Estimation of Genetic Parameters for Pork Belly Components in Yorkshire Pigs

  • This study was conducted to estimate the genetic parameters for pork belly traits and muscles in Yorkshire pigs. Each pork belly was cut into nine parts perpendicular to the thoracic vertebrae (6th to 14th). Traits of belly muscles including the deep pectoral, latissimus dorsi, cutaneous trunci, rectus abdominis, external and internal abdominal oblique from 382 purebred pigs were recorded and analyzed using SAS Package (9.1) and Derivative-free restricted maximum likelihood methods. Heritability estimates for belly traits ranged from 0.27 to 0.49, while they were 0.12 to 0.66 for belly muscles. Moderate to high heritability estimates were noted in belly weight (0.33), belly length (0.28), and belly width (0.49). In belly muscles, the latissimus dorsi and deep pectoral, which are located only in the 6th to 9th vertebrae sections, were found to have heritability estimates ranging from 0.21 to 0.29 and 0.23 to 0.35, respectively. Strong heritability estimates were observed in the 7th to 13th sections of cutaneous trunci muscle ranging from 0.42 to 0.66. Genetic correlations of latissimus dorsi m. with belly length were positive (0.50), while cutaneous trunci m. with belly weight also revealed a positive relationship that ranged from 0.35 to 0.47. The estimated genetic parameters indicate that belly weight can be improved by genetic selection. Differences in the levels of heritability occurred among various parameters of Yorkshire pork belly, which should be considered when performing selection to improve pork belly quality. Moreover, these results can provide valuable information that can be used as the basis for further investigations to improve pork belly.

Effects of Dual-task Training on Chronic Stroke Patients' Balance and Upper Extremity Function (이중과제 훈련이 만성기 뇌졸중 환자의 균형 및 상지기능에 미치는 영향)

  • The aim of this study was to investigate the effects of dual-task training on balance and upper extremity function with chronic stroke. For seventeen persons with chronic stroke, we executed dual-task training for 4 weeks, five times per week, and 30 minutes each time. Before and after the dual-task training, the patients were tested with Berg Balance Scale(BBS) and BTS FreeEMG 300(BTS FreeEMG 300, BTS, Italy). After the dual-task training, the subjects showed significant changes in the score of BBS(p<.05). And, the subjects' shoulder and elbow movement had negative values of change after the intervention, indicating that there was a positive change. The results of this study show that dual-task training for persons with chronic stroke is a useful therapeutic approach by enhancing the their quality of life through improving balance and upper extremity function.

Erythromycin Withdrawal Time in Olive Flounder (Paralichthys olivaceus) after Oral Administration (양식 넙치(Paralichthys olivaceus)에 대한 Erythromycin 휴약기간 설정)

  • The withdrawal time of erythromycin in cultured olive flounder was investigated to ensure the food safety of the fish treated with erythromycin. The oral administration of erythromycin was carried out using the experimental diet containing erythromycin (200 mg/kg) daily dose of 40 mg/kg body weight. The 45 day experimental period was broken into 7 days of habituation, 8 days of medication and 30 days of additional feeding without antibiotics. The erythromycin concentration in the flounder muscle had been increased gradually with medication. After 5 days of medication, the concentration increased to its maximum level of 6.05 mg/kg. After discontinuing the antibiotic, the erythromycin concentration decreased drastically and day 9 was below 0.1 mg/kg. The erythromycin concentration had slowly declined from the 6th to the 20th day after medication and disappeared completely after 25 days. From these results, the time needed to reduce the erythromycin level to the 0.2 mg/kg limit adopted by the ED and Japan was suspected to be 4-6 days. Therefore, a reasonable withdrawal time following ED and Japanese regulatory guidelines for erythromycin in the cultured flounder could be estimated to be 10 days.

