Effect of LED LightIrradiation on the Mycelial Growth and Fruit Body Development of Hypsizygus Marmoreus (LED 광원이 느티만가닥버섯 균의 균사 생장과 자실체 생육에 미치는 영향)

  • A edible mushroom, Hypsizygus marmoreus is commercially cultivated. However, the researches of cultivation and physiological characteristics were not conducted in Korea. In this study, we conducted on artificial cultivation of H. marmoreus and elucidated the effect of light on the mycelial growth and fruit body development using LED light sources with different wavelength; blue (peak wave length 460nm), green(peak wave length 530nm), yellow(peak wave length 590nm), red(peak wave length 630nm), and white as positive control. Mycelial growth of H. marmoreus strains were inhibited about 30~40% in inhibition ratio under the illumination with blue, green, yellow LED light. However, red LED light was not inhibited. Elongation of stipe was effective under the long wave length such as yellow and red light. Abnormal fruit body was produced under the long wavelength and dark. However, development of pileus was effective under the short wavelength such as green and blue light. Also, as a result of cultivation with mixed light for high quality and harvest, many effective numbers and yields of fruiting bodies were obtained in the mixed treatment of blue and white light, and pileus developed well.

  • The trp2 gene encoding trifunctional enzyme in Coprinus cinereus was investigated the expression in the heterothallic mushroom species. To identify the homology of trp2 gene to Schizophyllum commune and Pleurotus ostreatus, southern hybridization was performed with plasmid pHIONA8 containing C. cinereus trp2 gene as a probe, which resulted in a strong signal indicating an homologous sequence to the chromosomal DNA of S. commune. About 50 transformants per dish was appeared in the complementation test by pHIONA8 using S. commune tryptophan auxotroph as host. In the mating test between transformants and other mating type alleles, the fruiting body of S. commune was formed at $30^{\circ}C$ in $2{\sim}3$ weeks.

Studies on the Development of Casing Materials Using Sawdust Bottle Culture in Cultivated Mushroom, Agaricus bisporus (병재배 폐톱밥을 이용한 양송이 복토재료 개발에 관한 연구)

  • After bottle culture of Pleurotus ostreatus, sawdust was taken out from the bottle and accumulated in the middle of March, and then composted. As the result, Y value was decreased rapidly 30 days after composting, and it was decreased slowly after 30 days. It is considered that 118 days is required for composting, however, it is possible to use for casing material after at least 48 days composting. The pH and total nitrogen content of sawdust based on composting period had tendency to increase as composting was processed. Total carbon and C/N rate had tendency to decrease as time went on. Based on the rate of 10, 30 and 50%, each sawdust was added to clay loam used as casing material for culturing A. bisporrus. Among various treatments, the mycelial growth of A. bisporus was more favorable in the treatment of 30% sawdust than in the single treatment of clay loam. Based on the date necessary for primodium formation of A. bisporus, the primodium formation in the treatment of 30% sawdust was reduced to about 5 days as compared with that of any other treatments When 30% sawdust was added to clay loam used as casing material for culturing A. bisporus, the yield of its fruiting body was increased to 28%.

  • Brown blotch disease of oyster mushrooms is caused by tolaasin and its analog peptide toxins which are produced by Pseudomonas tolaasii. Tolaasin peptides form pores in the plasma membrane and destroy the fruiting body structure of mushroom. Lysis of red blood cells, hemolysis, can be occurred by cytotoxic activity of tolaasin. The hemolytic activity of tolaasin is inhibited by metal ions, such as Zn2+ and Ni2+. When Gadolinium ion was added, a biphasic effect was observed on tolaasin-induced hemolysis, an increase in hemolysis at submillimolar concentrations and an inhibition at millimolar concentrations. The mechanism of gadolinium ion-induced inhibition of tolaasin activity may not be similar to those of the inhibitions by other metal ions. Since gadolinium ion has been reported to change a lateral pressure of lipid membrane by binding to the negative charges of membrane lipids, it may not directly work on the tolaasin channel gating, but rather decrease the stability of tolaasin channel by increasing firmness of membrane.

