-mycorrhizal seedlings. When the mycorrhizal seedlings were transplanted in slope land, survival rate and dry weight were 62% and 850 mg/seedling, respectively. On the other hand, survival rate and dry weight of non-mycorrhizal seedlings were 11% and 430 mg/seedling, respectively.

  • To evaluate potential of VA mycorrhizal fungi for promoting growth of nitrogen-fixing trees and efficiency of fungal inoculation in relation to soil fertility and dual inoculation with bacteria, Alnus firma was grown for six months in pots with steam-sterilized soil after inoculation with Glomus mosseae, and Robinia pseudoacacia was grown in the fumigated field after inoculation with native Glomus sp. and Gigaspora sp. In unfertilized Alnus plants, 27% increase in dry weight(or 18% in height) was observed by Glomus inoculation, while plants inoculated with both VA mycorrhiza and actinomycete(crushed nodule inoculum) showed synergistic effect of 83% dry weight increase over uninoculated plants. In fertilized Alnus plants, mycorrhizal inoculation alone or dual inoculation with actinomycete resulted in depression of height and dry weight of plants. In case of Robinia, dual inoculation stimulated height growth by 23% (or dry weight by 25%) over the control in unfertilized field, while 13% more height growth(or 21% more dry weight) was observed in fertilized field. It is concluded that VA mycorrhizae, especially Glomus mosseae, have a potential for growth enhancement in Alnus, that synergistic effect of dual inoculation(mycorrhiza + actinomycete) exists in both Alnus and Robinia, and that responses of these plants to VA mycorrhiza are more pronounced in unfertilized soil.

Increased Salinity Tolerance of Cowpea Plants by Dual Inoculation of an Arbuscular Mycorrhizal Fungus Glomus clarum and a Nitrogen-fixer Azospirillum brasilense

  • Pot greenhouse experiments were carried out to attempt to increase the salinity tolerance of one of the most popular legume of the world; cowpea; by using dual inoculation of an Am fungus Glomus clarum and a nitrogen-fixer Azospirillum brasilense. The effect of these beneficial microbes, as single- or dual inoculation-treatments, was assessed in sterilized loamy sand soil at five NaCl levels ($0.0{\sim}7.\;2ds/m$) in irrigating water. The results of this study revealed that percentage of mycorrhizal infection, plant height, dry weight, nodule number, protein content, nitrogenase and phosphatase activities, as well as nutrient elements N, P, K, Ca, Mg were significantly decreased by increasing salinity level in non-mycorrhized plants in absence of NFB. Plants inoculated with NFB showed higher nodule numbers, protein content, nitrogen concentration and nitrogenase activities than those of non-inoculated at all salinity levels. Mycorrhized plants exhibited better improvement in all measurements than that of non-mycorrhized ones at all salinity levels, especially, in the presence of NFB. The concentration of $Na^+$ was significantly accumulated in cowpea plants by rising salinity except in shoots of mycorrhizal plants which had $K^+/Na^+$ ratios higher than other treatments. This study indicated that dual inoculation with Am fungi and N-fixer Azospirillum can support both needs for N and P, excess of NaCl and will be useful in terms of soil recovery in saline area.

Application of Arbuscular Mycorrhizal Fungi during the Acclimatization of Alpinia purpurata to Induce Tolerance to Meloidogyne arenaria

  • An experiment was conducted to evaluate the tolerance of micropropagated and mycorrhized alpinia plants to the parasite Meloidogyne arenaria. The experimental design was completely randomized with a factorial arrangement of four inoculation treatments with arbuscular mycorrhizal fungi (AMF) (Gigaspora albida, Claroideoglomus etunicatum, Acaulospora longula, and a non-inoculated control) in the presence or absence of M. arenaria with five replicates. The following characteristics were evaluated after 270 days of mycorrhization and 170 days of M. arenaria inoculation: height, number of leaves and tillers, fresh mass of aerial and subterranean parts, dry mass of aerial parts, foliar area, nutritional content, mycorrhizal colonization, AMF sporulation, and the number of galls, egg masses, and eggs. The results indicated a significant interaction between the treatments for AMF spore density, total mycorrhizal colonization, and nutrient content (Zn, Na, and N), while the remaining parameters were influenced by either AMF or nematodes. Plants inoculated with A. longula or C. etunicatum exhibited greater growth than the control. Lower N content was observed in plants inoculated with AMF, while Zn and Na were found in larger quantities in plants inoculated with C. etunicatum. Fewer galls were observed on mycorrhized plants, and egg mass production and the number of eggs were lower in plants inoculated with G. albida. Plants inoculated with A. longula showed a higher percentage of total mycorrhizal colonization in the presence of the nematode. Therefore, the association of micropropagated alpinia plants and A. longula enhanced tolerance to parasitism by M. arenaria.
