  • Cylindvocarpon destmtalns isolate CY-92-01, pathogen of root-rot of Panax ginseng showed t the maximum mycelial growth on the Czapek solution agar among the thirteen kinds of media. Five isolates (Isolate CY-92-01, CY-92-03, CY-92-07, CY-94-01, CY-94-02) of C. destructan from various growth stages of p. ginseng recovered from several geographical sites also showed maximum growth in the Czapek-Dox broth compared with potato dextrose broth and V-8 juice broth. Rapid growth rate was maintained until 12 days after inoculation on the Czapek-Dox broth and mycelial weight was somewhat constant until 20 days. After 30 days of incubation, the mycelial weight began to decrease. The fungal growth occurred from 5$^{\circ}C$ to $25^{\circ}C$ and optimum temperature for growth was 2$0^{\circ}C$. Mycelial weight orderly decreased at 15, 25, 10, and 5$^{\circ}C$. Quantitative measurement was impossible at 5$^{\circ}C$. No fungal growth was occurred at the temperature higher than 3$0^{\circ}C$. Growth was observed at all tested pH ranges from 2.8 to 8.0. Optimum pH for growth was 4.0~5.0 followed by pH 3.3~3.5 and 5.4~6.0. The least growth occurred at pH 2.8.

  • The effects of carbon and nitrogen sources on the mycelial growth and sporulation of microconidia and chlamydospores of five isolates of Cylindrocarpon destructans (Zinssm.) Scholten causing root rot of Panax ginseng were studied. For the carbon sources, fructose, glucose, maltose, and sucrose in Czapek-Dox broth showed good mycelial growth of 178∼201 mg in dry weight compared with 64 mg of the control. The best carbon sources tested for conidial formation were sucrose and maltose with 2.75 and 3.03 log conidia/ml, respectively. For the nitrogen sources, aspartic acid, NaNO3, KNO3, arginine, threonine, and leucine increased mycelial growth of the fungi to 208∼231 mg in dry weight without significant difference (p=0.05) among them. Meanwhile the growth with cystine was poor (26.3 mg dry weight), and no conidium and chlamydospore were formed. Maximum microconidial formation was observed in the media with NaNO3 and KNO3 as 3.37 and 3.35 log conidia/ml, and for the chlamydospore formation the (NH4)2SO4-containing medium and the nitrogen-absent medium were the best as 3.40 and 3.57 log chlamydospores/ml, respectively. No conidium was found in the medium without nitrogen sources, in which chlamydospore formation increased 6 times more than in the nitrogen-amended medium. However, deletion of carbon source in the medium did not affect on the formation of conidia and chlamydospores of C. destructans.

Characteristics of mycelial growth and fruit body of Sparassis latifolia strains and selection of suitable incubation conditions in liquid spawn

  • Sparassis latifolia is called "Cauliflower Mushroom" and is known as an edible mushroom that has high content of ${\beta}$-glucan. Recently, artificial cultivation of S. latifolia has been done by bottle, plastic bag and wood cultivation in Korea. However it is not widely used because there are low incubation ratio and yield. For the high efficiency of production, we aim to find the superior strains and media for better mycelial and fruit body growth. First, we analyzed the genetic relationship among 31 strains and divided five groups with three kinds of URP primers. And then ten strains were selected from five groups based on the experiment of mycelial growth. The suitability of media for mycelial growth was different according to media type. The suitable solid and liquid media for mycelial growth of S. latifolia isolates were PDA and M2, respectively. In addition, with regard to C/N ratio, the mycelial growth increased even until C/N 160. Second, we investigated the production of fruitbody of the strains by plastic bag cultivation. The substrate was mixed with larch sawdust, corn flour, and wheat flour (8:1:1, v/v). Moisture content of substrate was controlled by about 60% with 10% molasses solution. Out of 31 strains, 19 strains formed primordia. The eight strains produced more than 140g/1kg in fresh weight. Third, molasses culture media was selected for the mycelial growth. And molasses suitable sugar content and input aeration were around 8Brix% and 0.3~0.6vvm, respectively. The longer the incubation period is, the more dried weight of mycelia increased, but medium volume decreased. Therefore, the best incubation period was 9 to 11 days depending on strains. In the future, research project entitled development of culture system and new variety for stable production of S. latifolia will be considered as a new item.