Proteomic Comparison between Japanese Black and Holstein Cattle by Two-dimensional Gel Electrophoresis and Identification of Proteins

  • Differences of meat qualities between Japanese Black and Holstein have been known in Japan, however, the causative proteins and/or the genetic background have been unclear. The aim of this study was to identify candidate proteins causing differences of the meat qualities between the two breeds. Using technique of two-dimensional gel electrophoresis, protein profiling was compared from samples of the longissimus dorsi muscle and subcutaneous adipose tissue. Five protein spots were observed with different expression levels between breeds. By using LC-MS/MS analysis and Mascot program, three of them were identified as ankyrin repeat protein 2, phosphoylated myosin light chain 2 and mimecan protein. Subsequently, we compared the DNA coding sequences of three proteins between breeds to find any nucleotide substitution. However, there was no notable mutation which could affect pI or molecular mass of the proteins. The identified proteins may be responsible for different characteristics of the meat qualities between Japanese Black and Holstein cattle.

cDNA Microarray Analysis of Differential Gene Expression in Boar Testes during the Prepubertal Period

  • In an attempt to understand the biochemical mechanism for the synthesis of the anabolic steroid, 19-nortestosterone, produced by prepubertal boar testes and its physiological role, normalized complementary DNA (cDNA) from boar testes was generated. DNA sequencing of 2,016 randomly selected clones yielded 794,116 base pairs of high quality nucleotide sequence. Computer-assisted assembly of the nucleotide sequence of each clone resulted in 423 contigs and 403 singletons including several genes for steroidogenic enzymes and molecules related to steroid metabolism. Analysis of gene expression pattern by use of the presently-fabricated cDNA microarray identified a number of genes that were differentially expressed during the postnatal development period in boar testes. Two genes of unknown function were identified to be highly expressed in the testis of 2-weeks-old neonatal boar. In addition, the sequencing of open reading frames of these genes revealed their homology with human alpha hemoglobin and Homo sapiens hypothetical LOC643669, transcript variant 1. Moreover, the transcripts of these genes were also detected in porcine muscle and adipocytes, in addition to Leydig cells of pigs.

Effect of feeding Korean red pine bark extract on the levels of fatty acid and cholesterol in chicken meats (닭고기의 콜레스테롤과 지방산 함량에 관한 소나무 껍질추출물의 급여효과)

  • This study was conducted to evaluate the effects of dietary Korean red pine bark extract as an antibiotic replacements on cholesterol, fatty acids and the shelf-life of chicken meat. To accomplish this, chickens were fed the optimal level of red pine bark extract that was found to replace antibiotics in the diet of broilers. A total of 180 male broilers(Ross strain 308) were divided into three treated groups, T1(control group), T2(8 ppm of avilamycin) and T3(65 ppm of red pine bark extract per kg diet). The lipid content was reduced by 24.67% and 20.49% in T3 group, while the cholesterol level also decreased significantly in the T3 group by 20.49% and 20.55% when compared to the T1 and T2 groups, respectively. In addition, the saturated fatty acid level was lower in the T3 group than in the T1 and T2 groups, while the unsaturated fatty acid level of the T3 group was significantly higher than those of the other groups. The TBARS value of chicken thigh muscle containing its skin on the 7th day of low temperature storage was significantly lower by 23.86% and 21.17% in the T3 group than in the T1 and T2 groups, respectively. Evaluation of the color of the meat revealed that the $L^*$value (lightness) and $b^*$value(yellowness) were higher in the T3 group than in the T1 and the T2 groups, but that the pH was significantly lower in the T3. Based on the results of this study, the addition of 65 ppm red pine bark extract to the diet of broilers should improve their meat quality with respect to the lipid contents and shelf-life when compared to the addition of antibiotics.

A study on menopausal symptoms and health needs among middle aged women. (중년여성의 갱년기 증상과 건강요구에 대한 기초조사)

  • During the middle age of a woman's life cycle. several health changes and problems occur. Therefore. middle aged women must manage their health and maintain quality life by coping with bodily changes. However. today there is not enough research and health programs for middle aged women. Data from the study will be used for health promotion program development of middle aged women. Data was collected from January 21 to 24. 1995 by telephone interview. Four hundred middle aged women between 40 and 59 years old and living in Chon Ju City were interviewed. The results of this study were as follow: 1. Frequency rate of menopausal symptoms was 38.4%. The most serious menopausal symptom was psychosomatic symptom. Mean score orders of experienced symptoms were back pain, joint pain(1.80). nervousness(1.80). general weakness(1.67). 2. The most important problems as perceived by the clients were children(45.8%), health (24.0%) and economics(7.8%). The most serious health problems were concerning the muscle-skeletal system(45%) such as arthritis. spinal disk problems and osteoporosis. Adult diseases04.S%) such .as hypertension and diabetes were also health concerns. Health management activities reported were exercise(22.5%), social activity02%) and inactivity(53%)' 33% of clients were interested in health groups and they wanted a program of health education, exercise and social activity to be provided. 3. General characteristic variables were significantly related to the level of menopausal symptoms as follows: age(t=-2.06, p=0.040), status of marriage(t=-3.56, p=0.000), educational level (F=4.35. p=0.05) and menopausal status(t=4.37, p=0.000).