Characterization and Control of Vascellum curtisii (Berkeley)Kreisel Causing the Fairy Ring Arcs in the Golf Course in Korea (골프코스에서 페어리링의 원인이되는 Vascellum curtisii의 특징과 방계)

  • We have found the clusters of tiny spiny puffball-like mushrooms growing gregariously in fairy ring (arcs) rimmed by a zone of darker green grass in the golf courses. Macroscopic as well as microscopic characters were examined for the morphology of fruiting body. Exoperidium is thin and densely spiny with minute fibrillae at early stage. The connivent spines were soft and quite persistent. In age, the fibrillae scrumble away with a powdery coating, which leaves white endoperidium becoming pale brown. It's interior was white and fleshy at first, but turns into an olive-colored dust as the gleba, the spore-producing tissue, develops to maturity and loaded with olive-brown spore mass. Then, distinct apical pore developed on the endoperidium. Rudimentary subgleba(sterile base) was narrow, chambered, delineated from the gleba by a membrane in young material. These characters suggested this fungus is a Vascellum, a member of the family Lycoperdaceae. The shapes of the spores were globose, echinulate, $3{\sim}3.5{\mu}m$ in diameter, thick-walled, and olive brown. Capillitial threads were $8-9{\mu}m$ wide, mostly colorless in KOH solution and thin-walled, which designated as "paracapillitium". This is an another character that distinguishes this mushroom from Lycoperdon spp. The spines developed on exoperidium were characteristically connivent; their apices joined together in a point, leaving a space below, which gives the appearance of vault to each group of usually 5 to 6 fibrillae. Based on the above characters, this fungus is identified as Vascellum curtisii (Berkeley). The characters distinguishable this from Lycoperdon pulcherrimum, and Vascellum pretense are discussed in detail. Control trial was also attempted. Strong vertical raking(SVR) followed by applying 500x detergent solution (Spark, Aekyung Co. Seoul) resulted in excellent control over any other treatments. In this plot, fruiting body was not developed throughout the end of mushroom growing season.

Inhibitory Effect of Ni2+ on the Tolaasin-induced Hemolysis (톨라신의 용혈활성에 대한 Ni2+의 저해효과)

  • The bacterial toxin, tolaasin, causes brown blotch disease on the cultivated mushrooms by collapsing fungal and fruiting body structure of mushroom. Cytotoxicity of tolaasin was evaluated by measuring hemolytic activity because tolaasins form membrane pores on the red blood cells and destroy cell structure. While we investigated the inhibitions of hemolytic activity of tolaasin by $Zn^{2+}$ and $Cd^{2+}$, we found that $Ni^{2+}$ is another antagonist to block the toxicity of tolaasin. $Ni^{2+}$ inhibited the tolaasin-induced hemolysis in a dose-dependent manner and its Ki value was $\sim10$ mM, implying that the inhibitory effect of $Ni^{2+}$ is stronger than that of $Cd^{2+}$. The hemolytic activity was completely inhibited by $Ni^{2+}$ at the concentration higher than 50 mM. The effect of $Ni^{2+}$ was reversible since it was removed by the addition of EDTA. When the tolaasin-induced hemolysis was suppressed by the addition of 20 mM $Ni^{2+}$, the subsequent addition of EDIA immediately initiated the hemolysis. Although the mechanism of $Ni^{2+}$ -induced inhibition on tolaasin toxicity is not known, $Ni^{2+}$ could inhibit any of fallowing processes of tolaasin action, membrane binding, molecular multimerization, pore formation, and massive ion transport through the membrane pore. Our results indicate that $Ni^{2+}$ inhibits the pore activity of tolaasin, the last step of the toxic process.