Sclerotium cepivorum의 균사생장 및 균핵 생성에 대한 수분압의 영향 (Effect of Water Potential on Mycelial Growth and Production of Sclerotia of Sclerotium cepivorum)

  • 마늘 흑색 썩음균핵병인Sclerotium cepivorum에 대한 수분압과 온도의 영향을 얄아보기 위하여 PDA와 PDB에 NaCI, KCI, 설탕 및 polyethylene glyool 6,000으로 수분압을 조절한 수 15, 20, $25^{\circ}C$에서 균사생장 및 균핵 생성량을 조사하였다. 균사신장은 -1,970에서 -2,240 J/Kg까지는 크게 영향을 받지는 않았으나, =2,240 J/Kg 이하에서는 균사신장이 급격히 저하되었다. -450에서 -2,240 J/Kg까지의 수분압에서는 기본배지에서보다 건물중이 약간 증가하였다. 수분압 감소에 따른 건물중에의 영향은 균사신장에서 보다는 작았다. PEG를 이용해 수분압 조절했을 경우 균사신장과 건물중 모두 다른 용질을 사용했을 경우보다 크게 감소했다. 균사신장은 -1,000 J/Kg에서, 건물중은 -2,240 J/Kg에서 약 50%가 각각 감소했다. 수분압 감소에 따른 균핵 형성에 대한 영향은 균사생장에서 보다 더욱 컸다. -450에서 -910 J/Kg 사이에서 균핵형성이 50% 이상 줄었으며, -2,240 J/Kg 이하에서는 균핵이 형성되지 못했다. 균사신장과 균핵형성은 기본배지에서는 $20^{\circ}C$dptj 양호하였으나, 수분압이 감소하면서 $25^{\circ}C$에서 더 좋은 경향을ㄹ 보였다. NaCl, KCI 설탕 및 PEG에 따라 차이는 있었으나, 균시생장과 균핵 형성은 비슷한 경향의 증감을 보였다.

  • Twenty plant extracts were tested against mycelial growth, sclerotium formatiom and dry weight of mycelium with sclerotia of Sclerotium rolfsii Sacc. The highest(90 mm) mycelial growth was measured in Adhatoda vasica, Tegetes erecta, Allium cepa, and Curcuma longa. The lowest(25 mm) was in Azadirachta indica. No mycelial growth was found in any concentration of cow, buffalo, and goat urine. The highest(90 mm) and the lowest(15 mm) mycelial growth were measured in Biomil and Urea, respectively. No mycelial growth was observed in Zinc. The highest(60 mm) and the lowest(2 mm) mycelial growth were recorded in Macuprex(Dodine; 65% WP) and Boron(100% Boric acid and 17% Boron) respectively. Mycelial growth was totally inhibited in Rovral(Iprodione; 50% WP).

  • 홍국의 혈압강화효능과의 관련성이 시사된 GABA 및 균체량 그리고 홍국이 나타내는 가장 큰 특성 중의 하나인 색도가 서로 다른 각 홍국추출물을 SHR에 경구투여하고 투여 1시간 후부터의 7시간 후까지 혈압강하효과에 미치는 영향을 측정하였다. 모든 홍국 투여군에서 혈압강하효과가 나타났으며 이 효과는 GABA나 균체량 또는 색도와 상관관계를 나타내지 않았다.

Phellinus linteus의 액체배양을 위한 새로운 합성배지의 개발 (Development of a New Synthetic Medium Composition for the Submerged Culture of Phellinus linteus)

  • A new synthetic medium was developed for the submerged mycelial cultures of Phellinus linteus. The medium for maximum mycelial growth of Phellinus linteus (3 days incubation, 28$^{\circ}C$, pH 5) consisted of (per 1 L): glucose, 90 g peptone, 10 g soluble starch, 10 g yeast extract, 3 g KH2PO4, 1 g MgSO4.7H2O, 1 g and CaCl2, 0.1 g. The concentrations of glucose, peptone, yeast extract, KH2PO4, MgSO4.7H2O, and CaCl2 were examined in the ranges of 10~90 g/L, 0~10 g/L, 0~15 g/L, 0~2 g/L, 0~1 g/L, and 0~0.5 g/L, respectively. The dry weight of mycelium in 3 days increased to 16.79 mg/mL using the new synthetic medium. The optimum temperature for mycelial growth of Phellinus linteus was 28$^{\circ}C$. The concentrations of KH2OP4, CaCl2, and yeast extract, which gave the maximum mycelial growth of Phellinus linteus, existed in the concentration ranges examined in this study. But, in the cases of other compositions (MgSO4.7H2O, peptone, and glucose), the mycelial growth of Phellinus linteus increased with the concentration in the ranges.