ENZYME ACTIVITY AND ACIDITY IN VINEGAR PICKLING OF ANCHOVY (멸치 Vinegar 절임시의 산도와 효소 활성)

  • The fellowing results are obtained by experiment on the variation of enzyme activity during pickling anchovy (Engraulis japenicus) in vinegar. 1) Protease activity increased during brine-salting. It decreased for a short time by tile addition of acetic acid and gradually changed at an invariable ratio during pickling. 2) Lipase activity decreased during brine salting but increased temporarily by the addition of vinegar, after this no fixed tendency could he observed during pickling whereas acid value abviously increased. 3) VBN (Volatile basic nitrogen) and NaCl content quickly increased during brine salting. During pickling VBN increased slowly while NaCl content scarcely varied. 4) The bacterial plate count decreased suddenly when the anchovy were placed in brine but it increased during brine salting and decreased again when anchovy were pickled and the number of count remained the same thereafter. 5) To maintain preservation effects, the acidity of the vinegar must be maintained more than $2\%$ in the muscle. 6) Based on this experimental condition, it transpires that 8 days of immersion In brine produces sufficient salt concentration in the muscles. From the results of VBN, acidity, amino nitrogen, lipase activity, and organoleptic tests, the quality of pickle was still preserved after 47 days and taste of the pickle seemed best on the thirty second day of the storage.

The Effects of an Exercise Program on Physical Fitness, Cardiopulmonary Function and Life Satisfaction for Adult Women (운동프로그램이 성인여성의 체력, 심폐기능 및 생활만족도에 미치는 효과)

  • Purpose: This study was to examine the effects of an exercise program on physical fitness, cardiopulmonary function and life satisfaction for adult women. Method: The exercise program combined dance and resistance training. The subject group consisted of 114 women aged between 33 and 60. Three 8-week sessions consisted of 55-80% HR max for 60-90 minutes a day and 3 times a week from March to November, 2004. Data were collected through pre- and post-exercise tests before and after each session. Data were collected with dynamometer, sphygmomamometer, spirometer and structured questionnaires. Data were analyzed employing descriptive statistics and paired t-test with SPSS/PC(10.0version) program. Results: There were significantly positive changes in muscle strength, flexibility, balance quality, forced vital capacity and life satisfaction, but no significantly positive changes in agility and blood pressure. Conclusion: This study showed that an exercise program has partially positive effects for adult women. The results of this study suggest that there should be programs of continuous exercise at community health centers for adult women's health.

Analysis of Hanwoo Loin Proteome by 2-D Gel Electrophoresis and Peptide Mass Fingerprinting

  • A proteomic map of Hanwoo loin was obtained using 2-D SDS-PAGE and mass spectrometric analysis: 27 bovine proteins plus 2 proteins having similarities to other mammal proteins out of 52 proteins analyzed. The identified proteins consisted of 50 % basic house keeping proteins involved in metabolism, 30% muscle proteins, and other miscellaneous proteins. Many proteins on the 2-D gel with different molecular weights and isoelectric points were identified as same proteins due to posttranslational modification. As many of the identified house keeping proteins showed the high sequence similarities to other mammal equivalent proteins, searching the mammal databases could confirm the annotation. The preliminary identification of the proteome in bovine loin tissue could reveal the functions of proteins at over 50 % of chance with high fidelities. Using the established loin proteome map, proteomic difference between 1 yr and 2 yr Hanwoo loin tissues were compared on 2D gel. Regardless of the difficulty normalizing protein concentrations and sample-to-sample variations, three unidentified proteins and myoglobin were selected as up-regulated proteins during the fat deposition period. This study contributes to a move thorough and holistic understanding of beef meat, helping to build the basis for future identification of new markers for good quality meat.