Characterization of a New Cultivar "Dan Bi" by Mono-mono Hybridization in Pleurotus eryngii (단핵균주간 교잡에 의한 큰느타리버섯 신품종 "단비"의 특성)

  • A new cultivar "Dan Bi" of Pleurotus eryngii was developed by the method of mono-mono crossing between monokaryotic strains derived from KNR2312 and KNR2596. The parental strains, KNR2312 and KNR2596, are characterized by the property of high quality and a small number of primordia formation, respectively. The optimum temperature of mycelial growth was 25 and that of fruiting body development was $15{\sim}16^{\circ}C$. The period of harvesting including primordia formation was 0.7~1.3 days later than that of control strain Knneutari No. 3 in the culling cultivation. The color of pileus and stipe surface was neutral-brown and pure white, respectively. The shape of pileus was dome and has a scale like as cobweb. The yield was $93{\pm}9.7$ g per 850 cc of plastic bottle. Analysis of the genetic characteristics of the new commercial variety "Dan Bi" showed a different profile as that of the control strain, Knneutari No. 3, when RAPD (Random Amplified Polymorphic DNA) primer #8005 was used. This new variety "Dan Bi" of Pleurotus eryngii is characterized by a small number of primordia formation after scratching.

The production of media and optimal additive rate using the cultivation media wastes of Pleurotus eryngii (큰느타리버섯 폐배지 이용 배지 제조 및 적정 첨가비율)

  • After bottle culture of Pleurotus eryngii, media were taken out the bottle and normally utilized compost. However, nutritional elements were remained in the waste media. This study was carried out to investigate the reusable possibility and the optimal additive rate of waste media in an artificial cultivation of Pleurotus eryngii. The pH had tendency to decrease as the waste media was added from 6.0 to 4.8. Based on the additive rate of 10, 20, 30, 40, and 50%, each treatment waste media was added to new media for culturing Pleurotus eryngii. Among various treatments, the mycelial growth and primordia formation of Pleurotus eryngii were more favorable in the addition of 10-30% waste median than in the addition of 40 and 50%. The yield of its fruiting body was increased slightly in the treatment of 10-30% waste media.

Inhibitive Effect of Mouse Sarcoma 180 by Crude Polysaccharide Extracted from Fruiting Body of Aramillaria mellea (뽕나무버섯(Armillaria mellea)의 자실체에서 추출한 조다당류가 생쥐의 Sarcoma 180에 미치는 억제효과)

  • Armillaria mellea, one of edible and medicinal mushroom belonging to Tricholomataceae of Basid-iomycota, has been known to have outstanding inhibitive effects on the sarcoma 180 and Erhrlich carcinoma of mice. Neutral salt soluble (0.9% NaCl), hot water soluble and methanol soluble substances (hereinafter referred to Fr. NaCl, Fr, HW and Fr. MeOH, respectively) were extracted from the mushroom. In vitro cytotoxicity tests, crude polysaccharide were not cytotoxic against cancer cell lines such as NIH3T3 and Sarcoma 180 at the concentration of $1000{\mu}g/ml$. Intraperitoneal injection with crude polysaccharides exhibited life prolongation effect of $60{\sim}67.5%$ in mice inoculated with Sarcoma 180, respectively. Fr, NaCl improved the immunopotentiation activity of B lymphocyte by increasing the alkaline phosphatase activity by $1.8{\sim}3.0$ folds, respectively. In case of Fr. NaCl, the numbers of peritoneal exudate cells and circulating leukocytes were increased by 10 and 2 folds, respectively.

Breeding and cultural characteristics of a newly bred Lentinula edodes strain, 'Bambithyang' (표고 신품종 '밤빛향'의 육성 및 특성)

  • A new cultivar, 'Bambithyang', was bred from monokaryotic strains of Sanbaekhyang and SANJO 707ho using the mono-mono hybridization method. Its incubation period was determined to be 100 days and its optimum temperature for fruit body flushing, 11-20℃. The mushrooms had a convex cap (diameter, 69.6 mm; thickness, 15.2 mm) and stipes in their center. The color of their upper sides was chestnut brown; their gills were arranged in a ripple style and had medium density. Their scales were cream-colored and fully distributed; the stipes were cream-colored and cylindrical (thick upper part). They also had cream colored fluffs and sporadic fruiting bodies. Zone lines were present when they were cultivated with mother and father varieties. Their stipe lengths and caps were 9% shorter and 16.9% thicker, respectively, than those of Sanbaekhyang.