Mycelial growth of Lentinula edodes in response to different mixing time, pressure intensity, and substrate porosity

  • Biological efficiency (BE), the ratio of fresh mushrooms harvested per dry substrate weight, expressed as the percentage of Lentinula edodes, also known as shiitake, was determined using the 'Sanjo 701' strain stored in the Department of Mushroom at the Korea National College of Agriculture and Fisheries. The mycelia were grown in glass columns with varying levels of moisture content and varying mixing periods of 0.5, 1, 2, and 3 hours. The substrate was sterilized using a steam pressure autoclave sterilizer at normal and high pressure to avoid contamination. The results showed that mycelial growth (126 mm/15 days) was optimized at 55% moisture content. The best mycelial growth of 117 mm/15 days was obtained with 2 hours of mixing time. Normal pressure sterilization yielded better results with mycelial growth of 96 mm/15 days at $100^{\circ}C$ compared to 88 mm /15 days with sterilization at $121^{\circ}C$. Mycelial density was higher, i.e. 3(+++), with normal pressure sterilization compared to 2(++) with high pressure sterilization. Furthermore, sawdust mixed with 5% woodchips increased the substrate porosity and yielded higher mycelial growth. Thus, we demonstrated that the optimum harvest or potential increased yield of shiitake can be obtained by modulating moisture content, mixing time, and substrate porosity.

  • 1. Cassava의 전분을 이용하여 검정곰팡이(Aspergillus niger)를 $30^{\circ}C$에서 4일 동안 진탕배양 하였을 때 배지 중 Cassava의 전분의 함량이 4%, 6%, 8% 및 10%일 때 96시간 배양된 각각의 균체량은 13.35mg/ml, 15.78mg/ml, 14.48mg/ml 및 9.93mg/ml이었다. 2. Cassava의 전분을 이용하여 검정곰팡이를 $30^{\circ}C$에서 4일 동안 진탕배양하여 얻어진 균체를 Lowry 방법에 의하여 그 균체단백을 측정한 바 배지 중의 Cassava의 전분의 함량이 4%, 6%, 8% 및 10%일 때 각각의 균체단백은 2.6mg/ml, 2.7mg/ml, 2.5mg/ml 및 1.7mg/ml이었으며 6% 함유구에서 균체단백의 양이 가장 많았다. 3. 배지 중의 Cassava의 전분량을 6%로 맞추고 질소원의 종류를 $NaNO_3,\;(NH_4)_2SO_4$ 및 Urea를 사용하고 질소량을 Czapeck-Dox 배지에 있는 $NaNO_3$의 질소량의 50%, 100%, 200%, 400% 및 $800^{\circ}C$되게 변화시키면서 진탕배양한 바 균체생산량에는 차이가 없었다.

Viral Effects of a dsRNA Mycovirus (PoV-ASI2792) on the Vegetative Growth of the Edible Mushroom Pleurotus ostreatus

  • A double-stranded RNA (dsRNA) mycovirus was detected in malformed fruiting bodies of Pleurotus ostreatus strain ASI2792, one of bottle cultivated commercial strains of the edible oyster mushroom. The partial RNA-dependent RNA polymerase (RdRp) gene of the P. ostreatus ASI2792 mycovirus (PoV-ASI2792) was cloned, and a cDNA sequences alignment revealed that the sequence was identical to the RdRp gene of a known PoSV found in the P. ostreatus strain. To investigate the symptoms of PoV-ASI2792 infection by comparing the isogenic virus-free P. ostreatus strains with a virus-infected strain, isogenic virus-cured P. ostreatus strains were obtained by the mycelial fragmentation method for virus curing. The absence of virus was verified with gel electrophoresis after dsRNA-specific virus purification and Northern blot analysis using a partial RdRp cDNA of PoV-ASI2792. The growth rate and mycelial dry weight of virus-infected P. ostreatus strain with PoV-ASI2792 mycovirus were compared to those of three virus-free isogenic strains on 10 different media. The virus-cured strains showed distinctly higher mycelial growth rates and dry weights on all kinds of experimental culture media, with at least a 2.2-fold higher mycelial growth rate on mushroom complete media (MCM) and Hamada media, and a 2.7-fold higher mycelial dry weight on MCM and yeast-malt-glucose agar media than those of the virus-infected strain. These results suggest that the infection of PoV mycovirus has a deleterious effect on the vegetative growth of P. ostreatus.
